제주 자생 식물 추출물의 항산화 및 항노화에 관한 연구(II) (Antioxidative and Antiaging Effects of Jeju Native Plant Extracts(II))

  • 선구 연구로부터 저자들은 제주 자생 식물 추출물의 항산화 및 세포보호 효과에 대한 결과의 일부를 이미 보고하였으며, 본 연구에서는 제주도에서 자생하는 나머지 37종의 식물 추출물에 대한 항산화, 미백 그리고 주름생성억제 효과를 측정하였다. 항산화 효과는 1,1-diphenyl-2-picrylhydrazyl radical(DPPH)을 이용한 free radical 소거 활성 측정, $Fe^{3+}-EDTA/H_2O_2$ 계에서 생성된 활성산소종으로 인한 luminol의 화학발광을 이용한 소거 활성, 사람 적혈구를 대상으로 하여 rose-bengal로 증감된 활성산소에 대한 세포보호 효과를 측정하였다. 미백 및 주름억제 효과측정으로는 각각 tyrosinase, elastase의 활성 저해 효과를 측정하였다. 실험결과, free radical의 소거 활성($FSC_{50}$)은 소귀나무 수피(Myrica rubra, 5 ${\mu}g/mL$), 광대싸리 수피(Securinega suffruticosa, 8 ${\mu}g/mL$)에서 높게 나타났고, 활성산소 소거 활성($OSC_{50}$)은 상수리나무 잎(Quercus acutissima)과 광대싸리 수피(Securinega suffruticosa)에서 0.009 ${\mu}g/mL$로 높게 나타났으며, 세포보호 효과(${\tau}_{50}$)는 50 ${\mu}g/mL$에서 광대싸리 수피(Securinega suffruticosa, 895 min), 버드나무 줄기(Salix koreensis, 640 min)에서 크게 나타났다. 200 ${\mu}g/mL$에서 tyrosinase의 활성 저해 효과($IC_{50}$)는 소귀나무 수피(Myrica rubra, 77.8%), elastase의 활성 저해 효과($IC_{50}$)는 버드나무 줄기(Salix koreensis, 76.2%)에서 큰 효과가 나타났다. 결론적으로 광대싸리 수피, 소귀나무 수피, 상수리나무 잎, 버드나무 줄기, 동백나무 잎/줄기 추출물은 기능성 화장품 제조를 위한 원료로 사용하기에 충분한 가능성이 있음을 확인할 수 있었으며, 향후에 제품화를 위하여 좀 더 다양한 연구들이 필요하다고 사료된다.

In Vivo Antifungal Activities of the Methanol Extracts of Invasive Plant Species Against Plant Pathogenic Fungi

  • Plants are the promising reservoirs for natural products with their diverse secondary metabolites. Many invasive plants have been introduced in Korea, which adversely affect on the native ecosystem but holds difficulty removing them due to their proliferation. In this study, we evaluated disease control efficacy of methanol extracts from four invasive plant species against 7 representative crop pathogens. Methanol extract of Phytolacca americana effectively suppressed rice blast, tomato gray mold, and tomato late blight in a dose dependent manner. The methanol extract of Amorpha fruticosa also exhibited potent antifungal activity against pepper anthracnose in a concentration dependent way. These data suggest that the extracts of P. americana and A. fruticosa can be developed as plant disease protection agents against rice blast, tomato gray mold, tomato late blight, and pepper anthracnose. Furthermore, more extensive research will be required to identify and isolate active compounds from problematic invasive plant species to develop valuable agrochemicals.

Preliminary screening to assess the antimicrobial activities of extracts of evergreen woody species from South Korea against Staphylococcus aureus

  • This study aimed to screen for plants with antimicrobial potential among the evergreen woody species of South Korea that are used for horticulture and landscaping and to provide basic information about plants with proven antimicrobial activity to underpin future research. The plant materials were extracted under various conditions, and the antimicrobial activities of the extracts were evaluated by agar diffusion assay. The screening tests demonstrated that the crude extracts of 43 species had inhibitory effects against S. aureus. The inhibitory activities of four species (Elaeocarpus sylvestris, Camellia japonica, Cleyera japonica, and Quercus salicina) were relatively higher than that of the synthetic antimicrobial agents methylparaben and phenoxyethanol. The highest inhibitory activity was observed with the leaf extracts (extracted with methanol for 30 minutes) of E. sylvestris, based on induction of the largest inhibition zone of 23.3 mm in size. In addition, solvent fractions of E. sylvestris were evaluated. The largest inhibitory zone of 23.1 mm was observed for the n-butanol fraction, which is likely to contain effective compounds that exhibit inhibitory activity against S. aureus. In contrast, n-hexane and residual aqueous fractions showed no antimicrobial activity. Overall, our findings confirm that evergreen woody plants native to South Korea have potential antimicrobial activity.

자생 식물로 부터 Cyclin-dependent Kinase 4/Cyclin D1 저해물질의 탐색 (Searching of Cyclin-Dependent Kinase 4/Cyclin D1 Enzyme Inhibition Materials from the Native Plants)

  • 인체 세포의 세포주기를 조절하는 Cyclin-dependent Kinase(CDK) 중 CDK4/Cyclin D1 효소 저해 물질을 찾기 위하여 국내에 자생하는 37과 87종의 식물 추출물을 탐색한 결과, 50%$(5\;{\mu}g/ml)$ 이상의 CDK4/Cyclin D1 호소 저해활성을 보인 것은 단삼(Salvia miltiorrhiza), 노박덩굴(Celastrus orbiculatus), 삼백초(Saurus chinensis), 목단(Paeonia suffruticosa) 및 오이풀(Sanguisorba officinalis)이었다. 이들 중에서 목단과 오이풀 추출물이 가장 높은 56%의 저해율을 각각 나타내었다. 또한 추출물에서 활성물질을 추출 정제하여 2차원 NMR 등을 포함한 각종 기기분석을 통하여 cryptotanshinone으로 동정하였다.

꿀풀 추출물의 유산균에 대한 생육과 항산화 활성 및 어류 병원성 미생물에 대한 항균활성 (Growth and Antioxidant Activity on Lactic Acid Bacteria and Antimicrobial Activity on Fish Pathogenic Bacteria By Prunella vulgaris var. aleutica Fernald Extracts)

  • 본 연구는 제주도에서 채집된 꿀풀을 꽃과 잎 부위별로 나누어 각 부위를 열수 추출하여 각 추출액을 농도별로 첨가하여 E. faecium KCCM 12118, L. rhamnosus KCCM 32826, L. plantarum KCCM 11542, P. pentosaceus KCCM 40464 균종에 대하여 생육에 미치는 영향과 그 배양액을 가지고 DPPH 라디칼 소거활성 및 Hydroxyl radical 소거활성 그리고 어류질병 미생물에 대한 항균활성을 조사하였다. 꿀풀의 꽃과 잎 추출액을 농도별로 첨가하여 4종의 lactic acid bacteria를 72시간 배양하면서 생육활성을 보았을 때 10%를 첨가하여 48시간 배양 하였을 때가 가장 좋은 생육조건임을 알 수가 있었으며, 또한 lactic acid bacteria 생육 시 꿀풀 꽃과 잎 추출액 첨가가 lactic acid bacteria 증식에 효과가 있음을 나타내었다. DPPH 라디칼 소거활성에서는 꿀풀 꽃 추출액을 10% 첨가하여 배양하였을 때 합성 항산화제인 BHA(90%)와 BHT(81%)의 EDA와 비교하였을 때 BHA와 유사하거나 조금 높은 활성을 BHT보다는 높은 라디칼 소거활성을 나타내었다. Hydroxyl radical 소거활성은 5% 꿀풀 각 부위의 추출액을 첨가한 배양액에서부터 BHT보다 높은 소거활성을 나타내기 시작하여 10%를 첨가한 배양액에서는 BHA보다 높은 소거 활성을 나타내었다.

Phenolic constituents and biological activities of leaf extracts of traditional medicinal plant Plectranthus amboinicus Benth (Lamiaceae)

  • Plectranthus amboinicus Benth (Lamiaceae) is a medicinal plant native to India, and its leaves are widely used in several traditional medicinal preparations. The purpose of this study was to detect and quantify phenolics present in ethyl acetate and acetone extracts of P. amboinicus leaves, and evaluate their antioxidant, antibacterial, antimutagenic and anticancer activities. The HPLC chromatograms of crude leaf extracts indicated the presence of phenolics like caffeic acid, coumaric acid, rutin, quercetin and gallic acid, which were present in the range of 0.01 - 1.41 mg/g in ethyl acetate and 0.03 - 1.93 mg/g in the acetone extract. The acetone extract showed statistically (p < 0.05) higher antioxidant activity ($IC_{50}$, 99.59 ${\mu}g/ml$) than ethyl acetate extract ($IC_{50}$, 149.96 ${\mu}g/ml$). Statistically (p < 0.05) higher antimutagenicity was shown by acetone extract (46.16%) as compare to ethyl acetate extract (12.16%) at 500 ${\mu}g/plate$ concentration. The acetone extract showed higher antibacterial activity than ethyl acetate extract, and both the extracts showed highest activity against B. cereus (375 and 625 ${\mu}g/ml$, respectively) and lowest activity against Y. enterocolitica (1000 and 1125 ${\mu}g/ml$, respectively). Both the extracts also showed inhibitory effect on cancer cell lines HCT-15 and MCF-7. These results suggest that the leaves of P. amboinicus possess various biological activities, and validate the traditional use of the leaves of P. amboinicus against cold, infection and ulceration.

국내 자생 동백나무(Camellia japonica L.) 추출물의 항산화 및 항미생물 활성 (Antioxidant and Antimicrobial Activities of The Extracts from Native Camellia japonica in Korea)

  • 동백나무의 잎과 꽃의 식용자원화를 위한 일환으로서 동백의 부위별 성분분석, DPPH radical 소거능에 의한 항산화활성, 및 항미생물 활성을 조사하였다. 수분 함량은 꽃과 어린 잎 부위에서 70% 이상의 함유율을 보였고, 조단백질의 함량은 꽃을 제외한 나머지 부위에서 3%이상이었으며, 지방 함량은 씨부위에서 23.08%로 많이 함유되어 있었다. 회분의 함량은 수피 부위에서 2.97%와 탄수화물의 함량은 에서 높게 나타났다. 항산화활성은 어린잎(RC$_{50}$=30.37$\mu\textrm{g}$/mL), 꽃봉오리 (RC$_{50}$=52.97$\mu\textrm{g}$/mL)와 꽃(RC$_{50}$=59.48 $\mu\textrm{g}$/mL)이 높은 활성을 보였으며 합성 항산화제인 BHT의 RC$_{50}$ 값이 584.04$\mu\textrm{g}$/mL의 활성 보다 높은 항산화 활성을 가지고 있음을 확인하였다. 동백을 각 부위별로 추출하여 병원성균과 식중독균, 식품과 관련이 있는 세균 및 효모 등 4균주에 대하여 항미생물 활성을 검토한 결과, 동백의 수피 부위가 P. vulgaris와 B. subtilis에 대한 높은 항균성을 나타내었으며, 특히, 어린잎은 C. albicans와 T. beigelii 모두에 대하여 매우 강한 활성을 나타내었다.을 나타내었다.

Screening of the Physiological Activity of Solvent Extracts of Paulownia coreana Bark and Antioxidative Effect of the Extracts on an Edible Oil

  • Paulownia coreana is a medicinal, edible and industrial plant with the largest leaf, and is native to Korea. We evaluated the reducing power activities, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities (RSAs), nitrite scavenging activities (NSAs) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation RSAs by solvent extraction of P. coreana bark by using $50^{\circ}C$ hot water and $25^{\circ}C$ methanol. The antioxidative effect of P. coreana bark extract on corn seed oil was evaluated using the Rancimat test. The extraction yields on dry weight basis with 15 folds of hot water and methanol were 23.88% and 5.30%, respectively; further the flavonoid content in the hot water extract was over 2.5 times more than that in the methanol extract. The DPPH RSA of the methanol extract was substantially higher than that of the water extract, whereas the NSA of the water extract was higher than that of the methanol extract at pH 1.2. The ABTS RSAs of the two extracts were almost the same as that of ascorbic acid and 2,6-ditertiarybutyl hydroxytoluene. The two extracts of P. coreana bark in this study were found to slightly improve the oxidation stability of corn seed oil.

카카오 열수 추출물 투여의 마우스 면역세포 활성효과 (Effect of Theobroma cacao L. Extracts on Mouse Spleen Immune Cells Activation)

  • Theobroma cacao L., a fruit of cacao trees, is a perennial plant, which belongs to Sterculiaceae, and is native to the Amazon in South Africa. It also has been known for its various biologically active effects, such as anti-oxidation, anti-cancer, and anti-bacterial. The spleen cell proliferations of mice were measured at 48 hours after treatment of Theobroma cacao L. water extracts in seven concentrations(0, 5, 10, 50, 100, 250, 500 and 1,000 ㎍/mL) an ELISA assay. The production of cytokine (IL-1β, TNF-α, IFN-γ), is secreted by macrophages stimulated with LPS, was detected by ELISA assay using the cytokine kit. From the results of in vitro study, both splenocytes and cytokine production activated by peritoneal macrophages have increased when water extracts were supplemented in the range between 250 and 500 ㎍/mL concentration. Notably, splenocytes production has a signigicant proliferation at 500 ㎍/mL concentration. The result from this research suggests that supplementation with Theobroma cacao L. water extracts may enhance the immune function by stimulating the splenocyte proliferation and improving the cytokine production activating macrophage in vitro.

Antiproliferative Effects of Native Plants on Prostate Cancer Cells

  • As part of the research for the natural products about prostate-related disease, this study screened 159 plant species from 46 families, which included a total of 213 different kinds of local native plants and these plants were tested for the ability to inhibit LNCaP proliferation, an androgen-sensitive prostate cancer cell line, and DU145 proliferation, which is a more aggressive androgen-insensitive prostate cancer cell line. The results indicated that nineteen of 213 types of plants exhibited antiproliferative activity (cell viability < 30%, $500{\mu}g/mL$) on the growth of androgen-sensitive LNCaP cell lines, and five of them exhibited DU145 cell antiproliferative activity (cell viability < 30%, $500{\mu}g/mL$). The methanol extracts of Eurya emarginata (stems), Gleditsia japonica var. koraiensis (leaves), Photinia glabra (leaves) and Elaeagnus macrophylla (leaves) showed antiproliferative activity on both the androgen-sensitive LNCaP cells (cell viability < 30%) and androgen-insensitive DU145 cells (cell viability > 100%). The study also found that the methanol extracts of Styrax japonica (fruits), Aralia continentalis (leaves), Fagus crenata var. multinervis (stems), Thuja orientalis (stems) and Poncirus trifoliate (branches) presented the strongest activity and demonstrated potent antiproliferative activity on both cell lines (LNCaP and DU145 cell viability < 30%).
