Dietary Manipulation and Increase in Plasma Unsaturated Fatty Acids in Sheep

  • Forty three 7-month old, Barbados Black $Belly{\times}Malin$ crossbred sheep were used for the trial. They were allotted into three treatment groups fed varying levels of oil palm (Elaeis guineensis) frond pellets and commercial sheep pellets. Treatment diets were 80% commercial pellet+20 % (% w/w) oil palm frond pellet (CON group, n=15), 50% commercial pellet+50% oil palm frond pellet (% w/w) (HAF group, n=14) and 80% oil palm frond pellet+20% (% w/w) commercial pellet (OPF group, n=14). The plasma fatty acid profiles from these animals were compared before and after 14 weeks of feeding. Results showed that total unsaturated fatty acid content in the CON group had increased by 10% (p<0.01) from the pre-treatment values. All three treatment groups had significantly different plasma n-6 and n-3 polyunsaturated fatty acid contents at the end of the trial. In fact, the CON group had significantly (p<0.01) more n-6 polyunsaturated fatty acid content compared to its own initial values, and also the values from the HAF and OPF groups. However there was a significant (p<0.01) decline in plasma n-3 polyunsaturated fatty acids in all groups. The final total unsaturated to saturated fatty acid content ratio was significantly (p<0.01) highest in the CON group, demonstrating the high plasma unsaturated fatty acid content in these animals. This study shows the plasma unsaturated fatty acids in sheep can be increased by dietary manipulation.

Fatty Acid Composition of 20 Subtropical Fish Species from Jeju Island (제주도산 아열대성 어류 20종의 지방산조성 특징)

  • Lipid levels and fatty acid compositions of 20 species of subtropical fish caught off the coast of Jeju Island, Korea were determined. The fish were categorized according to total lipid (TL) content as follows; lean fish ($TL=1.19{\pm}0.32%$, n=10 species), low-fat fish ($TL=3.02{\pm}0.44%$, n=6 species) and medium-fat fish ($TL=6.87{\pm}1.23%$, n=4 species). The prominent fatty acids were saturated fatty acids (SFAs), at 16:0 and 18:0 monounsaturated fatty acids (MUFAs), at 18:1n-9 and 16:1n-7, and polyunsaturated fatty acids (PUFAs), at 22:6n-3, 20:5n-3 and/or 20:4n-6. Across all species, the percentages of PUFAs and SFAs were $38.0{\pm}7.89%$ and $34.3{\pm}3.08%$, respectively, with no significant difference between both groups (P<0.05), but showed higher proportions than that of MUFA group ($27.8{\pm}7.23%$) (P<0.05). The proportion of n-3 PUFAs was $27.1{\pm}8.68%$ across all fish species, similar to levels in other fishes from Korea. These results suggest that subtropical fish from Jeju Island are a good source of n-3 PUFAs for humans.

The Effect of Acyl Chain Structure on the Hydrolysis of Fatty Acids from Fish Oil by Lipase-OF 360,000 (아실체인 구조가 효소 Lipase-OF 360,000에 의한 지방산의 가수분해 특성에 미치는 영향)

  • The hydrolysis characteristics of each fatty acid composing the fish oil by means of the lipase from Candida cylindracea was investigated. The saturated fatty acids, C14:0, C16:0 and C18:0, and the unsaturated fatty acids with one double bond, C16:1, C18:1(n-7), C18:(n-9), C20:1 and C22:1 were more easily hydrolyzed than the $\omega$-3 polyunsaturated fatty acids. when the number of carbon of the $\omega$-3 fatty acids was same but that of double bond was different, the hydrolysis of the $\omega$-3 fatty acids having lower number of double bond was more rapidly carried out. When the degree of polyunsaturation was same but the number of carbon was different, the lipase acted more rapidly upon the $\omega$-3 fatty acids with lower number of carbon. Docosahexaenoic acid(DHA) was most highly concentrated in the glyceride mixture after 120hr hydrolysis among the various $\omega$-3 polyunsaturated fatty acids. The weight percentage of DHA to the $\omega$-3 fatty acids in the fish oil was 31.87% but that in the glyceride mixture after 120hr hydrolysis was 51.89%.

Taurine Concentrations Are Closely Associated with Fatty Acids Concentrations in Breast Milk from Koreans (한국인 모유의 지방산 조성과 타우린 농도의 상관관계에 관한 연구)

  • Kong chain $\omega$3 & $\omega$6 fatty acids and taurine have been suggested to have structural and /or functional roles in the brain. The purpose of this study was to evaluate the changes in the percentage of fatty acids and taurine concentration in human milk obtained from Korean mothers(n=37) as lactation proceeds from birth to 5wks postpartum , and to determine the correlation between taurine concentration and fatty acid composition in breast milk. Level of fatty acids and taurine in breast milk were measured by GLC and HPLC, respectively. The percentages of medium -chain saturated fatty acids(10 : 0, 12 : 0 & 14: 0) were increased significantly , while percentages of long-chain saturated fatty acids(22 :0 & 24 : 0), and most of the long chain $\omega$6 and $\omega$3 fatty acids(20 : 3$\omega$6 , 20 :4$\omega$6, 22 :4$\omega$6 , 22 :5$\omega$6, 22 :5$\omega$3 & 22 : 6$\omega$3) were decreased significantly in mature milk (5 wks postpartum ) as compared to the colostrum. Taurine concentrations in colostrum and mature milk from Korean mothers were 549 $\pm$58 and 233 $\pm$41nmol/ml , respectively. Taurine concentrations in human milk was negatively correlated with the percentages of medium -chain saturated fatty acids ( 10 : 0 &12 : 0), and positively correlated with the percentages of long-chain saturated fatty acids( 16 :0, 20 : 0, 22 : 0& 24 : 0) and long-chain monounsaturated fatty acids( 20 : 1, 22 : 1 & 24 ; 1). Taurine concentration in human milk was positively correlated with elongation indices of both $\omega$6 and $\omega$3 fatty acids, and negatively correlated with the desaturation index of $\omega$6 fatty acids. These close relationships between fatty acids compositions and taurine level in human milk are worth pursing further investigation, especially with regard to their common roles in retina and brain development.

A genome-wide association study for the fatty acid composition of breast meat in an F2 crossbred chicken population

  • The composition of fatty acids determines the flavor and quality of meat. Flavor compounds are generated during the cooking process by the decomposition of volatile fatty acids via lipid oxidation. A number of research on candidate genes related to fatty acid content in livestock species have been published. The majority of these studies focused on pigs and cattle; the association between fatty acid composition and meat quality in chickens has rarely been reported. Therefore, this study investigated candidate genes associated with fatty acid composition in chickens. A genome-wide association study (GWAS) was performed on 767 individuals from an F2 crossbred population of Yeonsan Ogye and White Leghorn chickens. The Illumina chicken 60K significant single-nucleotide polymorphism (SNP) genotype data and 30 fatty acids (%) in the breast meat of animals slaughtered at 10 weeks of age were analyzed. SNPs were shown to be significant in 15 traits: C10:0, C14:0, C18:0, C18:1n-7, C18:1n-9, C18:2n-6, C20:0, C20:2, C20:3n-6, C20:4n-6, C20:5n-3, C24:0, C24:1n-9, monounsaturated fatty acids (MUFA) and polyunsaturated fatty acids (PUFA). These SNPs were mostly located on chromosome 10 and around the following genes: ACSS3, BTG1, MCEE, PPARGC1A, ACSL4, ELOVL4, CYB5R4, ME1, and TRPM1. Both oleic acid and arachidonic acid contained the candidate genes: MCEE and TRPM1. These two fatty acids are antagonistic to each other and have been identified as traits that contribute to the production of volatile fatty acids. The results of this study improve our understanding of the genetic mechanisms through which fatty acids in chicken affect the meat flavor.

Proximate and Fatty Acid Compositions of Commercial Domestic and Imported Melania Snails Semisulscospira sp. (시판 국내산 및 수입산 다슬기(Semisulscospira sp.)의 일반성분 및 지방산 조성)

  • The proximate, fatty acid and dimethyl acetal (DMA) compositions of domestic and imported melania snails Semisulcospira sp. being sold commercially were studied. The protein content ranged from 8.09% (China) to 11.3% (North Korea). The lipid content ranged from 0.84 (China) to 2.36% (North Korea). Domestic melania snail had protein (10.5%) and lipid (1.02%) contents intermediate between those of the two imported melania snails. The prominent fatty acids were 16:0 and 18:0 saturated fatty acids, 16:1n-7, 20:1n-11, 18:1n-9, 18:1n-7 and 16:1n-9 monounsaturated fatty acids, and 20:4n-6, 20:5n-3 (eicosapentaenoic acid, EPA), 22:2 non-methylene-interrupted diene (NMID), 18:3n-3, 18:2n-6 and 22:5n-3 (docosapentaenoic acid, DPA) polyunsaturated fatty acids (PUFA). The percentage of n-3 PUFA was higher in domestic (19.6%) and North Korean (imported, 19.5%) snails than in that from China (imported, 16.8%), while the n-6 PUFA level was higher in snails from China (21.2%) and domestic snails (14.7%) than in that from North Korea (9.87%). The total DMA, i.e., 18:0 DMA, 16:0 DMA and 18:1 DMA was 7.30, 7.27, and 4.86% domestic snail and those from China and North Korea, respectively. These results suggest that melania snail is a source of n-3 PUFA, and DMA derived from plasmalogen.

Dietary intake of fat and fatty acids by 1-5-year-old children in Korea: a cross-sectional study based on data from the sixth Korea National Health and Nutrition Examination Survey

  • BACKGROUND/OBJECTIVES: We examined dietary fat intake and the major food sources by young children in Korea. SUBJECTS/METHODS: A total of 1,041 children aged 1-5 years were identified from the 2013-2015 Korea National Health and Nutrition Examination Survey. Data on total fat and fatty acid intake were obtained by a single 24-h dietary recall. Food sources were identified based on the amounts of total fat and fatty acids consumption according to each food. Fat and fatty acid intakes and their food sources were presented by age group (1-2-y, n = 401; 3-5-y, n = 640). Fat and fatty acid intakes were also evaluated according to socioeconomic characteristics. RESULTS: The mean intake of fat was $27.1{\pm}0.8g$ in the 1-2-y group and $35.5{\pm}0.7g$ in the 3-5-y group, and about 23% of the total energy was obtained from fat in both age groups. The mean intake of saturated fatty acids (SFA) was $10.5{\pm}0.3g$ in the 1-2-y group and $12.7{\pm}0.3g$ in the 3-5-y group, with the 1-2-y group obtaining more energy from SFA than the 3-5-y group (9.2% vs. 8.3%). The mean intake of polyunsaturated fatty acids (PUFA) was $6.3{\pm}0.1g$ in the total subjects, with $0.8{\pm}0.03g$ of n-3 fatty acids and $5.5{\pm}0.1g$ of n-6 fatty acids being consumed. Milk, pork, and eggs were major food sources of total fat, SFA, and monounsaturated fatty acids, and soybean oil was the main contributor to PUFA in both age groups. In the 1-2-y group, children in rural areas had significantly higher intake of PUFA and n-3 fatty acids than did those in urban areas. CONCLUSIONS: Our findings provide current information on dietary fat intake among young Korean children and could be used to establish dietary strategies for improvement of health status.

Nutritional Analysis of Purple Com Husk Extract

  • Seakso 5, a maize hybrid, was applied for variety in 2021 by Gangwon Agricultural Research and Extension Services in Korea. This variety was bred to produce a purple extract of com husk. It shows purple color in the husk and cob and contains abundant anthocyanins. In this study, to obtain basic data on purple com 'Seakso 5' husk extract (PCHE), a husk extract was prepared and the contents of vitamins, amino acids and fatty acids were analyzed. The Seakso 5 husks were extracted with 30% alcohol and concentrated, after adding dextrin to the concentrate, it was spray-dried to prepare an extract. The PCHE were analyzed for vitamins, amino acids, and fatty acids by referring to the Food Code. As a result, three types of vitamins were detected, and their contents were vitamin B1 0.14 mg/100g, vitamin B2 2.30 mg/100g, and vitamin C 11.32 mg/100g. A total of 15 amino acids were detected in the PCHE, including aspartic acid and glutamic acid, and the content of aspartic acid was the highest at 3.02 mg/g. Simultaneous analysis of fatty acids Among 37 types, saturated fatty acids were palmitic acid (C16:0), stearic acid (C18:0), monounsaturated fatty acids were oleic acid (C18:1(n-9)), polyunsaturated fatty acids were linoleic acid(C18:2(n-6)c) and a-Linolenic Acid (C18:3(n-3)) were detected. Among the detected fatty acids, the content of palmitic acid was the highest at 40.75 mg/100g. In the future, These analysis results will be used as reference data for temporary food ingredient applications by the Ministry of Food and Drug Safety.

Comparisons of Feeding Ecology of Euphausia pacifica from Korean Waters Using Lipid Composition (한국 근해의 난바다곤쟁이 Euphausia pacifica의 지방 조성에 의한 섭식 생태 비교)

  • Dietary lipid biomarkers (fatty acids, fatty alcohols, and sterols) in adult specimens were analyzed to compare and understand the feeding ecology of the euphausiid, Euphausia pacifica, from three geographically and environmentally diverse Korean waters (Yellow Sea, East China Sea, and East Sea). Total lipid content of E. pacifica from Korean waters was about 10% dry weight (DW) with a dominance of phospholipids (>46.9% of total lipid content), which are known as membrane components. A saturated fatty acid, C16:0, a monounsaturated fatty acid, C18:1(n-9), and two polyunsaturated fatty acids, C20:5(n-3) and 22:6(n-3), were most abundant (>60% of total fatty acids) in the fatty acid composition. Some of the fatty acids showed slight differences among regions although no significant compositional changes of fatty acids were detected between these regions. Phytol, originating from the side chain of chlorophyll and indicative of active feeding on phytoplankton, was detected all samples. Trace amounts of various fatty alcohols were also detected in E. pacifica. Specifically, krill from the Yellow Sea showed relatively high amounts of longchain monounsaturated fatty alcohols (i.e. 20:1 and 22:1), generally found in herbivorous copepods. Three different kinds of sterols were detected in E. pacifica. The most dominant of these sterols was cholest-5-en-$3{\beta}$-ol (cholesterol). The lipid compositions and ratios of fatty acid trophic markers are indicative of herbivory in E. pacifica from the Yellow Sea and East Sea (mainly feeding on dinoflagellates and diatoms, respectively). The lipid compositions and ratios of fatty acid trophic markers are indicative of carnivory or omnivory in E. pacifica from the East China Sea, mainly feeding on microzooplankton such as protozoa. In conclusion, lipid biomarkers provide useful information about krill feeding type. However, further analyses and experiments (i.e. gut content analysis, in situ grazing experiment, etc.) are needed to better understand the feeding ecology of E. pacifica in various marine environments.

Effect of Quality and Quantity of Dietary Fats on the Status of Tocopherol and Lipid Peroxidation of Plasma and Tissue in Rats (식이지방의 종류와 수준에 따라 쥐의 혈장과 조직의 Tocopherol 및 지질과산화상태에 미치는 영향)

  • The study was to compare the effect of dietary fatty acids on fatty acid profile in tissue and the status of tocopherol and lipid peroxidation, and superoxide dismutase and glutathione peroxidase activities at two fat levels. Male Sprague Dawley rats weighing average 350g(17 weeks) were fed either low fat(LF, 4.3% w/w, 10% kcal) or high fat(HF, 20.8%, w/w, 40% kcal)diet for 6 weeks. The fats used were beef tallow as a source of saturated fatty acid, corn oil for n-6 linoleic acid, perilla oil for n-3 $\alpha$-linolenic acid and fish oil for n-3 eiocosapentatenoic acid(EPA) and n-3 docosahexaenoic acid(DHA). Palsma tocopherol was significantly reduced by fish oil compared to beef tallow at body fat level. However, there was no significant effect on the levels of plasma MDA, RBC MDA and tocopherol, and RBC hempolysis by the type and amount of dietary fat. The peroxidizibility index of fatty acid profile in plasma and liver was increased and liver MDA level was significantly increased by fish oil when dietary fat level was increased. The activities of SOD and GSHPx tended to be increased by perilla oil and fish oil at both fat oil significantly reduced the incorpration of c20:4 and increased the incorporation of c20:5 into liver compared to corn oil. The incorporation of n-3 fatty acids into tissue by perilla oil rich in $\alpha$-linolenic acid was significantly higher tan corn oil and its effect was improved with higher amount of perilla oil in diet by high fat diet. Overall, the lipid peroxidation of tissue could be prevented by tocopherol supplementation when dietary fat level was low in diet. However, at high fat diet, tocopherol supplementation might not be enough to prevent the lipid peroxidation in tissue since the potential for lipid peroxidation was tended to be increased with higher incorporation of higher unsaturated n-3 fatty acids into tissue. Therefore, it could not be recommended to consume large amount of fish oil even with excess amount of tocopherol supplemented to the high fat diet.
