Influence of the Lysine to Protein Ratio in Practical Diets on the Efficiency of Nitrogen Use in Growing Pigs

  • Twelve gilts were used to investigate the effect of lysine to protein ratio (5.2 g lysine/100 g CP vs. 6.7 g lysine/100 g CP) in practical diets on nitrogen retention and the efficiency of utilization in growing pigs. Treatments involved 2 levels of dietary lysine (5.2 or 6.7 g/100 g CP) and 3 levels of dietary crude protein (11, 14 and 17% in diet). Nitrogen retention was greatest when pigs were fed the control diet containing 17% protein. Nitrogen retention progressively increased as dietary protein increased (p < 0.01), but it was not affected by lysine concentration (g/100 g CP). Apparent biological value (ABV, nitrogen retained/apparently digestible nitrogen) was estimated to be ~50% at the maximum nitrogen retention. ABV was not affected by lysine concentration, but declined (p < 0.05) as the dietary protein level increased. The efficiency of intake N used for maximum nitrogen retention was approximately 44%. One gram of lysine supported approximately 9 to 10 g apparent protein accretion (nitrogen retention ${\times}$ 6.25/lysine intake) in pigs fed control diets. The efficiency of lysine utilization for protein accretion was lower in pigs fed high-lysine diets (6.7 g lysine/l00 g CP) so that 1 g of lysine accounted for 7 to 8 g of protein accretion in these pigs (p < 0.01). The lysine required to support maximum nitrogen retention in pigs fed high-lysine diets was higher than that in pigs fed control diets, which suggests that lysine was over-fortified relative to crude protein, since practical diets can not be formulated without excess of some amino acids. In summary the concentration of 5.2 g total lysine/100 g CP in diet is more appropriate for corn-soybean diets than the commonly suggested the content of 6.7 g total lysine/100 g CP.

Achieving a Nitrogen Balance for Japanese Domestic Livestock Waste: Testing the Scenario of Planting Feed Grain in Land Left Fallow

  • In this study, we assess the recent changes in the amount of excretion by the livestock industry, and discuss the effects of increasing the ratio of cultivated land on the reduction of surplus nitrogen from a cost-performance perspective. Nitrogen has contributed to acidification of ecosystems and nitrate concentrations in groundwater, especially in Europe. Therefore, we assessed the level of nitrogen waste from the domestic Japanese livestock industry, including cattle, swine and poultry during the period 1987-2001. This assessment assumed that 40% of the nitrogen from the domestic livestock industry was emitted as gas into the air and that 60% of the nitrogen was contained in manure used on domestic cultivated land. Nitrogen excreted from livestock, excluding gas emission, decreased by 11% from 0.504 million tons to 0.447 million tons during 1993-2001. Thus, the peak period of nitrogen excretion from livestock is already past in Japan. However, the area of cultivated land under management also decreased during 1990-2000. In addition, the area of paddy and upland fields left unplanted for a year increased during 1990-2000. Therefore, if all manure from the domestic livestock industry had been utilized on the fields as organic fertilizer, but not on arable land left uncultivated for the past year, the nitrogen per net area of cultivated land would have increased by 5%, from 125 to 131 N kg/ha, during 1990-2000. To reduce the nitrogen ratio on cultivated land through the planting of feed grain to utilize the nitrogen, a comparison of the cost performance of feed grains indicated that barley would be more suitable than wheat, rice or soybean. Had barley been planted in 100% of the land left fallow for the past year in 2000, 4% (20,000 tons) of the nitrogen from livestock waste would have been used in the harvest, and the nitrogen per land unit would have not increased but decreased from 125 to 121 N kg/ha during the same decade. Furthermore, when converted into Total Digestible Nutrients, 7% of imported feed corn could have been replaced with the harvested barley in 2000. Planting barley on this fallow land had three benefits; reducing the risk of manure overload on the land, slowing down the decrease in cultivated land, and raising the feed self-sufficiency ratio. Thus, it would be beneficial to plant feed grain such as barley in land left fallow for the past year through utilization of manure.

동해 남부 해역의 수괴 분포와 용존 무기 및 유기 영양염의 분포 특성: 2011년 9월 관측자료를 중심으로 (Distribution of Water Masses and Distribution Characteristics of Dissolved Inorganic and Organic Nutrients in the Southern Part of the East Sea of Korea: Focus on the Observed Data in September, 2011)

  • 2011년 9월에 동해의 수괴 분포와 용존 무기 및 유기 영양염의 분포 특성을 파악하였다. 수온, 염분, 용존산소의 분포를 통하여 연구해역의 수괴 기원은 WM(water mass)-I, WM-II, WM-III, WM-IV 등 4개의 대표적인 수괴로 구분되었으며, 그 성격은 각각 대마난류표층수, 대마난류중층수, 북한한류수, 동해고유수와 유사하였다. 용존 영양염의 경우, 용존 무기 질소(DIN; dissolved inorganic nitrogen)와 용존 무기 인(DIP; dissolved inorganic phosphorus)은 WM-IV에서 가장 높았으며, WM-III, WM-II, WM-I 순으로 나타났다. 반면에 용존 유기 질소(DON; dissolved organic nitrogen)와 용존 유기 인(DOP; dissolved organic phosphorus)은 무기 영양염과 상반되는 분포를 보였다. 연구해역에서 수괴 전체에 대한 DIN : DIP 비는 약 15.8로 Redfield ratio(16)에 근접한 수치를 보이고 있으나, 혼합층의 경우 5.3으로 무기질소가 식물플랑크톤 성장의 제한 요인으로 작용할 수 있는 것으로 보였다. 하지만 무기 질소가 제한된 혼합층에서 DON은 용존 총 질소(DTN; dissolved total nitrogen) 중 약 70%를 구성하였다. 따라서 풍부한 DON은 동해에서 식물플랑크톤의 성장을 위한 중요한 영양염 공급원으로 판단된다.

  • This study was performed to investigate the effect of dietary levels on protein metabolism in eight healthy Korean adult females. The 20-day metabolic study consisted of 2 day adaptation period and three 6-day experimental. Three experimental diets were low protein (LP : protein 44g), high protein(HP : protein 85g) and high animal protein (HAP : protein-84g). The apparent absorption and balance on nitrogen were significantly higher in high protein than in low protein diet. Nitrogen, absorption rate was about 75% for low protein and about 85% for high protein intake. The mean values of nitrogen balance were -1.28% for low protein and 0.78% for high protein diet. All the subjects were in negative nitrogen balance at the low protein intake while they were in positive nitrogen balance at the high protein intake. The mean daily urinary nitrogen excretion increased with increased level of protein intake. Urea nitrogen was the largest part of the urinary nitrogen. The ratio of urea nitrogen to total urinary nitrogen increased significantly for 79 to 85% as protein intake was doubled.

부분질산화와 혐기성 암모늄산화를 이용한 돈사폐수처리 (Piggery Waste Treatment using Partial Nitritation and Anaerobic Ammonium Oxidation)

  • Nitrogen removal with the combined SHARON (Single reactor system for high ammonium removal over nitrite)ANAMMOX (Anaerobic ammonium oxidation) process using the effluent of ADEPT (Anaerobic digestion elutriated phased treatment) slurry reactor with very low C/N ratio for piggery waste treatment was investigated. For the preceding SHARON reactor, ammonium nitrogen loading and removal rate were $0.97kg\;NH_4-N/m^3_{reactor}/day$ and $0.68kg\;NH_4-N/m^3_{reactor}/day$ respectively. In steady state, bicarbonate alkalinity consumption for ammonium nitrogen converted to $NO_2-N$ or $NO_3-N$ was 8.4 gram per gram ammonium nitrogen. The successive ANAMMOX reactor was fed with the effluent from SHARON reactor. The loading and removal rate of the soluble nitrogen defined as the sum total of $NH_4-N$, $NO_2-N$ and $NO_3-N$ in ANAMMOX reactor were $1.36kg\;soluble\;N/m^3_{reactor}/day$ and $0.7kg\;soluble\;N/m^3_{reactor}/day$, respectively. The average $NO_2-N/NH_4-N$ removal ratio by ANAMMOX was 2.41. Fluorescence in situ hybridization (FISH) analysis verified that Candidatus Kuenenia stuttgartiensis were dominate, which means that they played an important role of nitrogen removal in ANAMMOX reactor.

Temporal Variations in Isotope Ratios and Concentrations of Nitrate-nitrogen in Groundwater as Affected by Chemical Fertilizer and Livestock Manure

  • Isotope ratio ($^{15}N/^{14}N$) and nitrate-nitrogen concentration in groundwater were measured to investigate the effect of chemical fertilizer and livestock manure on temporal variations in nitrate-nitrogen concentration and to estimate the contribution of fertilizer and manure to groundwater contamination by nitrate. Four study wells from a rural area in Kyonggi province were selected. One well was located on an upper site from a livestock feedlot, and the others were situated at lower sites from the feedlot. The ${\delta}^{15}N$ values were analyzed by a stable isotope ratio mass spectrometer (Micromass, VG Optima IRMS). Reproducibility of the method and precision of the mass spectrometer were below 1.0 and 0.1‰, respectively Even though study wells were located at the same area, nitrate-nitrogen concentrations and ${\delta}^{15}N$ values differed and fluctuated during the sampling period. The ${\delta}^{15}N$ values of well located at upper site from the feedlot were extremely variable (-1.48~20.80‰). The ranges of ${\delta}^{15}N$ value of three wells situated at lower sites from the feedlot were 11.83~20.73 (ave. 16.11), 8.90~11.73 (ave.11.01), and 5.29~12.73‰ (ave. 8.21‰) with increasing distance from the feedlot. The average values of contribution proportion of nitrogen derived from livestock manure to nitrate-nitrogen in groundwater were 79% for the well closet to the feedlot, 44% for the well most distant from the feedlot, and 56% for the well in between the two wells.

소화성 궤양 흰쥐에서 체내 질소이용율 증진을 위한 체내 질소원에 관한 연구 - 단백질과 단백질 가수분해물의 비율을 중심으로 - (A Study on the Nitrogen Sources for the Enhancement of the Nitrogen Bioavailability in Rats with Peptic Ulcer -The Ratio of Casein and Casein Hydrolysate-)

  • This study aimed to verify the nutritional and curative effects of protein hydroysate and optimal ratio between protein and protein hydroysate as nitrogen source in rats with cysteamine-induced duodenal ulcer. Duodenal ulcer rat model was established by intraperitoneal injections of cysteamine. Sprague-Dawley, female rats weighing approximately 200g were intrapertionealy injected twice cysteamine(13mg/100g BW) at intervals of 3hours per day. This procedure was repeated 3 times at intervals of 3 days. Animals fed on 10% casein diet for injection periods. After last injection, 5 kinds of kiets (the ratio of casein and casein hydrolysate was 100 : 0(C100), 75 : 25(CH 25), 50 : 50(CH 50), 25 : 75(CH 75), 0 : 100(CH 100)) were given. The rate were sacrificed after feeding diet, 1, 3, 5 days. Ulcer index, hexosamine content of stomach and duodeum, gastric motility, trypsin activity, blood glutathione, plasma total protein, albumin, amino-N, urinalry urea nitrogen, creatinine, hydroxyproline and retention rate of nitrogen were analyzed for nutritional effects of diet treatments. There were no differences among diet groups in the view of the growth and diet treatments. There difference of ulcer curation by diet was appeared after 3 days. The ulcer indexes of C100 and CH 25 of 3, 5 days were significantly higher than those of CH 50, CH 75 and CH 100. This result was the same as hexosamine content of stomach, plasma protein, albumin concentration and nitrogen retention rate. The more casein hydrolysate diet had, the lower trypsin activity was. The more casein gydroysate diet had, the higher excretion of hydroxyproline was. These results show that protein hydrolysate can be applied in diet therapy for the patients with gastronitestinal ulcer. It suggests that it has curative effect of diet when nitogen sources include at least over than 50% of protein hydrolysate.

멜라민과의 기계적 혼합을 통한 탄소나노튜브의 선택적 질소 도핑 (Selective Nitrogen Doping of Carbon Nanotubes Through Different Mechanical Mixing Methods with Melamine)

  • 질소 도핑에 의한 Pyridinic-N, Pyrrolic-N, Graphitic-N과 같은 결합 구성의 형성은 탄소나노소재의 물리적 특성을 조절하거나 새로운 물성을 부여하는 중요한 역할을 한다. 본 연구에서는 멜라민을 도펀트 소스로 혼합하여 탄소 나노튜브(CNT)의 질소 결합 구성을 조절하는 간단하고 비용 효율적인 접근 방식을 제안한다. Stirring, Bath sonication 및 Tip sonication과 같은 세 가지 기계적 방법을 통해 멜라민을 탄소나노튜브와 혼합한 후 열처리를 통해 질소 도핑을 진행하였다. CNT 대비 멜라민 비율이 높을수록 Pyrrolic-N의 비율이 높게 나타났으며 Stirring을 통해 혼합한 경우 가장 높은 Pyridinic-N의 비율을 나타내었다. 본 연구에서 제안한 방법은 기계적인 혼합 방법을 이용해 탄소나노튜브에 다양한 종류의 질소 도펀트를 선택적으로 형성할 수 있는 손쉬운 방법으로, 향후 질소가 도핑된 탄소나노물질의 응용을 용이하게 할 것으로 기대된다.

Cucumber Growth and Nitrogen Uptake as Affected by Solution Temperature and NO3-:NH4+ Ratios during the Seedling

  • The effect of solution temperature and nitrogen form on cucumber (Cucumis sativus L.) growth, photosynthesis and nitrogen metabolism was investigated in hydroponic culture. Cucumber plants were grown for 35 days in a greenhouse at three constant solution temperatures ($15^{\circ}C$, $20^{\circ}C$, and $25^{\circ}C$) within a natural aerial temperature ($15-30^{\circ}C$). Four nitrate:ammonium ($NO{_3}^-:NH{_4}^+$) ratios (10:0, 8:2, 5:5, and 2:8 $mmol{\cdot}L^{-1}$) at constant nitrogen (N) concentration of $10mmol{\cdot}L^{-1}$ were applied within each solution temperature treatment. Results showed an increasing solution temperature enhanced plant growth (height, dry weight, and leaf area) in most N treatments. Dry weight accumulation was greatest at the 10:0 $NO{_3}^-:NH{_4}^+$ ratio in the $15^{\circ}C$ solution, the 5:5 ratio in the $20^{\circ}C$ solution and the 8:2 ratio in the $25^{\circ}C$ solution. Photosynthetic rate (Pn) response to solution temperature and $NO{_3}^-:NH{_4}^+$ ratio was similar to that of plant growth. Probably, the photosynthate shortage played a role in the reduced biomass formation. Increasing solution temperature enhanced the nitrate reductase (NR) activity, and further reduced shoots nitrate content. Our results indicate that the optimal ratio of nitrate to ammonium that promotes growth in hydroponic cucumber varies with solution temperature.

유기물부하가 낮은 하수의 전달탈질공법에 의한 탈질방안 (The Study on denitrification of low organic loading sewage by pre-denitrification process)

  • This study was conducted to analyze the operating conditions of predenitrification process to improve the treatment efficiency in low organic loading sewage plant in use today, and to investigate the treatment efficiency of pilot plant added night soil as well as the nitrogen removal characteristics of pilot plant added carbon sources. In the operation under the condition of $BOD_{5}$ sludge load 0.03-0.28kg $BOD_{5}$/kg VSS/d and oxic ammoniac nitrogen sludge load 0.02-0.24 $kgNH_{4}^{+}$-N/kg MLVSS/d, nitrification efficiency is higher than 95%. In order to achieve 70% nitrogen removal at the T-N sludge loading 0.06kg T-N/kg VSSㆍd and the SRT 6~11 days, optimum operating factors were revealed to $CODc_{r}$/T-N ratio 9, recycle ratio 2.6, and denitrification volume ratio 0.33. At this time, denitrification capacity was approximately 0.09 kg $NO_{3}^{-}$-N/kg $CODc_{r}$; specific nitrification rate was 3.4mg $NH_{4}^{+}$-N/g MLVSS/hr; and specific denitrification rate was 4.8mg $NO_{3}^{-}$-N/g MLVSS/hr.
