Effect of Essential and Nonessential Amino Acids in North Carolina State University (NCSU)-23 Medium on Development of Porcine In vitro Fertilized Embryos

  • The present study was conducted to examine the effect of different levels of essential and nonessential amino acid in NCSU-23 medium on the in vitro-produced porcine embryo as it develops from the zygote to the blastocyst stage. Four experiments were performed, each with a completely randomized design involving 5 to 8 replications of treatments. In order to know the effect of nonessential amino acids in NCSU-23 medium, 0, 5, 10 and $20{\mu}/ml$ MEM were supplemented there to, (Exp. 1) and the medium was supplemented with same level of essential amino acids (Exp. 2). The combined effect of nonessential (0, 5, 10 and $20{\mu}/ml$ MEM) and essential amino acids (0, 5, 10 and $10{\mu}/ml$ MEM) in NCSU-23 medium (Exp. 3), first 72 h with non-essential amino acids (at 0, 5, 10 and $20{\mu}/ml$ MEM), and last 4 d with essential amino acids with the same level as NEAA (Exp. 4) were examined. The embryo development was monitored and the quality of blastocysts was evaluated by counting the number of total cells and determining the ratio of inner cell mass (ICM) to trophoectoderm (TE) cells. When Eagle's nonessential amino acids (MEM) added to NCSU-23 medium, it significantly increased the likelihood of development to the 2- to 4-cell stage and subsequent blastocyst development. Supplementation of different levels of essential amino acids in the NCSU-23 medium decreased cleavage rate, rate of morula and blastocyst development and the number of ICMs. In the case of the combined effect of essential and nonessential amino acids, better and significant results were found for blastocysts, hatching blastocysts and for ICM numbers which were also dose dependent. With respect to the biphasic effect of nonessential and essential amino acids, nonessential amino acids increased cleavage whereas essential amino acids increased the total cell number. Neither the nonessential nor the essential group of amino acids, on their own, affected blastocyst cell number or the differentiation of cells in the blastocyst. In conclusion, this study determined the role of nonessential and essential amino acids in the culture of the porcine embryo and showed that the embryo requires different levels of amino acids as it develops from the zygote to the blastocyst stage.

한국 성인 남자 및 여자의 혈청 유리 아미노산의 정상치 (Serum Free Amino Acid Levels in Korean Adult Males and Females)

  • 20-39세 사이의 성인 남자 34명과 여자 35명에 대하여 혈청 유리 아미노산의 정상치를 구하였다. 성인 남녀 다 같이 threonine(일부glutamine이 포함)alanine, lysine이 아미노산 중에서 약 40%를 차지하고 있었으며, 총 필수 아미노산의 함량은 총비필수 아미노산의 함량보다 유의하게 낮았다. 혈청아미노산 조성에서 뚜렷한 성별의 차이가 나타났는데 성인 남자에서는 alanine, cystine, valine, isoleucine, leucine, tyrosine, tryptophan, threonine, methionine, proline 등이 , 여자에서는 lysine, histidine이 각각 유의하게 높은 수준을 보였다. 총 필수 아미노산의 함량 그리고 총비필수 아미노산 및 총 아미노산에 대한 총필수 아미노산의 몰비도 성인 남자가 유의성 있는 높은 수준을 나타내었다. 아울러 총필수 아미노산에 대한 각 필수 아미노산의 몰비를 구하여 성별로 비교하였다.

고나트륨과 저나트륨 식이시 성인 여성의 혈중 아미노산 농도변화에 관한 연구 (Effects of High and Low Sodium Diet on Plasma Amino Acid Levels in Korean Adult Women)

  • This study was performed to determine the effect of dietary Na levels on plasma amino acid levels. Plasma amino acid levels were compared in 20 adult healthy women subjects who were given high Na diet (290.48 mEq/day : NaCl 17g) or low Na diet (51.26 mEq/day : NaCl 3g) for subsequent 6 days. Plasma essential amino acids levels were significantly decreased (36%) while plasma non-essential amino acids levels were significantly increased (22%) when subjects were given low Na diet (p<0.001). Among essential amino acids, threonine was decreased (74%) significantly when subjects were given low Na diet (p<0.01). Among nonessential amino acids, serine (49%), proline (20%) and aspartic acid (14%) were increased (p<0.01), while arginine (48%) and glutamic acid (27%) were decreased (p<0.001). In conclusion, dietary Na contents seemed to be an important factor to affect plasma amino acid levels. It would be appropriate to decrease the dietary Na intakes level considering the various clinical effects of dietary Na on the body fluid. For the patients who need low Na diet, it would be suggested that the level of dietary proteins should be carefully considered along with dietary Na manipulation. Central Functions of Amino Acids for the Stress Response in Chicks

  • The nutritional significance of essential amino acids, as well as non-essential amino acids, is well documented in poultry production with regards to growth performance and protein accretion. However, the function of amino acids in the stress response is still unclear. L-Pipecolic acid, a L-lysine metabolite in the brain, induced a hypnotic and sedative effect acting via the ${\gamma}$- aminobutyric acid receptors. L-Arginine also induced a sedative effect via its metabolism to L-ornithine. In addition, three-carbon nonessential amino acids like L-alanine, L-serine and L-cysteine also induced sedative effects. These facts suggest that the requirement for amino acids in both essential and non-essential types may require reconsideration to add the concept of stress amelioration in the future.

THE EFFECT OF DIFFERENT CARBOHYDRATE AND NITROGEN SOURCES ON THE UTILIZATION OF AMINO ACIDS IN THE SMALL INTESTINE OF SHEEP

  • This experiment was conducted to examine the effects of different carbohydrate and nitrogen source upon the utilization of amino acids in the small intestine of sheep. The results obtained are as follows: 1) For the quantities of total amino acid-N(TAA-N), essential amino acid-N(EAA-N) and nonessential amino acid-N(NEAA-N) passing at the duodenum and ileum except NEAA-N passing at the ileum were no significant differences (p>0.05) between diets. The quantities of NEAA-N passing at the ileum for the diets containing meat and bone meal were significantly higher (p>0.05) than the diets containing soybean meal. The mean values for the proportionate disappearance apparently digested TAA-N, EAA-N and NEAA-N within the small intestine for four diets were $0.692{\pm}0.0449$, $0.702{\pm}0.0132$ and $0.682{\pm}0.726$, respectively. 2) There were no significant differences (p>0.05) in duodenal individual amino acid flow between diets with the exception of aspartic acid and glycine. The amounts of each amino acid in duodenal digesta, expressed as a proportion of the amounts ingested for the four diets, were shown that there were net gains of EAA with the exception of arginine and NEAA with the exception of glutamic acid, glycine and praline prior to the small intestine. 3) Within the small intestine, there were no significant losses of each EAA (p>0.05) but significant losses of aspartic acid and glycine of NEAA between diets (p<0.01). The mean values of the proportionate losses of methionine, alanine and lysine within the small intestine were $0.816{\pm}0.04$, $0.767{\pm}0.04$ and $0.732{\pm}0.01$, respectively.

Seasonal Variation in the Nutritional Content of Mideodeok Styela clava

  • We evaluated changes in the nutritional composition of Mideodeok Styela clava harvested during the months of January, March, and May 2005. Glutamic acid, aspartic acid, arginine, leucine, and lysine were the most dominant amino acids present. Ratios of essential amino acids to nonessential amino acids were comparable to those of many fish species, with values ranging from 0.55 to 0.61 and 0.66 to 0.67 for muscle of Mideodeok from Geoje and Tongyeong, respectively. Mideodeok seems to be a suitable source of important fatty acids as it contains high levels of polyunsaturated fatty acids. Eicosapentanoic acid EPA; 20:5n-3 and docosahexanoic acid DHA; 22:6n-3 were the most dominant fatty acids, ranging from 20.0 to 22.3% and 16.5 to 17.9% in muscle, and 20.3 to 23.2% and 15.2 to 18.8% in tunic, respectively. The total mineral fraction of Mideodeok was 22.2-27.3% of dry matter. Sodium, calcium, magnesium, and potassium were the most dominant minerals in both muscle and tunic.

  • This study was conducted to investigate the contents of dietary fiber (DF), compositions of fatty acids in lipid fraction and amino acids in salt-soluble protein from the functional food source such as Platycodi radix, perilla seed, evening primrose seed and aloe vera. The contents of dietary fiber, neutral detergent fiber (N.D.F) , acid detergent fiber( A.D.F) . kignin, hemicellulose an dcellulose in evening primorse seed were higher than those of other samples, except the content of cellulose .The ration of polyunsaturated /saturated (P/S) fatty acid in total lipids was 6.31 in perilla seed, which was higher than those of other samples. The content of linolenic acid (n-3) in perilla seed was 55.47%. The content of linoleic acid (n-6) in evening primrose seed was 71.88% , which was higher than those of other samples. The fatty acid composition in neutral lipids were the same as those of total lipids. The PUFA contents of fatty acid in glycolipids were 61.76% in perilla seed. And also, the ratio of n-6/n-3 in evening primrose seed was 15.19. The fatty acid compositions in phospholipids were the sameas those of glycolipids. The contents of PUFA in Platycodi radix were 62.96% . The essential amino acid contents of salt-soluble protein were 47 mole % in Platycodi radix , which was slightly higher than those other samples. The ration of essential amino acid /nonessential amino acid (E/N) was 0.9 and 0.66 in Platycodi radix and aloe vera, respectively.

Chemical Composition of Pinus koraiensis Seed and Its Biological Activity

  • This study was to investigate the chemical composition and biological activities of Pinus Koraiensis seed. The oil, moisture, ash, crude protein, and carbohydrate contents of P. Koraiensis seed were 58.21, 7.84, 1.56, 14.26, and 18.13%, respectively. The ratios of essential amino acid and nonessential amino acid against total amino acids were 36.6 and 60.3%, respectively. The saponification value of seed oil was 166.8 mg KOH/g oil. Among various fatty acids, the linoleic acid content was the highest, 35.5%, which was approximately 72.6% of polyunsaturated fatty acid. The ethylacetate extract of P. Koraiensis seed had the highest DPPH radical scavenging activity (62.8%) at 7.0 mg/mL, followed by hexane extract, methanol extract, and hot water extract. The maximum nitrite scavenging activity was obtained 59.3% at pH 1.2. The total phenolic concentration of ethylacetate extract was 98.7 mg/g, approximately 4.8 folds higher than that of the hot water extract. The maximum inhibition activities of elastase using ethylacetate extract and collagenase using hexane extract were 58.8 and 40.7%, respectively. These results indicate that P. koraiensis seed extract could be applied to present the possibilities of industrial applications for the developments of cosmetics.

점등체계가 육계 닭고기의 아미노산과 지방산 조성에 미치는 영향 (Effect of lighting regimes on amino acid and fatty acid contents of broiler chicken meat)

  • The study was carried out to investigate the effect of lighting regimes on performance and meat quality of broiler. A total of 912 chicks was divided into 2 groups with male (3 replications/group, 34 heads/replication) and female (3 replications/group, 42 heads/replication). Treatments were control (C, natural lighting), T1 treatment (24L), and T2 treatment (1~2d, 24L; 2~4d, 23L+1D; 4~16d, 16L+8D; 16~21d, 18L+6D; 21~42d, 23L+1D) by lighting regimes. Lysine content of male chicken meats at T2 treatment was lower compared to that of other treatments (p<0.05). There was no significant difference in all nonessential amino acid (AA) among treatments. Methionine content of female chicken meats at T2 treatment was lower compared to that of other treatments (p<0.05). The rate of C16:0, C16:1n7, and C18:1n9 of male chicken meat at T2 treatment was higher than that of other treatments (p<0.05), while C18:2n6 was lower at T2 treatment than other treatment (p<0.05). Mono-saturated fatty acid was the highest at T2 treatment, however, polysaturated fatty acid was the lowest at that treatment (p<0.05). There was no considerable difference in fatty acid composition of female chicken meat among treatments. Finally, lighting regime didn' affect on amino acid and fatty acid contents, and used possibly on energy reduction.

가다랑어 내장의 효율적인 이용방법 개발에 관한 연구 (A Study on Developement of Effective Utilization Method of Skipjack Tuna Viscera)

  • 가다랑어 내장의 효율적 이용방법을 개발하기 위하여 내장을 자숙한 후 압착분리한 잔사에 Aspergillus oryzae를 이용하여 발효시킴과 동시에 이때 분리된 추출액을 원심분리하여 유분을 제거한 후 농축시켜 발효중인 잔사에 다시 첨가하는 개량식 발효어분 제조법(FFMA)을 개발하여 발효시 추출액을 첨가하지 않는 가등(加藤)법(FFMN) 및 비발효법인 재래법(NFM)과 비교 연구결과는 다음과 같다 발효어분에 있어서 일반성분의 변화는 3가지 방법간에는 큰 차이는 볼 수 없었고, 칼슘의 함량이 개량법에서 161.0 mg%로 가등(加藤)법과 재래법의 147.0 mg% 및 148.0 mg%보다 높았다. $Vitamin\;B_{1},\;B_{2}$ 및 C의 함량은 개량법에서 각각 1.76, 2.18 및 43.10 mg%로 가등(加藤)법의 1.373, 1.383 및 35.6 mg%, 재래법의 0.732, 0.669 및 14.20 mg%보다 각기 높게 나타났다. 과산화물가는 개량법에서 33.6으로 가등(加藤)법의 29.0보다 약간 높았으나 재래법의 126.0보다는 훨씬 낮았다. 개량식 발효어분에서 추출된 추출물과 가다랑어 내장을 그대로 추출된 추출물의 유리아미노산 함량은 총 유리아미노산에서는 발효 어분에서 3957 mg/100g로 내장의 경우보다 1.9배 정도 높았다. 총 유리필수아미노산의 경우도 발효어분에서 884 mg/100g로 내장의 경우보다 1.2배 정도 높았고, 총 유리비필수아미노산의 경우도 발효어분에서 2733 mg/100g로 내장의 경우보다 2.5배 높았다. 개별 유리아미노산의 경우 발효어분을 사용한 경우가 내장을 사용한 경우보다 필수아미노산의 경우 threonine, methionine, lysine 등의 함량이 높았고 비필수아미노산은 거의 모두가 월등히 높게 나타났다.
