Comparison of the fecal microbiota with high- and low performance race horses

  • Exercise plays an important role in regulating energy homeostasis, which affects the diversity of the intestinal microbial community in humans and animals. To the best of the authors' knowledge, few studies have reported the associations between horse gut microbiota along with their predicted metabolic activities and the athletic ability of Jeju horses and Thoroughbreds living in Korea. This study was conducted to investigate the association between the gut microbiota and athletic performance in horses. This study sequenced the V3 and V4 hypervariable regions of the partial 16S rRNA genes obtained from racehorse fecal samples and compared the fecal microbiota between high- and low-performance Jeju horses and Thoroughbreds. Forty-nine fecal samples were divided into four groups: high-performance Jeju horses (HJ, n = 13), low-performance Jeju horses (LJ, n = 17), high-performance Thoroughbreds (HT, n = 9), and low-performance Thoroughbreds (LT, n = 10). The high-performance horse groups had a higher diversity of the bacterial community than the low-performance horse groups. Two common functional metabolic activities of the hindgut microbiota (i.e., tryptophan and succinate syntheses) were observed between the low-performance horse groups, indicating dysbiosis of gut microbiota and fatigue from exercise. On the other hand, high-performance horse groups showed enriched production of polyamines, butyrate, and vitamin K. The racing performance may be associated with the composition of the intestinal microbiota of Jeju horses and Thoroughbreds in Korea.

GC-MS 를 이용한 전통민속소주의 향기성분 분석과 다변량통계해석 (Determination and Multivariate Analysis of Flavour Components in the Korean Folk Sojues Using GC-MS)

  • 우리나라 전통민속소주를 비롯한 중국산 카오리양츄, 스카치위스키, 일본산 소주틀 polydivinyl benzene으로 고체상추출 농축한후 GC와 GC-MS로 향기성분을 분석 하고 다변량통계해석을 적용하였다. 우리나라 민속소주에는 끊는점이 비교적 높은 ethyl succinate, ethyl pelargonate 와 같은 에스테르 향기성분함량이 높아 뒷맛을 느끼게 하는데 비하여 카오리양츄는 끊는점이 낮아 휘발성이 강한 ethyl acetate, ethyl butyrate등의 함량이 많아 처음맛이 강렬하게 느껴진다고 생각된다. 민속소주에는 furfural의 함량이 비교적 많았으며 전통민속소주에는 isopentyl acohol의 함량이 isobutyl acohol 또는 n-propyl alcohol 의 함량보다 상대적으로 많은데 비하여 카오리양츄는 n-propyl alcohol함량이 높온 차이를 보였다. 다변량통계해석학의 한 기범인 주성분분석을 적용한결과 민속소주는 제1주성분점수가 비슷하였으나 제2주성분점수는 편차를 보였으므로 전통민속소주로서외 공통점이 있으면서도 각 시료별로는 고유한 특성이 인정되었다. 주성분분석과 판별분석을 통하여 중류주 상호간의 동질성과 이질성을 파악하므로서 분류동정 뿐만아니라 품질평가나 개선에 매우 유익한 객관적 정보를 도출해낼 수 있었다.

식물성유 첨가가 면양의 반추의 발효특성과 소화율 및 지방산염 형성에 미치는 영향 (Effects of Supplemental Vegetable Oils on Ruminal Characteristics, Digestibility and Formation of Fatty Acid Soaps in Sheep)

  • 본 시험은 oil 첨가시 알팔파건초를 기질로 하여 반추위 내에서 유리된 지방산과 양이온이 결합하여 형성되는 지방산염, 반추위 발효성상 및 소화율에 미치는 영향에 대하여 살펴보고자 시험을 실시하였다. 면양 3두를 이용하여 $\circled1$ oil 무첨기구, $\circled2$ 대두유 7% 첨기구 및 $\circled3$ 옥수수유 7% 첨가구로 하여 3${\times}$3 라틴방각법으로 수행하였다. 면양에게는 알팔파건초와 농후사료를 70 : 30의 비율로 급여하였다. 위액은 오전사료 급여 전 1시간, 사료 급여 후 1, 3, 6 및 9시간째 채취하였다. pH는 사료급여 후 3시간까지 감소하다가 그 이후에는 증가하였다{P<0.0001). $NH_3$-N는 pH 와는 반대의 경향을 나타내었다(P<0.0001). 총 휘발성 지방산, acetate, propionate 및 butyrate의 농도에는 변화가 없었다. A/P 비율은 배양시간이 증가함에 따라 감소하다가 9시간에서는 사료급여 1시간 수준이었다{P<0.0001). 건물, total-N, 조섬유, 회분, 기용무질소물, NDF 및 ADF의 소화율은 차이가 없었다. FAS의 형성은 사료급여 1시간에 비하여 사료급여 후에 증가하였으며(P<0.05), 대조구에 비해 oil 첨가구가 높았다(P<0.05). 장쇄지방산이 주로 지방산염을 형성하였다. 그래서 oil 첨가가 반추위 발효특생과 소화율에 미치는 유해한 영향을 방지할 수 있었던 것으로 사료 된다.

Application of lactic acid bacteria producing antifungal substance and carboxylesterase on whole crop rice silage with different dry matter

  • Objective: This study was conducted to investigate effects of antifungal substance and carboxylesterase-producing inoculant on fermentation indices and rumen degradation kinetics of whole crop rice (WCR) silage ensiled at different dry matter (DM) contents. Methods: Dual-purpose inoculants, Lactobacillus brevis 5M2 and Lactobacillus buchneri 6M1, confirmed both activities of antifungal and carboxylesterase in the previous study. The WCR at mature stage was chopped, and then wilted to obtain three different DM contents consisting of 35.4%, 43.6%, and 51.5%. All WCR forages were applied distilled water (CON) or mixed inoculants with 1:1 ratio at 1×105 colony forming unit/g (INO), and ensiled into 20 L mini silo (5 kg) in quadruplicates for 108 d. Results: The INO silages had lower lactate (p<0.001) and butyrate (p = 0.022) with higher acetate (p<0.001) and propionate (p<0.001) than those of CON silages. Ammonia-N (p<0.001), lactate (tendency; p = 0.068), acetate (p = 0.030), and butyrate (p<0.001) concentrations of INO silages decreased linearly with increasing DM content of WCR forage. The INO silages presented higher lactic acid bacteria (p<0.001) with lower molds (p<0.001) than those of CON silages. Yeasts (p = 0.042) and molds (p = 0.046) of WCR silages decreased linearly with increasing DM content of WCR forage. In the rumen, INO silages had higher the total degradable fraction (p<0.001), total volatile fatty acid (tendency; p = 0.097), and acetate (p = 0.007), but lower the fractional degradation rate (p = 0.011) and propionate (p<0.001) than those of CON silage. The total degradable fraction (p<0.001), total volatile fatty acid (p = 0.001), iso-butyrate (p = 0.036), and valerate (p = 0.008) decreased linearly with increasing DM content of WCR forage, while the lag phase (p<0.001) was increased linearly. Conclusion: This study concluded that application of dual-purpose inoculants on WCR silage confirmed antifungal and carboxylesterase activities by inhibiting mold and improving rumen digestibility, while increase of wilting times decreased organic acids production and rumen digestibility.

Effect of Protein Sources on Rumen Microbial Protein Synthesis Using Rumen Simulated Continuous Culture System

  • A rumen simulated continuous culture (RSCC) system was used to study the influence of supplementation of the three different types of protein sources such as urea, casein and soy protein on rumen microbial synthesis in terms of rumen microbial synchronization. The urea treatment showed the highest pH value. Ammonia nitrogen concentration was rapidly increased after feeding and not significantly different in the urea treatment (13.53 mg/100 ml). Protozoa numbers were not significantly different for soy protein and casein treatment compared to urea treatments during incubation. The average concentration of total VFA (mMol) was not detected with significant difference among treatments, but iso-butyrate production showed the highest for soy protein treatment among treatments (p<0.001). The lowest concentration in total iso-acids (iso-butyrate and iso-valerate) production was observed in urea treatment. The soy protein treatment showed no significantly change in acetate/propionate. The amounts of dry matter (DM) out flow showed no significant difference among treatments. Organic matter (OM) flow was the highest for urea treatments and the lowest for casein treatment (p<0.03). The nitrogen flow for casein treatment was not significantly different from other treatments. The efficiency of microbial protein synthesis in terms of microbial nitrogen (MN) synthesis (g MN/kg ADOM) digested in the rumen was highest for casein treatment (58.53 g MN/kg ADOM) compared to soy protein and urea (p<0.05). This result suggests that rumen ammonia releasing rate may influence on microbial protein synthesis in the rumen.

Effects of Sorghum Hybrid and Grain Supplementation of Silage-Based Diets on Nutrient Digestibilities and Passage Rates and Ruminal Metabolism in Growing Steers

  • Six medium-framed steers, fitted with ruminal cannulas, were utilized in a $6{\times}6$ Latin square design with a $3{\times}2$ arrangement of treatments to determine the effects of sorghum hybrid and grain supplementation on nutrient digestibilities and passage rates and ruminal metabolism of silage-based diets fed to growing steers. The diets consisted of three wholes-plant silages (a high grain-containing, grain sorghum and middle-season, moderate grain-containing, and late-season, low grain-containing forage sorghums), each fed with or without 25% rolled grain sorghum. No significant interactions occurred between sorghum hybrid and grain supplementation for the digestion or passage rate criteria measured. Ruminal butyrate concentration was the only fermentation characteristic affected by a hybrid ${\times}$ grain supplementation interaction. The grain sorghum silage diets had the highest DM, OM, and ADF digestibilities; the late-season silage diets, the lowest. Digestibility of NDF tended to be highest (p < 0.10) for the grain sorghum silage, whereas starch digestibility was not affected by sorghum hybrid. Ruminal ammonia, acetate, propionate, butyrate, and total VFA concentrations were highest for the grain sorghum silage diets. Grain supplementation increased DM and OM digestibilities, but had no effect on digestibilities of NDF, ADF, and starch. Ruminal pH was decreased, but total VFA concentration and acetate : propionate ratio were not affected by grain supplementation.

Effects of Branched-chain Amino Acids on In vitro Ruminal Fermentation of Wheat Straw

  • This study investigates the effects of three branched-chain amino acids (BCAA; valine, leucine, and isoleucine) on the in vitro ruminal fermentation of wheat straw using batch cultures of mixed ruminal microorganisms. BCAA were added to the buffered ruminal fluid at a concentration of 0, 2, 4, 7, or 10 mmol/L. After 72 h of anaerobic incubation, pH, volatile fatty acids (VFA), and ammonia nitrogen ($NH_3$-N) in the ruminal fluid were determined. Dry matter (DM) and neutral detergent fiber (NDF) degradability were calculated after determining the DM and NDF in the original material and in the residue after incubation. The addition of valine, leucine, or isoleucine increased the total VFA yields ($p{\leq}0.001$). However, the total VFA yields did not increase with the increase of BCAA supplement level. Total branched-chain VFA yields linearly increased as the supplemental amount of BCAA increased (p<0.001). The molar proportions of acetate and propionate decreased, whereas that of butyrate increased with the addition of valine and isoleucine (p<0.05). Moreover, the proportions of propionate and butyrate decreased (p<0.01) with the addition of leucine. Meanwhile, the molar proportions of isobutyrate were increased and linearly decreased (p<0.001) by valine and leucine, respectively. The addition of leucine or isoleucine resulted in a linear (p<0.001) increase in the molar proportions of isovalerate. The degradability of NDF achieved the maximum when valine or isoleucine was added at 2 mmol/L. The results suggest that low concentrations of BCAA (2 mmol/L) allow more efficient regulation of ruminal fermentation in vitro, as indicated by higher VFA yield and NDF degradability. Therefore, the optimum initial dose of BCAA for in vitro ruminal fermentation is 2 mmol/L.

유전자재조합 CHO 세포에서 Histone Deacetylase Inhibitor를 이용한Albumin-erythropoietin 생산성 증진 (Enhanced Production of Albumin-erythropoietin by Histone Deacetylase Inhibitors in Recombinant CHO Cells)

  • Chinese hamster ovary (CHO) cells are the most widely used mammalian host for the commercial production of recombinant proteins. However, they show relatively low yields of recombinant proteins in comparison with microbial cells. Various strategies have been tried to overcome this drawback. The acetyl moieties are attached to the N-terminus of histone by histone acetyltransferase (HAT) while histone deacetylase (HDAC) removes histone-bound acetyl groups. HDAC inhibitor (HDACi), such as sodium butyrate, sodium propionate and valproic acid, can enhance specific productivity of CHO cells. Human albumin-erythropoietin (Alb-EPO) is a novel 105 kDa protein comprising recombinant human EPO fused to human albumin. In this study, we examined the effects of HDACi on the production of Alb-EPO in CHO cells with various concentrations in the range of 0-1 mM. The results showed that sodium butyrate was found to be the best HDACi for enhancing productivity. It enhanced not only the production of Alb-EPO but also the apoptosis of recombinant CHO cells.

Anti-tumor Promoting Activity of Some Malaysian Traditional Vegetables (Ulam)

  • Ethanolic extracts of different parts of 10 local traditional vegetables (ulam) (Amaranthus gangeticus, Jussiaea linifolia, Eugenia polyantha, Trapa incisa, Trichosanthes anquina, Mangifera indica, Pachyrrhirus erosus, Barringtonia mcarostachya, Carica papaya, and Coleus tuberosus) were screened for in vitro antitumor promoting activity using the inhibition test of Epstein-Barr virus (EBV) activation in Raji cells induced by phorbol 12-myristate 13-acetate and sodium-n-butyrate. All the extracts were found to have strong inhibition activity toward EBV-activation, except for leaf extract of T. anquina. The extracts were non-cytotoxic to the Raji cells except for the extracts of A. gangeticus (leaves), B. macrostachya (leaves), E. polyantha (young leaves), and J. linifolia (leaves) where the viability of the cells were decreased significantly.

Cyclohexanol 이용성 세균의 분리 및 특성 (Isolation and Characterization of Cyclohexanol-utilizing Bacteria)

  • A bacterium, which can utilize cyclohexanol as a sole source of carbon and energy, was isolated from sludge in sewage of Ulsan Industrial Complex for Petrochemicals, Korea and identified as Rhodococcus sp. TK6. The growth conditions of the bacteria were investigated in cyclohexanol containing media. The bacteria utilized cyclohexanol, cyclohexanone, cyclohexane-1,2=diol, cyclopentanol, cyclopentanone, and $\varepsilon$-caprolactone but not cyclohexane, cyclohexane-1,2-dione, and cyclooctanone. The bacteria were able to utilize alcohols such as ethanol, 1-propanol, 1-butanol, 1-pentanol, 1-hexanol, 2-methyl-1-propanol, 3-methyl-1-butanol, 2-propanol, and 2-butanol as well as cyclohexanol, organic acids such as adipate, propionate, butyrate, valerate, n-caproate, and 6-hydroxycaproate, and aromatic compounds such as phenol, salicylate, p-hydroxbenzoate, and benzoate as a sole source of carbon and energy. Cyclohexanone as a degradation product of cyclohexanol by Rhodococcus sp. TK6 was determined with gas chromatography.
