ADHESIVENESS EVALUATION OF ACTIVATED PLATELET USING Arg-Gly-Asp-Phe(RGDF)-IMMOBILIZED SURFACE

  • The adhesion of activated and normal platelets to fibrinogen requires the receptor binding site of GPIIb/IIIa. These recognition sites exists in the A ${\alpha}$ chain(RGDS at 572-575 and RGDF at 95-98) and the carboxy-terminal of ${\gamma}$ chain (HHLGGAKQAGDV at 400-411) of fibrinogen. In this study, we developed RGDF-immobilized surface to detect the unctional state of platelet. RGDF-immobilized surface was prepared on the glass using photolithographic technology. Platelet adhesion to RGDF-immobilized surface was observed by staining platelets with mepacrine using a fluorescence microscope using mepacrine. Using the RGDF peptide of fragment E, we observed that the platelets pretreated with PGE1 interacted incompletely with RGDF-immobilized surface, whereas ADP activated platelets interacted with the surface extensively. These results show that the distinct selectivity of RGDF-immobilized micro-patterned surface can be used to detect the unctional state of platelets.

  • Antimetastatic effects of Agaricus mixed prescription (AMP) were studied in the respect of blood-borne metastasis. For this aim, cytotoxicity against various cancer cells and normal cells, Chicken Chorioallantoic membrane (CAM) assay, cancer cell adhesion assay, platelet aggregation assay, pulmonary colonization, life span of S-180 implanted mice, and cytokine release assay were evaluated, respectively. The results were summarized as follows; AMP did not exert any cytotoxicity against all cell lines with IC50 of 25mg/ml on B16BL6. AMP disrupted formation of CAM at 1mg/ml. AMP was suppressive in adhesion assay of B16BL6. AMP also inhibited tumor induced platelet aggregation. In pulmonary colonization assay by B16BL6, the number of colonies in the lungs was significantly decreased in sample group than in control group. In animal study with S-180, the life span of AMP treated group was extended than that of control group. IL-12 was effectively increased in AMP treated group in cytokine release assay. Taken together, AMP can be possibly applied to cancer or metastasis.

  • Infantile hypertrophic pyloric stenosis (IHPS) a common childhood disorders characterized by nonbilious projectile vomiting, an olive shaped mass in the right upper quadrant of the abdomen and visible gastric peristaltic wave in the upper abdomen. Its etiology and pathogenesis are not clear but abnormal nerve distribution of the pylorus has been $postulated^{2-6}$. We performed immunocytochemical staning to the pyloric muscle from 10 IHPS and 3 controls patients, utilizing specific monoclonal antibody to NCAM(neural cell adhesion molecule). In IHPS patients, the number of NCAM protein immunoreactive nerve fibers were less than that in normal subjects. Auerbach myenteric plexuse was well developed and interbundle nerve plexuse was present but nerve fibers supplying individual muscle cells in smooth muscle bundles were poorly developed. These results indicate reduction of innervation in smooth muscles in IHPS patients that possibly contributes to the pathogenesis of IHPS.

  • Effects of sliding speed, applied load, and thickness of PMMA (Poly Methyl Methacrylate) coating layers on their dry sliding frictional and wear behavior were investigated. Sliding wear tests were carried out using a pin-on-disk wear tester. The PMMA layer was coated on Si wafer by a spin coating process with two different thicknesses, $1.5\mu\textrm{m}$ and $0.8\mu\textrm{m}$. AISI 52100 bearing steel balls were used as a counterpart of the PMMA coating during the wear. Normal applied load and sliding speed were varied. Wear mechanisms of the coatings were investigated by examining worn surfaces using an SEM. Friction coefficient of the coatings decreased with the increase of the applied load. Both adhesion and deformation of the coating determined the coefficient. The thicker PMMA layer with the thickness of $1.5mutextrm{m}$ showed lower friction coefficient than the thinner layer under most test conditions. Effects of sliding speed and applied load on the frictional behavior were varied depending on the thickness of the coating layer.

Antimetastatic effect of several crude drugs with antitumor activity on B16-Fo and A549 cells( II ) (항암활성 수종생약의 B16-Fo와 A549 암세포에 대한 항전이 효과( II ))

  • Ethyl ether fractions of Ferula Resina(EFR) and Lithospermi Radix(ELR) and hexane fraction of Salviae miltiorrhizae Radix(HSR) exerted an inhibitory effect on cell adhesion of A549 to extracelluar matrix most effectively in vitro cell adhesion assay. Thus, with above fractions for the evaluation of antitumor activity, T/C% was monitored in ICR bearing S-180 and for antimetastatic effect, pulmonary colonization assay, weight of organs, changes of WBC and platelet were studied. In pulmonary colonization assay incidence rate to control was 73 % 42 %, 14 % in ELR, HSR and EFR-treated groups repsectively. Gain of lung weight was significantly inhibited in all groups while spleen weight was significantly reduced only in SMR group, but no changes in kidney and liver as compared with control. Number of platelet was significantly increased in all groups to normal range as compared with thrombocytopenic contol. WBC was significantly reduced only in LR group. These results suggest that ethyl ether fraction of Ferula Resina has more effective antimetastatic activity.

CBP7 Interferes with the Multicellular Development of Dictyostelium Cells by Inhibiting Chemoattractant-Mediated Cell Aggregation

  • Calcium ions are involved in the regulation of diverse cellular processes. Fourteen genes encoding calcium binding proteins have been identified in Dictyostelium. CBP7, one of the 14 CBPs, is composed of 169 amino acids and contains four EF-hand motifs. Here, we investigated the roles of CBP7 in the development and cell migration of Dictyostelium cells and found that high levels of CBP7 exerted a negative effect on cells aggregation during development, possibly by inhibiting chemoattractant-directed cell migration. While cells lacking CBP7 exhibited normal development and chemotaxis similar that of wild-type cells, CBP7 overexpressing cells completely lost their chemotactic abilities to move toward increasing cAMP concentrations. This resulted in inhibition of cellular aggregation, a process required for forming multicellular organisms during development. Low levels of cytosolic free calcium were observed in CBP7 overexpressing cells, which was likely the underlying cause of their lack of chemotaxis. Our results demonstrate that CBP7 plays an important role in cell spreading and cell-substrate adhesion. cbp7 null cells showed decreased cell size and cell-substrate adhesion. The present study contributes to further understanding the role of calcium signaling in regulation of cell migration and development.

The Cytotoxic Effect of 3,4,5-Trihydroxybenzoic Acid and Related Compounds against Oral Cancer Cell Lines (3,4,5-Trihydroxybenzoic Acid와 관련 화합물의 구강암 세포주에 대한 세포독성효과)

  • Objectives : In order to evaluate the cytotoxicity of 3,4,5-trihydroxybenzoic acid and related compounds on the growth of normal cell lines and human oral epithelioid cell line, cell viability, cell adhesion ability, and morphological changes of cells were examined. Methods : We measured the cytotoxicity of 3,4,5-trihydroxybenzoic acid and related compounds with 3-[4,5-dimethyl-thiazol-2-yl]-2,5-diphenyltetrazolium bromide-[MTT), and 2,3-bis-[2-methoxy-4-nitro-5-sulfophenyl]-2H-tetrazolium -5-caboxanilide-[XTT) methods. Results : The cytotoxicity of 3,4,5-trihydroxybenzoic acid($IC_{50}$, $2,552.40\;{\mu}M$) was low according to the toxic criteria. Cytotoxic effect of 3,4,5-trihydroxybenzoic acid and related compounds against $IC_{50}$ value in cell morphology increased in a concentration-dependent manner. In light microscopy, $100\;{\mu}M$ 3,4,5-trihydroxy-benzoic acid showed th highest cytotoxic activity. Conclusions : These results suggest that 3,4,5-trihydroxybenzoic acid may have a potential anticancer activity.

Solid Lubrication Characteristics of DLC Coated Alumina Seals in High Temperature

  • Plasma immersion ion beam deposition (PIIBD) technique is a cost-effective process for the deposition of diamond like carbon thin film, the possible solid lubricant on large surface and a complex shape. We used PIIB process for the preparation of DLC thin film on $Al_2O_3$ with deposition conditions of deposition temperature range $200^{\circ}C$, working gas pressure of 1.310-1Pa. DLC thin films were coated by $C_2H_2$ ion beam deposition on $Al_2O_3$ after the ion bombardment of SiH4 as the bonding layer. Energetic bombardment of $C_2H_2$ ions during the DLC deposition to ceramic materials generated mixed layers at the DLC-Si interface which enhanced the interface to be highly bonded. Wear test showed that the low coefficient of friction of around 0.05 with normal load 2.9N and proved the advantage of the low energy ion bombardment in PIIBD process which improved the tribological properties of DLC thin film coated alumina ceramic. Furthermore, PIIBD was recognized as a useful surface modification technique for the deposition of DLC thin film on the irregular shape components, such as molds, and for the improvement of wear and adhesion problems of the DLC thin film, high temperature solid lubricant.

Characteristics of MOCVD Cobalt on ALD Tantalum Nitride Layer Using $H_2/NH_3$ Gas as a Reactant

  • Microprocessor technology now relies on copper for most of its electrical interconnections. Because of the high diffusivity of copper, Atomic layer deposition (ALD) $TaN_x$ is used as a diffusion barrier to prevent copper diffusion into the Si or $SiO_2$. Another problem with copper is that it has weak adhesion to most materials. Strong adhesion to copper is an essential characteristic for the new barrier layer because copper films prepared by electroplating peel off easily in the damascene process. Thus adhesion-enhancing layer of cobalt is placed between the $TaN_x$ and the copper. Because, cobalt has strong adhesion to the copper layer and possible seedless electro-plating of copper. Until now, metal film has generally been deposited by physical vapor deposition. However, one draw-back of this method is poor step coverage in applications of ultralarge-scale integration metallization technology. Metal organic chemical vapor deposition (MOCVD) is a good approach to address this problem. In addition, the MOCVD method has several advantages, such as conformal coverage, uniform deposition over large substrate areas and less substrate damage. For this reasons, cobalt films have been studied using MOCVD and various metal-organic precursors. In this study, we used $C_{12}H_{10}O_6(Co)_2$ (dicobalt hexacarbonyl tert-butylacetylene, CCTBA) as a cobalt precursor because of its high vapor pressure and volatility, a liquid state and its excellent thermal stability under normal conditions. Furthermore, the cobalt film was also deposited at various $H_2/NH_3$ gas ratio(1, 1:1,2,6,8) producing pure cobalt thin films with excellent conformality. Compared to MOCVD cobalt using $H_2$ gas as a reactant, the cobalt thin film deposited by MOCVD using $H_2$ with $NH_3$ showed a low roughness, a low resistivity, and a low carbon impurity. It was found that Co/$TaN_x$ film can achieve a low resistivity of $90{\mu}{\Omega}-cm$, a low root-mean-square roughness of 0.97 nm at a growth temperature of $150^{\circ}C$ and a low carbon impurity of 4~6% carbon concentration.

Synthetic Hydrogel as an Implant Material with Enhanced Biocompatibility and Biostability (생체적합성 및 생체안정성이 향상된 이식용 합성 수화젤)

  • Hydrogels with enhanced biocompatibility and biostability were prepared by copolymerization of 2-hydroxyethyl methacrylate (HEMA) and sodium methacrylate (SMA) at high monomer concentration to replace a sponge which has limited applications as an implant material. It was found that incorporation of SMA moiety suppressed cytotoxicity. P(HEMA-co-SMA) hydrogel prepared at SMA feed ratio of 0.05 showed minimal cytotoxicity as compared with a normal cell culture plate. The adhesion and the spreading of cells were preferred on the surface of the hydrogel prepared with SMA feed ratio of 0.01. On the other hand, the hydrogel prepared with SMA feed ratio of 0.05 showed lower cell adhesion. Histological findings revealed no evidence of significant foreign body reaction in the tissues around the copolymer hydrogels. Conclusively, it is suggested that the hydrogels prepared by copolymerization of HEMA and SMA at high monomer concentration are strong candidates for an implant material with excellent biocompatibility and biostability.
