An In Vitro Assay to Screen for Translation Inhibitors

  • Protein synthesis is the ultimate outcome of gene expression which, in turn, is regulated by several translation factors. We attempted to identify substances that can inhibit the translation process in vitro when the outcome protein is luciferase. To this end, we developed a sensitive cell-free protein synthesis assay using luciferase as the reporter. The synthesis of luciferase increased proportionately as mRNA was added to a $15-{\mu}l$reaction medium in concentrations raging from 5 ng to 500 ng. The maximum amount of luciferase was synthesized when the media were incubated at $25^{\circ}C$ for 40 min. The concentration of each compound that inhibited luciferase production by 50% ($IC_{50}$) was calculated. Hygromycin, puromycin, and cycloheximide yielded an $IC_{50}$ of 0.008, 0.8, and $0.7{\mu}g/ml$, respectively. A filtrate of Streptomyces spp. isolates inhibited protein synthesis up to S-fold when added to the in vitro translation assay mixture.

Properties of the Fusants of Lactobacillus acidophilus 88 and Lactobacillus casei subsp. casei KCTC 1121

  • Protoplast fusion between L. casei KCTC 1121 and L. acidophilus 88 was attempted to obtain improved strains. The fusants produced a bacteriocin against indicator strains, making a smaller inhibition zone compared to that of L. acidophilus 88. After culturing for 2 months on selective medium, the selected fusants were still stable without segregation. Fusants showed higher lipase activity compared to those of the two parent strains. Fusant No.4, 11, and 15 exhibited excellent lactic acid productivity. Fusant No.4 and 15 exhibited improved proteolysis ability compared to the two parent strains. Whereas L. casei possessed both ${\beta}-galactosidase$ and $phospho-{\beta}-galactosidase$ activities, and L. acidophilus 88 had only ${\beta}-galactosidase$ activity, the fusants had both the intermediate enzyme activities. Cell size of the fusants was greater than that of the parents.

Trichoderma koningii의 種間 原形質體 融合에 대한 硏究 (Intraspecific Protoplast Fusion in Trichoderma koningii)

  • The conditions for the protoplast fusion of auxotrophic mutants of Trichoderma koningii were determined. A preparation of commercial enzyme Driselase was used successfully to isolate protoplasts from the 18 hr old mycelium of T. koningii. The yields of protoplasts production were ranged from $0.3{\times}10^8$ to $2.5{\times}10^8$ protoplasts per mg of damp mycelium of various auxotrophic mutant strains. The regeneration frequencies from $9.3{\times}10^{-3}\;to\;2.0{\times}10^{-1}$ were obtained when the protoplasts from auxotrophic mutants were plated on the malt extract medium containing 0.6M $MgSO_4$, and 2% agar, and the optimal concentration of PEG for protoplst fusion was 30%. Exposure of protoplasts to PEG for 10 min was found to be sufficient to induce high frequency heterokaryon formation. Optimal pH of fusion mixture was determined as 5.5, and 1 mM of calcium chloride in fusion mixture was found to be sufficient to enhance protoplast fusion frequency. Under optimal condition, the fusion frequency of the cross between protoplasts from various auxotrophic mutants were $1.6{\times}10^{-2}\;and\;4.1{\times}10^{-2}$.

S5 Lipase : An Organic Solvent Tolerant Enzyme

  • In this study, an organic solvent tolerant bacterial strain was isolated. This strain was identified as Pseudomonas sp. strain S5, and was shown to degrade BTEX (Benzene, Toluene, Ethyl-Benzene, and Xylene). Strain S5 generates an organic solvent-tolerant lipase in the late logarithmic phase of growth. Maximum lipase production was exhibited when peptone was utilized as the sole nitrogen source. Addition of any of the selected carbon sources to the medium resulted in a significant reduction of enzyme production. Lower lipase generation was noted when an inorganic nitrogen source was used as the sole nitrogen source. This bacterium hydrolyzed all tested triglycerides and the highest levels of pro-duction were observed when olive oil was used as a natural triglyceride. Basal medium containing Tween 60 enhanced lipase production to the most significant degree. The absence of magnesium ions ($Mg^{2+}$) in the basal medium was also shown to stimulate lipase production. Meanwhile, an alkaline earth metal ion, $Na^+$, was found to stimulate the production of S5 lipase.

  • Medium-chain-length polyhydroxyalkanoates (MCL-PHAs), which have constituents with a typical chain length of $C_{6}-C_{14}$, are polyesters that are synthesized and accumulated in a wide variety of Gram-negative bacteria, mainly pseudomonads. These biopolyesters are promising materials for various applications because they have useful mechanical properties and are biodegradable and biocompatible. The versatile metabolic capacity of some Pseudomonas spp. enables them to synthesize MCL-PHAs that contain various functional substituents; these MCL-PHAs are of great interest because these functional groups can improve the physical properties of the polymers, allowing the creation of tailor-made products. Moreover, some functional substituents can be modified by chemical reactions to obtain more useful groups that can extend the potential applications of MCL-PHAs as environmentally friendly polymers and functional biomaterials for use in biomedical fields. Although MCL-PHAs are water-insoluble, hydrophobic polymers, they can be degraded by microorganisms that produce extracellular MCL-PHA depolymerase. MCL-PHA-degraders are relatively uncommon in natural environments and, to date, only a limited number of MCL-PHA depolymerases have been investigated at the molecular level. All known MCL-PHA depolymerases share a highly significant similarity in amino acid sequences, as well as several enzymatic characteristics. This paper reviews recent advances in our knowledge of MCL-PHAs, with particular emphasis on the findings by our research group.

Selection of Herbicide Tolerant Cell lines from $\gamma$-ray-Irradiated Cell Cultures in Rice (Oryza sativa L. cv. Ilpumbyeo)

  • Herbicide tolerant rice (Oryza sativa L. cv. Ilpumbyeo) cell lines were selected from $\gamma$-ray-irradiated anther-derived cell cultures. The anther-derived cell clusters were small (300 to 400 ${\mu}{\textrm}{m}$ in diameter) and uniform ones that were screened by miracloth filtering. The cell suspensions were very efficient to plate one layer onto agar medium and to screen target cell lines. Herbicide tolerant cell lines were selected by 5 mg/L cyhalofop butyl (CHB) treatment by using the small cell suspensions on agar N6 medium containing 1 mg/L 2,4-D and 0.2 mg/L kinetin. Of the cell lines, one line (CHB-1) showed stable tolerance at 10 mg/L concentration after 6-month culture without herbicide suspension. Growth stability of CHB-1 was similar to that of control cell line on 10 mg/L CHB containing medium. In this experiment we established herbicide tolerant cell line selection system by using anther-derived uniform-cell suspensions with $\gamma$-ray-irradiation.

Streptomyces griseoplanus SL20209에 의한 Aminopeptidase M 저해제의 생산 조건

  • Maximum amount of the aminopeptidase M inhibitors produced by Streptomyces griseoplanus SL20209 in 500 ml-Erlenmeyer flask was accumulated after cultivation for 3 days at 28$\circ$C, thereafter the amount of inhibitors decreased slowly with a pH change to alkaline. Arabinose, xylose, mannose and soluble starch were good carbon sources for the production of the inhibitors. On the other hand, glucose was only good for the cell growth but potently inhibited the production of inhibitors. Natural organic nitrogen sources such as soybean meal, fish meal, gluten meal and peanut powder were good for the production of inhibitors, however, soytone, peptone and inorganic nitrogens such as NH$_{4}$C1 and NH$_{4}$NO$_{3}$ were poor. Inclusion of yeast extract (0.5%, w/v) or K$_{2}$HPO$_{4}$ (0.05%) into the production medium increased the production of inhibitors by accelerating cell growth. The production of inhibitors was slightly increased on the medium containing CaCO$_{3}$ (0.3%) and zeolite (0.5%), respectively. Optimal temperature and initial pH range for the production ot inhibitors were determined to be 28$\circ$C and 6.0-7.0, respectively. Employing two improved production media consisting of 3% arabinose or soluble starch, 2.5% soybean meal, 0.5% yeast extract, 0.05% K$_{2}$HP0$_{4}$, 0.1% CaCO$_{3}$ and 0.3% zeolite (pH 6.8), 1.8-fold increase in the amount of inhibitors was achieved, comparing with the basal medium used.

Varietal Difference Based on Efficiency of Rice Anther Floating Culture

  • To evaluate the efficiency of anther floating culture according to the maturing group, the varietal difference and classification of fifty varieties was conducted in N6 liquid medium containing 1mg $l^{-1}$ NAA, 0.25 mg $l^{-1}$ kinetin. The efficiency of callus induction was widely ranged from 0 to 113.4%, but the mean callus induction was not significantly different among maturing groups. The efficiency of anther floating culture showed the highest variation in early-maturing group among three maturing groups. The varieties with the best callus induction were Sambaegbyeo and Jinbuolbyeo, while the recalcitrant variety was Obongbyeo in early-maturing group. The efficiency of plant regeneration showed the highest trends in late-maturing group among three maturing groups. The fifty varieties were classified into three groups (distance=0.78) by cluster analysis based on the callus formation and plant regeneration. Group including only two varieties, Shinunbongbyeo and Sambaegbyeo had the excellent androgenic efficiency, and the medium efficiency of Group was included thirty-six varieties. Whereas twelve varieties, including three Tongil varieties were fell into the bad efficiency of Group. Especially, Tongil varieties containing Japonica rice, Obongbyeo were the recalcitrant genotypes for the anther floating culture.

Modeling of Breast Cancer Prognostic Factors Using a Parametric Log-Logistic Model in Fars Province, Southern Iran

  • In general, breast cancer is the most common malignancy among women in developed as well as some developing countries, often being the second leading cause of cancer mortality after lung cancer. Using a parametric log-logistic model to consider the effects of prognostic factors, the present study focused on the 5-year survival of women with the diagnosis of breast cancer in Southern Iran. A total of 1,148 women who were diagnosed with primary invasive breast cancer from January 2001 to January 2005 were included and divided into three prognosis groups: poor, medium, and good. The survival times as well as the hazard rates of the three different groups were compared. The log-logistic model was employed as the best parametric model which could explain survival times. The hazard rates of the poor and the medium prognosis groups were respectively 13 and 3 times greater than in the good prognosis group. Also, the difference between the overall survival rates of the poor and the medium prognosis groups was highly significant in comparison to the good prognosis group. Use of the parametric log-logistic model - also a proportional odds model - allowed assessment of the natural process of the disease based on hazard and identification of trends.

Geomechanical analysis of elastic parameters of the solid core of the Earth

  • It follows from the basic principles of mechanics of deformable solids relating to the strength, stability and propagation of elastic waves that the Earth's inner core cannot exist in the form of a spherical structure in the assumed thermobaric conditions and calculation values of physico-mechanical parameters. Pressure level reaches a value that is significantly greater than the theoretical limit of medium strength in the model approximations at the surface of the sphere of the inner core. On the other hand, equilibrium state of the sphere is unstable on the geometric forming at much lower loads under the influence of the "dead" surface loads. In case of the action of "follower" loads, the assumed pressure value on the surface of the sphere is comparable with the value of the critical load of "internal" instability. In these cases, due to the instability of the equilibrium state, propagation of homogeneous deformations becomes uneven in the sphere. Moreover, the elastic waves with actual velocity cannot propagate in such conditions in solid medium. Violation of these fundamental conditions of mechanics required in determining the physical and mechanical properties of the medium should be taken into account in the integrated interpretations of seismic and laboratory (experimental) data. In this case, application of the linear theory of elasticity and elastic waves does not ensure the reliability of results on the structure and composition of the Earth's core despite compliance with the required integral conditions on the mass, moment of inertia and natural oscillations of the Earth.
