잠재성 Herpes Simplex Virus의 재활성화에 대한 n-Butyrate의 효과 (Effect of n-Butyrate on the In Vitro Reactivation of Latent Herpes Simplex Virus)

  • 4-carbon fatty acid인 n-Butyrate(n-BTA)는 herpes virus의 일종인 Epstein-Bat virus(EBV)에 작용해서 잠복형인 EBV를 활성형태로 유도시키는 것으로 알려져 있다. 고로 본 실험에서는 mouse의 삼차신경절에 잠복하고 있는 HSV의 재활성화에 대한 n-BTA의 효과를 실험 관찰하였다. Pentobarbital로 마취시킨 mouse의 양쪽눈 각막을 30 gauge 주사바늘로 scarify한 후에 type I HSV(HSV-1) $10{\mu}1(1{\times}10^5$ plaque-forming units)를 각각 점안 감염시켰다. virus를 감염시킨 4주 후에 mouse의 삼차신경절을 적출하여 시험관 내에서 조직배양을 시행하였다. 조직배양시에 0. 1, 0. 25, 0.5. 1.0 그리고 2.0mM농도의 n-BTA를 첨가하였으며 1일, 2일, 3일간 각각 배양한 후 신경절을 연마하여 연마액내의 HSV-1 titer를Vero cell monolayer에서 plaque assay로 측정하였다. 1) n-BTA첨가군은 잠재성 HSV가 대조군에 비하여 현저하게 빨리 재활성화 되었고 재활성화되는 virus의 양도 현저히 증가되었다. 2) 24시간을 계속해서 n-BTA 각 농도를 첨가해서 배양할 군은 n-BTA 6시간 첨가 배양하고 새로운 배양액으로 갈아서 18시간 배양한 군에 비해 잠재성 virus의 재활성화가 현저히 증가되었다. 3) Gang1ionic latent HSV-1의 재 활성화에 영향을 미치는 각 농도의 n-BTA는 Vero cell의 monolayer에서의 HSV-1의 번식에는 아무런 영향을 미치지 않았다.

Enhancing Butyrate Production, Ruminal Fermentation and Microbial Population through Supplementation with Clostridium saccharobutylicum

  • Butyrate is known to play a significant role in energy metabolism and regulating genomic activities that influence rumen nutrition utilization and function. Thus, this study investigated the effects of an isolated butyrate-producing bacteria, Clostridium saccharobutylicum, in rumen butyrate production, fermentation parameters and microbial population in Holstein-Friesian cow. An isolated butyrate-producing bacterium from the ruminal fluid of a Holstein-Friesian cow was identified and characterized as Clostridium saccharobutylicum RNAL841125 using 16S rRNA gene sequencing and phylogenetic analyses. The bacterium was evaluated on its effects as supplement on in vitro rumen fermentation and microbial population. Supplementation with $10^6CFU/ml$ Clostridium saccharobutylicum increased (p < 0.05) microbial crude protein, butyrate and total volatile fatty acids concentration but had no significant effect on $NH_3-N$ at 24 h incubation. Butyrate and total VFA concentrations were higher (p < 0.05) in supplementation with $10^6CFU/ml$ Clostridium saccharobutylicum compared with control, with no differences observed for total gas production, $NH_3-N$ and propionate concentration. However, as the inclusion rate (CFU/ml) of C. saccharobutylicum was increased, reduction of rumen fermentation values was observed. Furthermore, butyrate-producing bacteria and Fibrobacter succinogenes population in the rumen increased in response with supplementation of C. saccharobutylicum, while no differences in the population in total bacteria, protozoa and fungi were observed among treatments. Overall, our study suggests that supplementation with $10^6CFU/ml$ C. saccharobutylicum has the potential to improve ruminal fermentation through increased concentrations of butyrate and total volatile fatty acid, and enhanced population of butyrate-producing bacteria and cellulolytic bacteria F. succinogenes.

The Effect of Nitrogen Fertilization to the Sward on Guineagrass (Panicum maximum Jacq cv. Gatton) Silage Fermentation

  • To investigate the effect of nitrogen fertilization on the quality of tropical grass silage, guinea grass grown with 3 types of nitrogen fertilizers, namely, urea, ammonium sulfate, and compound fertilizer 804, at 2 fertilization levels, 0.5 and 2.5 kg $Na^{-1}$ (0.5 N and 2.5 N, respectively), was subjected to silage fermentation. Silage fertilized with 0.5 N showed butyrate-dominant fermentation, irrespective of the type of fertilizer used. On the other hand, fermentation of silage fertilized with 2.5 N was significantly affected by the type of fertilizer used; fertilization with ammonium sulfate and compound fertilizer 804 resulted in silage that contained a large amount of butyrate and no lactate; this silage was considered to be of a significantly low quality as compared with silage fertilized with 0.5 N. Among silage fertilized with 2.5 N, the desirable butyrate-free fermentation was found only in urea-fertilized silage, which had the best quality. Grass material fertilized with a high level of urea accumulated a relatively high concentration of nitrate nitrogen (0.22% dry matter). Our results presented here suggest that nitrogen fertilizer management could affect the quality of tropical grass silage and that a relatively high concentration of nitrate in silage may promote butyrate-free fermentation even in tropical grass silage.

Effect of Butyric Acid on Performance, Gastrointestinal Tract Health and Carcass Characteristics in Broiler Chickens

  • An experiment was conducted to study the effect of graded levels of butyric acid (butyrate) on performance, gastrointestinal tract health and carcass characteristics in young broiler chickens. Control starter (0-3 wk) and finisher (4-5 wk) diets were formulated to contain 2,900 kcal ME/kg and 22% CP, and 3,000 kcal ME/kg and 20% CP, respectively. Subsequently, four other experimental diets were formulated to contain 0.05% antibiotic (furazolidone) or 0.2, 0.4 and 0.6% butyric acid. Each diet was fed at random to 8 replicates of 6 chicks each throughout the experimental period (0-5 wk). The results showed that 0.4% butyrate in the diet was similar to antibiotic in maintaining body weight gain and reducing E. coli numbers but superior for feed conversion ratio. No added advantage on these parameters was obtained by enhancing the concentration of butyrate from 0.4 to 0.6% in the diet. Feed intake and mortality were not influenced by the dietary treatments. A reduction in pH of the upper GI tract (crop, proventiculus and gizzard) was observed by inclusion of butyrate in the diets of broilers compared to either control or antibiotic-fed group. Butyrate at 0.4% was more effective in reducing the pH than 0.2% butyrate. Within the lower GI tract, 0.4 and 0.6% butyrate was effective in lowering pH in the duodenum, but no effect was found in either the jejunum or ileum. The villus length and crypt depth in the duodenum increased significantly in all the butyrate treated diets irrespective of the level tested. Carcass yield was higher and abdominal fat content was lower significantly in all the butyrate treatment groups compared to the control or antibiotic group. From these findings, it is concluded that 0.4% butyric acid supplementation maintained performance, intestinal tract health, and villi development and carcass quality in broiler chickens.

Histone Deacetylase Inhibitors Induce the Differentiation of Eosinophilic Leukemia EoL-1 Cells into Eosinophils

  • EoL-1 cells differentiate into eosinophils in the presence of n-butyrate, but the mechanism has remained to be elucidated. Because n-butyrate can inhibit histone deacetylases, we hypothesized that the inhibition of histone deacetylases induces the differentiation of EoL-1 cells into eosinophils. In this study, using n-butyrate and two other histone deacetylase inhibitors, apicidin and trichostatin A, we have analyzed the relationship between the inhibition of histone deacetylases and the differentiation into eosinophils in EoL-1 cells. It was demonstrated that apicidin and n-butyrate induced a continuous acetylation of histones H4 and H3, inhibited the proliferation of EoL-1 cells, and induced the expression of markers for mature eosinophils such as integrin ${\beta}7$, CCR1, and CCR3 on EoL-1 cells, while trichostatin A evoked a transient acetylation of his tones and induced no differentiation into eosinophils. These findings suggest that the continuous inhibition of histone deacetylases in EoL-1 cells induces the differentiation into mature eosinophils.

  • Acetate, propionate, butyrate are the major soluble volatile fatty acids metabolites of fermented food waste leachates. This work investigate the effects of volatile fatty acid on the growth rate and $NH_4-N$, $PO_4-P$ removal efficiency of mixotrophic microalgae Chlorella vulgaris to treat digested food waste leachates. The results showed that acetate, propionate and butyrate were efficiently utilized by Chlorella vulgaris and microalgae growth was higher than control condition. Similar trends were observed upon $NH_4-N$ and $PO_4-P$ consumption. Volatile fatty acids promoted Chlorella vulgaris growth, and nutrient removal efficiencies were highest when acetate was used, and butyrate and propionate showed second and third. From this work it could be said that using mixotrophic microalgae, in this work Chlorella vulgaris, fermented food waste leachates can be treated with high efficiencies.

Effects of Chemical Composition and Temperature for the Production of Volatile Fatty Acids During Anaerobic Decomposition Process of Marine Sinking Particles

  • Anaerobic decomposition experiments were performed to know the effect of chemical composition and temperature for the production of volatile fatty acids (VFAs) from marine sinking particles. Sinking particles were obtained with sediment traps set in Aburatsubo Inlet, Kanagawa Prefecture, Japan, in February, May and August. Sinking particles collected in May were composed of higher fraction of chl. a than the other two months. February and May samples were used to perform the decomposition experiments. VFAs production rates were higher in May sample than February. The production rates increased with increase of incubation temperature, and order of production rates of four VFAs were acetate>n-butyrate>propionate>iso-butyrate at $10^{\circ}C\;and\;20^{\circ}C$. At $28^{\circ}C$, the production rate of propionate was higher than n-butyrate. Based on these results, it is considered that production of VFAs from sinking particles during anaerobic decomposition depends on the chemical composition and temperature.

  • Complexes of the type[Co(T)(L-pro)], where T is the quadridentate $N_2O_2$-type ligand, N,N'-dimethylethylenediamine-N,N'-diacetate or N,N'-dimethylethylenediamine-N,N'-di-$\alpha$-butyrate, have been prepared. The complexes were separated into the two stereoisomers, $\Delta$-s-cis-[Co(T)(L-pro)] and $\Lambda$-s-cis-[Co(T)(L-pro)]. They were characterized by their proton magnetic resonance, absorption and circular dichroism spectra, elemental analyses.

Slow Release Substrate를 이용한 Geobacter lovleyi의 TCE 탈염소화 연구 (The Study of TCE Dechlorination using Geobacter lovleyi with Slow Release Substrate Applied)

  • 본 연구에서는 Slow Release Substrate(SRS)로 사용되는 TBOS의 분해특성과 TBOS 분해생성물인 acetate와 butyrate를 적용한 탈염소화 효율을 파악하고자 하였다. 회분식 실험은 GC/FID를 이용하여 TCE 및 cis-dichloroethene(cis-DCE), 1-butanol, TBOS를 분석하였으며, acetate와 butylate는 HPLC를 이용하여 분석하였다. 혐기성 가수분해 반응을 통해 1M의 TBOS는 4M의 1-butanol로 전환 및 축적되었으며, 가수분해율은 $0.186{\mu}M/day$로 나타났다. 또한, 1-butanol은 퇴적토 내 토착균주에 의해 acetate와 butyrate로 분해되었다. 이 결과 TBOS는 자연에서의 탈염소화 공정에서 SRS로 사용하기에 적합한 것으로 판단된다. Acetate와 butyrate를 전자공여체로 적용한 TCE 탈염소화 반응은 초기 TCE 농도가 낮아짐에 따라 탈염소화 효율은 높아지는 것으로 나타났다. 또한, acetate를 적용한 탈염소화 반응의 1차 반응 상수가 butyrate를 적용한 경우보다 높게 나타났다. 이는 탈염소화 반응에서 Geobacter lovleyi의 기질친화도 및 생분해성, 그리고 다양한 기질에 대한 적응도의 영향으로 판단된다. 그러나 Geobacter lovleyi의 TCE 탈염 소화 반응에 따른 cis-DCE의 축적이 발생할 경우 Geobacter lovleyi의 탈염소화 능력이 감소하는 것으로 나타났다. 결론적으로 SRS는 Geobacter lovleyi를 이용한 TCE 탈염소화 공정 향상에 도움이 될 것이며, 이에 따라 발생되는 cis-DCE는 영가철 같은 환원성 금속이나 공존 가능한 탈염소화 미생물을 이용한 처리가 함께 필요할 것으로 판단된다.
