Effect of Supplement nutrition on the Mycelial Growth of Lentinus edodes

  • Mycelial growth of L. edodes by supplement nutrition of softwood was studied on a sawdust medium. The sawdust used was from the following softwood species : Larix leptolepis, Pinus densiflora and Pinus koraiensis. The added nutritions consisted of carbon nutritions(sucrose, active carbon, xylose, glucose, paper pellet), nitrogen nutritions(potassium nitrate, ammonium chloride, asparagine, glutamic acid) and vegetable oil(rice bran oil). The sawdust medium was a mixture of 76% sawdust, 20% rice bran, 3% carbon nutrition, 0.4% nitrogen nutrition and 0.6% calcium carbonate. Following addition of carbon and nitrogen nutritions on the sawdust medium proved most suitable : L. leptolepis (glucose, glutamic acid), P. densiflora (active carbon, asparagine) and P. koraiensis (xylose, glutamic acid). The highest mycelial growth was obtained from sawdust medium of optimum condition with 97% of L. leptolepis, 110% of P. densiflora and 98% of P. koraiensis. This study has provided useful preliminary information for the cultivation of L. edodes.

  • An on-line FIA system in place of traditional Conway method was used for quicker and simpler analysis of volatile basic nitrogen of pork. The on-line system was developed by connecting an FIA apparatus with an ammonia electrode as a detector to a personal computer. The data from Conway method and from the on-line FIA system were in good agreement and the correlation coefficient was 0.981. One sample could be analyzed in about 4 min.

고추장 저장 연장에 대한 연구 (Studies on the Extension of the Shelf-life of Kochujang during Storage)

  • 본 연구에서는 고추장의 저장 중 이화학적 및 관능적 변화를 통해 효과적인 항갈색화제, 유기산을 선별하며 유통기간 연장에 대해 알아보고자 하였다. 45$^{\circ}C$와 55$^{\circ}C$에서 항갈색화제 0.5% ascorbic acid일 때 control과 유의적 차이가 있었기에 ascorbic acid를 갈변 억제제로 선정했다 또한, 45$^{\circ}C$와 55$^{\circ}C$에서 0.03% ascorbic acid일 때 control과 유의적 차이가 있어, 갈변 억제제로 선정했다. 45$^{\circ}C$와 55$^{\circ}C$에서 유기산 0.5% citric acid 일 때 control과 유의적 차이가 있었기에 citric acid를 갈변 억제제로 선정했다. 또한, 45$^{\circ}C$와 55$^{\circ}C$에서 0.03% ascorbic acid에 0.06% citric acid일 때 control과 유의적 차이가 있어, 효과적인 갈변억제 농도로 선정했다. 0.03% ascorbic acid와 0.06% citric acid를 첨가한 고추장 유통기간을 알아보기 위하여 고추장의 아미노태 질소, pH, 적정산도 및 표면색도 등을 실험한 결과에서 아미노태 질소가 관능적 변화와 상관관계(r=-0.8987)가 높았기에, 고추장의 유통기간 설정 하는 품질특성으로 삼았다. 저 장기간에 따른 아미노태 질소값의 변화율은 1차 반응식으로 해석되었으며, 0.03% ascorbic acid와 0.06% citric acid 첨가물이 유통기간 예측에서 고추장 대조구 보다 45$^{\circ}C$일 때 69%, 55$^{\circ}C$일 때 56% 연장 할 수 있었다.

채소(菜蔬) 및 과실중(果實中) 질산염(窒酸鹽)과 아질산염(亞窒酸鹽)의 축적(蓄積)에 관(關)한 연구(硏究) (Studies on the Accumulation of Nitrite and Niarite in Vegetables and Fruits)

  • Vegetables and fruits purchased from several markets in Seoul from July to October in 1977 were analyzed to know the level of nitrate-and nitrite-nitrogen accumulation in relation to a public health. Radishes and chinese cabbages utilized mainly as pickled vegetables in Korea resulted in the highest concentration of nitrate-nitrogen. Some of the levels for radishes and chinese cabbages were notably high and exceeded a recommended upper limit of 300 ppm $No_{3}-N$, and thus these levels would render these samples unsafe for use. The levels in some of vegetables other than radish and chinese cabbage, e.g., spinach, lettuce, green onion, cabbage were relatively high and considered to be unsafe for use in feeding infants, where as those of green pepper, bean sprouts and parsely were very low and safe. And also the levells in fruits were very low and safe. Nitrite-nitrogen contents in all tested vegetables and fruits ranged to trace and appeared not to be accumulated in fresh vegetables and fruits. Stems and roots of radishes and chinese cabbages accumulated approximately 2 fold more nitrate-nitrogen than leaves in 5 samples of each vegetable tested.

Energy Balance by Carbon and Nitrogen Balance Technique in White Leghorn and Rhode Island Red Hens Fed Maize- and Broken Rice-Based Diets

  • Carbon (C) and nitrogen (N) balance technique was used to determine energy balance in Rhode Island Red (RIR) and White Leghorn (WL) laying hens fed maize-and broken rice (BR)- based diets. Carbon and nitrogen intake and outgo were determined for three days on ad libitum fed diets followed by 2/3 of ad libitum intake for next three days. Carbon analysis was done by using four 'U' tubes in which carbon dioxide released during bomb calorimetry was absorbed on drierite in tube 1 and 2 whereas tube 3 and 4 contained sodalime self indicating granule. Carbon in $CO_2$ was determined by an open circuit respiration system. Energy retention (E, kcal) was calculated as E = 12.386 C (g) - 4.631 N (g). By regressing metabolisable energy (ME) intake on energy balance, maintenance ME requirement of RIR was 128 whereas, that of WL hens was $144kcal/kg\;W^{0.75}/d$. Effciency of utilization of ME for maintenance from BR-based diet in RIR hens was equal but in WL hens it was 11% less than maize-based diet.

Volatile Compounds in Oyster Hydrolysate Produced by Commercial Protease

  • Volatile compounds in raw oyster and oyster hydrolysate produced with protease were compared by vacuum simultaneous steam distillation-solvent extraction/gas chromatography/mass spectrometry. Sixty-two volatile compounds were detected in both samples. Of these, 57 were positively identified, composed mainly of aldehydes(12), ketones(9), alcohols(14), nitrogen-containing compounds(9), acids(6), terpenes(4), and miscellneous compounds(8). Levels of acids decreased after hydrolysis, whereas several other compounds such as aldehydes, ketones, and nitrogen containing compounds increased. Pyrazines, found in high abundance, were only detected in oyster hydrolysate.

단백질 섭취기준: 단백질 필요량과 추정 방법 및 단백질에너지 적정비율 (Dietary Reference Intakes for Protein: Protein Requirement and Estimation Method, AMDR (Amount of Macronutrient Distribution Range), for Protein)

  • This study assessed the current EAR, RDA, and AMDR for protein, which were set in 2005 and revised in 2010 as the DRIs for Koreans. A classical approach to establish the EAR for protein has been the nitrogen balance method. This method has practical limitations and problems in statistical analysis by giving over estimations of nitrogen balance. Thus, the present EAR for protein might be lower than the true requirement. Recent reevaluations of nitrogen balance studies by bilinear regression analysis and the IAAO method have indicated that the EAR of 0.66 g/kg bw/d should be increased by 39% to give 0.92 g/kg bw/d. The AMDR for protein in the Korean DRIs was set at 7-10%, which covers almost the entire population's protein intake. Since the 5th percentile of Korean protein intake is close to 10% of energy and due to the beneficial effects of protein beyond the maintenance of nitrogen equilibrium, the lower range of 7% needs to be increased up to 10%. For practical meal arrangement, 15% of energy as protein, which is close to the average protein intake of Koreans, seems to be proper, although the value is almost two times the EAR.

Effect of Level of Leucaena leucocephala in the Diets of Jamunapari Goats on carbon Nitrogen and Energy Balances

  • Fifteen intact male Jamunapari goats, average body weight $22.0{\pm}1.18kg$ were divided into three groups of 5 animals in each to investigate the effects of feeding leucaena on energy retention and distribution of retained energy. Leucaena leaves and twigs provided 0%, 25% and 50% of CP in the rations of animals in $L_1$ (control), $L_2$ and $L_3$ groups, respectively. Energy balances were determined in an open circuit respiration chamber from gaseous exchange and nitrogen carbon balances. Energy retentions calculated from gaseous exchange data were 181.6, 190.0 and 172.8 kJ/kg $W^{0.75}/d$ and from carbon-nitrogen balances were 178.2, 199.5 and 171.1 kJ/kg $W^{0.75}/d$ in $L_1$, $L_2$ and $L_3$ groups, respectively. No significant difference was observed among the groups in both the methods. The retention of nitrogen and energy in the form of protein was similar in different treatment groups. Similarly, no significant effect was observed on energy retention in the form of fat and total energy retention due to incorporation of leucaena in the diets.

단백질(蛋白質) 최저요구량(最低要求量) 측정(測定)에 관한 연구(測定) (A Study on Estimation of Minimum Protein Requirement)

  • This study was carried out to estimate the minimum protein requirement for the Korean college girls, and to see the effect of carbohydrate intake on the minimum protein requirement in the rat. In the first experiment, three students attending Ewha Womans University were given protein free diet for 7 days while caloric intake were adjusted to maintain constant body weight. Endogenous nitrogen excretion was measured to estimate their minimum protein requirement. Second experiment was carried out in 3 parts to find out the effect of carbohydrate intake on the minimum protein requirement using 15 female, 50 days old rats weighing 120g. Inexperiment 2-1, the rats were fed protein free diet for 9 days and endogenous nitrogen excretion was measured to determine the minimum protein requirement of the rats. In experiment 2-2, the minimum casein requirement of the rats needed to maintain nitrogen equilibrium was estimated by the least squares regression method feeding 3 different levels of casein determined from the result of experiment 2-1. Finally, in experiment 2-3, the rats were given the minimum casein requirement for 7 days with 3 different levels of carbohydrate to find out the effect of carbohydrate intake on the nitrogen balance. The results of this study were as follows. Experiment I. Daily endogenous nitrogen losses of 3 college girls were 1.89g in urine, 1.10g in feces and 0.0108g from skin since the total daily endogenous nitrogen loss was 3.0g and the total daily nitrogen intake was 0.52g, the minimum nitrogen requirement of these subjects was about 2.48g/day (15.5g protein/day). Experiment II. Experiment 2-1: daily endogenous nitrogen losses of the rats fed protein free diet were $22.63{\pm}0.48\;mg$ through urine and $13.12{\pm}0.47\;mg$ in feces. The total daily endogenous nitrogen ouput was $35.71{\pm}1.27\;mg$ and the minimum protein requirement of the rats was 225mg/day in this study. Experiment 2-2: the minimum casein requirement needed to maintain the rats in nitrogen equilibrium was determined to be 340mg/day. Experiment 2-3: the rats were given 350mg/day casein with 3 different levels of carbohydrate (12,26 or 46 Cal by sugar-cornstarch mixture). The nitrogen balances were measured to be +0.0021, +0.0073 and +0.0143 respectively and there were significant differences among 3 groups.
