OKN을 유발하는 단축 Mesencephalic 구조에 GABA Antagonist를 미량 주입할 때의 닭의 OKN 방향적 불균형성에 관한 특수효과 (Specific Effects on Monocular OKN Directional Asymmetry of Unilateral Microinjections of GABA Antagonist into the Mesencephalic Structures in the Chicken)

  • GABA antagonist인 SR 95531이 닭의 pretectum nuclei (nucleus Superficialis Synencephali nSS) 또는 nBOR (nucleus Ectomammilaris nEM)속에 미량 주입되었다. 단안의 optokinetic nystagmus는 한쪽 뇌의 내부에 약품을 주입하기 전후에 탐색 코일 테크닉에 의해서 기록되었다. 한쪽 nSS 또는 nEM 속에 SR 95531를 미량 주입하면 자극의 양방향에 대하여 반대쪽 눈에 의해서 유도된 단안의 OKN속도 게인이 가역적으로 증가하였다. 이때 nSS속에 주입하면 N-T 성분의 속도 게인보다 T-N 성분의 속도 게인을 더욱 강하게 증가시킴으로써 방향적 불균형성이 더욱 증가되었다. 한편 nEM 속에 약품을 주입하게 되면 T-N 성분의 속도 게인보다 N-T 성분의 속조 게인이 더욱 강하게 증가함으로써 방향적 불균형성을 제거했다. 이것으로부터 우리는 nSS 특히 N-T 자극에 방응에 단안의 OKN이 관련되어 있고, nEM은 N-T 자극에 대한 OKN 반응에 더욱 관련되어 있는 것으로 유추할 수 있었다. 이 결과는 그 약품이 중뇌 수준에서 OKN에 관련된 GABAergic 메카니즘의 억제작용을 제거한다는 것을 암시한다. 미량 주입된 핵에 대하여 같은쪽 눈에 의해서 유도된 OKN의 게인의 증가는 수평적 OKN에 관련된 이들 중뇌 구조들 사이에 강한 상호작용이 존재한다는 것으로 설명된다.

3D 바이오프린팅과 무지지체 조직공학 기술 기반 추간판 복합 조직 제작 (Fabrication of Tissue Engineered Intervertebral Disc Using Enable 3D bio-printing and Scaffod-Free technologies)

  • Intervertebral disc(IVD) mainly consists of Annulus fibrosus(AF) and Nucleus pulposus(NP), playing a role of distributing a mechanical load on vertebral body. IVD tissue engineering has been developed the methods to achieve anatomic morphology and restoration of biological function. The goal of present study is to identify the possibilities for creating a substitute of IVD the morphology and biological functions are the same as undamaged complete IVD. To fabricate the AF and NP combine biphasic IVD tissue, AF tissue scaffolds have been printed by 3D bio-printing system with natural biomaterials and NP tissues have been prepared by scaffold-free culture system. We evaluated whether the combined structure of 3D printed AF scaffold and scaffold-free NP tissue construct could support the architecture and cell functions as IVD tissue. 3D printed AF scaffolds were printed with 60 degree angle stripe patterned lamella structure(the inner-diameter is 5mm, outer-diameter is 10 mm and height is 3 mm). In the cytotoxicity test, the 3D printed AF scaffold showed good cell compatibility. The results of histological and immunohistochemical staining also showed the newly synthesized collagens and glycosaminoglycans, which are specific makers of AF tissue. And scaffold-free NP tissue actively synthesized glycosaminoglycans and type 2 collagen, which are the major components of NP tissue. When we combined two engineered tissues to realize the IVD, combined biphasic tissues showed a good integration between the two tissues. In conclusion, this study describes the fabrication of Engineered biphasic IVD tissue by using enable techniques of tissue engineering. This fabricated biphasic tissue would be used as a model system for the study of the native IVD tissue. In the future, it may have the potential to replace the damaged IVD in the future.

  • The VLBI Space Observatory Program (VSOP) is a worldwide project of one radio telescope in space with many ground radio telescopes. The concerted space VLBI network enables us to reveal high resolution and high quality images of radio sources. The space radio telescope is anticipated to be launched in January/February 1997, and collaborative observations have been coordinated. The basic parameters of the system and present status are given.

  • A new restoration method of chest X-ray image (dual project filter) was proposed to improve SNR (signal to noise ratio) characteristics. In this method, a priori information of system and anatomical structure and statistics of projected object are used in the design of filter. Dual projection filter varies its parameters, adapting to the local regions of chest(lungregion, mediasternum, subdiaphragm) and the structure of chest (bone, tissue, blood vessel, bronchia). The performance of Dual Projection Filter was 0.1-0.2dB better than Dual Sensor Wiener Filter, which was used for initial estimate of Dual Porjection Filter.

  • The Korean railroad has developed as a foundation of nation's nucleus network. Nowadays Korea has entered into a new era because of the operation of KTX(Korea Train Express). As an introduction of KTX, the Korean railroad system has advanced Korean style high speed rail(HSR-350x), which is based on transfer and own technology. It is expected that the Korean rail road system has the leading design and production ability. Korea is planning to develop its own high speed railroad system until 2007. So the present task is to initiate a strategic export method for the development and value-added of Korean high speed railroad system. Furthermore this study proposes the administrative and technical policy for the basic technical research and creativeness predominance.

Regulator of G-Protein Signaling 4 (RGS4) Controls Morphine Reward by Glutamate Receptor Activation in the Nucleus Accumbens of Mouse Brain

  • Crosstalk between G-protein signaling and glutamatergic transmission within the brain reward circuits is critical for long-term emotional effects (depression and anxiety), cravings, and negative withdrawal symptoms associated with opioid addiction. A previous study showed that Regulator of G-protein signaling 4 (RGS4) may be implicated in opiate action in the nucleus accumbens (NAc). However, the mechanism of the NAc-specific RGS4 actions that induce the behavioral responses to opiates remains largely unknown. The present study used a short hairpin RNA (shRNA)-mediated knock-down of RGS4 in the NAc of the mouse brain to investigate the relationship between the activation of ionotropic glutamate receptors and RGS4 in the NAc during morphine reward. Additionally, the shRNA-mediated RGS4 knock-down was implemented in NAc/striatal primary-cultured neurons to investigate the role that striatal neurons have in the morphine-induced activation of ionotropic glutamate receptors. The results of this study show that the NAc-specific knock-down of RGS4 significantly increased the behaviors associated with morphine and did so by phosphorylation of the GluR1 (Ser831) and NR2A (Tyr1325) glutamate receptors in the NAc. Furthermore, the knock-down of RGS4 enhanced the phosphorylation of the GluR1 and NR2A glutamate receptors in the primary NAc/striatal neurons during spontaneous morphine withdrawal. These findings show a novel molecular mechanism of RGS4 in glutamatergic transmission that underlies the negative symptoms associated with morphine administration.

니코틴중독에 대한 족삼리 전침자극 및 황련의 작용기전 (Effect of Acupuncture and Coptidis Rhizoma on Repeated Nicotine-induced Behavioral Sensitization in the rats)

  • We have previously demonstrated that repeated injections of nicotine produced an increase in locomotor activity, dopamine(DA), release and c-Fos expression in the nucleus accumbens, one of the major projection areas of the central DA system. Acupuncture as a therapeutic intervention is widely used for the treatment of many functional disorders such as substance abuse and mental dysfunction. And many studies have shown that Coptidis Rhizoma has a suppressive effect on the central nervous system (CNS) and can affect the neurotransmitter systems in the CNS. In order to investigate whether acupuncture and Coptidis Rhizoma have an influence on nicotine-induced reinforcing and behavioral effects, we examined the effect of zusanli(ST36) and Coptidis Rhizoma on repeated nicotine-induced locomotor activity, and zusanli(ST36) on c-Fos expression as an important maker of postsynaptic neuronal activity in nucleus accumbens. Male SD rats received Coptidis Rhizoma (100mg/kg, p.o.) 30 min before injections of nicotine (0.4 mg/kg, s.c.) for 7 days. Rats were followed withdrawal for 3 days and one challenge for 1 day. Systemic challenge with nicotine produced a much larger increase in locomotor activity. Pretreatment with acupuncture at zusanli(ST36, 100Hz) and Coptidis Rhizoma decreased in nicotine-induced locomotor activity. These results demonstrated that reduction in locomotor activity by acupuncture at zusanli(ST36, 100Hz) and Coptidis Rhizoma may be mediated by reduction of dopamine release. Our results suggest that acupuncture at zusanli(ST36, 100Hz) and Coptidis Rhizoma may have therapeutic effect on nicotine addiction.

Do Opioid Receptors Play a Role in Blood Pressure Regulation?

  • The potential role of endogenous opioid peptides (EOPS) in cardiovascular regulation has only recently been entertained. EOPS have been localized in brain, spinal cord, autonomic ganglia, particularly the adrenal gland, and many other peripheral tissues. There are at least five major types of opioid receptors; namely ${\mu},\;{\delta},\;k,\;{\sigma},\;and\;{\varepsilon}$ and Experimental evidence indicates that cardiovascular actions of the peptide are mediated primarily by ${\mu},\;{\delta}$ and k receptors, and that these receptor types may be allosterically coupled. In anesthetized rabbits met-enkephalin decreased blood pressure and heart rate, which closely paralleled a reduction in sympathetic discharge. Naloxone, but not naloxone methobromide, antagonized these effects, which suggests a central site of action of met-enkephalin. A number of autonomic agents, particularly adrenergic ${\alpha}$-and, ${\beta}-agonists$ and antagonists modify the cardiovascular actions of met-enkephalin. Experiments in reserpine-treated and adrenalectomized rats provide no evidence of sympathetic nervous system involvement in the pressor responses to intravenous injection of opioid peptides, but rather suggest a direct peripheral action. Finally, activation of a beta-endorphinergic pathway projecting from the arcuate nucleus to the nucleus tractos solitarii in rats can cause naloxone reversible hypotension and bradycardia. There is evidence to implicate this pathway in antihypertensive drug action and in the modulation of baroreflex activity.

Keratinocyte Proliferation in Aged Rat Skin by High Voltage Pulsed Current Stimulation

  • The purpose of this study was to determine the effect of high voltage pulsed current (HVPC) stimulation on proliferative activities of basal keratinocytes by measured nucleolar organizer region (NOR) expression and thickness of spinous layer in aged rat skin. Fifty-one weeks old twelve male Sprague-Dawley rats (300∼350g) were divided into control and HVPC stimulation groups. Each animal's hair on the back were removed. The HVPC stimulation group received an negative monophasic twin peak pulsed current stimulation with 50 V, while the control group was given the same treatment without electricity. The rats were sacrificed after 3 weeks. The biopsy specimens were fixed in formalin, embedded in paraffin and stained with hematoxyline-eosin and silver nitrate. The thickness of basal to granular layer of the epidennis were measured using a light microscope and computerized image analysis system. The number of argyrophilic nucleolar organizer region (AgNOR) were counted using a light microscope and computerized image analysis system and calculated as the mean number of AgNOR per nucleus in the basal keratinocyte. By using a Student's t-test, an increase in the thickness of basal-spinous layer (P<0.001) of epidermis can be observed in HVPC stimulation rats as compared with the control rats, whereas the thickness of the granular layer is not affected. A Student's t-test showed a significantly higher mean NOR number per nucleus of the basal keratinocyte in the HVPC stimulation rats than control rats (P<0.001). There was significantly positive correlation between the NOR number and the thickness of basal-spinous layer (r=0.80, P<0.05). These results suggest that the HVPC stimulation may increase the thickness of spinous layer in the epidennis due to increased proliferative activities of basal keratinocytes in epidennis in aged rat skin.

Brain Mechanisms of Cognitive, Emotional and Behavioral Aspects of Taste

  • Taste is associated with hedonic evaluation as well as recognition of quality and intensity. Taste information is sent to the cortical gustatory area in a chemotopical manner to be processed for discrimination of taste quality. It is also conveyed to the reward system and feeding center via the prefrontal cortices. The amygdala, which receives taste inputs, also influences reward and feeding. In terms of neuroactive substances, palatability is closely related to benzodiazepine derivatives and $\beta$-endorphin, both of which facilitate consumption of food and fluid. The reward system contains the ventral tegmental area, nucleus accumbens and ventral pallidum and finally sends information to the lateral hypothalamic area, the feeding center. The dopaminergic system originating from the ventral tegmental area mediates the motivation to consume palatable food. The actual ingestive behavior is promoted by the orexigenic neuropeptides from the hypothalamus. Even palatable food can become aversive and avoided as a consequence of postingestional unpleasant experience such as malaise. The brain mechanism of these aspects of taste is elucidated.
