Morphological Characteristics of Granular Sludge in dPAO SBR (dPAO SBR 입상슬러지의 형태학적 특성)

  • The morphological characteristics of granules, which were generated in lab-scale sequencing batch reactor (SBR) for simultaneous nitrogen and phosphorus removal with denitrifying phosphorus accumulating organism (dPAO) were identified. Granular sludge was fully developed in the anaerobic-anoxic (An-Ax) SBR after 180 days of SBR operation. The average diameter of granular sludge was 2.2 mm and rod-type organisms dominated in the granules. In addition, about 1.0 mm of white precipitate was observed in the core of the granule, and the material was confirmed that it is very similar to hydroxyapatite $(HAP;\;Ca_5(PO_4)_3(OH))$ by X-ray diffraction) analysis.

  • For the purpose of explaining the mechanism of tooth discoloration caused by tetracycline (TC), a study was conducted by setting up the following in vitro system.   • PLLA grafted Hydroxyapatite / polylactide (g-HA/PLA) composites were prepared by three grafting methods. The modified particles (p-HA) were dispersed more uniformly in the PLLA matrix than pure n-HA. The p-HA/PLLA composites exhibited better mechanical properties and thermal stability than the n-HA/PLLA composites. The composites also demonstrated improved cell compatibility due to the good biocompatibility of the HAP nanoparticles and the more uniform distribution of the PLLA-grafted HAP nanoparticles on the film surface. All of these results indicated that the p-HAP/PLLA nano-composites might have a promising medical application in bone repair and in bone tissue-engineering.

  • This study investigated on the adsorption of nonpoint pollution source using the Sand, hydroxyapatite(HAP), Zeolite and mixed culture. The adsorption of nonpoint pollution source on Sand, hydroxyapatite(HAP), Zeolite and mixed culture was investigated during a series of batch adsorption experiments. After the batch absorption experiments analysed COD, T-N, T-P on adsorption water. The experimental data was analysed using the pseudo-first-order adsorption kinetic models. Langmuir and Freundlich isotherm models were tested for their applicability. The maximum adsorbed amount $(Q_{max})$ of COD were found to be sand 0.0511mg/g, HAP 0.1905mg/g, Zeolite 1.0366mg/g and Mixed media 0.7444mg/g. The maximum adsorbed amount $(Q_{max})$ of T-N were found to be sand 0.0159mg/g, HAP 0.0537mg/g, Zeolite 0.5496mg/g and Mixed media 0.1374mg/g. The maximum adsorbed amount $(Q_{max})$ of T-P were found to be sand 0.0202mg/g, HAP 0.1342mg/g, Zeolite 0.0462mg/g and Mixed media 0.1180mg/g. As a result, the mixed media was effective to remove nonpoint pollution source.

The Identification of Type II DNA Topoisomerase-Associated Protein Kinase Activity from Regenerating Rat Liver (재생 쥐간에서 분리한 DNA topoisomerase II에 결합된 protein kinase 활성)

  • We have found a protein kinase activity that is tightly associated with type II DNA topoisomerase purified from regenerating rat liver. The activities of protein kinase and topoisomerase II were not separable when the enzyme was subjected to analytical chromatographies (Hydroxyapatite, phosphocellulose, and double strand DNA cellulose) and glycerol gradient sedimentation. The kinase activity from purified rat topoisomerase II was also inactivated by the topoisomerase II inhibitors such as N-ethylmaleimide or novobiocin. The evidences, however, do not rule out a possibility that the kinase activity resides in a polypeptide other than the topoisomerase II protein. The topoisomerase II-associated protein kinase required Mg++ for its activity, and this requirement was not substituted by any other mono- or divalent ions. Histone H1 act as a strong stimulator and a good substrate for the kinase activity and other histones and ${\alpha}$-casein could not substitute the effect of histone H1.

PQQ-Dependent Organic Acid Production and Effect on Common Bean Growth by Rhizobium tropici CIAT 899

  • Rhizobium tropici CIAT 899 is capable of synthesizing inactive apo-glucose dehydrogenase (GDH). To become an active holo enzyme, the GDH requires a cofactor, PQQ. When R. tropici CIAT 899 was grown in a broth culture medium containing hydroxyapatite and pyrrolo quinoline quinone (PQQ), pH decreased while the concentration of soluble P increased. The solubilization of hydroxyapatite was associated with the production of gluconic acid and 2-ketogluconic acids. The organic acid production and P solubilization were greatly enhanced when the bacterium was grown with air supply. Effect of R. tropici CIAT 899 with (CI+PQQ) and without PQQ (CI) on the common bean growth was examined. Shoot and root weight, and N and P contents in CI+PQQ treatment, were significantly higher than those in control and CI treatment. Nodule weight and acetylene reducing activities were also significantly higher in CI+PQQ treatment than in other treatments.

Recovery of high quality external carbon sources using crystallization from pretreated excess activated sludge by alkali and ozone (알칼리-오존 동시 전처리된 잉여슬러지로부터 결정화를 이용한 고품질 외부탄 소원 회수)

  • In this research, recovery of high quality organics from excess activated sludge and its potential as a external carbon sources for BNR process was studied. By simultaneous treatment of alkali and ozone, TSS concentration was reduced by 32%, and RBDCOD fraction was increased by 76.2%, and major constitute of produced organic were acetic acid and propionic acid. Also, nitrogen and phosphorus were greatly solubilized. However, because acid-hydrolyzable phosphorus(AHP) was major part of solubilized phosphorus, $NH_4{^+}-N$ and $PO_4{^3}-P$ concentration were insufficient for effective formation of crystal like as MAP(Magnesium Ammonium Phosphate) and hydroxyapatite. By placing BPR reactor before alkali-ozone treatment reactor, $PO_4{^3}-P$ concentration in pretreated sludge was increased by 1.8 times, and improved potential of phosphorus recovery by crystallization. In experiment of crystallization, hydroxyapatite formation was more easily applied than MAP. By hydroxyapatite formation, $SCOD/PO_4-P$ ratio was greatly increased from 32.7 at control to 141.9 at $Ca^{2+}/PO{_4}^{3-}-P$ mole ratio of 2.4. The results based on this study indicated that the proposed system configuration has potential to reduce the excess sludge production, to recover phosphorus in usable forms as well as utilize organics as a external carbon source in BNR process.

Effect of three nanobiomaterials on microhardness of bleached enamel

  • Objectives: The aim of this in vitro study was to evaluate the effect of incorporating three different nanobiomaterials into bleaching material on microhardness of bleached enamel. Materials and Methods: The crowns of 24 extracted sound human molars were sectioned. Sixty enamel specimens ($2{\times}3{\times}4 mm$) were selected and divided into five groups (n = 12): Group 1 received no bleaching procedure (control); Group 2 underwent bleaching with a 40% hydrogen peroxide (HP) gel; Groups 3, 4, and 5 were bleached with a 40% HP gel modified by incorporation of bioactive glass (BAG), amorphous calcium phosphate (ACP) and hydroxyapatite (HA), respectively. The enamel microhardness was evaluated. The differences in Knoop microhardness data of each group were analyzed by one-way ANOVA, followed by post hoc Tukey tests. Results: Significant differences were observed between the study groups. The enamel microhardness changes in Groups 1, 3, 4, and 5 were significantly lower than that of Group 2 (p < 0.001). Conclusions: Within the limitations of this study, it can be concluded that incorporation of each one of the three tested biomaterials as remineralizing agents might be effective in decreasing enamel microhardness changes subsequent to in-office bleaching.

Dentinal Tubules Occluding Effect Using Nonthermal Atmospheric Plasma

  • Nonthermal atmospheric plasma has been studied for its many biomedical effects, such as tooth bleaching, wound healing, and coagulation. In this study, the effects of dentinal tubules occlusion were investigated using fluoride-carboxymethyl cellulose (F-CMC) gel, nano-sized hydroxyapatite (n-HA), and nonthermal atmospheric plasma. Human dentin specimens were divided to 5 groups (group C, HA, HAF, HAP, and HAFP). Group HA was treated with n-HA, group HAF was treated with n-HA after a F-CMC gel application, group HAP was treated with n-HA after a plasma treatment and group HAFP was treated with n-HA after a plasma and F-CMC gel treatment. The occlusion of dentinal tubules was investigated using scanning electron microscopy (SEM) and energy dispersive x-ray spectroscopy (EDS), which shows Ca/P ratio. In the EDS results, a higher Ca/P ratio was shown in the groups including n-HA than in the control group. The specimens of group HAP and HAFP had a higher Ca/P ratio in retentivity. In the SEM results, there was not a significant difference in the amount of times applied. Therefore, this study suggests F-CMC gel and n-HA treatment using nonthermal atmospheric plasma will be a new treatment method for decreasing hypersensitivity.
