To develop mushroom varieties for cultivating at low temperature on oak logs, a strain with a low fruiting body generation temperature was crossed with Di-mon to select for a line with excellent properties. Selection was followed by cultivation testing. From these studies, Heunghwa 1ho was identified. The optimum temperature for cultivating Heunghwa 1ho, was 13.3℃. The fruiting temperature range was 6.4~20.2℃, identical to that of the parent strain. Growth at 25℃ for 7 days achieved optimal mycelial growth of 61.9±2.10 mm, superior to growth of the parent strain at this temperature. The cap shape of Heunghwa 1ho was convex, cap diameter was 57.8±8.31 mm, and cap color was brown. Heunghwa 1ho showed similar genetic traits to those of the parental strain. However, dry weight (20.1 kg/m3) and cap diameter and color are superior to those of the parent strain. The 3 year fresh oak mushroom yield was 113.8 kg/m3, superior to the respective yields of the parent strains JMI 10047 and JMI 90021 (92.5 kg/m3 and 66.4 kg/m3).
These studies were investigated for effect of concentration of sodium chloride on occurrence and growth of fruitbody in oyster mushroom, Pleurotus ostreatus. When the mycelia were inoculated and cultured on the PDA plate added with the different concentrations of sodium chloride, the growth of them were not affected at the concentration of 0.5%, started to decrease at 1.0% as compared with the PDA plate without sodium chloride, and they did not grow at 5.0%. When tested at the column filled with sawdust, the tendency was similar, but the decreasing rate of mycelial growth was gentle. In sawdust bottle cultivation sprinkled with the different concentration of sodium chloride solution at the different stage, the productivity and quality of fruitbody started to decrease at 3.0% of the solution and the yield at the treatment of 10.0% solution was only 47% of that in non-treated bottles. Treated at the different stages of mushroom development, treatment just after mushroom sprout did not show any different from that just after scraping spawn from the complete grown bottles. The sprinkle treatment at this stage looked better than that of the soaking with the solution before mushroom sprout. The yield at this treatment with 10% solution showed little damage with that of 90% of non-treated bottle. Sprinkle treatment during mushroom growth stage did not affect and showed almost same to non-treated bottle except the treatment with 10.0% solution. The treatment of this stage showed better results of growing characters than the treatment after and before mushroom sprouting even 10% solution. Morphological characteristics of oyster mushroom do not distinguish to treatment of different stage and concentration of sodium chloride solution. The analyzing data of quality on underwater usually used for oyster mushroom in Inchon, Yeongjong and Kanghwa area showed over-concentration to the standard for edible water and agricultural water. But it was not confirmed whether this water quality could affect to mushroom cultivation.

Recently sawdust cultivation of Shiitake mushroom (Lentinula edodes) is increasing. It is important to make mycelia to be brown on the substrate surface. This browned surface in sawdust cultivation plays an important role like as artificial bark of the oak log, which protects the other pests and suppresses water evaporation in the substrate. In order to isolate genes which related to brown color formation, differential display method was used. Two cDNA fragments obtained by DD-PCR were 1.2 and 1.6kb and these were expressed in white colored mycelia from L. edodes, but not brown colored mycelia. Partial sequencing of these cDNA fragments showed that the 1.6kb cDNA had 100% identity with the microsatellites gene from Dugenia polichroa. However, the other 1.2kb cDNA fragment had poly T tail on 3' region of partial open reading frame on 5' region. The new primer designed based on the sequence of 1.2kb cDNA was constructed. RT-PCR analysis using the newly designed 0.12kb cDNA specific primer showed that the gene was only expressed in white color mycelia, but not in brown color mycelia. Sequence analysis of 5' region of this 1.2kb cDNA revealed that this gene contained partial open reading frame consisted of 110 amino acid. Homology search using DNASIS database showed that this gene had high sequence homology of 66.7% in DNA level and 69.2 % in amino acid level with dTDP-glucose 4,6-dehydratases gene from Arabidopsis thaliata. The dTDP-glucose 4,6-dehydratases gene was known to be function to have tolerance with oxidation stress. These results strongly suggest that this gene isolated from white mycelia of L. edodes might have a function of repressor against mycelia browning. Therefore I designated this gene as BCR (Brown Color Repressor) gene.
This study was conducted to provide basic data for setting a standard medium for winter mushroom cultivation. Investigation of medium composition in winter mushroom farms in Korea revealed that the types of medium used for each farm were slightly different and that the mixing ratio of the medium also varied. All farmers used corncob, rice bran, and beet pulp as the main media, and calcium carbonate and ground oyster shells were used at 1.0~3.8% to adjust the pH of the medium. Analysis of the physicochemical properties of the mixed media showed nitrogen content of 1.28~1.52%, carbon content of 45.7~48.5%, and C/N ratio of 30.7~37.9. The content of inorganic components was significantly different in each farm, depending on the type and amount of minerals in the mixed medium. Mycelial growth was the fastest at Farm 2, reaching 6.5 cm within 34 days of culture, followed by Farm 4 at 5.7 cm, whereas Farm 1, 3, and 5 showed similar growth. Mycelial density was high without any significant difference between farms. Survey of yield according to medium composition in each farm showed the highest yield in Farm 2 at 173.4 g/bottle, followed by Farm 1 at 168.4 g/bottle, whereas Farm 3 and 4 showed similar yield. However, Farm 4 had the lowest yield at 145.4 g/bottle. Therefore, mushroom yield was different depending on the medium composition in the farm, and thus selection of a medium that can be used universally in multiple mushroom farms was considered possible.
This study was conducted to diversify the cultivation of mushroom items and develop a competitive variety of Hypsizigus marmoreus. We focused on developing Hypsizigus marmoreus with lower bitterness, likable shape, and high yield. We have collected and tested characteristics of genetic resources from domestic and abroad since 2019. Breeding (2019), characterization (2020), productivity test (2021), and farm demonstration test (2021) have been sequentially conducted. We bred the new variety 'Yunseul' with unique traits. The optimal temperature for mycelial and fruit body growth were 22-25℃ and 15-18℃, respectively. It was similar to the control variety (Mangadak-2Ho) in the pileus form (hemispherical shape) and the cultivation period. However, it was thinner and longer than the control variety with the pileus diameter, thickness, stipe diameter, and length being 19.9 mm, 7.0 mm, 9.4 mm, and 86.3 mm, respectively. The effective number of fruit bodies was 47.8 in bottle cultivation, which was more than that of the control variety. The yield was 197.4 g/bottle (1,100 cc), which was 30% higher than the yield of the control variety, 151.9 g/bottle (1,100 cc). The parent and control varieties were also incubated alongside the new variety. The somatic incompatibility line was distinct. The band pattern in the mycelial DNA PCR reaction was different from that of the parent and control varieties, confirming the hybrid species.
The separation of the bacteria inhibiting Trichoderma sp. mold, the strain causing blue mold disease that occurs frequently when cultivating mushroom while carrying out the efficient fermentation of mushroom medium, from the growth was done. In about 200 strains isolated primarily from fungus garden samples, 6 strains were secondly isolated, which had fast growth rates and a clear zone on the plate medium of SM, AM, and CM. Among the 6 strains isolated, the C-1 strain showed high enzymatic activity of cellulase, amylase, and protease, and strong antibacterial activity for the T. virens and T. harzianum, selected finally. The selected C-1 strain was identified as Paenibacillus polymyxaby the result of the identification by Bergey's Manual of Systematic Bacteriology and the analysis of the nucleotide sequence of 16S rRNA, and named as P. polymyxa CK-1. In reviewing the growth conditions of the P. polymyxa CK-1 strain, the optimum cultivation temperature was $45^{\circ}C$, and the optimum pH for growth was in the range of 6.0~7.0. Appropriate incubation time of P. polymyxa CK-1 for the growth inhibition of the fungus T. virens and T. harzianum was 22 to 36 hours. And the fungal growth was not observed, even when leaving two molds inoculated on each petri dishes, which were treated with 24 hour culture solution of P. polymyxa CK-1 strain for 10 days. As a result of studying the thermal stability of the antagonists produced by the P. polymyxa CK-1 strain, no mycelial growth of the two fungi was observed in the test group treated for 20 minutes at $60^{\circ}C$ and $100^{\circ}C$, but mycelial growth was slightly observed in the test group treated for 20 minutes at $121^{\circ}C$. As aresult of reviewing the impact of the P. polymyxa CK-1 culture medium on mushroom mycelial growth, it showed no effect on a variety of mushroom mycelial growth including enoki mushroom and shiitake mushroom.
This study was carried out to investigate effect of NaCl treatment on growth oyster mushroom and Trichoderma spp. on PDA and in rice straws. We also investigated the possibility of oyster mushroom cultivation using rice straw harvested from the reclaimed land having been drained sea water. Mycelial growth of oyster mushroom was increased by treatment of 0.2% NaCl but decreased by treatment of the higher concentration of NaCl. In the case of the mycelial growth on PDA of Trichoderma spp., no change was found in 0.5%~1.0% NaCl, but decreased in the range of 1.0%~3.0% NaCl and drastically decreased at 5.0% NaCl. In the rice straws treated with different concentration of NaCl solution, mycelial growth of oyster mushroom showed almost same result compare to PDA. The spore formation of Trichoderma mould was almost same in both 0% and 0.3% NaCl, decreased in 0.5% and was not found spores in the higher concentration of NaCl. As increasing salt concentration in the rice straws, the NaO contents were increased. The $K_2O$ contents were decreased before and after sterilization. The moisture content of rice straws showed no difference by treatment of 3.0% NaCl but decreased the moisture by treatment of 5.0% NaCl. No pH change was found in the rice straws treated with NaCl. Mushroom yield in the rice straw of reclaimed land was a little higher than that of normal paddy land straw. The duration of primordium formation was not affected by NaCl concentration in rice straws. The yield of fruiting body in 0.3% NaCl treatment was 2,700kg, which was almost same to non-treated plot, but decreased in 0.5% NaCl. EC value of soaking water after submerging rice straw was higher than before, but NaO content was not changed at both condition.
Potato late blight caused by Phytophthora infestans Cooke is one of the major diseases in the cultivation of potatoes in Korea. Effect of chitosan preparations (SH-1 and SH-2) was evaluated on the inhibition of mycelial growth of P. infestans, and protective activity using detached potato leaf assay both in vivo and in the field test. SH-1 and SH-2 were showed protective activity of young plant with control values more than 95% potato late blight by inoculation with pathogens under growth chamber conditions. Mycelial growth was inhibited the radial growth over 74% at a concentration of $300{\mu}g/ml$ of both SH-1 and SH-2. Spraying with SH-1 and SH-2 on the leaves for detached leaf assay reduced disease development. The content of total polyphenol in stem was significantly increased by SH-1 and SH-2 application in the field. In field experiments, foliar application with both SH-1 and SH-2 were significantly reduced the development of late blight on potato plants. Control of late blight disease was obtained with control values of 72% and 53% by application of 1% SH-1 and SH-2, respectively, with 4 times at 7 days interval, and reduced with similar disease control values by application with 3 times at 14 days interval compared with untreated control. SH-1 and SH-2 applications increased the fresh weight of potato, and higher grade potatoes were also increased. The results showed that SH-1 and SH-2 applications can be used as eco-friendly natural fungicide for organic farming for the increase of yields and control of late blight.
For effective control of insect pest which can outbreak in a field during crop cultivation, various control agents such as eco-friendly agricultural materials (EFAM) including microbial control agents and chemical pesticides have been applied at fields and these control agents may be treated simultaneous or sequential in the same field to suppress diverse pests and diseases. The agents may influence each other and control efficacy may also differ from interactions. Therefore we need to test compatibility of microbial control agents with other agricultural agents. In this study, we investigated influence of pre-treated EFAMs, which are registered for whitefly control in greenhouse, on germination, mycelial growth and control efficacy of Isaria javanica isolate against sweet potato whitefly. The results showed that a mixture of paraffin oil+cinnamon oil among 4 EFAMs highly reduced germination ($8.9{\pm}1.3%{\sim}24.5{\pm}0.9%$) and mycelial growth ($0.81{\pm}0.01cm{\sim}0cm$) of I. javanica. To investigate the effects of the treatment interval between EFAMs and I. javanica on sweet potato whitefly control, four different EFAMs were pre-treated 0, 1, 4, and 7 days before applying I. javanica. Pre-treatment of four EFAMs inhibited insecticidal activity of I. javanica against sweet potato whitefly. Therefore when EFAMs and a mycopesticide using I. javanica spray simultaneous or sequential, application of EFAMs need more than 7 days interval after treatment of mycopesticide at field.

The 'Heuktari', a new mid-high temperature adaptable variety of oyster mushroom for the bottle culture, was bred by mating with monokaryons isolated from 'P11056' and 'MT07156'. The optimum temperature for the mycelial growth was $23{\sim}26^{\circ}C$ on PDA medium and that for the primordia formation and the growth of fruiting body of 'Heuktari' was $18{\sim}19^{\circ}C$ on sawdust substrate. In case of bottle cultivation, the period of mycelial growth was required about 30 days. In addition, the period of primordia formation and growth of fruiting body was 4 days and 5 days, respectively. In the characteristics of fruiting body, shape and color of pilei were round type and dark grayish brown, stipe color was white color and stipe shape was short and thick. The yield of fruiting bodies was 180 g/900 ml bottle which was 15% higher than that of Suhan-1ho. The gumminess and brittleness of stipe tissue were 110% and 140% stronger than those of Suhan-1ho, respectively.
