CK2 is a serine threonine kinase that participates in a variety of cellular processes with more than 300 defined substrates. This critical enzyme is known to be upregulated in cancers, but the role of this upregulation in carcinogenesis is not yet fully understood but c-myc, one of the defined CK2 substrates, is a well-known proto-oncogene that is normally essential in developmental process but is also involved in tumor development. We evaluated the optimal enzyme and substrate concentrations for CK2 activity in both neoplastic and non-neoplastic human lung tissues using the c-$myc^{424-434}$ peptide (EQKLISEEDL) as a substrate. The activities measured for the neoplastic tissue were 600-750 U/mg protein while those for the control tissue was in the range of 650-800 U/mg. $K_m$ value for c-myc peptide was determined as $0.33{\mu}M$ in non-neoplastic tissue and $0.18{\mu}M$ in neoplastic tissue. In this study, we did not observe an increased activity in the neoplastic tissue when compared with the non-neoplastic lung tissue, but we recorded two times higher affinity for c-$myc^{424-434}$ in cancer tissue. Considering the metabolic position of c-$myc^{424-434}$, our results suggest that phosphorylation by CK2 may be important in dimerization and thus it might affect the regulation of c-myc in cancer tissues.
Objectives: This study was conducted to examine the effects of nutrition education with a dietary guidebook for children on dietary attitude, nutrition knowledge and nutrient intakes. Methods: The subjects were 54 higher grade elementary students (27 educated vs. 27 non-educated). The educated group was provided individual and/or group lessons (40 min/lesson/week, 4 week) using a dietary guidebook of Children developed by The Korean Society of Community Nutrition (KSCN) & Korean Food and Drug Administration (KFDA). The contents were Balanced Diet, Smart Food Choices, Cooking a Healthy Snack and Building a Healthy Body. We examined the differences in nutrition knowledge, dietary attitudes and dietary intake between the educated group and non-educated group. Results: After education, the educated group improved dietary attitude, nutrition knowledge and qualitative nutrient intakes compared to the non-educated group. Specifically, among dietary attitudes, 'taking a meal with salty and spicy food' increased, while among nutrition knowledge, 'functions of protein', 'functions of fat', 'foods with carbohydrates', 'foods with fat', 'foods with vitamins', and 'foods with minerals' were increased. Nutrition adequacy ratio (NAR) scores for vitamin C, iron, and zinc were increased. Conclusions: Nutrition education using a dietary guidebook for children developed by the KSCN & KFDA had positive effects on nutrition knowledge and qualitative nutrient intakes. These findings suggest that nutrition education focused on personalized daily energy and nutrient requirements may improve dietary attitude and quantitative nutrient intakes of children.
본 연구의 목적은 학생들의 일상적 소비의 맥락 속에서 인간-비인간, 장소에 대한 이해의 관계적 전환에 기반하여 윤리적 지리교육의 가능성을 모색해보는 데에 있다. 이를 위해 먼저 학생들이 소비하는 상품 네트워크가 탈 국지화되면서 타자에 대한 책임과 배려의 윤리가 축소되는 상황에 대하여 살펴보았다. 이어서 학생들이 소비와 윤리의 문제를 책임과 배려의 관계적 윤리라는 관점을 통하여 바라볼 수 있는 안목의 토대로서 장소와 인간-비인간의 관계적 이해라는 관점을 제시한다. 마지막으로 휴대폰 커넥션을 사례로 소비, 관계적 지리, 책임과 배려의 윤리의 관계를 살펴보았다. 윤리적 지리교육의 역할은 학생들이 자신들의 삶 속에서 부재하는 현존으로서 다양한 인간 및 비인간들과 연계감을 가지고 이들에 대하여 더 많은 책임과 배려를 제공할 수 있는 인식적, 실천적 능력을 가지도록 하는데 있다는 점을 주장한다.
DIY(Do It Yourself) 중심의 창작 활동이 확산되면서 일반 사용자들이 손쉽게 아이디어를 구체화 할 수 있는 환경이 마련되고 있다. 3D 프린터를 비롯한 디지털 기술과 도구의 대중화가 급속도로 진행되고 있는 반면, CNC 조각기의 경우 전문 지식의 부족과 복잡한 사용 절차로 인해 사용이 원활하지 않다. 이에 본 연구에서는 3D 형상 모델 파일만 선택하면 자동으로 가공이 가능한 CNC 조각기의 시스템을 개발하였다. 피삭재와 공구의 크기 및 종류를 제한하고, 생산성의 배제를 통해 공정 계획 및 공구경로 생성, 피삭재 고정, 피삭재 좌표계 파악을 자동화하였다. 그 결과 비숙련자도 혼자 충분히 사용 가능한 CNC 조각기의 시스템이 개발되었다. 이를 통해 기존보다 넓은 범위에서 다양한 사용자들이 사용할 수 있는 CNC 조각기의 발전이 기대된다.
The purpose of this study was to investigate perception and awareness of yaksun and willingness to eat yaksun in the future, and to examine the factors affecting intake behavior of yaksun of adults in Daegu and Gyeongbuk region. The data were collected in April, 2011 through a self-administered questionnaire. Chi-square test, t test, and binary logistic regression were conducted by SPSS Windows V.19.0. The results were as follows. Almost sixty-six percent of the respondents answered that they knew yaksun. The perception level of yaksun for the intake respondents was higher than that of non-intake respondents. Additionally, many respondents agreed to varied menu development and convenient-food development of yaksun. Intake respondents showed higher level of agreement for the varied menu development of yaksun than non-intake respondents. On the other hand, four fifths of the respondents showed that they would be willing to eat yaksun in the future. The level of willingness to eat yaksun in the future for the intake respondents was higher than that of non-intake respondents. Additionally, the respondents who knew yaksun showed higher level of willingness to eat it than those who did not know it. Finally, the result of binary logistic regression analysis for intake behavior of yaksun showed that gender, educational level, and the awareness of yaksun were significant. On the basis of these results, not only the promotion of yaksun to consumers but also the development of convenient foods, varied menus for different age groups, and menus with cheap prices are needed to raise yaksun popularity.
Three phase hollow fiber-liquid phase microextraction (HF-LPME), which is faster, simpler and uses a more environmentally friendly sample-preparation technique, was developed for the analysis of Non-Steroidal Anti-Inflammatory Drugs (NSAIDs) in human urine. For the effective simultaneous extraction/concentration of NSAIDs by three phase HF-LPME, parameters (such as extraction organic solvent, pH of donor/acceptor phase, stirring speed, salting-out effect, sample temperature, and extraction time) which influence the extraction efficiency were optimized. NSAIDs were extracted and concentrated from 4 mL of aqueous solution at pH 3 (donor phase) into dihexyl ether immobilized in the wall pores of a porous hollow fiber, and then extracted into the acceptor phase at pH 13 located in the lumen of the hollow fiber. After the extraction, 5 ${\mu}L$ of the acceptor phase was directly injected into the HPLC/UV system. Simultaneous chromatographic separation of seven NSAIDs was achieved on an Eclipse XDB-C18 (4.6 mm i.d. ${\times}$ 150 mm length, 5 ${\mu}m$ particle size) column using isocratic elution with 0.1% formic acid and methanol (30:70) at a HPLC-UV/Vis system. Under optimized conditions (extraction solvent, dihexyl ether; $pH_{donor}$, 3; $pH_{acceptor}$, 13; stirring speed, 1500 rpm; NaCl salt, 10%; sample temperature, $60^{\circ}C$; and extraction time, 45 min), enrichment factors (EF) were between 59 and 260. The limit of detection (LOD) and limit of quantitation (LOQ) in the spiked urine matrix were in the concentration range of 5-15 ng/mL and 15-45 ng/mL, respectively. The relative recovery and precision obtained were between 58 and 136% and below 15.7% RSD, respectively. The calibration curve was linear within the range of 0.015-0.96 ng/mL with the square of the correlation coefficient being more than 0.997. The established method can be used to analyse of NSAIDs of low concentration (ng/mL) in urine.
Background: This study was designed to investigate the role of the hepatocyte growth factor (HGF) with regards to differentiation of somatic stem cells originating from the human umbilical cord blood (UCB) into hepatic lineage cells in vitro culture system. Methods: Mononuclear cells from UCB were cultured with and without HGF based on the fibroblast growth factor (FGF)-1, FGF-2, and stem cell factor. The cultured cells were confirmed by immunofluorescent staining analysis with albumin (ALB), cytokeratin-19 (CK-19), and proliferating cell nuclear antigen (PCNA) MoAb. ALB and CK-18 mRNA were also evaluated by reverse transcription-polymerase chain reaction. In order to observe changes in proliferating capacity with respect to the cultured period, CFSE with affinity to proliferating cells were tagged and later underwent flow cytometry. Results: In the HGF-treated group, cultured cells had a large oval shaped appearance with adherent, but easily detachable characteristics. In the HGF-non treated group, these cells were spindle-shaped with strong adherent characteristics. Expressions of ALB and CK-19 were evident in HGF-treated group compared to non-expression of those in to HGF-non treated group. Dual immunostaining analysis of the ALB producing cells showed presence of PCNA in their nuclei, and ALB and CK-18 mRNA were detected on the 21st day of cultured cells in the HGF-treated group. Conclusion: Our findings suggest that HGF has a pivotal role in differentiating somatic stem cells of human UCB into hepatic lineage cells in vitro.
본 연구는 특전부사관 우수자원 홍보를 통해 인력을 획득하는 연구의 문제에서 출발하여 다음 4가지 홍보 전략을 연구하였다. 첫째, 페이스북의 홍보 전략 매개체이다. 둘째, 유튜브의 홍보 전략 매개체이다. 셋째, 블로그의 홍보 전략 매개체이다. 넷째, Web site의 홍보 전략 매개체이다. 따라서 특전부사관 우수자원 획득을 위한 전략적인 매개체를 활용하여 홍보 활동을 통한 우수자원을 획득하는 데 있다. 더 나아가 특전 부사관 우수인력을 획득하는데 4가지 홍보 전략과 외국군 우수인력 획득사례를 통해 본 연구를 고찰하고자 한다. 결국, 본 연구는 전략적 매개체를 활용하여 특전부사관 우수자원 획득을 위해 모병 활동을 하는 데 기초적인 시사점을 제공할 수 있다.
Gefitinib, a first generation epidermal growth factor receptor tyrosine kinase inhibitor (EGFR TKI), provides obvious clinical benefit in patients with EGFR-mutant non-small cell lung cancer (NSCLC). However, patients ultimately develop gefitinib resistance which mainly caused by EGFR T790M secondary mutation. In the current study, we investigated whether the root extract of Scutellaria baicalensis (SB) overcomes gefitinib resistance. Gefitinib-resistant H1975 human NSCLC cells (EGFR L858R/T790M double mutant) were treated with gefitinib and/or ethanol extract of SB (ESB) to evaluate the effect of ESB on the gefitinib sensitivity. The cell viability was measured by MTT assay and trypan blue exclusion assay. The colony-forming ability was evaluated by anchorage-dependent colony formation assay. Combined treatment with gefitinib and ESB markedly decreased the cell viability and colony formation than single treatment with gefitinib or ESB in H1975 cells. In addition, cells treated with both gefitinib and ESB exhibited a significant increase of sub-G1 DNA content which indicates apoptotic cells compared with those treated with gefitinib or ESB alone. As a molecular mechanism, combined treatment with gefitinib and ESB strongly downregulated the phosphorylation of ERK and JNK than single treatment with gefitinib or ESB. Taken together, our results demonstrate that ESB sensitizes H1975 cells to gefitinib treatment. We cautiously propose that ESB can be used in combination with gefitinib for the advanced NSCLC patients with acquired resistance to EGFR TKIs.
This study was intended to investigate the effect on the human body such as subcutaneous fat thickness the circumference of extremities etc. of the clothing training of putting on thin clothes periodically from the cold period. The subjects were divided into the clothing training group and the non-training group, The training group was asked to wear cool clothes in daily life and to wear the training clothes of T-shirts with half-length sleeves and pants and perform the clothing training for two hours daily three times a week in a cold environment over the period from November to February. The non-training group was asked to lead a life wearing comfortable clothes. Then a comparative experiment was conducted at 15$\pm$1$^{\circ}C$, 50$\pm$5% R, H and 0.25m/sec before and after the clothing training. After the clothing training regardless of gender subcutaneous fat thickness was more increased and total clothing weight per the surface area of the body was decreased in the training group than the non-training group. The training group showed lower skin temperature in the limbs and lower average skin temperature than the non-training group irrespective of gender which proved the effect of the clothing training. The training group was shown to have attendancy toward a greater sense of warmth and a less sense of discomfort which proved the effect of the clothing training.
