  • Treating bulk superalloy scrap with molten zinc has been studled to facililate recycling and recovery- of cobalt.Superalloys investigated were the cobalt-base Mar-M-509 and X45 and the nickel-base Rene 80. Charges withZnlscrap ratlos of 1.5-6.5 were heated to 750-9002 far 1-7.5 hours in a nitrogen atmosphere. The moltenzinc dissolved superalloy scrap and zinc was removed by vacuum distillation at 850-Wk for 4-6 hours. Ithas been concluded that the optimum conditions of decomposition for Mar-M-509 and Rene 80 \"ere dissolutiontemperature of about 850k, Znlscrap ratlo of about 5, and dissalution time of about 5.5 hours. The zinc-treatedsuperalloy prouducts were friable and reacted rapidly with acid solutions. Leaching 9mm pieces of unalloyedMar-M-509 or Rene 80 with 5 times the stolchlometric amount oi 6N HCI at 90t ior 3 hours dissolved about1.5-7.270, while leachmg of the minus 20-mesh products dissolved about 89.0-93.0%.ved about 89.0-93.0%.

Cryopreservation of Citrus limon (L.) Burm. F Shoot Tips Using a Droplet-vitrification Method

  • This study describes the successful establishment of a cryopreservation protocol for Citrus limon cultivars: 'Frost Eureka limon' and 'Cook Eureka limon', using a droplet-vitrification method. The shoot tips that were excised from in vitro grown seedlings of the two cultivars were preserved in liquid nitrogen (LN) and successfully regenerated into whole plants. Excised shoot tips were pre-cultured for 1 or 2 days in 0.3 M and 0.5 M sucrose solutions at $25^{\circ}C$ and incubated in a loading solution (LS) composed of 17.5% glycerol + 17.5% sucrose in Murashige and Skoog (MS) medium for 40 min at $25^{\circ}C$. Prior to direct immersion in LN for 1 h, the shoot tips were dehydrated with plant vitrification solution 2 (PVS2) at $0^{\circ}C$ or PVS3 at $25^{\circ}C$. The frozen shoot tips were re-warmed and unloaded with 1.2 M sucrose in $\text\tiny{^1/_2}$ MS for 30 min at $25^{\circ}C$. Shoot tips were post-cultured overnight on survival medium and then micrografted onto 'trifoliate orange' (Poncirus trifoliate (L.) Raf. seedling rootstocks for recovery and to produce whole plants. The highest regrowth rates were 53.5% and 50.3% for cryopreserved shoot tips of 'Frost Eureka limon' and 'Cook Eureka limon', respectively, when pre-cultured in 0.3 M and 0.5 M sucrose concentrations in a sequencing manner, with LS and treated with PVS2 for 60 min at $0^{\circ}C$. We also investigated whether the ammonium ion concentration on post-culture medium affected the viability of the cryopreserved Citrus shoot tips. The viability of cooled samples, following culturing on woody plant media (WPM) containing $\text\tiny{^1/_4}$ ammonium nitrate overnight before micrografting, was the highest (70.3%) in 'Frost Eureka limon'. The study described here is a cost-effective and safe method to conserve Citrus fruit cultivars, for the improvement and large-scale multiplication of fruit plants and for breeding disease resistance.

A Case of Renal Cortical Necrosis in a 15-year-old Boy with Acute Kidney Injury

  • Renal cortical necrosis (RCN) is patchy or diffuse ischemic destruction of the renal cortex caused by significantly reduced renal arterial perfusion. It is a rare cause of acute kidney injury (AKI) and is associated with high mortality. Here, we review the case of RCN in a 15-year-old boy who developed AKI. A 15-year-old boy was referred to our hospital from a local hospital due to a sharp decrease in his renal function. He presented with acute flank pain, nausea with vomiting, and oliguria for the past two days. He had taken a single dose of antihistamine for nasal congestion. At our hospital, his peak blood pressure was 148/83 mmHg and he had a high body mass index of $32.9kg/m^2$. The laboratory data showed a blood urea nitrogen (BUN) of 28.4 mg/dL, a creatinine of 4.26 mg/dL, and a glomerular filtration rate estimated from the serum cystatin C of $20.2mL/min/1.73m^2$. Proteinuria (spot urine protein to creatinine ratio 1.66) with pyuria was observed. Kidney sonography showed parenchymal swelling and increased renal echogenicity. Due to rapidly progressing nephritis, steroid pulse therapy (750 mg/IV) was done on the second day of his admission and the patient showed complete recovery with normal renal function. However, the kidney biopsy findings revealed renal cortical hemorrhagic necrosis. Multifocal, relatively well-circumscribed, hemorrhagic necrotic areas (about 25%) were detected in the tubulointerstitium. Although RCN is an unusual cause of AKI, especially in children, pediatricians should consider the possibility of RCN when evaluating patients with rapidly decreasing renal function.

An Adaptive Local Management Approach Cannot Overcome Large-Scale Trends: A Long-Term Case-Study for Saxifraga hirculus Conservation

  • Saxifraga hirculus is classified as a vulnerable plant species in Europe, and it is confined to base-rich flushes in the British uplands. However, a lack of available information about its conservation status hampers the development of adaptive strategies for its in-situ conservation, especially with respect to grazing. To assess the effectiveness of sheep grazing in maintaining viable populations of S. hirculus, we compared the community dynamics of the vegetation in a base-rich flush over 44 years in two plots: one sheep-grazed under business-as-usual sheep grazing densities and the other fenced to exclude grazing. The plots were established in 1972, and the abundances of all vascular plants, bryophytes, and litter were measured at six intervals until 2016. Our results showed that although the presence of S. hirculus was maintained in both plots over the 44 years, it declined and reached a minimum between 1995 and 2010, when it was close to extinction. Since 2013, Saxifraga has recovered only slightly. Interestingly, the S. hirculus response appeared to be independent of grazing treatment, but it mirrored wider changes in the vegetation composition and structure within the flush over the 44 years. These changes are similar to others reported in broader uplands that have been attributed to a combination of reduced nitrogen and sulfur deposition and global warming. Thus, the simple adaptive management approach of "just managing" sheep grazing appeared ineffectual for preserving the S. hirculus population. S. hirculus showed signs of recovery at the end of the study period within this base-rich flush.

Anti-fatigue activity of dripped spent hens chicken essence in ICR mice

  • Objective: Chicken essence and branched chain amino acid (BCAA) supplementation has been recognized to significantly relieve fatigue. To obtain chicken essence with high amounts of BCAA, spent hens herein was used to prepare dripped chicken essence (SCE) and compared with commercial dripped chicken essence (CCE) for in vivo anti-fatigue effect. Methods: To determine the effect on anti-fatigue by dripped chicken essence, the exhaustive swimming was performed. Thirty-two 7-week ICR mice were divided into four groups, which included the control group (CG), CCE, SCE-1X and SCE-2X. The mice were given daily oral administration (0.012 mL/g body weight/d). The fatigue index analysis was conducted weekly. Results: The results showed that SCE had a higher BCAA level as expected, and mice treated with dripped chicken essence (CCE and SCE) could significantly improve exercise performance. The lower blood lactate level, blood urea nitrogen level and creatine phosphokinase activity were found in the supplement of SCE group compared with the CCE group, which suggested that the SCE possessed strong anti-fatigue ability. This could possibly be due to the higher content of BCAA. Conclusion: In this study, SCE promoted recovery from physical fatigue in mice and elevated endurance ability. Among them, the double dose (SCE-2X) showed the strongest anti-fatigue ability. Taken together, spent chickens could be a good source of chicken essence to improve the effect of anti-fatigue.

Assessment of Carbon Storage Capacity of Stands in Abandoned Coal Mine Forest Rehabilitation Areas over time for its Development of Management Strategy (폐탄광 산림복구지 관리방안 도출을 위한 산림복구 후 시간경과에 따른 임분탄소저장량 평가)

  • The objective of this study was to develop a management strategy for the recovery of carbon storage capacity of abandoned coal mine forest rehabilitation area. For the purpose, the biomass and stand carbon storage over time after the forest rehabilitation by tree type for Betula platyphylla, Pinus densiflora, and Alnus hirsuta trees which are major tree species widely planted for the forest rehabilitation in the abandoned coal mine were calculated, and compared them with general forest. The carbon storage in abandoned coal mine forest rehabilitation areas was lower than that in general forests, and based on tree species, Pinus densiflora stored 48.9%, Alnus hirsuta 41.1%, and Betula platyphylla 27.0%. This low carbon storage is thought to be caused by poor growth because soil chemical properties, such as low TOC and total nitrogen content, in the soil of abandoned coal mine forest rehabilitation areas, were adverse to vegetation growth compared to those in general forests. DBH, stand biomass, and stand carbon storage tended to increase after forest rehabilitation over time, whereas stand density decreased. Stand' biomass and carbon storage increased as DBH and stand density increased, but there was a negative correlation between stand density and DBH. Therefore, after forest rehabilitation, growth status should be monitored, an appropriate growth space for trees should be maintained by thinning and pruning, and the soil chemical properties such as fertilization must be managed. It is expected that the carbon storage capacity the forest rehabilitation area could be restored to a level similar to that of general forests.

Effects of Slow Freezing on Development of Blastomeres Separated from Mouse Preimplantation Embryos (완만동결이 생쥐 초기배 난할세포의 발달에 미치는 영향)

  • The development of single blastomeres isolated from 2-, 4- and 8-cell mouse embryos and the ability of such blastomeres to survive slow freezing were studied. Of 223, 60 and 188 single blastomeres isolated from 2-, 4- and 8-cell mouse embryos, respectively, 111 blastomeres (49.8%) from 2-cell embryos, 12 blastomeres (20.0%) from 4-cell embryos and blastomeres (16.5) from 8-cell embryos developed into blastocysts after culture for 96 hrs. The recovery rate was 54.2% (65/120), 46.4% (13/28) and 24.3% (17/70) of blastomeres derived from 2-, 4- and 8-cell embryos following freezing and thawing and the survival of frozen-thawed blastomeres was 27.1% (16/59), 36.4% (4/11) and 17.6% (3/17), and respectively. The apparently six normal fetuses were obtained from frozen-thawed blastomere from 2-cell embryos after transferring into the recipients. These results indicate that mouse btastomeres isolated from preimplatation stage embryos can survive storage in liquid nitrogen following slow freezing.

  • Objectives: The aim of the present systematic review was to investigate the cryopreservation process of dental pulp mesenchymal stromal cells and whether cryopreservation is effective in promoting cell viability and recovery. Materials and Methods: This systematic review was developed in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement and the research question was determined using the population, exposure, comparison, and outcomes strategy. Electronic searches were conducted in the PubMed, Cochrane Library, Science Direct, LILACS, and SciELO databases and in the gray literature (dissertations and thesis databases and Google Scholar) for relevant articles published up to March 2019. Clinical trial studies performed with dental pulp of human permanent or primary teeth, containing concrete information regarding the cryopreservation stages, and with cryopreservation performed for a period of at least 1 week were included in this study. Results: The search strategy resulted in the retrieval of 185 publications. After the application of the eligibility criteria, 21 articles were selected for a qualitative analysis. Conclusions: The cryopreservation process must be carried out in 6 stages: tooth disinfection, pulp extraction, cell isolation, cell proliferation, cryopreservation, and thawing. In addition, it can be inferred that the use of dimethyl sulfoxide, programmable freezing, and storage in liquid nitrogen are associated with a high rate of cell viability after thawing and a high rate of cell proliferation in both primary and permanent teeth.

Analysis of caffeine in aqueous sample by hollow fiber-liquid microextraction (HF-LPME) (HF-LPME를 이용한 수용액 시료중의 카페인 분석)

  • A method for the determination of trace amount of caffeine in urine and various drink samples using hollow fiber-liquid phase microextraction (HF-LPME) and capillary gas chromatograph/nitrogen phosphorus detector (GC/NPD) has been established. HF-LPME method has been optimized with respect to several experimental parameters including the effects of the hollow fiber length, extraction solvent, stirring mode, pH and salt concentration for the determination of caffeine from aqueous samples. The correlation coefficient of calibration curve for caffeine was 0.9994. The average recovery was 102%(n=3). The established method is feasible for the determination of trace amounts of caffeine in several aqueous sample. The limit of detection (LOD) and the limit of quantitation (LOQ) have been found to be 2.5 and 10 ng/mL, respectively. The established HF-LPME method for the analysis of caffeine from aqueous sample can be used for the determination of biological, food and environmental samples.

Efficient Cryopreservation of in vitro Grown Shoot Tips of Pear (Pyrus spp.) by Droplet-vitrification

  • In this study, cryopreservation by droplet-vitrification was applied to pear (Pyrus spp.) germplasm. We focused on the development and assessment of various strategies for the selection of suitable tissue, osmoprotection, and dehydration. We also evaluated post-thaw recovery of cryopreserved explants by droplet-vitrification. Preferentially, we tested the effects of preculture and loading treatments to determine which tissues were more suitable, either the apical shoot tips or the axillary buds. Apical shoot tips showed the better regrowth rate than in vitro axillary buds. The most effective techniques for cryopreservation were as follows. Shoots from in vitro seedlings which had been cultured for about 5-6 weeks were cold-hardened at 4℃ for one week, excised shoot tips were precultured on liquid MS medium including 0.3 M sucrose for 31 hours and 0.7 M sucrose for 17 hours, osmoprotected in loading solution (LS) for 40 min, and then cryoprotected in dehydration solution (PVS3) for 90 min. In addition, we found that regrowth rates of explants on regrowth medium after exposure to liquid nitrogen (LN) were higher than those on MS medium. Results indicated that the highest regrowth percentage was 95.6% for 'Bartlett' cultivar and 68.9% for 'BaeYun No.3' cultivar. Consequently, apical shoot tips of two pear cultivars, 'Bartlett' (P. communis) and 'BaeYun No.3' (P. pyrifolia), were successfully cryopreserved by droplet-vitrification. Results of this study show that the enhanced droplet-vitrification method described in the present study could be used as an effective means for long-term storage of pear genetic resources.
