Development of a Novel Yeast Strain Which Ferments Soy Sauce by Protoplast Fusion

  • In order to develop a novel yeast which produces the charateristic aroma of soy sauce, a protoplast fusion between Zygosaccharomyces rouxii WFS4 and Torulopsis versatilis IAM 4993 was carried out. Auxotrophic mutants as selective markers were obtained from Zygosaccharomyces rouxii and Torulopsis versatilis by treatment of N-methyl-N -nitro-N-nitrosoguanidine. The conditions of the protoplast formation and the regeneration for fusion were examined. The protoplast fusion using polyethylene glycol 4000 led to the fusion frequency of $4~5{\times}10^{-7}\;cells/ml$. Among fusants, a fusant ST723-F31 presented the best results in terms of the aromaticity of fragrance, the growth pattern, the resistance against salt and the degree of growth according to pH. It makes easy to control the production and the balance of aroma components so that it gives a good flavor, shortens the fermentation period and, simplifies the preparation process when using a bioreactor into which fusant is immobilized.

  • 생물 계면활성제의 개발을 위해 MNNG(N-Methyl-N-Nitro- NitrMoguanidjne), EMS, UV radiator 등 random mutation을 유도하여 가장 고기능 유화제 생산 균주를 선별하여 다양한 탄소원에 따른 생육도와 유화활성, 유화 활성에 미치는 pH 등 생물 계면활성제의 생산에 관한 조사를 한 결과 Bunker A에서는 유화활성이 원래 균주보다 최고 약 6배까지 증가했으며 표면장력 또한 40.3dyne/cm 에서 34.0dyne/cm 으로 크게 감소되었다.

  • 대두 경남 장려품종인 장백, 광교, 강림 및 남천등의 근류로부터 Rhizobium japonicum을 분리하여 그 중 질소고정력이 R. japonicum 61A 76이나 3 I1110보다 강한 JB 101을 선별하였다. 선별된 JB 101을 N-methy1-N'-nitro-N-nitrosoguanidine 처리 및 UV조사에 많은 돌연변이체를 얻고 이들로 부터 경남 장려품종인 장백에 접종시근류 생성력 및 질소고정력등 JB 101에 비하여 훨씬 높을 뿐만아니라 SM 35에 비해서도 다소 높은 JB 65를 선별하여 이 균주의 근류생성력, 질소고정력, 고정된 암모니아가 질소고정에 미치는 영향, hydrogenase 활성등 공생관련 제 특성과 아울러 이 균주 단백질의 아미노산 조성, 2D-polyacrylamide gel을 이용한 전기영동등의 생화학적 특성도 아울러 조사하였다.

Comet Assay를 이용한 전통발효식품인 배추김치의 항유전 독성효과 (The Antigenotoxic Effects of Korean Native Fermented Food, Baechu Kimchi Using Comet Assay)

  • This study carried out to eluciate the cancer chemoprevention of Korean native fermented food, baechu kimchi using Comet assay (in other words, single cell microgel electrophoresis). For this purpose, baechu kimchi was fractionated by water, n-hexane, chloroform and ethyl acetate. 5 strains of dominant fermented bacteria were isolated from baechu kimchi. The water fraction, n-hexane fraction, chloroform fraction, ethyl acetate fraction and water insoluble fractions showed no antigenotoxicitie in non-tumoral normal 3T3 cells. Among 5 bacteria isolates from baechu kimchi, two isolates bacteria 1 and 2 strongly inhibited genotoxicity of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in non-tumoral normal 3T3 cells (p<0.05). Bacteria 3, 4 and 5 were also not antigenotoxic.

  • The regulation of DNA repair genes expression was investigated using fused genes, in which the promoter of repair genes was hybridized with the lacZ structural gene. The activities of beta-galactosidase expressed from the fused gense were highly increased when the host cells were exposed to methylating agents, such as methyl methansulfonate (MMS), N-methyl-N'-nitro-nitrosoguanidine (MNNG) and methyl nitrosourea (MNU). On the other hand, the enzyme activities from the fused genes were not induced when the cells were treated with ethylating or nonalkylating agents, such as ethyl methansulfonate (EMS), 4-nitroquinoline-1-oxide (4NQO), Bleomycin, and Benzo(a)pyrene (BP).

녹황색 채소류의 돌연변이유발 억제 및 AZ-521 위암세포의 성장 저해효과 (Inhibitory Effect of Green-Yellow Vegetables on the Mutagenicity in Salmonella Assay System and on the Growth of AZ-521 Human Gastric Cancer Cells)

  • N-methyl-N'-nitro-N-nitrosoguanid me (MNNG)와 N-nitrosodimethylamine (NDMA)의 돌연변이 유발성 및 AZ-52l 사람의 위암세포의 성장에 미치는 녹황색 채소류의 저해 효과를 검토하였다. 26종의 시험 채소중 24종(92%)이 MNNG에 대한 항돌연변이성이 있었으며 (p<0.01, 0.05) 80%이상 저해 효과를 보인 시료는 들깻잎, 배추, 꽃양배추, 상치, 갓, 미나리, 들미나리, 당근, 우엉이었다. 한편 NDMA에 대해서도 실험채소중 85%가 항돌연변이성을 나타내었는데 (p<0.01, 0.05) 들깻잎, 배추, 케일, 콩나물, 양파는 80%이상의 저해 효과를 나타내었다. 위암세포의 성장 저해 효과에서는 들미나리와 들깻잎이 가장 저해 효과가 컸으며(97~100%) 콩나물과 미나리가 다음으로 효과가 컸었다(p< 0.001). 또한 브로콜리, 쑥갓, 풋고추, 고추잎, 시금치, 양배추, 고구마도 통계적인 유의성을 나타내며 암세포 성장을 억제하는 효과가 있었다(p< 0.01, 0.05).

Antimutagenic Activity of the Methanol Extract and Compounds of Angelica keiskei in the Salmonella Assay System

  • The methanol extract of aerial part of Angelica keiskei Koidzumi exhibited a strong antimutagenic activity against aflatoxin $B_1\;(AFB_1)$. N-methyl-N'-nitro-N-nitrosoguanidine and 4-nitroquinoline-1-oxide in the Ames test with Salmonella typhimurium TA100. Cynaroside, isolated front ethylacetate fraction of the methanol extract Over silica gel, inhibited the mutagenicity of $AFB_1$ with an inhibition value of 96% at 1.0 mg/plate concentration and 87% at 0.5 mg/plate concentration. Other compounds, hyperoside, sucrose and luteolin-7-rutinoside, isolated from ethylactate or n-butanol fraction, also showed antimutagenic effect.

  • 전보에서 분리 확보한 lysine 생산균주 Corynebacterium sp. S-27-12와 Brevibacterium flavum ATCC 15168 및 Micrococcus glutamicus ATCC 13032를 친주로 하여 자외선, $Co^{60}$및 N-methyl-N'-nitro-N-nitrosoguanidine treatment를 처리, 90여주의 변이주를 분리하고 이들의 lysine 생산능을 비교하여 lysine 생산이 8mg/ml 이상인 균주 6주를 선정하였다. 이들 중 leucine 요구주 Brev. flavum U46-N59의 lysine 축적조건을 검토한 결과를 종합하여 포도당 100, urea 10, CSL 40, $KH_2PO_4\;2;\;K_2HPO_4,\;0.5;\;MgSO_4.\;7H_2O,\;0.4;\;antifoam\;S-57,\;1g;\;Fe_2(SO_4)_3.XH-2O,\;10;\;MnCl_2,\;4H_2O,\;10mg;\;biotin,\;30;\;thiamine-HCl,\;100{\mu}g;$ 증류수 1 l로 된 pH7.5의 합성배지에 Brev. flavum U46-N59를 $28^{\circ}C$에서 4일간 플라스크(20ml/500ml)에 진탕배양했을때(180진탕/분) 최대로 21.6mg/ml의 lysine을 생산하였다. Penicilin은 40U/ml의 농도가 되도록 배양 36시간 후에 첨가하였다.

발효콩 추출물의 항돌연변이원성 및 세포독성 효과 (Antimutagenic and Cytotoxicity Effects of Fermented Soybean Extract)

  • This study was carried out to determine the antimutagenic and anticancer effects of fermented soybean using Ames test and cytotoxicity, respectively. The ethyl acetate fraction (200 g/plate) of fermented soybean in the Salmonella typhimurium TA100 strain showed 86.6% of inhibition rate against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG). In addition, the suppression of ethyl acetate fraction with same concentration of fermented soybean in the Salmonella typhimurium TA98 and TAI00 strains showed 82.4% and 90.8% inhibition against 3-amino-l,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-l), respectively. The cytotoxicity effects of fermented soybean against the cell lines with human lung carcinoma (A549), human gastric carcinoma (AGS) and human breast adenocarcinoma (MCF-7) were inhibited with the increase of the extract concentration. The treatment of 1.0 mg/mL ethyl acetate fraction of fermented soybean showed strong cytotoxicities of 71.6%, 91.5% and 80.7% against A549, AGS and MCF-7, respectively.

어성초즙 및 추출물의 항돌연변이 효과 (Antimutagenic Effects of the Juice and Boiling Water Extract of houttuynia cordata Thunb)

  • The inhibitory effects of the juice and boiling water extract of Houttuyina cordata Thunb., which has been known as a traditional folk antitumor agent, on the genotxicity induced by mutagens were investigated . For this purpose, Salmonella typhimurium TA98 and TA100, and the transheterozygous (mwh/+) third larvae of Drosophila melanogaster were used. The juice of fresh H. cordata 9JHC) and boiling H. cordata extract (BHC) showed antimutagenicities toared alfatoxin B₁ in S. typhimurium TA 98 and TA100. The JHC's inhibitory effect increased by 76% to 98% as the adding concentrations increased, and the BHC had a 86-97% inhibitory effect. however, in the case of N-methyl-N'-nitro-N nitrosoguanidine (MNNG), the JHC's inhibitory effect was 52-60% irrespectively of its concentrations and the BHC had a low inhibitory effect. The JHC and BHC slightly reduced the somatic chromosomal mutagenicity by MNNG in the wing hair spot test system (mwh/+) of D. melanogaster, which suggests that they can inhibit gene mutation , deletion and mitotic chromosomal recomgination.
