Effect of flaking on the digestibility of corn in ruminants

  • In this study, we aimed to assess the effect of flaking on the nutrient digestibility of corn grain in ruminants. In this regard, in vitro rumen fermentation, in situ rumen degradability, and in vivo metabolic experiments were performed. The automated gas production technique was used for the in vitro fermentation experiments. Six types of corn flakes with various degrees of gelatinization (32%, 41%, 48%, 66%, 86%, and 89%) were ground and incubated in rumen fluid to measure rumen fermentation characteristics and digestion rate. The in situ degradability of ground corn, whole corn, and corn flakes with 62% and 66% gelatinization was measured by incubation in the rumen of two cannulated Holstein cows. In vivo metabolic experiments were performed using 12 crossbred goats (29.8 ± 4.37 kg) using a 3 × 3 Latin square design. The dietary treatments consisted of ground corn and flaked corn with 48% or 62% gelatinization. In vitro experiments showed that as the degree of gelatinization increased, the digestion rate increased linearly, while the discrete lag time decreased linearly (p < 0.05). The effective rumen dry matter degradability, determined by in situ fermentation, was 37%p lower in corn flakes than ground corn, assuming a passage rate of 6%/h (p < 0.01), and there was no difference between the two flakes. In the in vivo experiment, there was no difference in dry matter intake, average daily gain, feed efficiency, and nitrogen utilization among the treatment groups (p > 0.05); however, the crude fat digestibility was lower for corn flakes than for ground corn (p < 0.05). To summarize, the rate of fermentation of corn flakes increased as the degree of gelatinization increased. However, non-ground corn flakes had lower rumen digestibility and did not improve in vivo apparent nutrient digestibility, compared with ground corn. In contrast to the assumption that flaked corn provides more energy to ruminant animals than ground corn, we conclude that the digestibility and energy value of corn flakes are lower than those of ground corn if mastication does not sufficiently reduce the particle size of corn flakes.

  • Measurement of DCP is considered inadequate and unsatisfactory means of assessing the protein value of the diet as no distinction is made between the digestion in ferestomach and in the small intestine. Protein meals should be classified on the basis of rumen degradable protein (RDP) and rumen undegradable protein (UDP). Usually, protein meals naturally available with high level of UDP or bypass protein value should be preferred for incorporation in the diet of lactating and growing animals. However, if such resources are non-available or are expensive, protein meals having high degradability can be carefully subjected to heat or formaldehyde treatment to achieve desired level of rumen bypassability. Various studies conducted the world over have revealed that bypass protein feeding to ruminants, especially when animals are fed on crop residue based basal diet, help increasing feed conversion efficiency in growing and lactating ruminants.

  • In this study we performed test to compare and analyze the chloride diffusion properties in concrete according to organic coating materials used at concrete structures widely and inorganic coating materials as eco-materials in recently. The results of study were shown as follow ; 1) Chloride diffusion coefficient was low as low as and that of organic and inorganic painted was lower than non-painting 2) In case of inorganic coating material applied in this study, it should be possible to evaluate the chloride diffusion properties by CTH method. Because presumption value by CTH method is similar with real estimate value by digestion experiment.

Elevation Factors of Fibrinogen in the Elderly Koreans

  • Plasma fibrinogen is risk factor of vascular disease including stroke, ischemic heart disease, atherosclerosis and thrombosis. Many studies have confirmed that high plasma fibrinogen levels are related with age, obesity, cholesterol, alcohol consumption, and genotype. This study was carried out to investigate the effect of fibrinogen genotype and other characteristics on the plasma fibrinogen levels in the elderly Koreans. For this study the blood samples were collected from 178 healthy elderly Koreans (102 males and 76 females, $55{\sim}80$ year olds). The blood samples were analyzed by smoking status, cholesterol levels, genotype, age, exercise, drinking, and gender. The plasma fibrinogen was assayed by clotting method, cholesterol being assayed by cholesterol oxidase method. The $\beta$-fibrinogen genotype was detected by PCR of relevant region and digestion with Alu I. The alleres with the restriction site and the non cleavable alleres were designated $A_1$ and $A_2$. In conclusion, genotype $A_1A_2$ and exercise are increased and associated with plasma fibrinogen levels. But, there were no significant differences by smoking, gender, age, drinking and cholesterol.

도축 폐혈액 단백질을 이용한 유산균체의 생산

  • For the utilization of animal blood produced in slaughter for the cultivation of lactic acid bacteria, the nitrogen sources in a complex(MRS) medium were replaced by blood plasma proteins. Focusing the purpose on the industrial production of a probiotics, the hydrolytic activities of three industrially applicable proteases were compared for the effective digestion of the proteins, and Alcalase(the product of Novo Nordisk) was selected with comparatively high activity. The growth of Streptococcus thermophilus KCCM12020 was best among the four strains of lactic acid bacteria tested. With Alcalase-digested proteins in the medium, the growth rates and the final cell concentrations were higher than those with non-digested proteins. The cell mass produced in the medium containing blood proteins as nitrogen sources, $2.5{\times}10^9$ CFU/ml, was significantly high and about 70% of that in MRS medium, showing a great possibility for the utilization of animal blood proteins as economic nitrogen sources in the production of cell mass of lactic acid bacteria.

  • Enteropathogenic Yersina enterocolitica is an important cause of human and animal disease. Phenotypic and genotypic characteristics currently used to identify Yersinia enterocolitica are not necessarily sufficient to differentiate pathogenic from non-pathogenic strains or to analyze the epidemiology of yersiniae at a molecular level. To improve the characterization of Yersinia enterocolitica, A total of 65 isolates of Yersinia enterocolitica were examined with bioserotyping, antibiotic susceptibilities, PFGE, PCR-ribotyping. Genomic DNA pattern generated by PFGE are highly specific for different strains of an organism and have significant value in epidemiologic investigations. The PFGE analysis of Not I-digested chromosomal DNA of Y. enterocolitica were performed with a CHEF Mapper(Bio-Rad, USA). Not I generated 19 restriction endonuclease digestion profiles(REDP). PCR-ribotyping, performed with primers complementry to conserved regions of 16S and 23S rRNA gene, generated 13 ribotypes. PCR-ribotyping can be considered a good technich for subtyping strains of Y.enterocolitica.

Moreover, Textile dying is a chemically intensive process and consumes large quantities of water.   • To see the effect of magnesium on adhesion to natural zeolites, a series of batch tests were performed in this research. Mixed bacteria were sampled from the digestion tank at a local sewage treatment plant in Seoul. Magnesium-zeolites were synthesized by mixing natural zeolites with 0.096 M, 0.24 M, and 0.48 M of MgCl2 solution. For comparison, manganese and trivalent ferric zeolites were also prepared. Two grams of 0.2 mm $\sim$ 0.3 mm sized zeolites(non-treated, Mg, Mn and Fe(III) treated zeolites) and 20 mL of water were mixed in a Corex 25 mL tube. Five milliliters of culture solution including bacteria was added to the tube. The tubes were equilibrated in a shaking incubator at mesophilic temperature $(30{\pm}2^{\circ}C)$. The bacterial concentrations were measured with a Microluminometer (New Horizons 3550i) and total organic carbon (TOC) spectrophotometer (Multi NC-3100).

Effect of Digestibility of Soybean Peptides on the Concentration of Serum Cholesterol in Rats (대두 펩타이드와 소화율이 흰쥐의 혈청 콜레스테롤 농도에 미치는 영향)

  • In order to investigate the hypocholesteremic effect of soybean perptides, soybean protein(ISP), casein(CNP), their peptic hydrolyzates fractionated by acid precipitation at different pH's(SHT, SH8, SH6, SH4, CHT, CH5, CH4) and amino acid mixtures of the same composition as the proteins(SAA, CAA) were fed to rats and the concentration of serum cholesterol was measured. Then in vitro digestibility and molecular weight distribution of the peptides by pepticpancreatic hydrolysis was measured by FPLC. The lower the in vitro digestibility of peptides is, the lower the concentration of serum cholesterol becomes(r=0.986) and the higher the ratio of macropeptides is, the lower the concentration of serum cholesterol becomes(r=-0.932) in rats. These results suggest that the in vitro digestibility of peptides has close relationship to the concentration of serum cholesterol in rats and non-digestible meacropeptides or polypeptides especially more than 1 kDa, formed through digestion in gut, may lower the serum cholesterol in rats.

Studies on Heated Protein Quality Using Homoarginine Method

  • To determine the quality of heated protein, in vitro method, invluding lysine, lysionalanine, and fructose-lysine as well as homoarginine by guanidination of lysine, was assessed using heated casein with of without glucose. In vivo methods such as PER, digestibility and BV were also tried on homoarginine, lysinoalanine, fructoselysine, and lysine. The nonreactive lysine for huanidination was hardly digestive, while the non heat damaged lysine side chanis in the protein were accessible for guanidination as well as for the digestion. A linear correlation(${\gamma}$=0.80) was obstained between PER and digestibility of the analysed lysine. Digestibility of homoarginine was higher that of true protein. However, in the guanidinated heated casein with glucose, digestibility of homoarginine was significantly reduced. It is suggested that the homoarginine method may mislead to over- or underestimation of the damaged protein quality.
