A 16S rDNA polymerase chain reaction assay to detect Mycoplasma pulmonis in rats model

  • Murine mycoplasmosis, caused by Mycoplasma (M.) pulmonis, is a prominent disease in rodent animals. The aim of this study was to develop a sensitive and specific PCR assay to detect M. pulmonis in animals and to assess the suitability of this assay for the detection of mycoplasmal infection in rats experimentally infected with M. pulmonis. A new PCR assay using the M. pulmonis-specific primer pairs MPul-F and MPul-R was developed. The primers and probe for the assay were designed from regions in the 16S rRNA gene that are unique to M. pulmonis. The novel PCR assay was very specific and sensitive for M. pulmonis, detecting the equivalent of 5 pg of target template DNA. It detected only M. pulmonis and no other Mycoplasma species or other bacterial species. The newly developed PCR assay also effectively detected M. pulmonis infection in rats. These results suggest that this PCR assay using M. pulmonis-specific primer pairs of MPul-F and MPul-R will be useful and effective for monitoring M. pulmonis infection in animals.

  • Objectives: The purpose of this study is to find the effect of Keumsuyukgunjeongamibang and aromatherapy on a child diagnosed with mycoplasma pneumonia. Methods: The child had been treated with herbal medicine and aromatherapy for 8 days. In order to examine the improvement, chest X-ray and blood test were performed. Results: Various symptoms, such as, coughing, rhinorrhea, dyspnea, was disappeared. The series of 4 X-ray images and blood test proved that the patient has improved. Conclusions: The result of this study shows that administration of Keumsuyukgunjeongamibang and aromatherapy are effective in treatment of mycoplasma pneumonia.

  • The role of atypical bacterial infection in the pathogenesis of asthma is a subject of continuing debate. There is an increasing body of literature concerning the association between the atypical bacteria such as Mycoplasma pneumoniae (M. pneumoniae) and asthma pathogenesis. Moreover, many studies investigating such a link have been uncontrolled and have provided conflicting evidence, in part due to the difficulty in accurately diagnosing infection with these atypical pathogens. This manuscript will review the relationship between M. pneumoniae infection and asthma pathogenesis.

Generation and Characterization of a Monoclonal Antibody with Specificity for Mycoplasma arginini

  • Previously, we generated monoclonal antibodies (MAbs) that bound to the surface of human embryonic stem cells (hESCs) in an attempt to discover new hESC-specific surface markers. In this study, MAb 47-235 (IgG1, ${\kappa}$) was selected for further characterization. The MAb bound to the surface of undifferentiated hESCs but did not bind to mouse ESCs or mouse embryonic fibroblast cells in flow cytometric analysis. The antibody immunoprecipitated a 47 kDa protein from the lysates of cell surface-biotinylated hESCs. Identification of the protein by quadrupole time of flight tandem mass spectrometry revealed that 47-235 binds to Ag 243-5 protein of Mycoplasma arginini. BM-Cyclin treatment of the hESCs that reacted with 47-235 resulted in loss of mycoplasma DNA and the reactivity to 47-235. Nevertheless, the hESCs that were reactive to 47-235 maintained self-renewal and pluripotency and thus could be differentiated into three embryonic germ layers.

  • The Guillain-Barre Sydrome (GBS) is post-infectious autoimmune disease and it could be caused by auto-antibodies produced after infections. Mycoplasma pneumoniae is one of rare cause of GBS and known to be associated with antibody to galactocerebroside (GalC) which is a major neutral glycolipid constituent of myelin. We report a case of GBS with immunoglobulin M GalC antibody after M. pneumoniae infection.

  • We had experienced a case of pericarditis associated with Mycoplasma and Salmonella co-infection. This 5-year old boy presented with high fever and generalized tonic-clonic seizure. The diagnosis was based on the markedly increased serial titer of Widal test and cold agglutinin with Mycoplasma indirect hemmagglution test during hospitalization. The electrocardiogram revealed P-R interval prolongation and an echocardiogram revealed moderate pericardial effusion. The patient treated with ceftriaxone and erythromycin for 2 weeks, and responded to the treatment well. We report this case with brief review of the related literatures.

Exacerbation of Chronic Gastritis by Mycoplasma Infection through Inducing Inflammation

  • Mycoplasmas resemble H. pylori in production of ammonia and induction of inflammatory cytokines from immune and non-immne cells. In Republic of Korea infection rate of H. pylori is relatively high but only a proportion of them invite additional inflammation and progress into gastric cancers. Therefore, additional risk factors cannot be excluded. The presence and identification of mycoplasma were confirmed by semi -nested PCR and sequencing and the results were compared with pathological data. Fifty-six samples collected from Korean chronic gastritis patients were used for the study. Twenty-three (41.1%) were positive to mycoplasmas and all of them were identified as human mycoplasmas, M. faucium, M. fermentans, M. orale, M. salivarium and M. spermatophilum. Mycoplasma-infected chronic gastritis samples showed more severe, additional infiltration of neutrophils than non-infected samples and the difference was significant (P < 0.05). In conclusion human mycoplasma infection may playa role in progression of chronic gastritis to metaplasia by inducing additional inflammation.

돼지의 유행성폐렴 원인균(Mycoplasma hyopneumoniae)에 대한 항체가 분포도 조사 (Studies on Enzyme-Linked Immunosorbent Assay(ELISA) for Detection of antibody to Mycoplasma hyopneumoniae)

  • Mycoplasmal pneumonia of swine(MPS) cause by Mycoplasma hyopneumoniae has been recognized as a serious impediment to swine production due to chronic respiratory disorder which result in the weight loss and decreased feed conversion. The disease causes a great economic losses in pig industry by characterizing with high morbidity, low mortality, growth retardation and low feed efficiency. The present study was conducted to investigate the titers of antibody against M hyopneumoniae from the regional and seasonal groups of the slaughtered pigs by enzyme-linked immunosorbent assay(ELISA). The result have shown that the average seropositive rate of M hyopneumoniae infection was 84.6% . The regional seropositive rate in Korea showed 87.4% in Kyonggj, 83.4n in Kangwon, 89.2% in Chungnam and 77.6% in Chungbuk area, respectively. Also the seasonal seropositive rate was appeared as 78.6% in spring,90.1% in summer, 76.9% in autumn and 83.8% in winter, respectively.

Detection of Respiratory Viral Pathogens and Mycoplasma spp from Calves with Summer Pneumonia in Korea

  • Respiratory pathogens of calves including bovine parainfluenza type 3 virus (BPI3V), bovine respiratory syncytial virus (BRSV), infectious bovine rhinotracheitis virus (IBRV) and Mycoplasma spp is well-known for winter pathogens. However, there are no studies about summer pneumonia pathogens of calves in Korea. The aim of this study was to detect respiratory pathogens from calves with summer pneumonia. Eighty calves from 5 regions were chosen and their nasal swabs were used to detect respiratory pathogens with real-time PCR. Mycoplasma spp was major primary respiratory pathogens in calves with summer pneumonia. Although, the detection rates of respiratory viruses were very low, serological assays showed that respiratory viruses exist widely in farms.
