눈 운동 측정 및 분석을 이용한 추적 눈 운동 동안의 모양지각 왜곡에 대한 참조대상의 영향 검증 (Examination of the Effect of Reference on Shape Perception during Pursuit by Eye Movements Recording and Analysis)

  • 최근의 연구에서 Li, Brenner, Cornelissen과 Kim (2002)은 추적 눈 운동 동안의 2차원 모양지각이 망막에 형성된 이미지 내용을 그대로 반영한다는 것을 보여 주었는데, 이러한 연구는 2차원 모양판단에 있어서 망막 외 정보가 전혀 고려되지 않았음을 시사한다. 이와 같은 실험실 연구와 달리, 실제 생활에서 2차원 모양지각의 왜곡은 대부분의 경우에 발생하지 않는데, 가능한 한 가지 이유는 실험실 연구에서 사용된 자극의 경우에 참조대상이 존재하지 않는데 비하여, 실제 생환에서는 다양한 참조대상이 목표대상의 주변에 존재하기 때문이다. 목표대상과 참조대상의 상대적인 위치관계는 추적 눈 운동 동안에 망막에서도 그대로 유지되는데 시각체계가 이러한 안정적인 관계를 목표대상의 모양을 지각할 때에 이용할 가능성이 있다. 본 연구에서는 다양한 참조대상을 이용하여 이러한 가능성을 검증하였다. 흥미롭게도, 추적 눈 운동 동안에 참조대상이 목표대상 주위에 존재할 때에도 모양지각 왜곡이 관찰되었는데, 왜곡의 양은 참조대상이 존재하지 않을 때에 비하여 감소하였다. 참조대상의 효과는 참조대상과 목표대상의 거리에 비례하였다. 이러한 실험결과는 추적 눈 운동 동안의 2차원 모양지각에 참조대상 정보가 이용되지만, 참조대상에 대한 정보만으로는 실제생활에서의 안정적인 모양지각을 설명할 수 없음을 시사한다.

  • 본 연구에서는 기능적 자기공명영상 기법을 이용하여, 정상 성인이 한국어 틀린 믿음 과제를 수행하는 동안 활성화되는 뇌 영역을 알아보고자 하였다. 실험 1에서 일차 틀린 믿음 과제를 수행하는 동안 양측 설전소엽과 측두-두정 인접부, 좌반구의 하두정소엽, 후측 대상회, 중전두회 등의 뇌 활성화가 관찰되었다. 실험 2에서 이차 틀린 믿음 과제를 수행하는 동안에는 좌반구의 중전두회와 내전두회와 우반구의 설전소엽, 중전두회, 측두-두정 인접부 영역이 활성화되었다. 이런 결과는 성인이 마음이론 이야기를 이해하는 방식은 보편적임을 시사하는 것이다.

Reduced Gray Matter Volume of Auditory Cortical and Subcortical Areas in Congenitally Deaf Adolescents: A Voxel-Based Morphometric Study

  • Purpose: Several morphometric studies have been performed to investigate brain abnormalities in congenitally deaf people. But no report exists concerning structural brain abnormalities in congenitally deaf adolescents. We evaluated the regional volume changes in gray matter (GM) using voxel-based morphometry (VBM) in congenitally deaf adolescents. Materials and Methods: A VBM8 methodology was applied to the T1-weighted magnetic resonance imaging (MRI) scans of eight congenitally deaf adolescents (mean age, 15.6 years) and nine adolescents with normal hearing. All MRI scans were normalized to a template and then segmented, modulated, and smoothed. Smoothed GM data were tested statistically using analysis of covariance (controlled for age, gender, and intracranial cavity volume). Results: The mean values of age, gender, total volumes of GM, and total intracranial volume did not differ between the two groups. In the auditory centers, the left anterior Heschl's gyrus and both inferior colliculi showed decreased regional GM volume in the congenitally deaf adolescents. The GM volumes of the lingual gyri, nuclei accumbens, and left posterior thalamic reticular nucleus in the midbrain were also decreased. Conclusions: The results of the present study suggest that early deprivation of auditory stimulation in congenitally deaf adolescents might have caused significant underdevelopment of the auditory cortex (left Heschl's gyrus), subcortical auditory structures (inferior colliculi), auditory gain controllers (nucleus accumbens and thalamic reticular nucleus), and multisensory integration areas (inferior colliculi and lingual gyri). These defects might be related to the absence of general auditory perception, the auditory gating system of thalamocortical transmission, and failure in the maturation of the auditory-to-limbic connection and the auditorysomatosensory-visual interconnection.

Optimized Expression, Purification, and Rapid Detection of Recombinant Influenza Nucleoproteins Expressed in Sf9 Insect Cells

  • Accurate and rapid diagnosis of influenza infection is essential to enable early antiviral treatment and reduce the mortality associated with seasonal and epidemic infections. Immunochromatography is one of the most common methods used for the diagnosis of seasonal human influenza; however, it is less effective in diagnosing pandemic influenza virus. Currently, rapid diagnostic kits for pandemic influenza virus rely on the detection of nucleoprotein (NP) or hemagglutinin (HA). NP detection shows higher specificity and is more sensitive than HA detection. In this study, we time-dependently screened expression conditions, and herein report optimal conditions for the expression of recombinant nucleoprotein (rNP), which was 48 h after infection. In addition, we report the use of the expressed rNP in a rapid influenza diagnostic test (SGT i-flex Influenza A&B Test). We constructed expression vectors that synthesized rNP (antigen) of influenza A and B in insect cells (Sf9 cells), employed the purified rNP to the immunoassay test kit, and clearly distinguished NPs of influenza A and influenza B using this rapid influenza diagnostic kit. This approach may improve the development of rapid test kits for influenza using NP.

  • Hemophagocytic lymphohistiocytosis (HLH) is characterized by fever, splenomegaly, jaundice, and pathologic findings of hemophagocytosis in bone marrow or other tissues such as the lymph nodes and liver. Pleocytosis, or the presence of elevated protein levels in cerebrospinal fluid, could be helpful in diagnosing HLH. However, the pathologic diagnosis of the brain is not included in the diagnostic criteria for this condition. In the present report, we describe the case of a patient diagnosed with HLH, in whom the brain pathology, but not the bone marrow pathology, showed hemophagocytosis. As the diagnosis of HLH is difficult in many cases, a high level of suspicion is required. Moreover, the pathologic diagnosis of organs other than the bone marrow, liver, and lymph nodes may be a useful alternative.

치매의 한방치료와 한양방 협진치료에 관한 전문가 집단 설문연구 (The Study on the Korean Traditional Medical Treatment and System of Collaborative Practice Between Korean Traditional Medicine and Western Medicine for Dementia: Based on Analysis of Questionnaire Survey in Professional Group)

  • Objectives : Recently, the elderly population and dementia prevalence have been rapidly increasing in Korea. Dementia is a concept of syndrome composed of characteristic symptoms. However, there have been no approved treatments for dementia with proven efficacy. Therefore, we investigated Korean traditional treatment being used in clinic and collaborative practice for dementia in the professional group. Methods : We conducted a survey using questionnaires to professors of oriental neuropsychiatry for a month. Items of the questionnaire consisted of overall Korean traditional medical treatment for dementia, which included herbal medicine, acupuncture, moxibustion, cupping, psychotherapy and others. Results : 1. It was shown that Yukmijihwangtanggamibang, Jowiseungcheongtang, Seonghyangjeonggisan have been used the most in herbal medicine. 2. Chechim, Saamchimbeop, and scalp acupuncture have been used the most in acupuncture treatment. 3. In Korean traditional psychotherapy, Ijeongbyeongi, Jieongoronyobeop, Gigongyobeop have been used, and effective in anxiety, agitation, improvement of comprehension for caregiver and environment. 4. Most professors answered that collaborative practice between Korean traditional medicine and western medicine has been needed for dementia. They suggested that herbal medicine, followed by acupuncture, and moxibustion can be used with western medicine simultaneously. Conclusions : The survey revealed that professors use herbal medicine and acupuncture as main treatments for dementia. They share perspective regarding the need of long period to treat dementia patients, and there are needs of collaborative practice for dementia.

  • Objectives : From Scrophularia buergeriana water extract(SBW), has been used in vivo test for its beneficial effects on neuronal survival and neuroprotective functions, particularly in connection with APP-related dementias and Alzheimer's disease(AD). $A{\beta}$ oligomer derived from proteolytic processing of the ${\beta}$-amyloid precursor protein(APP), including the amyloid-${\beta}$ peptide($A{\beta}$), play a critical role in the pathogenesis of Alzheimer's dementia. Methods : Using drosophila APP model on APP-induced neuronal cytotoxicity, we demonstrated that SBW prevents neurotoxicity of $A{\beta}$ oligomer, which are the behavior, and possibly causative, feature of AD. We investigated the neuroprotective effects of SBW against the effects of oligomeric $A{\beta}$ and fly behaveior and life span by UAS-GRIM/APP-GAL within transgenic flies. Results and Conclusions : SBW repaired damage leading to the behaveior of APP-induced fly and delayed life span. These results suggest that neuronal damage in AD might be due to two factors: a direct $A{\beta}$ oligomer toxicity and multiple cellular and molecular neuroprotective mechanisms, including attenuation of apoptosis and direct inhibition of $A{\beta}$ oligomer, underlie the neuroprotective effects of SBW.

  • Objective : Recent studies indicate that the deposition of beta-amyloid ($A{\beta}$) is related in the pathogenesis of Alzheimer's disease (AD), but the underlying mechanism is still not clear. Method : To investigate the potential cellular functions of APP and LMK02, we use transgenic drosophila as a model was treated with either LMK02, and the effect in APP expression was determined by climbing assay. LMK02 have been shown to be neuroprotective in fly model systems. We asked whether dietary supplementation with LMK02 would influence behavior and AD-like pathology in a transgenic fly model. Result LMK02 water extract have attenuated fly death in vivo. LMK02-treated fly increased percentage of flight ability more longly and survival ratio more than controls. APP-GRIM drosophila treated with LMK02 had significantly less accumulation of APP deposition in the eye and brain as compared to control drosophila. Conclusion : These results suggest that LMK02 prevent APP-induced neurotoxicity through attenuating flies death induced by APP, and may be useful as potential therapeutic agents for AD.

Anti-hypertensive Effects of Electroacupuncture at ST36 In Spontaneously Hypertensive Rat

  • The present study investigated whether electroacupuncture (EA) can alleviate hypertension and concomitant disorders such as decreased kidney weight and elevation of serum creatinine in spontaneous hypertensive rats. EA (2 Hz, 3 mA, 10 min) was applied to Joksamni (ST36) once daily for 7 days. Body weight, blood pressure and heart rate were measured on Day 0, 2, 6, 8, 10 and 12, and kidney weight and serum creatinine levels were examined after sacrifice (on Day 12 after last examination). In the ST36 group, the blood pressure were significantly decreased from 6thdays and its effects lasted up to Day12 (up to 5days after cessation of acupuncture), compared to control. And, significant decreases of the heart rates after EA at ST36 were also observed on Day 2, 6, and 8. However, there were no significant differences in daily body weight, kidney weight and serum creatinine between acupuncture and control group. These results showed that EA at ST36 caused anti-hypertension by decreasing blood pressure and heart rates in spontaneous hypertenstive rats, although it failed to alleviate concomitant disorders such as the decreased kidney weight and elevation of serum creatinine shown in hypertensive rats.

Molecular Cloning and Characterization of Serine/Threonine Phosphatase from Rat Brain

  • A novel serine/threonine protein phosphatase with EF-hand motif, which belongs to PPEF family was partially cloned from rat brain cDNA by employing RT-PCR method. The size of the amplified clone was 1.6kbp. The amplified DNA was subcloned into pGEM-T-Easy vector and the resulting plasmid was maned as pGEM-rPPEF2. The nucleuotide sequence is shared by 88% with that of mouse PPEF-2 cDNA, and the deduced amino acid sequence reveal 92% homology with that of mouse PPEF-2 cDNA. The N-terminal region of the cloned rat brain PPEF contains a putative phosphatase catalytic domain (PP domain) and the C-terminal region contains multiple $Ca^{2+}$ binding sites (EF region). The putative catalytic domin (PP) and the EF-hand motif (EF) regions were subcloned into pGEX4T-1 and were overexpressed in E. coli DH5 as glutathione-S-transferase (GST) fusion proteins. Expression of the desired fusion protein was identified by SDS-PAGE and also by immunoblot analysis using monoclonal antibody against GST. The recombinant proteins were purified by glutathione-agarose chromatography. This report is first to demonstrate the cloning of PPEF family from rat brain tissues. The clone reported here would be invaluable for the investigation of the role of this new type-phosphatase in rat brain.
