Lactic Acid Bacterias Growth, Antioxidant Activities and Antimicrobial Activity on Fish Pathogenic Bacteria by Native Plant Extracts, Jeju Island (제주도 자생식물 추출물의 유산균에 대한 생육과 항산화 활성 및 어류 병원성 미생물에 대한 항균활성)

  • In this study was investigated the growth effect of native plant (Microlepia marginata(panzer Christ., Prunella vulgaris. aleutica Fernald, Perillafrutescens var. japonica Hara., Gleichenia japonica Spreng) extracts on various lactic acid bacterias, electron donating ability, total cell count and hydroxyl radical scavenging activity. The total cell count of Enterococcus faecium KCCM 12118, Lactobacillus rhamnosus KCCM 32826, Lactobacillus plantarum KCCM 11542, Pediococcus pentosaceus KCCM 40464 in the absence of native plant extracts (10%) at $37^{\circ}C$ after 48hr were $2.2{\times}10^9\;cfu/ml$, $2.1{\times}10^9\;cfu/ml$, $2.3{\times}10^9\;cfu/ml$, $2.2{\times}10^9\;cfu/ml$. On the other hand, the total cell count of E. faecium KCCM 12118, L. rhamnosus KCCM 32826, L. plantarum KCCM 11542, P. pentosaceus KCCM 40464 in the presence of native plant extracts (10%) at $37^{\circ}C$ after 48hr were $4.3{\times}10^9\;cfu/ml$, $4.3{\times}10^9\;cfu/ml$, $4.8{\times}10^9\;cfu/ml$, $3.9{\times}10^9\;cfu/ml$. The electron donating ability indicated to E. faecium KCCM 12118, L. rhamnosus KCCM 32826, L. plantarum KCCM 11542, P. pentosaceus KCCM 40464 added by 10% native plant extracts, respectively. when 10% native plant extracts were added lactic acid bacterias, the electron donating ability is the highest. Hydroxyl radical scavenging activity of E. faecium KCCM 12118, L. rhamnosus KCCM 32826, L. plantarum KCCM 11542, P. pentosaceus KCCM 40464 showed higher than that of control.

Effect on Wnt/β-catenin Pathway of Methanol Extracts from Native Plants in Korea (국내자생식물 메탄올추출물의 Wnt/β-catenin 신호전달체계에 대한 효과)

  • The Wnt/β-catenin signaling pathway is mandatory in adipogenesis. In this study, we investigated the applicability of functional materials for the treatment of obesity by examining Wnt/β-catenin pathway reporter activity related to adipocyte differentiation inhibiting with korean native plant extracts. The luciferase activity of HEK 293-TOP cells increased the reporter activity approximately 152% and 130% by treatment with Sanguisorba officinalis and Thuja orientalis, respectively. Ricinus communis were represented about 90% higher activity, two samples(Rosa rugosa and Sophorae Flos) showed 80% higher activity than the control. Three samples of plant extracts (Zanthoxylum piperitum, Pueraria thunbergiana, Solanum nigrum) were about 70% higher activity compared with the non-treated control. Cytotoxicity of plant extracts was not detected in the rat neural stem cells. These results suggest that the selected eight plant extracts are safe compounds. Our findings indicate that Wnt/β-catenin pathway reporter activity could be used for high throughput screening system. In addition, the plant extracts selected as candidates for adipocyte differentiation inhibiting may be potential therapeutic agents for obesity, it will be exploring the possibility of developing an anti-obesity materials through further experiments with selected plant extracts.

Neuroprotective Effects of Methanol Extracts of Jeju Native Plants on Hydrogen Peroxide-induced Cytotoxicity in SH-SY5Y Human Neuroblastoma Cells

  • Neuronal death is a common characteristic hallmark of a variety of neurodegenerative disorders including Alzheimer's disease and Parkinson's disease. However, there have been no effective drugs to successfully prevent neuronal death in those diseases, whereas oriental medicinal plants have to possess valuable therapeutic potentials to treat neurodegenerative diseases. In the present study, in an attempt to provide neuroprotective agents from natural plants, 80% methanol extracts of a wide range of medicinal plants, which are native to Jeju Island in Korea, were prepared and their protective effects on hydrogen peroxide-induced apoptotic cell death were examined. Among those tested, extracts from Smilax china and Saururus chinesis significantly decreased hydrogen peroxide-induced apoptotic cell death. The extracts attenuated hydrogen peroxide($H_2O_2$)-induced caspase-3 activation in a dose-dependent manner. Further, plant extracts restored $H_2O_2$-induced depletion of intracellular glutathione, a major endogenous antioxidant. The data suggest that Jeju native medicinal plants could potentially be used as therapeutic agents for treating or preventing neurodegenerative diseases in which oxidative stress is implicated.

Inhibitory activity of methanol extracts from different colored flowers on aldose reductase and HPLC-UV analysis of quercetin

  • The methanol extracts of flowers obtained from 67 plant species were screened for their inhibitory activity on aldose reductase (AR). Alnus japonica, Aster spathulifolius, Chionanthus retusus, Morus bombycis, Crysanthemum boreale, Persicaria tinctoria, Platycarya strobilacea, and Serratula coronata var. insularis exhibited potent aldose reductase inhibitory (ARI) activity. HPLC-UV analysis of quercetin, an AR inhibitory flavonoid, was performed on extracts showing strong ARI activity. Quercetin was detected in C. retusus, C. boreale, P. tinctoria, and S. coronata var. insularis at concentrations of 1.33, 1.56, 0.82, and $3.37mg\;g^{-1}$ extract, respectively, indicating that quercetin contributed to the ARI activity of these extracts. In the samples in which quercetin was absent, other compounds may be responsible for their potent ARI activity. These results serve as a basis for further studies regarding the bioactive components responsible for the inhibitory effects of various flower extracts on AR activity.

Effects of Methanolic Extracts from Some Native Plant Resources and Medicinal Plants on Early Seedling Growth of Rice(Oryza sativa L.) (국내 식물자원 및 약용식물 추출물이 벼 유묘 생장에 미치는 영향)

  • This study was conducted to identify plant growth promoting effects of native plant resources and medicinal plants. 65 kinds of plant extracts from 64 species were evaluated based on two rice(Oryza sativa) seedling bioassays for gibberellins and gibberellin-like substances. 12 extracts of Artemisia princeps stem and leaf, Cirsium japonicum var. maackii whole plant, Rhododendron mucronulatum branch and leaf, Phragmites communis stem, Coix lacryma-jobi var. mayuen whole plant, Lespedeza bicolor branch and leaf, Hydrangea serrata f. acuminata whole plant, Phlomis umbrosa whole plant, Glycyrrhiza uralensis Rhizome, G. uralensis stem and leaf, Angelica gigas root, and Cnidium officinale rhizome showed growth promotion of rice seedlings. Our results suggested that the parts of these plants could be the potential sources as farm-made liquid fertilizers for plant growth promotion.

Insecticidal activity of native plant extracts against Culix pipiens pallens and Musca domestica (자생식물 추출물의 모기 및 집파리에 대한 살충활성)

  • Methanol extracts of 9 kinds of native medicinal plants(Taraxacum platycarpum, leaf; Pinus densiflora, leaf; Artemisia prinseps, leaf; Allium tuberosum, leaf; Cassia obtussifolia, whole; Sophora angestifolia, root; Stemonae sessilifolia, root; Lonicera japonica stem, leaf, flower; and Clivia miniata) were investigated for insecticidal effect. Methanol extracts of Pinus densiflora leaves and Sophora angestifolia radix showed relatively good insecticidal activity against Culex pipiens pallens larvae. Strong larvicidal activity against the Musca domestica larvae was produced from methanol extracts of Taraxacum platycarpum leaves and Allium tuberosom leaves. while Stemonae radix showed moderate activity. All materials tested revealed little or weak insecticidal activity against M. domestica adults.

Repellent activity of methanol extracts of native plants against Aedes albopictus and Blateria germanica (자생식물의 메탄올 추출물이 흰줄숲모기 및 바퀴에 대한 기피효과)

  • Methanol extracts of nine native plants(Teraxacum platycerpum leaf, Pinus densiflora leaf, Artemisia prinseps, leaf, Allium tuberosum leaf, Cassia obtussifolia, whole plant, Sophora angestifolia root, Stemonae sessilifolia root, Lonicera japonica, stem, leaf, flower, and Clivia miniata leaf were tested for repellent activity against Aedes albopictus and Blateria germanica. The extracts of P. densiflora, T. platycerpum, A. tuberosum and L. japonica (flower) exhibited excellent repellent activity against A. albopictus, while only L. japonica (leaf) had some good activity against B. germanica.

Screening of Inhibition Activity of LPS-induced NO Production by Ethanol Extracts from Jeju Island Native Plants and Algae

  • Herbal medicines have been used as a basic means of clinical trial throughout history, and traditional medicines are targeted to seek functional components. To discover new cosmetic or food ingredients among numerous natural resources from Jeju island, we screened for inhibition activity against nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Although NO formation plays an important role to relax vascular muscles or eliminate tumors, NO produced excessively in inflammatory condition can cause metabolic diseases or inflammatory dysfunctions. Among 52 natural resources ethanol extracts, 5 extracts inhibited NO production over 25% compared to only LPS-treated control at the concentration of $100{\mu}g/mL$. In further study, we try to investigate other bio-activities and the phytochemicals of 5 different extracts as useful ingredients for cosmetics or functional foods.
