The optimization of ELISA for methamphetamine determination : the effect of immunogen, tracer and antibody purification method on the sensitivity

  • To obtain more sensitive immunoassay for methamphetamine (MA) determination, the optimum condition of enzyme-linked immunosorbent assay (ELISA) was investigated in regard to immunogens, antibody purification methods and coating tracers. Activated MA, N-(4-aminobutyl)methamphetamine (4-ABMA), was conjugated with bovine serum albumin (BSA) or keyhole limpet hemocyanin (KLH) and used as immunogen. The antibodies were purified by protein G chromatography or various immunoaffinity chromatography-linked MA-protein ligands, such as MA-BSA, MA-KLH or MA-ovalbumin (OVA). Each purified antibody was characterized by means of sensitivity and cross-reactivity using the three MA-protein coating tracers, MA-BSA, MA-KLH and MA-OVA. The best sensitivity of each antibody was acquired with the MA-OVA tracer although the tracer concentration and the antibody titer level at optimum condition were varied. The antibody with high titer level did not always yield good sensitivity. At optimum condition, immunoaffinity chromatography-purified antibodies were better for sensitivity and for specificity than protein G-purified antibodies. The cross-reactivity of the purified antibodies seemed to be affected by immunogen structure and showed somewhat different patterns according to the immunoaffinity ligand utilized. These data show that the antibody purification method as well as choice of coating tracer and immunogen is essential for the sensitivity and specificity of EIA; the optimum condition for assay should be discovered using various methods and combinations.

양전자단층촬영에 이용 가능한 새로운 심근 혈류 추적자 개발; F-18이 표지된 유기암모늄염의 합성과 체내분포에 관한 연구 (Synthesis and $^{18}F$ Labelling of Organic Ammonium Salts to New Cardiac Flow Tracer for PET and Their Biodistribution)

  • In order to develop a $^{18}F$-labelled myocardial perfusion agent(flow tracer) for PET, $^{18}F$-labelled organic ammonium cations were synthesized and evaluated in relation to their biodistribution. Five quaternary organic ammonium compounds were labelled with $^{18}F$ in a side chain with moderate to good yields by direct introduction of $^{18}F$-fluoride. Radiochemical yields have been achieved in 30-40min by the precursors (tosylates) in dimethylsulfoxide 15-60% (decay corrected). The reaction was found to be autocatalyzed. A remote controlled procedure was developed in these synthesis. $^{18}F$-Labelling and HPLC-purification of com-pounds needed about 60 min(Yield; 7-20%). Up to now the two compounds N-4-[$^{18}F$]fluorobutyl-pyridinium cation(1) and N, N dibenzyl-4(2-[$^{18}F$]fluoroethyl)piperidinium cation(2) were investigated in relation to their biodistribution in mice. Compound 1 showed at 1 min post injection the high uptake of 19.22% ID/g organ in the myocardium but a following fast decline to 1.12% ID/g organ after 40min. Uptake of compound 2 was after 1min in the heart 5.90% ID/g organ but after 40min at the relative high value of 4.33% ID/g organ. Heart:blood ratio for compound(1) at 1 min was 8.3, at 40 min 2.6 for compound II 2.0(1min) and 15.0(40 min). As data of compound 2 showed greater heart uptake, slower myocardial release, and higher heart: blood ratios, compound 2 is a good candidate for further evaluation.

  • Nitrification and denitrification are important processes in the landfill site as they are deeply related with degradation and stabilization of refuse. Also nitrous oxide ($N_2O$) which is released from both nitrification and denitrification is known as greenhouse gas (GHG). The purpose of this study was to clarify the process by which $N_2O$ produced using $^{15}N$ isotope. Nitrate which was labeled to 10.08% with $^{15}KNO_3$ was used and $N_2O$ was analyzed with GC mass. Results was that even also when $O_2$ of bulk was 15%, $N_2O$ was released from denitrification. And as concentrations of $O_2$ increase, sum of $N_2O$ was released from denitrification. And as concentrations of $O_2$ increase, sum of $N_2O$ and $N_2$ was decreased and ratios of $N_2O$ in the reduced gases were increased. FISH technics also adaped to confirm whether which of nitrifiers existed in the substrates. When NEU was used of which the target was ammonia oxidizing bacteria, nitrifier was not detected at all. So it was confirmed that during the reaction denitrification was dominant process. Total bacteria distributions which were detected by EUB probe explained that as $O_2$ increase the number of bacteria also increase, but between the 10-15% of $O_2$ there was no any differences.

Indoor Air Quality of Acidic Air Pollutants at a Private House in Seoul During the Spring Months

  • Acidic air pollutants were collected to characterize indoor air quality at a private house in Seoul during the spring period. All indoor and outdoor samples were measured simultaneously using an annular denuder system. The data set was collected on twelve different days with a 24-hr sampling period in April and May 1997. The chemical species measured were HN $O_3$, HN $O_2$, S $O_2$ and N $H_3$in the gas phase and P $M_{2.5}$ (dp 2.5 ${\mu}{\textrm}{m}$), S $O_4$$^{2-}$, N $O_3$$^{[-10]}$ and N $H_4$$^{+}$ in the particulate phase. Indoor concentrations of HN $O_2$, N $H_3$, and P $M_{2.5}$ were greater than outdoor levels. However, indoor concentrations of HN $O_3$, S $O_2$, N $O_3$$^{[-10]}$ and N $H_4$$^{+}$ were less than those found from outdoors. In the case of S $O_4$$^{2-}$, the indoor and outdoor concentrations were similar. Indoor concentrations of P $M_{2.5}$ , S $O_4$$^{2-}$ and N $O_3$$^{[-10]}$ were dependent upon the outdoor concentrations. A tracer-gas decay technique with sulfur hexafluoride (S $F_{6}$ ) as the tracer gas was used to estimate the air exchange rate of a private home in the spring. The average air exchange rate was computed to be 2.87 h $r^{-1}$ .X> .

토양 내 PCE 제거과정에서 가스 분배추적자기법을 이용한 공기노출 PCE의 잔류량 검출 (Measurement of Gas-Accessible PCE Saturation in Unsaturated Soil using Gas Tracers during the Removal of PCE)

  • In this laboratory study, the changes in gas-exposed perchloroethene (PCE) saturation in sand during a PCE removal process were measured using gaseous tracers. The flux of fresh air through a glass column packed with PCEcontaminated, partially water-saturated sand drove the removal of PCE from the column. During the removal of PCE, methane, n-pentane, difluoromethane and chloroform were used as the non-reactive, PCE-partitioning, water-partitioning, and PCE and water-partitioning tracers, respectively. N-pentane was used to detect the PCE fraction exposed to the mobile gas. At water saturation of 0.11, only 65% of the PCE was found to be exposed to the mobile gas prior to the removal of PCE, as calculated from the n-pentane retardation factor. More PCE than that detected by n-pentane was depleted from the column due to volatilization through the aqueous phase. However, the ratio of gas-exposed to total PCE decreased on the removal of PCE, implying gas-exposed PCE was preferentially removed by vaporization. These results suggest that the water-insoluble, PCE-partitioning tracer (n-pentane in this study), along with other tracers, can be used to investigate the changes in fluid (including nonaqueous phase liquid) saturation and the removal mechanism during the remediation process.

FUNS - Filaments, the Universal Nursery of Stars. I. Physical Properties of Filaments and Dense Cores in L1478

  • Formation of filaments and subsequent dense cores in ISM is one of the essential questions to address in star formation. To investigate this scenario in detail, we recently started a molecular line survey namely 'Filaments, the Universal Nursery of Stars (FUNS)' toward nearby filamentary clouds in Gould Belt using TRAO 14m single dish telescope equipped with a 16 multi-beam array. In the present work, we report the first look results of kinematics of a low mass star forming region L1478 of California molecular cloud. This region is found to be consisting of long filaments with a hub-filament structure. We performed On-The-Fly mapping observations covering ~1.1 square degree area of this region using C18O(1-0) as a low density tracer and 0.13 square degree area using N2H+(1-0) as a high density tracer, respectively. CS (2-1) and SO (32-21) were also used simultaneously to map ~290 square arcminute area of this region. We identified 10 filaments applying Dendrogram technique to C18O data-cube and 13 dense cores using FellWalker and N2H+ data set. Basic physical properties of filaments such as mass, length, width, velocity field, and velocity dispersion are derived. It is found that filaments in L~1478 are velocity coherent and supercritical. Especially the filaments which are highly supercritical are found to have dense cores detected in N2H+. Non-thermal velocity dispersions derived from C18O and N2H+ suggest that most of the dense cores are subsonic or transonic while the surrounding filaments are transonic or supersonic. We concluded that filaments in L~1478 are gravitationally unstable which might collapse to form dense cores and stars. We also suggest that formation mechanism can be different in individual filament depending on its morphology and environment.

새만금호 3차원 수리.수질모델(EFDC)의 수치격자 민감도 분석 (A Sensitivity Analysis on Numerical Grid Size of a Three-Dimensional Hydrodynamic and Water Quality Model (EFDC) for the Saemangeum Reservoir)

  • Multi-dimensional hydrodynamic and water quality models are widely used to simulate the physical and biogeochemical processes in the surface water systems such as reservoirs and estuaries. Most of the models have adopted the Eulerian grid modeling framework, mainly because it can reasonably simulate physical dynamics and chemical species concentrations throughout the entire model domain. Determining the optimum grid cell size is important when using the Eulerian grid-based three-dimensional water quality models because the characteristics of species are assumed uniform in each of the grid cells and chemical species are represented by concentration (mass per volume). The objective of this study was to examine the effect of grid-size of a three dimensional hydrodynamic and water quality model (EFDC) on hydrodynamics and mass transport in the Saemangeum Reservoir. Three grid resolutions, respectively representing coarse (CG), medium (MG), and fine (FG) grid cell sizes, were used for a sensitivity analysis. The simulation results of numerical tracer showed that the grid resolution affects on the flow path, mass transport, and mixing zone of upstream inflow, and results in a bias of temporal and spatial distribution of the tracer. With the CG, in particular, the model overestimates diffusion in the mixing zone, and fails to identify the gradient of concentrations between the inflow and the ambient water.

감쇠탱크 설계를 위한 체류시간 계산 방법에 관한 연구 (A Study of Residence Time Calculation Methods in Decay Tank Design)

  • In this study we apply and compare a variety of numerical methods for calculating residence time distribution in decay tanks, a major design component in the for reducing N-16 radioactivity. Our research group has used a streamlined method using user-defined particle numbers. However, this streamlined method has several problems, including low exiting particle ratios, particle diminishing, and unphysical time distribution, among others. We utilize three numerical methods to establish residence time and time distribution (streamlined, discrete phase method [DPM], and user defined scalar [UDS]) and subsequently compare the averaged results of each. The three tests demonstrate the flow features within the decay tanks, which are then numerically simulated to enable comparison. We conclude that although each simulation predicts similar time averages, the UDS methodology provides a smoother time distribution and tracer contour plots at specific times.

화강암 기원 잔적토양에서 염료추적자의 침투 유형에 관한 연구 (A Dye Tracer Study of Infiltration Pattern in a Residual Soil Developed from Granite)

  • 지표 및 심토환경에서 물과 용액의 유동특성을 파악하는 것은 토양 및 지하수의 오염을 저감하고 방지하거나 기 오염 지역을 조사하고 정화하는데 있어서 필수적인 요소이다. 본 연구에서는 이 비형광, 비독성의 식용색소인 청색1호(Brilliant Blue FCF, $C_{37}H_{34}N_{2}Na_{2}O_{9}S_{3}$)를 염료 추적자로 이용하여 화강암에서 발달된 잔적토양에서의 침투 유형 및 그 특성을 조사하였다. 디지털 촬영된 수직 및 수평 착색영상은 지형정보 영상처리 프로그램인 ER Mapper, Version6.2를 이용하여 염료착색범위(dye coverage)로 전환되었다. 균질기반유동(homogeneous matrix flow)은 미약 입상 구조를 보이는 A 층위에서 발생하였으며, 상대적으로 세립조직인 A층위와 조립조직인 C층위의 경계에서는 선침투류(先浸透流, fingering)의 특징을 보여주었다. 모암인 화강암 조직의 기원인 거정질 광맥(pegmatitic vein)과 식물 뿌리는 C 층위에서 선택적 유동(preferential flow)을 유도하였음을 확인하였다.

Investigation of the gas Dynamics in an Upflow OMVPE Reactor by Raman Spectroscopy

  • The gas dynamics in a stagnation point upflow OMVPE reactor were studied by Raman spectroscopy. The gas temperature was measured as a function of inlet gas velocity and aspect ratio for both H$_2$ and N$_2$ carrier gases. The centerline temperature gradient was latger at higher inlet velocities and with the use of N$_2$, and only weakly dependent on the aspect ratio. a tracer molecule, CH$_4$, was used to investigate the steady state behavior of reactants in the reactor, and the use of a sweeping flow was found to be a suitable method for preventing wall deposition. The transient switching response of the gas manifold was also investigated. Under certain conditions (low velocities, unmatched flows) recirculation flows were apparent. Numerical calculations of the reactor gas dynamics gave reasonable agreement with experimental results when detailed thermal boundary conditions were included.
