The present study describes the ameliorating effect of $Ca^{++}\;on\;Cd^{++}$ toxicity on the germination, early growth of mungbean seedlings, nitrogen assimilation enzyme. s-nitrate reductase (NR), nitrite reductase (NIR), anti-oxidant enzymes (POD, CAT and SOD) and on the accumulation of hydrogen peroxide and sulphydryls. $Cd^{++}$ inhibited seed germination and root and shoot length of seedlings. While NR activity was down- regulated, the activities of NIR, POD and SOD were up- regulated with $Cd^{++}$ treatment. $Cd^{++}$ treatment also increased the accumulation of sulphydryls and peroxides, which is reflective of increased thiol rich proteins and oxidative stress. $Ca^{++}$ reversed the toxic effects of $Cd^{++}$ on germination and on early growth of seedlings as well as on the enzyme activities, which were in turn differentially inhibited with a combined treatment with calcium specific chelator EGTA. The results indicate that the external application of $Ca^{++}$ may increase the tolerance capacity of plants to environmental pollutants by both up and down regulating metabolic activities. This experiment was conducted to investigate the influence of nitrogen fertilizer forms, fertilizer and herbicide application rates on growth of radish. The nitrate content and nitrate reductase activity of radish were analyzed along with the growing stage. Nitrate nitrogen was more efficient than ammonium nitrogen for radish. With increasing the fertilizer application rate, accumulated of nitrate content was increased. The amount of nitrate nitrogen was highest at 25days after seeding in petioles, and $32{\sim}39days$ after seeding in root. Nitrate content was decreased as sampling date was delayed, whereas the content increased in the root at early growing stage and then decreased. The nitrate cotent increased in the order of petioles, roots, and leaf blades and nitrate reductase activity increased petioles, leaf blades, and roots. The higher nitrogen fertilizer and herbicide application increased nitrate nitrogen accumulation in radish as compaired with control treatment and nitrate reductase activity showed similar trend.

A field experiment with 200, 400 and 600kg-N/ha/year application levels was carried out to study the nitrate nitrogen accumulation of sorghum sudangrass hybrids(Xtragraze II and Civa 1990) at Iksan College Farm in 1995. The nitrate nitrogen content of Xtragraze II and Civa 1990 was increased by the application of nitrogen and decreased as the plant matured, then the nitrate nitrogen content was below the toxic level of ruminant at the level of 200kg-N application during the whole growing period. The nitrate nitrogen content of Xtragraze II and Civa 1990 exceeded the safe level of ruminant at the level of 400kg-N application, and that in Xtragraze II decreased at the low level in the later stage of growth, but that in Civa 1990 was almost kept constantly at the same level. The nitrate nitrogen accumulation of Civa 1990 had a greater tendency than that of Xtragraze II. A sum exceeding 200kg-N does not necessarily result in increase the amount of nitrate nitrogen in sorghum sudangrass hybrids. It is suggested that 400kg-N application may results in toxic level of nitrate nitrogen, and special attention must be given in feeding them.
Water culture experiments were carried out to elucidate the effects of N availability, temperature and water potential of culture solution on the uptake and assimilation of N and dry matter accumulation by barley seedlings. N assimilation and dry matter accumulation at 3 to 4 leaves stage in barley plants were maximized at about 3.4 % of N concentration in leaf. N assimilation by barley plants increased with increase of nitrate concentration up to 80ppm in the solution. Over this level nitrate began to accumulated in the leaves and stems proportionally to the N availability in culture solution. Nitrate reductase activity increased in parallel with the increase in the concentration of reduced N in leaves. N uptake by barley plants decreased markedly when water potential reduced below -2 bar or when temperature dropped below 5$^{\circ}C$. These results suggest that the basal application rate of N, 60kg per hectare, for the barley crop needs to be re-examined under the concept of N use efficiency with taking into consideration of temperature and soil N availability because about a half of N accumulated in the leaves of barley plant before wintering is known to be lost by winter killing of above-ground part of the plant.
Nitrate can serve as a terminal electron acceptor in place of carbon dioxide and inhibit methane emission in the rumen and nitrate reducing bacteria might help enhance the reduction of nitrate/nitrite, which depends on the type of feed offered to animals. In this study the effects of three levels of sodium nitrate (0, 5, 10 mM) on fermentation of three diets varying in their wheat straw to concentrate ratio (700:300, low concentrate, LC; 500:500, medium concentrate, MC and 300:700, high concentrate, HC diet) were investigated in vitro using buffalo rumen liquor as inoculum. Nitrate reducing bacteria, isolated from the rumen of buffalo were tested as a probiotic to study if it could help in enhancing methane inhibition in vitro. Inclusion of sodium nitrate at 5 or 10 mM reduced (p<0.01) methane production (9.56, 7.93 vs. 21.76 ml/g DM; 12.20, 10.42 vs. 25.76 ml/g DM; 15.49, 12.33 vs. 26.86 ml/g DM) in LC, MC and HC diets, respectively. Inclusion of nitrate at both 5 and 10 mM also reduced (p<0.01) gas production in all the diets, but in vitro true digestibility (IVTD) of feed reduced (p<0.05) only in LC and MC diets. In the medium at 10 mM sodium nitrate level, there was 0.76 to 1.18 mM of residual nitrate and nitrite (p<0.01) also accumulated. In an attempt to eliminate residual nitrate and nitrite in the medium, the nitrate reducing bacteria were isolated from buffalo adapted to nitrate feeding and introduced individually (3 ml containing 1.2 to $2.3{\times}10^6$ cfu/ml) into in vitro incubations containing the MC diet with 10 mM sodium nitrate. Addition of live culture of NRBB 57 resulted in complete removal of nitrate and nitrite from the medium with a further reduction in methane and no effect on IVTD compared to the control treatments containing nitrate with autoclaved cultures or nitrate without any culture. The data revealed that nitrate reducing bacteria can be used as probiotic to prevent the accumulation of nitrite when sodium nitrate is used to reduce in vitro methane emissions.
After the release of facts that the high nitrate contents in vegetables by Korean organic farming, organic farmers were recommended by Korean Organic Farmer Association to apply 20t/ha compost while they applied before as much as they could, sometimes even more than 100t/ha. This study was aimed to check the safety of Korean organic product in terms of nitrate content. Current nitrate content in leaf and root vegetables by 3 different farming systems(organic farming, conventional farming, greenhouse farming) were monitored and furthermore it was compared with previously published another data in Korea to find out any changes among the monitored years(1993-1999). Current nitrate content in chinese cabbage, lettuce and kale became less compare to those of the beginning of monitoring, and especially those cultivated organic farming got the lowest value while previous organic vegetables at beginning of 90's showed the higher value than those cultivated in greenhouse. This decrease in organic vegetables was affected by less application rate of organic fertilizer than before at 20t/ha. But the nitrate level by organic farming was still higher compare to those cultivated by conventional farming in open field, since currently monitored organic vegetables were cultivated basically in greenhouse condition. It shows the organic farmer the necessity of transfer their farming site and condition from greenhouse to open field in order to decrease of its high level of nitrate caused long-term application and slow release characteristics of organic fertilizer. It was concluded the adoption of soil nitrate test to recommend a organic farmer the exact application rate for need of crop growth. Additionally the mechanism of lower nitrate accumulation in rice and fruit vegetables were also discussed in the paper.
In the edible parts of chinese cabbage, the NO-3 accumulation was higher in the outer leaves than in the inner leaves and it was higher in the leaf midrib than in the leaf blade. In Radish, it was higher in the aerial part of the root than in the underground part of root. NO-3 accumulation in edible parts of chinese cabbage of organic farming fertilized with compost 8t/10a was about 4 times higher than those of conventional farming with recommended mineral nitrogen; and was similar to those of conventional farming fertilized with twice the mineral nitrogen rate. But, NO-3 accumulation in radish of organic farming with 8t/10a compost was lower than those of conventional farming with recommended mineral nitrogen. It showed NO-3 content in the edible parts of vegetables should be considered one of several parpmeters to judge a real safety vegetable to be certified by government.
Salt accumulation in soils of greenhouse due to the massive application of nitrogen fertilizers causes salt stress on the various crops, a serious problem in domestic agriculture. Since the majority of the salinity is nitrate, the excess nitrate should be removed; therefore, a bacterial strain having high capacity of nitrate uptake and identified as Enterobacter amnigenus GG0461 was isolated from the soils of greenhouse. Optimum conditions for the bacterial growth and nitrate uptake were investigated. GG0461 was able to grow without nitrate; however, nitrate facilitated the growth. The rate of nitrate uptake increased at alkaline pH and both growth and nitrate uptake were maximal at pH 8-9. When the initial pH of culture medium was increased to pH 8 or 9, it was decreased to neutral upon bacterial growth and nitrate uptake. These results imply that the major factor mediating bacterial nitrate uptake is a nitrate/proton antiporter. The fact was supported by the effect of nitrate addition in the absence of nitrate, since the addition of nitrate greatly increased the nitrate uptake and rapidly decreased pH of media.
This study was performed to develop the biological treatment technology of wastewater polluted with heavy metals. Heavy metal-tolerant microorganisms, such as Pseudomonas putida, P. aeruginosa, P. chlororaphis and P. stutzeri possessing the ability to accumulate cadmium, lead, zinc and copper, respectively, were isolated from industrial wastewater and mine wastewater polluted with various heavy metals. The effect of several external factors, such as temperature, pH and heavy metal compounds on heavy metal accumulation in the cells was investigated. The amount of heavy metal accumulation into cells according to the kind of heavy metal compound was slightly increased in the case of the heavy metal compound with -nitrate group, but generally, there is little change according to the kind of compound in the amount of heavy metal accumulation. The amount of heavy metal accumulation according to the precultured time was increased in the case of the cell precultured for 24 hours, but generally the precultured time did not affect to the amount of heavy metal accumulation. Heavy metal accumulation into cells was affected by several external factors, such as temperature and pH. The optimum temperature and optimum pH of the accumulation of heavy metal into cells were $20{\sim}37^{\circ}C$ and pH $6{\sim}8$, respectively. By increasing the concentration of each heavy metal-tolerant microorganism in the solution, the total amount of heavy metal accumulated was increased, whereas the amount of heavy metal accumulated per cell(mg, heavy metal/g, dry cells) was decreased. These results indicated that the amount of heavy metal accumulated was not proportional to the concentration of microorganisms.
