Effects of Nitric Oxide Donor Supplementation on Copper Deficient Embryos and Nitric Oxide-Mediated Downstream Signaling (Nitric Oxide Donor 첨가가 구리 결핍 배아의 발달과 Nitric Oxide 하위 신호전달체계에 미치는 영향)

  • One suggested mechanism underlying copper (Cu) deficiency teratogenicity is a low availability of nitric oxide (NO), signaling molecule which is essential in developmental processes. Increased superoxide anions secondary to decreased activities of Cu-zinc superoxide dismutase (Cu-Zn SOD) in Cu deficiency can interact with NO to form peroxynitrite, which can nitrate proteins at tyrosine residues. In addition, peroxynitrite formation can limit NO bioavailability. We previously reported low NO availability and increased protein nitration in Cu deficient (Cu-) embryos. In the current study, we tested whether Cu deficiency alters downstream signaling of NO by assessing cyclic GMP (cGMP) and phosphorylated vasodilator-stimulating phosphoprotein (VASP) levels, and whether NO supplementation can affect these targets as well as protein nitration. Gestation day 8.5 embryos from Cu adequate (Cu+) or Cu- dams were collected and cultured in either Cu+ or Cu- media for 48 hr. A subset of embryos was cultured in Cu- media supplemented with a NO donor (DETA/NONOate; 20 ${\mu}M$) and/or Cu-Zn SOD. Cu-/Cu- embryos showed a higher incidence of embryonic and yolk sac abnormalities, low NO availability, blunted dose-response in NO concentrations to increasing doses of acetylcholine, low mRNA expression of endothelial nitric oxide synthase (eNOS), increased levels of 3-nitrotyrosine (3-NT) compared to Cu+/Cu+ controls. cGMP concentrations tended to be low in Cu-/Cu- embryos, and they were significantly lower in Cu-/Cu- yolk sacs than in controls. Levels of phosphorylated VASP at serine 239 (P-VASP) were similar in all groups. NO donor supplementation to the Cu- media ameliorated embryonic and yolk sac abnormalities, and resulted in increased levels of cGMP without altering levels of P-VASP and 3-NT. Taken together, these data support the concept that Cu deficiency limits NO availability and alters NO/cGMP-dependent signaling in Cu- embryos and yolk sacs, which contributes to Cu deficiency-induced abnormal development.

  • In this study, vertical dynamic analyses on the suspension bridges under seismic load are developed. Time domain analysis, random vibration analysis, and spectral analysis are formulated theoretically. The random nitration analysis is checked by numerical integration and the mathematical integration with correlation coefficient which include CQC and SRSS method in the conditions of white noise and filtered white noise. Beam, truss and frame elements are used in order to model the suspension bridge. Geometric stiffness due to dead load is considered for cable and tower.

Protective effect of 3-(naphthalen-2-yl(propoxy)methyl)azetidine hydrochloride on hypoxia-induced toxicity by suppressing microglial activation in BV-2 cells

  • We recently reported the anti-inflammatory effects of 3-(naphthalen-2-yl(propoxy)methyl)azetidine hydrochloride (KHG26792) on the ATP-induced activation of the NFAT and MAPK pathways through the P2X7 receptor in microglia. To further investigate the underlying mechanism of KHG26792, we studied its protective effects on hypoxia-induced toxicity in microglia. The administration of KHG26792 significantly reduced the hypoxia-induced expression and activity of caspase-3 in BV-2 microglial cells. KHG26792 also reduced hypoxia-induced inducible nitric oxide synthase protein expression, which correlated with reduced nitric oxide accumulation. In addition, KHG26792 attenuated hypoxia-induced protein nitration, reactive oxygen species production, and NADPH oxidase activity. These effects were accompanied by the suppression of hypoxia-induced protein expression of hypoxia-inducible factor 1-alpha and NADPH oxidase-2. Although the clinical relevance of our findings remains to be determined, these data results suggest that KHG26792 prevents hypoxia-induced toxicity by suppressing microglial activation.

Syntheses New Bis-Crown Ethers Containing Imine Group(III) (새로운 이민기를 포함하는 비스 크라운 에테르의 합성)

  • We report herein synthetic results obtained six new types of bis-benzocrown ethers containing imine group. Bis crown ethers1~3 are aminobenzo-15-crown-5-ether linked with terephthalaldehyde, isophthalaldehyde, phthaldehyde respectively by imine reaction. Bis crown ethers1~3 are different distances in each crown ether rings. Bis crown ether 4 has large cavity in crown ethers. Functionalized crown ether 5 is synthesized amonobenzo-15-crown-5-ether and terephthaladehyde same ratio at one to one. Bis crown ether 6 has photosensitive linkage between crown ethers. Bis crown ether 7 is prepared by amonobenzo-15-crown-5-ether and triethyl ortho formate. 4'-Nitrobenzo-crown ethers and 3',4'-dinitrobenzo-crown ethers were prepared by nitration of benzo crown ethers, obtained from the reaction of catechol and oligoethylene glycol ditosylate. Crown ethers containing aldehyde group were synthesized from the reaction of 3,4-dihydroxybenzaldehyde and corresponding ditosylate respectively. The synthesized crown ethers were characterized respectively by IR, NMR, GC-Mass.

Influence of Two Moving Masses on Dynamic Behavior of a Simply Supported Pipe Conveying Fluid Flow (두 이동질량이 단순지지 유체유동 파이프의 동특성에 미치는 영향)

  • A simply supported pipe conveying fluid and two moving masses upon it constitute this nitration system. The equation of motion is derived by using Lagrange's equation. The influence of the velocities of two moving masses, the distance between two moving masses, and the velocities of fluid flow in the pipe have been studied on the dynamic behavior of a simply supported pipe by numerical method. The velocities of fluid flow are considered with in its critical values of a simply supported pipe without moving masses upon It. Their coupling effects on the transverse vibration of a simply supported pipe are inspected too. As the velocity of two moving masses increases, the deflection of a simply supported pipe is increased and the frequency of transverse vibration of a simply supported pipe is not varied. In case of small distance between two masses, the maximum deflection of the pipe occur when the front mass arrive at midspan. Otherwise as the distance get larger, the position of the front masses where midspan deflection is maximum moves beyond the midpoint of a simply supported pipe. The deflection of a simply supported pipe is increased by coupling of the velocities of moving masses and fluid flow.

Condition Monitoring of Link Driving System with Clearance (간극이 있는 링크구동계의 상태진단)

  • There is a clearance between the parts of a machine due to design tolerance, manufacturing error, wear, looseness, or misalignment. If the clearance is large, the vibration and noise of the machine is generally large. Therefore, the analysis on the nitration and noise of a machine can tell the clearance of the machine, which reveals the condition of the machine, i.e., the existence of faults and the safety of the machine. The investigation of this kind of research should be on the basis of experimental results. A link mechanism with a clearance at a joint between the coupler and locker is made for the investigation of the condition monitoring of a machine due to clearance. The vibration and sound are measured from the link driving system during the operation. The signals are clarified using line enhancement technique. The noise removed signals are used to develop the dynamic model of the system for a model based fault diagnosis. Also this study showed that the clarified signals can be used for the calculation of the joint forces between the coupler and rocker and for the correlation between the vibration and sound levels and the clearance sizes.

  • In this paper the characteristics of vibration induced by subway trains running on track is discussed. The quantitative prediction of the vibration level and the countermeasure for reduction of necessary, is of importance for the better environment. It was made the constructed Bundang line as first step with the modified Young-Dan type to basis on the Japanese Young-Dan type. In this paper it was measured and analyzed to two region ($ \circled1$Susuh-Bokjung. $ \circled2$Chorim-Suhyun region) of this. at present, operational Bundang line when averaging velocity of train is 60 (km/h). As the response characteristics of frequency Induced by subway operation, it was confirmed that frequency band of neighborhood of 30~80 Hz in generally dominant. Also to assess the quantitative nitration as response level to be measured for each point of two region in subway operation, the vibrational response level was measured at the state to be not subway operation. And the level was approximately $\fraction one-fifth~\fraction one-ten$ level comparing to subway operation.

  • Although PAN has ong been considered to belong to one of the most characteristic photooxidants, its concentra-tions have rarely been measured in a systematic manner in Korea. The first in-situ Measurements of PAN were made in concert with simultaneous analysis of NO, NO$_2$, $O_3$ and UV-Visible radiation intensity from Seoul and dong-Hae during Spring and early Summer of 1999. The aim of our study was to evaluate the behavior of photo-chemical species in a highly polluted urban air and a relative clean one. PAN were determined using a semi-auto-mated gas chromatograph equipped with an electron captured detection system. Calibration of the PAN was done by synthesizing liquid standards using a strong acid nitration of the corresponding peracid. The PAN concentra-tions were found in the range of a few ppbv in the urban air ad persistently less than a ppbv in the clean rural airs. Diurnal variations of PAN and $O_3$ were also observed consistently during the experiment with maxima occurring during the late daylight hours. While the positive dependency of $O_3$ production and PAN concentrations were ob-served in most occasion, the dependency became deviated significantly under changes in air masses.

Synthesis and Electrochemical Spectroscopic Characterization of Benzophenone Derivatives (벤조페논 유도체의 합성과 전기화학 및 분광학특성에 관한 연구)

  • The benzophenone derivatives(4-$CH_3O$-4'-$NO_2$ and 3,4'-di-$NO_2$) are synthesized by the Fridel-Craft acylation and the nitration method. Electrochemical redox potentials of the benzophenone derivatives (4-$CH_3O$, H, 3-Cl, 3-$NO_2$, 4-$NO_2$, 4-$CH_3O$-4'-$NO_2$, 3,4'-di-$NO_2$) are measured by using cyclic voltammometry. In the relationship of summing Hammett value and redox potential, we find a proportional constant$(\rho)$ that shows a good relation with an electrochemical property and a reactivity of the benzophenone derivatives. The benzophenone substituted with the electron donating groups(4-$OCH_3$ and 4-$OCH_3-4'-NO_2$) are higher the energy in the LUMO level, then increasing a band-gap energy$(E_g)$, their $E_gs$ are obtained as a 3.94 eV and 3.59 eV, respectively.

Effects of Potassium Sulfamate on Synthesis of Pottassium Dinitramide (포타슘디나이트라아마이드의 합성에 대한 술팜산칼륨의 영향 연구)

  • Ammonium dinitramide (AND) is one of the most promising oxidizers in propellants without chlorinated pollution to the atmosphere in these days. Potassium sulfamate (PS) is a key substance in ADN synthesis as forming nitrates such as $-N(NO_2)_2$. In this paper, potassium sulfamate as a starting material for ADN synthesis was prepared in a lab scale through crystallization with ethanol solvent, and observed the effects on the yield and purity of KDN. The prepared potassium sulfamates were analyzed using FE-SEM, XRD, BET and TGA-DSC. The lab-made PS, which was ground to $20{\mu}m$ showed more beneficial than a commercial product achieving high yield and purity of the synthesized KDN. It would be associated closely with crystallinity, porosity and pore size of prepared PS.
