Chemical Constituents of Selaginella tamariscina (권백의 화학성분)

  • The chemical constituents of Selaginella tamariscina were studied. From the n-hexane, chloroform and n-butanol soluble fractions, three compounds were isolated by chromatographic purification process. They were identified as lutein, $3{\beta}-cholesterol$ and hinokiflavone by spectral anaylsis(IR, MS, UV, $^1H-NMR$, $^{13}C-NMR$ and DEPT). Lutein and $3{\beta}-cholesterol$ were first isolated compounds from Selaginellaceae and hinokiflavone was a reported compound from this plant.

  • DEAE-Cellulose, Sephadex column chromatography and polyacrylamide gel electrophoresis were used to purify acid phosphatase, aryl sulfatases, ${\beta}-glucuronidase$ and cathepsin D in n-butyl alcohol extracts of porcine leukocyte Iysosomes. The degree of purification was quite high for all enzymes studied and some could be identified by histochemical reactions.

  • In the course of continuous work on the leaves of Allium tuberosum (Liliaceae), compounds 1 and 2 were isolated from the ethylacetate and butanol-soluble fraction, and identified as N-p-coumaroyl tyramine and bis(p-hydroxyphenyl) ether, respectively, on the basis of spectral data and physicochemical results.

The Correlation of Lower Flash Point data with Activity Coefficient Models

  • Two popular activity coefficient models, Wilson and NRTL equations have been used to correlate the published flash point data on the n-propanol + propionic acid and n-butanol + propionic acid systems through the optimization method. The results of these correlation were compared with the results calculated by Raoult's law. The optimization method were found to be better than those based on the Raoult's law. The optimization method based on the Wilson equation described the published data more effectively than was the case when the optimization method was based upon the NRTL equation.

The Prediction of Lower Flash Points by Optimization Method

  • The flash point is the most widely used flammability property for the evaluation of the flammability hazard of combustible liquid mixtures. In this paper, the reported flash points for the the binary systems, ethylbenzene+n-butanol and ethylbenzene+n-hexanol were correlated by the optimization method. The optimization method based on the van Laar and Wilson equations were compared with the Raoult's law. The calculated values based on the optimization method were found to be better than those based on the Raoult's law.

  • The anti-oxidant activities of the various fractions from the herbs of Artemisia apiacea were investigated. The n-hexane and n-butanol fractions were found to cause significant free radical scavenging effects on DPPH, their scavenging potencies as indicated in IC50 values, being 230.1 and 183.7 mg/$m\ell$, respectively. Isolation of Antifungal Compound and Biocontrol Potential of Lysobacter antibioticus HS124 against Fusarium Crown Rot of Wheat

  • Fusarium graminearum is the main cause of substantial economic loss in wheat production. The aim of this study is to investigate biocontrol potential of Lysobacter antibioticus HS124 against F. graminearum and to purify an antifungal compound. In preliminary study, n-butanol crude extract revealed destructive alterations in the hyphal morphology of F. graminearum and almost degraded with $1,000{\mu}g\;mL^{-1}$ concentration. For further study, the antifungal compound extracted from the n-butanol crude extract of L. antibioticus HS124 was identified as N-Butyl-tetrahydro-5-oxofuran-2-carboxamide ($C_9H_{16}NO_3$) using NMR ($^1H-NMR$, $^{13}C-NMR$, $^1H-^1H\;COSY$, HMBC, and HMQC), and HR-ESI-MS analysis. To our knowledge, N-Butyl-tetrahydro-5-oxofuran-2-carboxamide may be a novel compound with molecular weight of 186.1130. The minimum inhibitory concentration value of antifungal compound was $62.5{\mu}g\;mL^{-1}$ against F. graminearum. In an in vivo pot experiment, crown rot disease from F. graminearum was inhibited when wheat seeds were treated with both HS124 culture and F. graminearum. Growth of wheat seedling was enhanced by treatment of HS124 compared to control. Our results suggest that L. antibioticus HS124 characterized in this study could be successfully used to control F. graminearum and could be used as an alternative to chemical fungicides in modern agriculture.

Isolation of Berberine from the Rhizome of $Coptis$ $chinensis$ by Centrifugal Partition Chromatography (향류분배 크로마토그라피법에 의한 황련($Coptis$ $chinensis$) 뿌리로 부터 Berberine의 분리)

  • $Coptis$ $chinensis$ Franch rhizome is one of the important traditional Korea medicines with anti-inflammatory, anti-bacterial, anti-hypertensive and anti-cancer properties. The methanol extract of rhizome from the $Coptis$ $chinensis$ rhizome was purified by using preparative centrifugal partition chromatography (CPC) and HPLC method. The optimum two-phase CPC solvent system was composed of n-butanol:acetic acid:water at a ratio of 4:1:5. Berberine (16.8 mg) was successfully isolated by CPC and HPLC. The chemical structure of the compound was identified by (1 H)$^1H$, $^{13}C$-NMR and ESI-MS spectral data analysis.

Anti-oxidant Activity of Saussurea lappa C.B. Clarke Roots

  • This study was performed to investigate the potential use of Saussurea lappa C.B. Clarke as a source of antioxidant agents. Various solvent fractionates from S. lappa C.B. Clarke roots were investigated for their anti-oxidative effectiveness. The contents of total phenolics and flavonoids were determined by the Folin-Ciocalteu's colorimetric and the aluminum nitrate method, respectively. Total phenolic and flavonoid contents of n-butanol soluble fractionates from S. lappa C.B. Clarke, 44.43 ${\mu}g$ gallic acid equilibrium (GAE)/g extract and 92.15 ${\mu}g$ quercetin equilibrium (QE)/g extract, respectively, were higher than those of other solvent fractionates. The n-butanol soluble fractionates of S. lappa C.B. Clarke (1,000 ppm) showed the strongest inhibitory potential on 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical and reducing power at 92.98% and 0.38, respectively. Thus, our data shows that the S. lappa C.B. Clarke plant may help prevent antioxidative stress.

Isolation of 1',3'-Dilinolenoyl-2'-Linoleoylglycerol with Tyrosinase Inhibitory Activity from Flammulina velutipes

  • This study was carried out to evaluate the inhibitory effect of Flammulina velutipes extracts on tyrosinase activity and to identify its biologically active component. The ethyl acetate and n-butanol extracts showed potent tyrosinase inhibitory activities. Subsequently, fractions of the n-butanol extract showed only a partial tyrosinase inhibitory activity. The most active compound of tyrosinase inhibitory activity was identified from the ethyl acetate extract as 1',3'-dilinolenoyl-2'-linoleoylglycerol (LnLLn) by comparing its mass, $^1H-$, and $^{13}C-NMR$spectral data with those previously reported in the literature. LnLLn showed tyrosinase inhibitory activity with an $IC_{50}$value of 16.1 ${\mu}g/ml$. These results suggest that the ethyl acetate extract of F. velutipes could be applicable for the development of a new whitening agent.
