Effects of Cutting Condition on Rooting and Growth of Cut Flower in Plug Cutting of Dendranthema grandiflorum 'Shinma' (국화 '신마'의 플러그 삽목시 삽수조건이 발근 및 절화생육에 미치는 영향)

  • This study was carried out to examine the effect of cutting condition on rooting and their subsequent growth of cut flower in plug cutting of 'Shinma', one of autumn-flowering chrysanthemums. According to the results, cutting with four or six leaves was effective in shoot growth and rooting than any other treatments, and also the length and thickness of cut flower were increased. In case of cutting with leafless, it showed the poorest shoot growth among all of the treatments and there was no root. The weight of shoot was heavier than any other treatments in case of the length of cutting (5~7 cm). And the weight of root and its number were better than any other treatments, in treatment of the length of cutting (5 cm). The growth of shoot and rooting were enhanced by increasing the thickness of cutting. In case of thickness of cutting, the cutting (3.2 mm tnickenss) showed more favorable shoot length of cut flower than any other treatment. Therefore, it is recommended that the 5 cm long and 3.2 mm thick cuttings with four leaves are used to improve their rooting and subsequent growth in plug cutting of Dendranthema grandiflorum 'Shinma'.

Nutritional Analysis of Wild Conyza canadensis L. Extract (야생 망초(Conyza canadensis L) 추출물의 영양성분 분석)

  • Background: It has been reported that various substances exist in wild Conyza canadensis, but I think that the overall compositional analysis is still insufficient. Purposes: This study was to set extraction conditions and search for various nutritional components in the wild C. canadensis. Methods: Various extracts were extracted and analyzed using different analytical instruments to suggest and study the conditions for component analysis. Results: As for the general nutritional components of C. canadensis leaf extract, carbohydrates were 46.4%, crude protein 26.5%, moisture 16.3%, raw meal 9.5%, and natural fat 1.2%. The dietary fiber content was 30.76%. Free fructose, glucose, sucrose, and maltose were separated, and the amount of fructose was as high as 4,144.2 mg/100 g. In addition, we found 64.87 mg/g of K, 4.09 mg/g of P, 1.77 mg/g of Mg, and 0.16 mg/g of disodium in the extracts. A total of 20 fatty acids have appeared. In addition, unsaturated fatty acids (octadecenoic acid, octadecadienoic acid, and linolenic acid) were detected. Linolenic acid was high at 54.1%. The saturated fatty acid content was 0.39 g/100 g, and the trans fatty acid content was 0.01 g/100 g. No cholesterol was found in the plant. Riboflavin was at 0.4 mg/100 g, with six essential amino acids and 24 amino acids. Conclusions: As a result of the primary research on the plant, various nutritional and functional ingredients exist in the extract, and natural healing uses are available.

Studies on the Taxonomical Characters in Populus tomentiglandulosa and P. glandulosa (Populus tomentiglandulosa와 P. glandulosa의 분류학적(分類學的) 연구(硏究))

  • Populus glandulosa and Populus tomentiglandulosa, which were known to be natural hybrids, were examined for morphological, physiological and karyological traits to illucidate its hybridity and taxonomical importance. The results abtained were as follows; 1. Survival rate in rooting of cuttings and grafting was different between the hybrids and their rooting abilities showed incomplete dominance. 2. Their leaf openings showed incomplete dominance. The leaf longevities of P. alba ${\times}$ glandulosa and P. tomentiglandulosa were stronger than the other hybrids. 3. There were differences in resistance to toxicity of $KClO_3$ between the hybrids. 4. Many external leaf characters of the hybrids also showed incomplete dominance. P. tomentiglandulosa was similar in those characters to P. alba ${\times}$ glandulosa while P. glandulosa was similar to hybrids crossed, reciprocally crossed or back-crossed between P. davidiana and P. alba. 5. Their numbers of male flower showed incomplete dominance or hybrid vigor. The numbers of P. tomentiglandulosa were similar to thosa of P. alba ${\times}$ glandulosa while those of P. glandulosa to those of P. alba ${\times}$ davidiana or P. davidiana ${\times}$ alba. 6. Morphology and band color of male catkin bract showed incomplete dominance. Those of P. glandulosa were similar to those of P. alba ${\times}$ davidiana while those of P. tomentiglandulosa to those of. P. alba ${\times}$ glandulosa. 7. There were differences in vascular bundle number and arrangement of petiole between the hybrids. 8. Differences in the anatomical traits of stem did not exist between the hybrids but those in wood fiber size existed. 9. The chromosomes of artificial hybrids, P. glandulosa and P. tomentiglandulosa showed irregular behavior in metaphase I and II. 10. All hybrids including P. glandulosa and P. tomentiglandulosa showed small number of P.M.C. with 19 II but many univalent chromosomes were exhibited in metaphase I. 11. All hybrids including P. glandulosa and P. tomentiglandulosa showed a little abnormal nuclear plates as laggard chromosome and chromosome bridge in anaphase I and II. 12. The frequency of pollen tetrad and fertile pollen was low in most of the hybrids including P. glandulosa and P. tomentiglandulosa.

Classification for Types of Damages Caused by Cold Stress at Different Young Spike Development Stages of Barley and Wheat (맥류의 유수발육기 저온장해유형과 피해시기 분류)

  • Although the young spike of barley (Hordeum vulgare L.) or wheat (Triticum aestivum L.) is known as the most susceptible part to spring cold injury, the risk of cold injury is apt to be ignored in most breeding program due to the importance of early maturity. Based on these aspects, the types and inducing time, temperature conditions for induction and effects of cold injury on growth and yield in this study were investigated under greenhouse and field conditions through three years (1997-1999). In natural condition, low temperature around -2.4∼$-10.2^{\circ}C$ caused the death of plant. Several cold injury types such as partial degeneration of spike, partial discoloration of leaf, spike and awn, discoloration of culm and white spike were observed at low temperature around $-3.1^{\circ}C$. Low temperature around -2.4∼$-8.6^{\circ}C$ and 1.3-$7.6^{\circ}C$ caused degeneration and sterility of spike, respectively. Most materials were prepared to the spikelet foundation stage, spikelet differentiation stage, development stage of flower organ, booting stage and heading stage, which were known having risk for cold injury in field condition. Although most of the controlled stages were sensitive to the induced low temperature, booting stage was the most sensitive stage for cold injury. All of growth stages which were treated-heading stage, booting stage, development stage of flower organ, spikelet differentiation stage, spikelet foundation stage-were responded to low temperature treatment but the symptoms revealed were very specific according to the growth stages. Ears of plant in heading stage were discolored to white. Ears of plant in booting stage were degenerated in all or part of one. Plants in spikelet differentiation stage were sterile in all or part of one. When tried to detect the specific differences between normal and cold injured plants in appearance, spike length, distance between spike and flag leaf and the first internode length could be the critical points for occurrence of spike death caused by cold injury. In barley, the elongation of spike was stopped on 3.2cm after occurrence of spike degeneration, 4.7cm after occurrence of partial degeneration of spike, 5.0cm after occurrence of white spike. In wheat, it was stopped on 1.6cm after occurrence of stem death, 3.3cm after occurrence of spike degeneration, 8.3cm after occurrence of partial degeneration of spike, 8.1cm after occurrence of white spike, 7.5cm after partial discoloration of leaf and 9.3cm after partial discoloration of spike. The obtained results from low temperature treatment induced in growth chamber were similar to the field experiment, Beacuse the death of spikes was more when low temperature was treated two times than one times, the temperature should be upgrade to -3$^{\circ}C$ in order to get the same condition with field test.

Antioxidant Activities of Bamboo (Sasa Borealis) Leaf Extract according to Extraction Solvent (추출용매에 따른 조릿대 잎 추출물의 항산화활성)

  • This study was carried out to investigate the antioxidant activity of bamboo (Sasa borealis) leaf extract by measuring electron donating ability, superoxide dismutase (SOD)-like activity, reducing power, and lipid peroxidation inhibitory activity. Two crude extracts by water or 70% EtOH and five fractions of n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous from the crude extract of 70% EtOH were prepared for this study. The crude extracts of water and 70% EtOH yielded 8.5% and 11.4%, respectively and the yields of n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous fractions were 5.1% to 0.6%. Total polyphenol contents of the water and the 70% EtOH crude extracts were not significantly different; however, their total flavonoid contents were significantly greater in the 70% EtOH than in the water crude extract. Total polyphenol contents were the highest in chloroform fraction followed by ethyl acetate and n-butanol fractions and total flavonoid contents were the highest in ethyl acetate fraction followed by chloroform and n-hexane fractions. The two crude extracts as well as the five fractions showed election donating ability, SOD-like ability, reducing power, and lipid peroxidation inhibitory activity. Most of the antioxidant activities of each crude extract or fractions increased proportionally with the concentration. These results indicate that bamboo (Sasa borealis) leaf extracts show antioxidant activities due to its substantial content of polyphenol including flavonoid. Thus, it could be concluded that crude extracts by water or 70% EtOH and the fractions from the 70% EtOH extract, especially chloroform, ethyl acetate and n-butanol, would be useful as natural antioxidant substances.

Effects of Tempearture and Food Source on Pupal Development, Adult Longevity and Oviposition of the Tobacco Cutworm, Spodoptera litura Fabricius (온도 및 기주조건이 담배거세나방의 용발육, 성충수명 및 산란에 미치는 영향)

  • This study was conducted to determine the effect of temperatures, 24"C, 28$^{\circ}$C and 32"C,and food sources on pupal development, adult longevity and oviposition of tobacco cutworm, Spodopteralitura Fabricius. Percent pupation of S. litura was became higher at higher temperature from 23% to 88%.And its percent pupation was highest on soybean leaf (SL), followed by perilla leaf (PL), sweet potato leaf(SPL) and artificial diet (AD). Pupal weight ranged from 0.28g on SPL to 0.40 g on PL and was tended toheavier with decreasing temperature. The mean pupal duration was 14, 10 and 7 days at 24"C, 28$^{\circ}$C and32"C, respectively. Percent emergence was in range of 21% to 89% with higher percent emergence as thetemperature increased and both 28$^{\circ}$C and 24$^{\circ}$C was highest on SL, followed by PL, SPL and AD, but at24$^{\circ}$C the order was SPL, PL, SL and then AD. Preoviposition duration was 3.2 days at 24"C, 2.8 days at28$^{\circ}$C and 2.5 days at 32$^{\circ}$C. Adult longevity became shorter as the temperature increased from 6.4 to 3.9days. Male longevity was longer than that of female. Adults lived longest when they were reared on PL,followed by SL, SPL and AD. Total number of eggs laid per female varied from 803 to 1,441 regardlessof the treatments, but those were significantly more on natural foods than on AD. Number of eggs per eggmass was 97.4 at 24$^{\circ}$C 155.8 at 28$^{\circ}$C and 104.7 at 32$^{\circ}$C. Number of egg mass was 12.0, 6.7 and 11.3 at24"C, 28$^{\circ}$C and 32"C, respectively.4"C, 28$^{\circ}$C and 32"C, respectively.

Anti-Diabetic, Alcohol-Metabolizing, and Hepatoprotective Activities of Moringa (Moringa oleifera Lam.) Leaf Extracts (모링가 잎 추출물의 항당뇨, 알코올 대사 및 간 보호 활성)

  • This study was carried out to investigate anti-inflammatory, anti-diabetic, alcohol metabolizing, and hepatoprotective effects of hot water (MOW) and 80% ethanol (MOE) extracts from moringa (Moringa oleifera Lam.) leaf. The total phenol content of MOW and MOE were 45.49 and 63.06 mg tannic acid equivalents/g, respectively. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activities of MOW and MOE were remarkably elevated in a dose-dependent manner, and about 60.8% and 71.3% at 1 mg/mL, respectively (P<0.01). Superoxide dismutase-like activities of MOW and MOE were 2.8% and 7.4% at 5 mg/mL, respectively (P<0.05). ${\alpha}-Glucosidase$ inhibitory activity also increased in a dose-dependent manner in both extracts, and MOE was higher about two times than MOW at 5 mg/mL (P<0.001). The effects of MOW and MOE on alcohol metabolizing activity were determined by measuring generation of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). ADH and ALDH activities significantly increased upon addition of MOW and MOE (P<0.05). Anti-inflammatory activity was examined in lipopolysaccharide-stimulated RAW 264.7 cells. Nitric oxide production was reduced to 32.1% and 81.2% by addition of MOW and MOE at 1 mg/mL, respectively (P<0.05). MOW and MOE showed significant protective effects against tacrine-induced cytotoxicity in Hep3B cells at $100{\mu}g/mL$. These results suggest that moringa leaf extracts have great potential as natural health products.

Occurrence of White Rust and Growth of Chrysanthemum 'Baekma' under Various Relative Humidity and Temperature Conditions in the Greenhouse (시설 내 상대습도 및 온도 조건에 따른 국화 '백마'의 생육과 흰녹병 발생)

  • This study was carried out to investigate the effect of relative humidity (70, 80, 90, and 95%) and to examine the interrelationship of temperature (15, 20, and $25^{\circ}C$) and relative humidity (60, 70, 80, and 90%) in the greenhouse on growth and occurrence of white rust in chrysanthemum 'Baekma'. The developmental stages of white rust were monitored from the initial occurrence to sporidium formation in teliospores. When the relative humidity of greenhouse was adjusted to 90% or more for 40 days, the occurrence rate of white rust, the number of infected leaves, and the number of symptoms per leaf were more than 97%, 10, and 55, respectively. However, in the treatment with 70% relative humidity, the occurrence rate of white rust dropped to 30% and only two spots of symptoms were observed with only 1 or 2 infected leaves per plant. The growth of cut flowers, such as fresh weight and leaf number, was the highest in 70% relative humidity. In the 90% relative humidity treatment, the first symptom of white spot appeared at 6 days after planting in chrysanthemum infected with white rust (stage 1), and then after one day, teliospores came out through the epidermal tissue of leaf (stage 2). The telium was formed with increasing teliospores after 1 to 2 days (stage 3), and then the promycelia developed from teliospores (stage 4). After a lapse of 4 to 5 days, sporidia formed on promycelia (stage 5). Regardless of relative humidity, white rust did not occur at all in treatments at $25^{\circ}C$. In treatments at $20^{\circ}C$, the number of sporidia was high regardless of relative humidity, but white rust did not occur in 60% and 70% relative humidity. As the relative humidity was higher, the number of sporidia and occurrence rate of white rust greatly increased, but the occurrence rate of white rust decreased to less than 14% in 60% and 70% relative humidity in treatment at $15^{\circ}C$.

Antimicrobial Activity of Niaouli (Melaleuca quinquenervia) Leaf Extracts against Skin Flora (피부 상재균에 대한 니아울리 잎 추출물의 항균활성)

  • In this study, the antimicrobial activity of niaouli leaf extracts was evaluated against skin flora. The skin flora used for experiments were three gram-positive bacteria such as Bacillus subtilis (B. subtilis), Staphylococcus aureus (S. aureus), Propionibacterium acnes (P. acnes), and two gram-negative, Escherichia coli (E. coli), Pseudomonas aeruginosa( P. aeruginosa), and the yeast, Plasmodium ovale (P. ovale). The bioassay applied for determining the antimicrobial effects of niouli leaf extracts or fraction included the disc diffusion assay and broth dilution assay. Minimum inhibitory concentration (MIC) values of 50% ethanol extract on B. subtilis, S. aureus, P. acnes, E. coli and P. aeruginosa were 0.25%, 0.50%, 1.00%, 0.13% and 0.25% respectively and the MIC values of water fraction were 0.25%, 0.25%, 4,00%, 0.25% and 0.25%. P. ovale did not show antimicrobial activities. The MIC values of methyl paraben used as positive control indicated 0.25%, 0.25%, 0.25%, 0.13% and 0.50%. Also, Minimum bactericidal concentration (MBC) values of 50% ethanol extract were 2.00%, 2.00%, 1.00%, 0.50% and 2.00% individually and the MBC values of water fraction were 0.50%, 0.25%, 4.00%, 0.50% and 1.00%. The MBC values of methyl paraben indicated 1.00%, 0.500%, 0.50%, 0.50% and 1.00%. These results showed that water fraction was as good as methyl paraben except for P. acnes. The 50% ethanol extract also showed activity similar with it. Thus, it is concluded that the 50% ethanol extract/fraction of niaouli could be applicable to cosmetics as a natural preservatives effective in antimicrobial activity against skin flora.

Effects of Simulated Acid Rain on Nutrient Contents of Pinus densiflora S. et Z. and Forsythia koreana Nak. Seedlings (인공산성우(人工酸性雨)가 소나무 및 개나리묘(苗)의 식물체내(植物體內) 함유성분(含有成分)에 미치는 영향(影響))

  • For the purpose of examining the effects of simulated acid rain on nutrient contents of plant tissues in Pintos densiflora seedlings and Forsythia koreana rooted cuttings, the experimental design of randomized block arrangement with three replications was implemented in the experimental field of Yesan National Agricultural Junior College. One-year-old Pinus densiflora seedlings and Forsythia koreana cuttings were planted in the pots filled the mixed soils(nursery soil : forest soil of siliceous sandy loam=1 : 1 v/v) in the early spring of 1986. The regime of artificial acid rain, in terms of spray frequency per month and spray amount at single treatment per plot, was simulated on the basis of climatological data averaged from 30 years records. Simulated acid rain(pH 2.0, pH 4.0, and pH 5.5 as control) containing sulfuric and nutric acid in the ratio of 3 : 2(chemical equivalant basis) diluted with ground water were treated on the experimental plants under condition of cutting off the natural precipitation with vinyl tunnel, during the growing season(May 1 to August 31) in 1986. The results obtained in this study were as follow : (1) As for the nitrogen contents in plant tissues, P. densiflora increased significantly in one-year-old stembranch and root tissues, and F. koreana increased significantly in leaf and root tissues, as the pH levels of acid rain decreased. (2) The available phosphate contents in root tissues of P. densiflora, and in leaf and root tissues of F. koreana were significantly decreased, as the pH levels of acid rain decreased. (3) $K_2O$, CaO and MgO contents in plant tissues were significantly decreased in the both species as the pH levels decreased. And the effects of acid rain on F. koreana were higher than those of P. densiflora. (4) Sulfur contents of plant tissues in the both species were increased at pH 2.0 treatment. There were significant differences among three acid rain treatments in leaf and root tissues of P. densiflora, and in all parts of F. koreana. (5) In the effects of simulated acid rain on the both species and the tested soils, in general, F. koreana revealed higher sensitiveness than P. densiflora, and the lower pH levels of simulated acid rain were treated, the more sharp reaction was showed.
