Acidification of pig slurry effects on ammonia and nitrous oxide emissions, nitrate leaching, and perennial ryegrass regrowth as estimated by 15N-urea flux

  • Objective: The present study aimed to assess the nitrogen (N) use efficiency of acidified pig slurry for regrowth yield and its environmental impacts on perennial ryegrass swards. Methods: The pH of digested pig slurry was adjusted to 5.0 or 7.0 by the addition of sulfuric acid and untreated as a control. The pig slurry urea of each treatment was labeled with $^{15}N$ urea and applied at a rate of 200 kg N/ha immediately after cutting. Soil and herbage samples were collected at 7, 14, and 56 d of regrowth. The flux of pig slurry-N to regrowth yield and soil N mineralization were analyzed, and N losses via $NH_3$, $N_2O$ emission and $NO_3{^-}$ leaching were also estimated. Results: The pH level of the applied slurry did not have a significant effect on herbage yield or N content of herbage at the end of regrowth, whereas the amount of N derived from pig slurry urea (NdfSU) was higher in both herbage and soils in pH-controlled plots. The $NH_4{^+}-N$ content and the amount of N derived from slurry urea into soil $NH_4{^+}$ fraction ($NdfSU-NH_4{^+}$) was significantly higher in in the pH 5 plot, whereas $NO_3{^-}$ and $NdfSU-NO_3{^-}$ were lower than in control plots over the entire regrowth period. Nitrification of $NH_4{^+}-N$ was delayed in soil amended with acidified slurry. Compared to non-pH-controlled pig slurry (i.e. control plots), application of acidified slurry reduced $NH_3$ emissions by 78.1%, $N_2O$ emissions by 78.9% and $NO_3{^-}$ leaching by 17.81% over the course of the experiment. Conclusion: Our results suggest that pig slurry acidification may represent an effective means of minimizing hazardous environmental impacts without depressing regrowth yield.

Diallyl Disulfide Prevents Cyclophosphamide-Induced Hemorrhagic Cystitis in Rats through the Inhibition of Oxidative Damage, MAPKs, and NF-κB Pathways

  • This study investigated the possible effects and molecular mechanisms of diallyl disulfide (DADS) against cyclophosphamide (CP)-induced hemorrhagic cystitis (HC) in rats. Inflammation response was assessed by histopathology and serum cytokines levels. We determined the protein expressions of nuclear transcription factor kappa-B (NF-${\kappa}B$), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), oxidative stress, urinary nitrite-nitrate, malondialdehyde (MDA), and 8-hydroxy-2'-deoxyguanosine (8-OHdG). Finally, we studied the involvement of mitogen-activated protein kinases (MAPKs) signaling in the protective effects of DADS against CP-induced HC. CP treatment caused a HC which was evidenced by an increase in histopathological changes, proinflammatory cytokines levels, urinary nitrite-nitrate level, and the protein expression of NF-${\kappa}B$, COX-2, iNOS, TNF-${\alpha}$, p-c-Jun N-terminal kinase (JNK), and p-extracellular signal regulated kinase (ERK). The significant decreases in glutathione content and glutathione-S-transferase and glutathione reductase activities, and the significant increase in MDA content and urinary MDA and 8-OHdG levels indicated that CP-induced bladder injury was mediated through oxidative DNA damage. In contrast, DADS pretreatment attenuated CP-induced HC, including histopathological lesion, serum cytokines levels, oxidative damage, and urinary oxidative DNA damage. DADS also caused significantly decreased the protein expressions of NF-${\kappa}B$, COX-2, iNOS, TNF-${\alpha}$, p-JNK, and p-ERK. These results indicate that DADS prevents CP-induced HC and that the protective effects of DADS may be due to its ability to regulate proinflammatory cytokines production by inhibition of NF-${\kappa}B$ and MAPKs expressions, and its potent anti-oxidative capability through reduction of oxidative DNA damage in the bladder.

Preparation of Crosslinked Polyvinylbenzylchloride Anion Exchange Composite Membranes using Fabric Substrates and Their Electrodialysis Application for Ion Removal (천지지체를 사용한 가교화된 폴리비닐벤질클로라이드 음이온교환 복합막의 제조와 전기투석을 이용한 이온 제거 특성연구)

  • A series of anion exchange composite membranes were prepared and characterized for electrodialysis processes used in the removal of nitrate nitrogen and ions in groundwater. The membranes were prepared as follows; first, fabric substrates were fully impregnated with monomer mixtures of vinylbenzylchloride (VBC), divinylbenzene (DVB), Styrene (ST) and $\alpha,\alpha$-Azobis(isobutyronitrile) (AIBN). Second, they were thermally polymerized to yield crosslinked poly (VBCST- DVB)/fabric composite membranes. Finally, the membranes were treated with trimethylamine (TMA) / acetone to give $-N^+(CH_3)_3^-$-containing poly(VBC-ST-DVB)/fabric membranes. The basic membrane properties such as ion exchange capacity (IEC), electric resistance and water content of the resulting membranes were measured as a function of VBC/DVB and TMA/Acetone content. As a result, the composite membranes showed lower electric resistance and higher IEC than commercial anion exchange membranes (AMX, Astom). Electrodialysis tests using the prepared membranes were carried out for the removal of various ions such as $NaNO_3$, $MgSO_4$ and NaF for 60 minutes. The results showed that the ions were removed below 1 mg/L within about 15 minutes which indicates that the anion exchange membranes prepared here could be applied to the electrodialysis process. as can be seen in the following that the ion conductivity values were almost no change after 15 minutes electrodialysis.

Protein Synthesis during Somatic Embryo Development and Artificial Seed Germination of Apium graveolens L. after Abscisic Acid or Cold Treatment (쎌러리(Apium graveolens L.)의 체세포배 발생 및 인공종자 발아에 있어서 앱시스산 및 저온처리에 의한 단백질 합성)

  • To understand the molecular mechanism of hardening process in somatic embryo development and artificial seed germination in celery (Apium graveolens L.), the changes of protein synthesis by ABA or cold teatment at early globular stage were examined. Protein content and nitrate reductase activity in ABA- or cold-treated somatic embryo and seedlings were higher than that in unheated ones. The protein content and nitrate reductase activity were more prominent in somatic embryos than in seedlings. From two-dimensional electrophoresis, several protein spots specific to ABA or cold treatment were identified: 30 KD, 32 KD, 171 KD and 205 KD at heart-shaped stage; and 29 KD, 33 KD, 37 KD, 38 KD, 41 KD, 55 KD, 66 KD, and 110 KD at cotyledonary stage were the most specifically synthesized However the synthesis of certain polypeptides were repressed at heart-shaped or cotyledonary stage: 42 KD, 44 KD, 59 KD, 64 KD, 101 KD, 104 KD, and 190 KD at heart-shaped stage; and 29 KD and 116 KD at cotyledonary stage. The protein pattern changes by ABA or cold treatment occurred simultaneously and mainly in acid-soluble proteins during somatic embryo development and artificial seed germination. Therefore it is suggested that the metabolic changes for adaptation to environmental change occur during somatic embryo development and the germination and growth of seedling from embryo.

Uptake, Assimilation and Translocation of Ammonium or Nitrate in Italian Ryegrass

  • To investigate the partitioning of newly absorbed N derived from NO$_3$- and NH$_4$$^{+}$, 6 mM $K^{15}$ NO$_3$ or 3 mM ($^{15}$ NH$_4$)$_2$ was fed continuously in Italian ryegrass (Lolium multiflrum L.) for 7 days. Nitrogen metabolites (nitrate, amino acid, soluble- and insoluble protein) were analyzed at the end of $^{15}$ N feeding. Dry weight in shoot, stubble and root was not significantly different between NO$_3$$^{[-10]}$ and NH$_4$$^{+}$ feeding. Total nitrogen content in all three organs was significantly higher in NH$_4$$^{+}$ than NO$_3$$^{[-10]}$ feeding. Sum on N content in reduced N fractions (amino acids + proteins) in shoot, stubble and roots in NH$_4$$^{+}$ feeding increased by 13.3, 12.5 and 35.4 %, respectively, compared to NO$_3$$^{[-10]}$ feeding. The Relative Specific Activity (RSA, percentage of newly absorbed $^{15}$ N relative to total N in a sample) values of amino acids and insoluble proteins were significantly higher in NH$_4$$^{+}$ feeding. Total amount of newly absorbed $^{15}$ N in NO$_3$$^{[-10]}$ and NO$_3$$^{[-10]}$ feeding was 52.3 and 69.5 mg/plant on dry matter basis, respectively. In both NH$_4$$^{+}$ and NO$_3$$^{[-10]}$ grown plants, most of the N was allocated to the shoot, 67.5% in NH$_4$$^{+}$ feeding and 58.8% NO$_3$$^{[-10]}$ feeding, respectively. The $^{15}$ N amount incorporated in the reduced N compounds (amino acids and proteins) in NH$_4$$^{+}$ grown plants significantly increased by 74.8% compared to NO$_3$$^{[-10]}$ grown plants. The increase of the $^{15}$ N amount assimilated to amino acids in NH$_4$$^{+}$ grown plants was remarkably higher in roots as more than 7.25 times compared to NO$_3$$^{[-10]}$ feeding. These results indicated that Italian ryegrass was much efficiently utilized NH$_4$$^{+}$-N for the synthesis of reduced N compounds.reduced N compounds.

Antioxidant Activities of Extracts from Leaves and Stems of Achyranthes japonica (쇠무릎 잎과 줄기 추출물의 항산화 활성 연구)

  • This study was conducted to investigate the physiological characteristics of water and ethanol extracts from leaves and stems of Achyranthes japonica. The highest contents of total polyphenol and flavonoid compounds were 58.27 and 42.22 mg/g in water extract from leaves, respectively. The protein content was the highest at 16.42 mg/g in water extract from leaves. Ethanol extract from stems showed the highest content of reducing sugars at 11.35 mg/g. In the measurement of electron donating ability (EDA), ethanol extract from stems showed the highest EDA at 93.41% at a concentration of 1.0 mg/mL. Superoxide dismutase-like activity of ethanol extract from leaves was the highest at 8.13% at a concentration of 1.0 mg/mL. In the analysis of nitrate scavenging activity, water extract from leaves showed the highest activity at 94.90% at pH 1.2, and the activity increased as concentration increased and pH decreased. In the measurement of xanthine oxidase inhibition, ethanol extract from stems showed the highest inhibitory activity at 66.67% at a concentration of 1.0 mg/mL. Especially, nitrate scavenging activities of water extract from leaves were the highest under all pH conditions. These results verify that extracts from leaves of A. japonica have strong antioxidant activity and can be used as an effective antioxidant source for nutraceutical foods, medicines, and cosmetic stuffs.

Phase Composition and Pore Structure of Sol-Gel Derived Zirconia Nanopowders (Sol-Gel법에 의해 제조된 Zirconia 나노분말의 결정상과 기공특성)

  • Yttria-stabilized zirconia(YSZ) nanopowders were prepared by sol-gel method using zirconium-n-butoxide(ZNB) and yttrium nitrate as precursors. In addition, the effect of water content added during the hydrolysis reaction of ZNB was investigated on the phase composition and pore structure of the product powders. The phase composition of YSZ nanopowders with calcination temperatures showed the same trend, irrespective of $H_2O$ amounts added during the hydrolysis reaction of ZNB. All powders dried at $100^{\circ}C$ were amorphous and transformed to cubic phase at $400^{\circ}C$, which converted to tetragonal phase at $1,000^{\circ}C$. Monoclinic phase also appeared at $1,000^{\circ}C$. The powders showed the mixture of tetragonal and monoclinic phases from $1,000^{\circ}C$ to $1,400^{\circ}C$. The pore size distributions of the dried powders prepared with small amounts of water(less than or equal to $H_2O/ZNB=20$) showed mesopores, while those prepared with large amounts of water(greater than or equal to $H_2O/ZNB=50$) exhibited micropores.

Changes in Cerebral Blood flow Following Fermented Garlic Extract Solution with High Content of Nitrite (흰쥐에서 고용량 아질산이온 함유 마늘 발효농축액에 의한 뇌혈류 변화)

  • Nitrate-nitrite-nitric oxide (NO) pathway is a major alternative source of NO and is essential for NO - dependent physiological functions in body. Food supplements having nitrate/nitrite can improve metabolic syndromes including hypertension through antioxidant activity or vasodilation. The purpose of this study was to observe the effects of fermented garlic (F. garlic) having high concentration of NO2- on changes in blood flow and nitric oxide synthesis in the cerebral cortex of rodents. The generation of nitric oxide detected by a chemi-luminescence detector was higher in F. Garlic compared with NaNO2 solution under artificial gastric juice with pH 2.0. Ether F. garlic or NaNO2 diluted with artificial cerebrospinal fluid was directly applied into around the needle probe of laser Doppler flow meter that was located on epidural surface of the cortex. Direct application of F. garlic resulted in increase of cerebral blood flow detected by a laser Doppler flow meter with a dose-dependent manner. Compared with NaNO2 solution, F. garlic produced changes in cerebral blood flow at lower concentration of NO2-. Pretreatment of methylene blue, a guanylyl cyclase inhibitor prevented upregulation of cerebral blood flow by the treatment of F. garlic. In addition, the application of F. garlic with 250, 500ppm of NO2- caused significantly the production of NO in the cortical tissue but NaNO2 solution with 500ppm of NO2- did not. In summary, these results suggested that F. garlic with high content of NO2- induce increase in cerebral blood flow through nitric oxide-dependent signal pathway.

Mineralization of organic materials applied to Korean ginseng (고려인삼(高麗人蔘) 유기질비료(有機質肥料)의 무기화(無機化))

  • 1. Ginseng field and virgin soils, to which Yakto, washed Yakto, litters (deciduous and coniferous trees) and bone meal were applied, were incubated at 60% of field capacity of the soils to shed light on the production of $NH_4-N$ and $NO_3-N$ through mineralization of the organic materials being the nitrogen sources for Korean ginseng. 2. Total nitrogen content mineralized in the soils, to which the following organic materials were applied, was:bone meal>Yakto${\gg}$litter (deciduous trees)> washed Yakto, litter (coniferous trees). The content increased with incubation time in all the virgin and ginseng field soils excepting only the virgin soils applied with washed Yakto and litters of which the contents decreased. A large part of nitrogen mineralized in the virgin and ginseng field soils to which Yakto was applied was nitrate form, while a half ginseng field soils and a considerable part in the other virgin soils was ammonium form in the other treatments. 3. $NH_4-N$ content produced in the virgin and ginseng field soils applied with bone meal increased with the time, while it decreased in the other treatments. And the content in the soils applied with bone meal was very high. 4. $NO_3-N$ content showed a continuous increase during the incubation time in all treatments of ginseng field soils and only in the virgin soil to which Yakto was applied, wherease it showed almost no change in the other treatments of virgin soils. And the content in the soils applied with Yakto was very high. 5. Yakto was expected to supply nitrate enough to meet amounts of nitrogen required by ginseng with different ages and also expected to increase in soil fertility, while bone meal was expected only to supply much nitrogen of which a considerable part was ammonium form.

Application Effects of Some Nitrogen Fertilizers Forms for the Growth and Yield of Rice Plant (몇가지 형태(形態)의 질소비료시비(窒素肥料施肥)가 수도(水稻)의 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • Nitrogen fertilizer effectiveness on rice production was studied to evaluate the different forms and sources. Seven kind of nitrogen fertilizers were applied in two levels, 15 and 30 kg per 10a on Jinjubyeo (Japonica type) in silt loam paddy soil of pot cultivation. The results were summerized as follows; 1. pH in soils was decreased with enhancement of ammonium sulfate application in $NH_4-N$, but it was increased with times after nitric-acid application and PH change in soil was not remarkable when $NO_3-N$ with accessory component was applied. 2. $NH_4-N$ contents in soil were the lowest at 2 weeks after application in N 15kg/10a regardless of different sources of nitrogen fertilizer. $NO_3-N$, in N 30kg/10a, was decreased continuously until 4 weeks, while $NH_4-N$, Urea-N were at minimum during 2-3 weeks. 3. Growth of culm length and straw weight applied with AN (Ammonium Nitrate), AS (Ammonium Sulfate) and urea were superior to the form of nitrate. While NA (Nitric Acid), PN (Potassium Nitrate) and CN (Calcium Nitrate) plot of the $NO_3-N$ was the dominant fertilizers for root elongation. 4. Brown rice yields were increased dominantly by $NH_4-N$ application such as AS or AP than $NO_3-N$ pot. But the yields in case of $NO_3-N$ application CN, PN and NA were decreased. 5. N, P, Mg and Mn content of straw ranked the effectiveness of nitrogen forms as $NH_4-N$, Urea-N and $NH_4-N+NO_3-N$, while K, Ca and $SiO_2$ content of straw in $NO_3-N$ fertilizer plot were high while N, P, Mg, Mn, Fe and Mg were low. 6. Increament of nitrogen absorption in straw was stimulated by enhancement of phosphorous absorption and the growth and yield of rice plant were increased. Absorption of N, P, Ca and Mg was decreased by CN application. Absorption of N, P and Mg also was decreased by $NO_3-N$ application and N, P, Mg or Ca content were seemed to simulated the growth and yield of rice plant. 7. $SiO_2$, Zn and Fe contents of the root at harvest stage were higher than those of the straw. N, P, Mg, Mn, Zn and Fe contents were high in $NH_4-N$ and Urea treatment. While K, ca and $SiO_2$ contents, however, were high in $NO_3-N$ treatment.
