Effects on Growth and Medicinal Ingredients of Platycodon grandiflorum Radix by Organic and Chemical Fertilizers (유기질 및 화학비료 시용수준이 도라지의 생육 및 약용성분에 미치는 영향)

  • This study was done to elucidate effects on organic and chemical fertilizers for growth characteristics, saponin content and antioxidant activity in Platycodin grandiflorum Ridix root. Chemical and mixed organic fertilizers were treated at levels of 50%, 100% and 200% based on nitrogen 3 kg/10a before transplant of Platycodin grandiflorum Ridix. As a result, when 50% mixed organic and chemical fertilizer plots were treated to P. grandiflorum Ridix root, the root length showed the highest scores of 26.9 and 26.5 cm, respectively. Root width exhibited the highest scores of 25.6 mm and 25.2 mm in 50% mixed organic and chemical fertilizer plots, respectively. The number of fine-roots and fresh weight presented the highest values in 50% mixed organic fertilizer plot as 26.7 and 50.4 g, respectively. The content of platycodin D3 was ranged in 183.4~321.7 mg/100 g, which the highest value was observed in 100% mixed organic fertilizer plot. The content of saponin except for deapioplatycodin D presented higher amount in the mixed organic fertilizer plots. Treatment of 200% chemical fertilizer presented to be withered in transplant cultivation of 3 year-old roots.

Effects of Saengjihwangeumja-gami on STZ-induced Diabetic Mice (생지황음자가미(生地黃飮子加味)가 Streptozotocin으로 유발된 당뇨생쥐에 미치는 영향(影響))

  • First, mice were dosed with 50mg/kg of streptozotocin(STZ) twice every 24 hours to cause high blood-sugar. Then, after three days, mice were injected with 100mg/kg of STZ again. Two different dosages of Saengjihwangeumja-gami were given to the experiment groups: SA group, 15mg/kg/day, and SB group, 90mg/kg/day, in order to determine the effects of Saengjihwangeumja-gami, which has been known to be good for DM(Diabetes Mellitus). By observing weight and blood-sugar level changes, blood tolerance, the numerical value of BUN(Blood Urea Nitrogen) and creatinine in blood, and through light-electronicmicroscopic and immunohistologic investigations of pancreas and kidneys, the following results were obtained: 1. The experiment groups showed a high suppressive effect of weight-loss. 2. The experiment groups' blood-sugar and blood tolerance showed an effective lowering of blood-sugar levels. 3. The experiment groups did not show any noticeable change in the numerical value of BUN and creatinine in blood compared with that of the control groups. 4. The experiment groups showed a higher Insulin positive reaction of pancreatic islets ${/beta}-cell$ than the control groups. 5. The experiment groups showed a higher immuno-reaction against IGF- II than the control groups. 6. Observation of apoptosis of the pancreatic islets showed that the cells of experiment groups were less injured compared with those of the control groups, and fewer apoptag-positive reaction cells were seen in experiment groups than in the control groups. 7. Uunder electron-microscopy, the insulin-containing granules in pancreatic islets ${/beta}-cells$ had increased more in the experiment groups than in the control groups. 8. Under light microscopy, the injury on the inner & outer membrane of the glomerulus and epithelial cells of capillaries and cells among vessels were fewer in the experiment groups than in the control groups. 9. More apoptag-positive reaction cells in the kidney were seen in the control groups than in the experiment groups. 10. PAS-positive reaction substances had increased more in the substrate among the vessels of a glomerulus belonging to the control group than those of the experiment group. 11. Uunder electron-microscopy, the nucleonic membrane, nucleoplasm and mitochondria of proximal and distal renal tubular were more injured in the control groups than in the experiment groups. In conclusion, strong evidence for the efficacy of Saengjihwangeumja-gami in lowering blood-sugar, and in recovery and generation of pancreatic tissues injured by DM was observed. Results suggest Saengjihwangeumja-gami is an effective treatment for DM. Further study of the principles of blood-sugar dropping effects of Saengjihwangeumja-gami are needed, as well as further study of recovery and regeneration of pancreatic tissues injured by DM.

Effect of Selected Environmental Factors on the Production of Geosmin in Phormidium Sp. (Phormidium sp.의 Geosmin 생산에 미치는 환경요인의 영향)

  • A method for quantitative and qualitative analysis of geosmin, odorant produced by several actinomycetes and cyanobacteria, was established and optimized. The effects of environmental conditions on the growth of Phormidium sp. NIVA-CYA7 were examined and the production and release of geosmin by the species was analyzed by using the purge and trap-gas chromatographic technique. One of the major advantages of the technique established in the present study is that the preparation of sample is simpler and purge time is shorter. Under the culture conditions (pH 7.9, $20^{\circ}C$, 120-140 $\mu$E/$m^2$/s and Z8 medium), Phormidium showed growth characteristics with a lag phase for 8 days and an exponential phase for 14 days followed by a stationary phase. Reduction of inorganic nitrogen concentrations in the culture medium from 250 to 100 or 25 $\mu$M brought no significant effect on the cell growth. However, the cell growth was significantly inhibited with decreasing concentrations of inorganic phosphorus from 25 to 10 or 2.5 $\mu$M. When the inorganic phosphorus concentration in the medium was lowered from 25 to 10 $\mu$M, the levels of geosmin in the organism expressed as percentages per unit TOC and chlorophyII-$\alpha$ increased by 35% and 68%, respectively. When the initial pH of the medium was 9.4, geosmin content was 0.0824 $\mu$g/mg C, which was 2-fold higher than that at pH 7.9 Consequently, the level of geosmin in Phormidium was found to vary with growth phases of the culture, external inorganic phosphorus concentration and external pH, while the release of geosmin was not significantly affected by the factors.

Influence of Different Levels of NaCIO, Lactic Acid and Acetic Acid on Meat Quality and Microbiological Changes of Duck Breast During Storage (오리 가슴육의 소독제(NaCIO) 및 유기산(Lactic acid, Acetic acid) 처리 수준이 저장 기간 중 품질 및 미생물 변화에 미치는 영향)

  • This study was performed to extend the shelf-life of duck breast treated with NaCIO(20, 50ppm), lactic acid(1, 2%) and acetic acid(1, 2%). Changes in microbial counts, storage characteristics and color values of duck breasts were determined during storage at 4℃ for 7 days. Although pH values were not different on the first day of storage. they increased up to 3rd days of storage and decreased gradually thereafter. Thiobarbituric acid reactive substances(TBARS) of duck breasts treated with lactic acid and acetic acids were lower TBARS than those with lactic acid on the 7th days of storage. Volatile basic nitrogen(VBN) didn't differ among the treatments(P>0.05). Although the microbial counts were increased in all treatments, acetic acid treatment had lower microbial counts among the treatments during storage. These results indicated that acetic acid would be the best treatment to extend the shelf-life of duck breasts among others.

The Studies of the low power Wavelengths of He-Ne IR laser on the changes of the serum biochemical components in burn rats (저 출력 레이저 파장에 따른 화상 쥐 혈청 생화학 성분 변화에 대한 연구)

  • This studies were to investigate the effects of low power wavelengths Helium-Neon Infra-Red(He-Ne IR)laser on the changes of the blood biochemical components in burn rats. The thirty Spraque-Dewely adult male rats were assigned to the 5 groups; the experimental groups(3), the burn control group(1) and the control group(1). There was made three degree burn by the 250mW IR on the back of each rats, from 3 days after being, burned, the experimental laser groups were irradiated low power wavelengths (292Hz, 1168Hz, 4672Hz) He-Ne IR laser for 5 minutes every day during the 7days. The results wee as follows: The activities of aspirate aminotransferase(AST), alanine aminotransferase(ALT), and lactate dehydrogenase(LDH), on treated with the wavelengths ((292Hz, 1168Hz, 4672Hz) laser groups during 5 min. for 7 days were significantly increased to the control group respectively, but those of total cholesterol(CHOL) and alkaline phosphatase(ALP) were significantly decreased to the a control group (p<0.05). there were significantly decreased on the activity of alanine aminotransferase and alkaline phosphatase on the all experimental groups to the burn control group and those of lactate dehydronase on treated with 292Hz and 4672Hz wavelength9 groups to the burn control group but, were significantly increased on those of urea nitrogen (292Hz and 1168Hz)(p<0.05). The activity of AST, LDH, on treated with 1168Hz wavelengths were significantly decreased of the 292Hz and 4672Hz wavelengths groups that of ALT on the treated with 1168Hz group was significantly increased to the 4672 Hz wavelength group but those of CHOL and LDH on the 1168Hz and 4672Hz wavelength groups were dignificatly decreased to the MHz wavelengths group. As above results, the changes of the activities of biochemical conponents in the serum levels on the healing process have meaningful effected of the role of low power wavelengths He-Ne IR laser.

N Use Efficiency and Nitrate Leaching by Fertilization Level and Film Mulching in Sesame Cultivated Upland

  • This study was conducted to evaluate the effect of slow release fertilizers (SRF), crotonylidene diurea (CDU) and latex coated urea (LCU), on nitrogen (N) use efficiency (NUE) and nitrate-N leaching in a silty clay loam soil under polyethylene film mulching (PFM) for sesame cultivation. In PFM plot, concentrations of $NO_3-N$ and $NH_4-N$ in SRF applied soils were less than that in the urea plot during the whole growing period. However, $NO_3-N$ and $NH_4-N$ in all the non-mulched plots (NM) were not significantly different. Urea-N in soil treated with SRF was higher than urea plot until 50 days after application and was comparable in all the treatments after 50 days. $NO_3-N$ concentrations in leached solution in 21 days after urea fertilization in PFM and NM were 26 mg $L^{-1}$ and 83 mg $L^{-1}$, respectively. However, $NO_3-N$ in leached solution at applied CDU and LCU was less than that of urea similar to nitrate concentration in soil. $NO_3-N$ in leached solution in applied CDU and LCU in 44 days after application was about 25% lower than that urea plot and PFM, while the $NO_3-N$ concentration of CDU and LCU treatment in NM did not changed. Application of SRF increased the yield of sesame and N recovery compared to urea and there was a little difference between SRF and N levels. In conclusion, application of 80% N level with SRF increased N recovery and reduced nitrate leaching without reduction of yields compared with urea application.

Assessment of Water Purification Plant Vegetation for Enhancement of Natural Purification in Mankyeong River (만경강 본류의 자연정화능 향상을 위한 식생학적 진단)

  • This study was conducted to get some information on plants abilities to enhance water purification and to find out away to conserve the ecosystem in Mankyeong river. Vegetation were surveyed at 4 sites pointing by 1:5,000 topographical map, from June 2001 through March 2002. T-N content in water were high in all sites of Mankyeong river, the average T-N levels were 8.59 and 17.23 mg/L, summer and winter, respectively. The average T-P level during summer was 0.47 mg/L but that was 1.79 mg/L during winter. The BOD level in Mankyeong upstream ranged from 0.95 to 2.57 mg/L which would be in I or II grade according to water quality criteria by Ministry of Environment but BOD level in Mankyeong downstream ranged from 6.87 to 9.72 mg/L. The plant species of river flora were found 251, 98 and 85, upstream midstream and down stream, respectively. Among the surveyed plants, Ceratophyllum demersum, submerged plant and Nuphar subinteperrimum took up higher contents of phosphate and nitrogen than other piano. The Phragmites communis and Zizania latifolia having much biomass were thought to be suitable plants for enhancement of e natural water purification.

Effects of Different Humic Acids on Growth and Fruit Quality of Tomato Plant (부식산 시용이 토마토의 생장과 과실품질에 미치는 영향)

  • In greenhouse farming, a variety of humic acids have been applied to improve soil conditions and plant growth. However, it is still unclear that how humic acids combined with chemical fertilizers affect growth and quality of fruit vegetable crops. This study was conducted to determine the combination effect of humic acids and chemical fertilizers on the growth and fruit quality of tomato (Lycopersicon esculentum MILL.) grown under greenhouse conditions. Three different formulation types of humic acid were used: liquid type A, liquid type B and solid type C. The tomato plants were grown in three treatment combination plots and in conventional fertilizer (CF) plot with recommended levels of nitrogen, phosphorus and potassium: HA combined with CF (HA+CF), HB combined with CF (HB+CF) and HC combined with CF (HC+CF). For most of growth characteristics (i.e. leaf number, internode length, maximum leaf length, leaf width and chlorophyll contents) determined in this experiment, no significant differences were observed between all combination treatments and CF. However, integrated fruit qualities (i.e. averaged weight, sugar contents and acidity) were slightly improved in the humic acid combined with CF treatments when compared with CF alone treatment. No phytotoxicity was observed with humic acid treatments. However, further studies will probably be needed to use widely and safely these humic acids, in order to ensure a maximizing growth, fruit yield and quality of tomato.

Application Effect of Organic Fertilizer and Chemical Fertilizer on the Watermelon Growth and Soil Chemical Properties in Greenhouse (유기질비료와 화학비료의 시용수준에 따른 시설수박 생육과 토양화학성의 변화)

  • BACKGROUND: Organic fertilizers in watermelon cultivation are widely used to supply nutrient and organic matter. This study was conducted to investigate the effects of application rate of organic fertilizer on the watermelon growth and soil chemical properties in greenhouse METHODS AND RESULTS: The organic fertilizers used in this experiment were mixed expeller cake (MEC) and mixed organic fertilizer (MOF). The treatments were conducted with 4 levels (1.0 N, 0.7 N, 0.5 N and 0.3 N) on the basis of soil testing nitrogen fertilization (STNF) using MEC or MOF as the basal dressing, and using chemical fertilizers (CF) as the additional dressing on the rest of STNF. These fertilizations were compared to CF 1.0 N (0.3 N as the basal and 0.7 N as the additional dressing) and non fertilization (NF). The leaf area of watermelon in treatment 0.5 N and 0.3 N using MEC or MOF was similar to CF treatment. The absorbed nutrient amounts by leaf, weight and sugar contents of fruit in the 0.5 N and 0.3 N treatments were higher than other treatments. In 0.5 N and 0.3 N treatments using MEC or MOF on the basis of STNF, soil chemical properties such as electrical conductivity (EC), available $P_2O_5$ and exchangeable K concentrations after experiment showed tendency to decreasing or similar level before experiment. CONCLUSION(s): These results suggest that the MEC or MOF application as the basal dressing at the 30~50% level of STNF and CF application as the additional dressing on the rest of STNF be best to maintain adequate nutrient of soil and to increase marketable yield for watermelon.

Influences of Thermal Effluents on the Epilithic Algal Community in Small Stream Originating from the Seokjung Hot Spring (온천 배수 유입에 따른 소형 하천의 생태계 변화와 회복에 관한 연구 -소형 하천에서 온천 배수가 부착조류 군집에 미치는 영향)

  • To study the influences of thermal effluents flowing from hot spring on epilithic algal community, seasonal survey was carried out at stream and its watersheds from Seokjeong hot spring in Chollabuk-Do, Korea. Totally 7 points were divided into three regions fur sampling of water and epilithic algae, such as the direct effected, uneffected and the mixed region, respectively. At the discharging points of effluents, a dark-green cyanobacterial mat were remarkably constructed, mainly by two cyanobacteria, Oscillatoria and Phormidium. The mat formation were more obvious at low temperature than any other season, and even result in disappear with downstream and season. Totally, one hundred and fifty-three taxa of epilithic algae were classified with 15 unidentified species. Among the, diatoms occupied 58% of total species, whereas cyanobacteria was 67% of total biomass, comparatively. In terms of stream direction, relative abundance of cyanobacteria was only limited in the upstream in cold season, and result in this pattern disappeared with season change. Although all physicochemical variables at the discharging points, was very high, compare to other points, they were quickly decreased downstream. Among them, some heavy metals were not detected or below the detection levels at downstream. Nitrate nitrogen increased with downstream, as well as phosphorus and sulfate have a similar trend throughout, while ammonia quickly decreased in the initial period of discharging effluents. This suggest that although the thermal effluent with high temperature and organic compounds could polluted the small study stream, various contributions such as flowing water, intake of uneffected streawater and collaboration of cyanobacterial mat and stream bottom gradually induces a stable water system.
