Mass rearing system for Neodryinus typhlocybae(Hymenoptera: Dryinidae) as a biological control agent of Metcalfa pruinosa (미국선녀벌레 천적인 선녀벌레집게벌 대량사육 체계)

  • The mass-rearing system for Neodryinus typhlocybae as a biological control agent of Metcalfa pruinosa was established. Depending on the density of host nymphs and plants, the average number of cocoons produced by the parasitoids was 5-8 and 70-150 cocoons per leaf and sapling of mulberry, respectively. There is a significant difference in cocoon length between females (6.10-6.46mm) and males (4.20-4.62mm). Sex determination of cocoons before emergence will be helpful for efficiently releasing this parasitoid in fields. The parasitic rate of N. typhlocybae at the semi-field condition was on average 13-17%. The release number of this parasitoid did not affect parasitism. Nevertheless, the population growth rate of M. pruinosa was reduced by increasing the release number of N. typhlocybae. The parasitoid offspring's sex and bivoltine were influenced by the host age. On young host nymphs, the bivoltine portion of parasitoid increased. When parasitized on 4th or 5th nymphs, the offspring's female ratio of N. typhlocybae increased. This result may be useful for potentially controlling mass rearing production of parasitoid.

Morphological Characteristics and Material Comparison of Native Orostachys species (Orostachys japonica (Maxim.) A. Berger, Orostachys minuta (Kom.) A. Berger, Orostachys chongsunensis Y. N. Lee) (자생 바위솔(바위솔, 정선바위솔, 좀바위솔)의 형태적 특성과 성분 비교)

  • Orostachys species grow from anywhere in Korea. This variety, which lives in the harsh areas of the beach and mountains, is known to have as much change as it was isolated. It has been used as a medicine for a long time, and now the flower is beautiful and has been developed for horticulture. As a result of morphological characteristics and component analysis of three Orostachys species, the following results were obtained. 1. Orostachys species showed two types of leaves according to the growing environment. O. japonica (Maxim.) A. Berger and O. minuta (Kom.) A. Berger were lanceolate, and O. chongsunensis Y. N. Lee was obovate. 2. The leaves were all green. However, some of the O. japonica (Maxim.) A. Berger had white powder. Some of the O. minuta (Kom.) A. Berger was green with red spots on the back. O. chongsunensis Y. N. Lee showed strong gray before flowering but weak green when flowering. 3. The thorns of the leaves were O. japonica (Maxim.) A. Berger and O. minuta (Kom.), but O. chongsunensis Y. N. Lee was not. 4. Whether it was a leaf dot or not, there were dots in O. japonica (Maxim.) A. Berger and there were dots on the O. minuta (Kom.) and the O. chongsunensis Y. N. Lee. 5. The flowers were all white, and the stamens were red in O. japonica (Maxim.) A. Berger and O. minuta (Kom.) A. Berger and white in O. chongsunensis Y. N. Lee. 6. It was confirmed that the ingredients in Orostachys species contained kaempferol, quercetin, flavonoid etc., which have high antioxidant activity.

Phenotypical Characteristics Investigation and Selection of Superior Individuals from Natural Habitats of Sageretia thea in South Korea (국내 자생 상동나무 표현형 특성조사 및 우량 개체 선발)

  • In this study, the growth, physical characteristics of fruit and soil chemistry were investigated by grouping S. thea populations. The soil for each group was sandy loam or sandy loam, and it was analyzed to be slightly acidic to neutral, with a pH ranging from 5.6 to 7.0. Additionally, the leaf size was highest in the G7 group, and overall fruit growth was highest in the G1 group, confirming differences between the groups. The sweetness ranged from 16.8° Brix to 12.3° Brix, indicating a higher sweetness compared to Vaccinium oldhamill, blueberry, and blackberry. The correlation analysis between soil chemistry and S. thea growth characteristics revealed a significant negative correlation between calcium (Ca) and fruit growth characteristics. Furthermore, a significant positive correlation was observed between sodium (Na) and fruit acidity, as well as between cation exchange capacity (CEC) and fruit hardness.

Evaluation of antioxidant activity, zebrafish embryo toxicity, and regenerative efficacy of Symphoricarpos albus (Symphoricarpos albus의 항산화능과 Zebrafish 배아 독성 및 재생 효능 평가)

  • This study compared and evaluated the antioxidant activities of Symphoricarpos albus(S. albus) extract and fermented extract. Antioxidant activity was measured by DPPH radical scavenging, FRAP, and ABTS. Concentrations were measured at 200, 100, 50, and 10 ㎍/mL, and antioxidant activity increased in a concentration-dependent manner. S. albus leaves fermented extracts had the highest antioxidant activity. And this study evaluated the safety and tail regeneration of S. albus extract using zebrafish model embryos. Zebrafish are in the spotlight as an alternative animal and can be used for cosmetic research. Zebrafish embryos were collected and evaluated for coagulation rate, hatching rate, and cardiotoxicity. As a result, it was toxic at concentrations above 100 ㎍/ml. The tail was cut and the regenerative effect was observed for 3 days. As a result, from 72 hours, S. albus 200ug/ml leaf extract showed a 17% regenerative effect compared to the control group. These results suggest that S. albus can be used as a natural material for antioxidant and regeneration for skin improvement.

Variation of Stomatal Traits of Natural Population of Quercus spp. (참나무 천연집단(天然集團)의 기공형질변이(氣孔形質變異))

  • The variation of stomatal density and stomatal length of four species of oaks was studied for the purpose of examining the differences among populations and among individual trees within population. Nine populations of Quercus mongolica, four populations of Q. serrata and Q. variabilis respectively, and three populations of Q. acutissima were selected in the natural stands of oaks distributed through the whole country. Twelve leaves were sampled from each of 20 trees from each population. The length of 20 stomata and ten replications of stomatal density were measured from collodion replicas of each leaf under a microscope. Average stomatal densities and lengths ranged through $600-1000/mm^2$ and $19-26{\mu}m$ respectively in all of the species studied. The stomatal densities and lengths presented significant differences statistically at the level of 1 or 5% among populations and among individual trees within population in all the species. Quercus mongolica, especially, showed large variation among populations, while Q. variabilis did very narrow variation compared to the other species. The coefficients of variation of stomatal densities and lengths among individual trees within population exhibited small values of 3.7-12.0% and 1.4-5.3% respectively in all the populations of the species. The average stomatal densities of Q. mongolica showed statistically significant correlation of multiple correlation coefficient of $R_{df{\cdot}2.6}=0.868^*$ and multiple regression equation of $Y=0.041X_1(G.M.T.S.)+0.489X_2(G.M.H.S.)+22.37$ with the sum of growing season mean daily temperature and the sum of growing season mean daily humidity of the stand studied. However the average stomatal lengths showed no relation with the same meteological variables. The figures of frequency distribution of the measurements of leaves or the mean values of individual trees did not show normal distribution curves in some populations. The curves, as well as the results of ANOVA, exhibited the differences among populations.

Study on the Biosynthetic Characteristics of Photosynthetic Pigments in Dodder(Cuscuta australis R. Br.) Plant (실새삼의 광합성색소 생합성특성에 관한 연구)

  • This study was conducted to investigate whether 1) photosynthetic pigments(chlorophylls and carotenoids) are formed in dodder plant(Cuscuta australis), 2) there are any characteristics in the pigment biosynthesis, compared to that of other normal plants, and 3) dodder responds to some herbicides having target site on chloroplast. 1. Chlorophyll content of dodder tendrill grown under a natural daylight was 9 times and 50 times lower than that of field bindweed stem and leaf, respectively. 2. The photosynthetic pigment contents varied in different tissues, being higher in a apical region than in a lower region of seedling or tendrill. Chlorophyll wasn't almost observed below the 4th internode from the upper. 3. Pigment contents were greatly dependent on light intensity so that there were 4 to 6 times difference among light conditions. When the shoot containning low pigment contents under natural light, was incubated in growth chamber with various light intensities, the pigment contents were increased by 3 times of initial contents at about 97${\mu}E$ $m^{-2}s^{-1}$PAR. While the change in pigment contents was not observed at above 450${\mu}E$ $m^{-2}s^{-1}$PAR 4. Exogenous supply of 5mM 5-aminolevulinic acid increased protochlorophyllide by 7 times and 1.4 times in the etiolated shoot from field bindweed rhizome and in dodder stem, respectively, showing that dodder relatively has a low response to 5-aminolevulinic acid. 5. Pigment loss was observed in the treatment of paraquat, norflurazon, oxyfluorfen and diuron, and protoporphyrin IX was accumulated by oxyfluorfen as in normal plants Based on above results, several chracteristics of pigment biosynthesis in dodder seem to be summerized as follows. Photosynthetic pigment biosynthesis in Cuscuta australis runs even in low level. The pigment contents is differentially distributed in different regions and their contents seem significantly to be controlled by light intensities. Especially, chloroplast rapidly tends to degenerate with the development of tissue. Some herbicides having target site on chloroplast induce damage to dodder stem but are unlikely to control it well in field, except paraquat, due to low chloroplast activity and parasitic mode of nutrition.

In vivo Antifungal Activity of Pyrrolnitrin Isolated from Burkholderia capacia EB215 with Antagonistic Activity Towards Colletotrichum Species (탄저병균에 대하여 길항작용을 보이는 Burkholderia cepacia EB215로부터 분리한 Pyrrolnitrin의 항균활성)

  • An endophytic bacterial strain EB215 that was isolated from cucumber (Cucumis sativus) roots displayed a potent in vivo antifungal activity against Colletotrichum species. The strain was identified as Burkholderia cepacia based on its physiological and biochemical characteristics, and 16S rDNA gene sequence. Optimal medium and incubation period for the production of antifungal substances by B. cepacia EB215 were nutrient broth (NB) and 3 days, respectively. An antifungal substance was isolated from the NB cultures of B. cepacia EB215 strain by centrifugation, n-hexane partitioning, silica gel column chromatography, preparative TLC, and in vitro bioassay. Its chemical structure was determined to be pyrrolnitrin by mass and NMR spectral analyses. Pyrrolnitrin showed potent disease control efficacy of more than 90% against pepper anthracnose (Colletotrichum coccodes), cucumber anthracnose (Colletotrichum orbiculare), rice blast (Magnaporthe grisea) and rice sheath blight (Corticium sasaki) even at a low concentration of $11.1\;{\mu}g/ml$. In addition, it effectively controlled the development of tomato gray mold (Botrytis cinerea) and wheat leaf rust (Puccinia recondita) at concentrations over $33.3\;{\mu}g/ml$. However, it had no antifungal activity against Phytophthora infestans on tomato plants. Further studies on the development of microbial fungicide using B. cepacia EB215 are in progress.

Effect of Night-break Period on Growth and Anthesis of Orostachys japonicus (암기중단 처리시간에 따른 바위솔의 생장과 개화)

  • Orostachys japonjcus, called Wasong in herb medicine, has been artificially cultivated as an anti-tumor medicinal. The experiment was done to examine the effect of night-break periods imposed immediately before its bolting time on its morphological, flowering-related characters and fraction dry weights. After a plant was grown in a 15cm plastic pot containing a 2:1 soil:Peat moss mixture for about 3 months, three different night-break periods (0.5, 1 and 2 hours) around midnight were treated from Aug. 24. to compare with the natural daylength. The plants were sampled 6 times by 2-week interval after the treatments. Plant height and inflorescence length of all the treatments inclined with time lapse after the treatment and were shorter in 2 hour night-break due to slow increment than in the other treatments, while stem diameter showed reverse result. All the treatments except 2 hour night -break were nearly same in fraction, shoot and total dry weights per plant; two hour night-break treatment had greater leaf and bract weight from 6 weeks, greater stem, shoot and total weights from 2 weeks and greater root weight from 4 weeks but did less floret weight after the treatment. Although florets on the inflorescence were formed in 2 hour night-break treatment, flowering florets and flowering plants never occurred. In the other treatments showed the similar response, however, more florets appeared from 2 weeks, flowering florets was sharply increased from 6 weeks and flowering plants were reached up to 100% from 6 to 8 weeks after the treatment. Inflorescence length or number of total florets per plant in 2 hour night-break was positive-correlated to all the fraction dry weights except that those of natural daylength was not done, meaning that its artificial cultivation should permit bolting to secure more shoot dry matter.

Regeneration of Seedlings under Different Vegetation Types and Effects of Allelopathy on Seedling Establishment of Abies koreana in the Banyabong Peak, Mt. Chiri (지리산(智異山) 구상나무림(林)에서 타감작용(他感作用)이 치수형성(稚樹形成)에 미치는 영향(影響))

  • This study was conducted to compare the regeneration of the seedlings under different vegetation types and to identify the presence of allelopathy in Abies koreana(Ak) natural forest in Banyabong Peak(elevation, 1715m), of Mt. Chiri. Twenty quadrats($10m{\times}10m$) were placed in May, 1996 to classify vegetation structure using TWINSPAN. Water-soluble extracts from leaves and soil humus of different vegetation types were collected to test their effects on both seed germination of Ak and mycelial growth of ectomycorrhizal fungi. Phenolic compounds from soil humus were quantified using HPLC. Among the four vegetation types, Sasa borealis(Sb) was found in both Ak-Quercus mongolica(Qm) and Ak-Rhododendron schlippenbachii(Rs) communities. Natural seeding of Ak was $230,000{\pm}90,000seeds/ha$ in 1995 and their germination rate was 25% in an ideal laboratory condition. Density of Ak seedlings less than 5cm in height was 52,000/ha in 1996, while that of seedlings taller than 5cm in height was only 4,000/ha. In the case of Ak-Qm community, density of Ak seedlings with Sb understory was only 7% of the density of seedlings with Rs understary, suggesting the inhibitory effect of Sb. The germination rate of Ak seeds was significantly reduced by leaf extracts of Sb, and Rhododendron mucronulatum var. ciliatum(Rm) and Ak. Soil humus extract of Ak-Qm-Sb subcommunity reduced germination of Ak seeds by 81% and also reduced by 19% the respiration of mycelia of ectomycorrhizal fumgus, Lactarius sp. Among the seven phenolic compounds identified from the soil humus, extract, Ak-Qm-Sb subcommunity contained significantly high content of p-hydroxybenzoic acid, vanillic acid, and syringic acid. Particularly, p-hydroxybenzoic acid was present at 4.2ppm in the Sb roots and at 16.5ppm in the Sb humus, suggesting that it could be the primary allelopathic compound in Abies koreana forests with Sasa borealis understory.

Biomass and Net Production of a Natural Quercus variabilis Forest and a Populus alba × P. glandulosa Plantation at Mt. Mohu Area in Chonnam (전남(全南) 모후산지역(母后山地域) 굴참나무천연림(天然林)과 현사시나무인공림(人工林)의 물질생산(物質生産)에 관(關)한 연구(硏究))

  • A natural Quercus variabilis forest and a Populus alba${\times}$P. glandulosa plantation in Mt. Mohu area were studied to investigate aboveground biomass and net production. A $20m{\times}30m$ quadrat was set up in each stand, and 10 sample trees each of Quercus variabilis and Populus alba ${\times}$ P. glandulosa were cut for dimension analysis. There was little difference in accuracy among three biomass regression models of logWt=A+BlogD, $logWt=A+BlogD^2H$, and logWt=A+BlogD+ClogH, where Wt. D, and H were dry weight, DBH, and height, respectively. Aboveground total biomass of Quercus variabilis stand was 31,275kg/ha, and that of Populus alba ${\times}$ P. glandulosa was 55,581kg/ha. In both of Quercus variabilis stand and Populus alba ${\times}$ P. glandulosa stand, the proportion of each tree component to abovegound total biomass was high in order of stem wood, branches, stem bark, and leaves. Quercus variabilis stand was higher in the proportion of stem bark, branches and leaves than Populus alba ${\times}$ P. glandulosa stand, while the former was lower in that of stem wood than the latter. Aboveground total net production of Quercus variabilis stand was 4,267kg/ha/yr., and that of Populus alba ${\times}$ P. glandulosa stand was 3,903kg/ha/yr. The proportion of each tree component to aboveground total net production of Quercus variabilis stand was high in order of leaves, stem wood, branches, and stem bark. That of Populus alba ${\times}$ P. glandulosa stand was high in order of stem wood, leaves, branches, and stem bark. Net assimilation rate and efficiency of leaf to produce stem of Quercus variabilis stand were 2.121 and 0.840, respectively. Those of Populus alba ${\times}$ P. glandulosa stand were 3.376 and 2.085, respectively. Though Populus alba${\times}$P. glandulosa stand was lower in aboveground total net production than Quercus variabilis stand, the former was higher in aboveground total biomass than the latter. The reason was that Populus alba${\times}$P. glandulosa stand was higher in net production of stem wood of accumulation organs than Quercus variablis stand.
