천궁 돌연변이 유발을 위한 최적 감마선 조사량 (Gamma-ray Irradiation on Radio Sensitivity in Cnidium officinale Makino)

  • Background: Cnidium officinale Makino have been used in traditional medicine in Northeast Asia. Although gamma-ray mutagenesis has been used to develop breeding resources with novel characteristics, research on the radiation sensitivity of C. officinale Makino is limited. Hence, the optimal gamma-ray dosage for mutation breeding in C. officinale Makino was investigated. Methods and Results: Seedstocks were exposed to doses of gamma rays (5 Gy - 50 Gy), and subsequently planted in a greenhouse. After 30 days of sowing, the survival rates and growth decreased rapidly at doses above 20 Gy, while all individuals died at 50 Gy. The median lethal dose (LD50) was 25.65 Gy, and the median reduction doses (RD50) for plant height, number of stems, and fresh weight were 12.81, 9.32, and 23.26 Gy, respectively. Post-irradiation levels of malondialdehyde (MDA), peroxidase (POD), and chlorophyll in the aerial parts of the plant were quantified using spectrophotometry. Relative to the controls, the levels of MDA and POD increased, while the level of chlorophyll decreased at doses ≥ 10 Gy, indicating cellular damage. Conclusions: A dose of 20 Gy was found to be optimal for mutation breeding in C. officinale Makino.

황색종 연초 돌연변이 계통 KF8832-85의 특성 (Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars. Sensitivity of Lavender to Proton, Electron, and Gamma Radiation

  • While ion beams are widely used in plant breeding, little is known about the sensitivity of Lavandula angustifolia (lavender) to ionizing radiation. To compare the biological effects of different types of ionizing radiation on the germination and survival rates of lavender, we exposed lavender seeds to gamma rays, 3 MeV electron beams, and 1.89 MeV proton ion beams. We observed that the seed germination rate decreased with increasing dosages of all three types of ionizing radiation. The malformation rate of lavender seedlings exposed to electron beams and gamma rays increased with increasing radiation dosage. By contrast, the effect of the accelerated proton beams on the malformation rate was negatively correlated with the dosage used. The survival rate of lavender seedlings exposed to the three types of ionizing radiation decreased in a dose-dependent manner. In addition, the survival rate of seedlings irradiated with proton and electron beams decreased more slowly than did that of seedlings irradiated with gamma rays. The half-lethal dose of gamma rays, electron beams, and proton beams was determined to be 48.1 Gy, 134.3 Gy, and 277.8 Gy, respectively, and the most suitable proton-ion energy for lavender seeds in terms of penetration depth was determined to be 5 MeV. These findings provide valuable information for the breeding of lavender by radiation mutation.

  • Differences of meat qualities between Japanese Black and Holstein have been known in Japan, however, the causative proteins and/or the genetic background have been unclear. The aim of this study was to identify candidate proteins causing differences of the meat qualities between the two breeds. Using technique of two-dimensional gel electrophoresis, protein profiling was compared from samples of the longissimus dorsi muscle and subcutaneous adipose tissue. Five protein spots were observed with different expression levels between breeds. By using LC-MS/MS analysis and Mascot program, three of them were identified as ankyrin repeat protein 2, phosphoylated myosin light chain 2 and mimecan protein. Subsequently, we compared the DNA coding sequences of three proteins between breeds to find any nucleotide substitution. However, there was no notable mutation which could affect pI or molecular mass of the proteins. The identified proteins may be responsible for different characteristics of the meat qualities between Japanese Black and Holstein cattle.

Genome-wide association study for loin muscle area of commercial crossbred pigs

  • Objective: Loin muscle area (LMA) is an important target trait of pig breeding. This study aimed to identify single nucleotide polymorphisms (SNPs) and genes associated with LMA in the Duroc×(Landrace×Yorkshire) crossbred pigs (DLY). Methods: A genome-wide association study was performed using the Illumina 50K chip to map the genetic marker and genes associated with LMA in 511 DLY pigs (255 boars and 256 sows). Results: After quality control, we detected 35,426 SNPs, including six SNPs significantly associated with LMA in pigs, with MARC0094338 and ASGA0072817 being the two key SNPs responsible for 1.77% and 2.48% of the phenotypic variance of LMA, respectively. Based on previous research, we determined two candidate genes (growth hormone receptor [GHR] and 3-oxoacid Co A-transferase 1 [OXCT1]) that are associated with fat deposition and muscle growth and found further additional genes (MYOCD, ARHGAP44, ELAC2, MAP2K4, FBXO4, FBLL1, RARS1, SLIT3, and RANK3) that are presumed to have an effect on LMA. Conclusion: This study contributes to the identification of the mutation that underlies quantitative trait loci associated with LMA and to future pig breeding programs based on marker-assisted selection. Further studies are needed to elucidate the role of the identified candidate genes in the physiological processes involved in LMA regulation.

Detection of 881A→881G Mutation in Tyrosinase Gene and Associations with the Black Ear Coat Color in Rabbits

  • The tyrosinase gene was selected as a candidate for uncovering genetic mechanism causing 'black ear' coat color in rabbits. A PCR-SSCP detection method was established for the $881^A{\rightarrow}881^G$ mutation located in the central region of the tyrosinase gene between the CuA and CuB binding region signatures, and this was confirmed by sequencing and alignment. Fully consistent associations between the SNP and 'black ear' coat color were observed by analysis in a "black ear" pedigree and on 61 unrelated individuals. This SNP can serve as a molecular marker for use in "back ear" wool rabbit breeding.

감마선 처리에 의한 장미 삽수의 발근 여부에 따른 생존율과 돌연변이 발생빈도 (Effect of Gamma-ray on Survival and Mutation Rates of Rooted Cuttings and Unrooted Cuttings in Rose)

  • 장미는 국화, 난과 더불어 3대 화훼작물에 속하며, 절화 및 분화, 조경, 꽃꽂이 등 활용범위가 광범위하다. 본 연구는 장미의 발근된 삽수와 발근 전 삽수에 감마선을 조사하여 생존율과 돌연변이 발생빈도, 변이 스펙트럼을 비교하기 위해 실시하였다. 실험재료로는 스프레이형 장미 3품종('러블리디아', '엘로우바베', '햇살')과 스탠다드형 장미 2품종('비탈', '아쿠아')를 사용하였으며, 감마선 조사는 발근된 삽수와 발근 전 삽수로 구분하여 한국원자력연구원 저준위 감마선 조사시설에서 70Gy로 조사하였다. 발근 전 삽수의 감마선 처리구는 발근 후 생존개체만 정식하였으며, 발근된 삽수의 감마선 처리구는 바로 정식하였다. 생존율과 돌연변이 발생빈도, 변이 스펙트럼은 정식 후 30주에 비교하였다. 그 결과 발근된 삽수의 감마선 처리구는 생존율과 돌연변이 발생빈도가 각각 39.4%~55.1%, 0.7%~7.4%로 나타났으며, 발근 전 삽수의 감마선 처리구는 각각 16.4%~50.8%, 0%~5.1%로 확인되었다. 결론적으로 발근된 삽수가 발근 전 삽수에 비해 감마선 조사 시 상대적으로 높은 생존율과 돌연변이 발생빈도를 보인다. 하지만 생존개체만을 대상으로 돌연변이 발생빈도를 비교할 경우 발근 삽수의 경우 1.8%~14.1%, 발근 전 삽수의 경우 0%~10%로 품종간의 차이는 다소 있으나 전체적으로 큰 차이가 없는 것을 확인할 수 있다. 또한, 발근 삽수와 미발근 삽수 모두 다양한 화색, 화형, 꽃잎 수의 변이가 확인되어 변이 스펙트럼에서도 큰 차이를 확인할 수 없었다. 미발근 삽수를 방사선 처리에 바로 이용해도 발근 삽수가 차이가 없으므로 앞으로 이용 가능할 것으로 판단된다.

황기 발아 및 생장에 미치는 감마선 조사 효과 (Effect of Gamma Irradiation on the Germination and Growth of Astragalus membranaceus)

  • 황기는 한국의 중요 약용작물로 한약재 및 식품의 용도로 이용되고 있으며. 재배시 가장 중요한 조건은 병해 및 각종 재해 등에 대한 안정성의 확보이다. 본 연구에서는 황기의 돌연변이 육종체계 확립 및 유용변이체를 육성하기 위한 목적으로, Y-선 처리에 의한 발아, 출현 및 생존율 등에 대한 감수성을 구명하여 각종 병해 및 재해에 안정성을 지닌 돌연변이체를 육성하기 위한 기초자료를 얻고자 하였다. 시험재료로는 포천 재래종을 이용하였고 파종 후 4일째부터의 발아율은 무처리에 비하여 고선량 (300 Gy 이상)에서 유의하게 낮아지는 것으로 나타났으며, 감마선량별 생존율은 무처리 대비 100 Gy 32%, 200Gy 43%, 300 Gy 63%, 400 Gy 이상에서는 72${\sim}$89%로 현저히 감소하였다. 감마선에 민감한 형질은 초장과 분지수였고, 둔감한 형질은 엽폭, 엽장, 엽수의 순으로 나타났으며, 초장, 분지수 및 생존율을 대상으로 한 황기의 생육 및 생존 반감 ($LD_{50}$)에 해당하는 선량은 $200{\sim}300$ Gy로 나타났다. 종합적으로 감마선 처리구의 발아, 생존율 및 생육차이를 감안해 보았을 때 황기 돌연변이 유기를 위한 적정 감마선량은 $200{\sim}300$ Gy로 판단되었다.

배유 돌연변이처리로 개발된 연질미 전분의 이화학적 특성 (Physicochemical and pasting properties of rice starches from soft rice varieties developed by endosperm mutation breeding)

  • 건식제분 쌀가루를 제조하기 위해 MNU로 돌연변이 처리하여 육종한 한가루와 신길의 가공적성을 확인하기 위하여 쌀의 80%를 차지하는 전분의 이화학적, 형태적 및 호화특성을 일반벼인 추청과 설갱 전분과 비교하였다. 신길전분의 아밀로스 함량은 27.1%로 나머지 쌀 전분은 16.5-19.5%이었다. 물결합능력과 팽윤력은 한가루 전분이 가장 높았고 전분입자의 크기($d_{50}$)도 가장 컸다. 전분입자의 형태는 설갱, 한가루, 신길전분의 전분입자 형태는 일부 한 면이 둥근 입자가 다면체 입자와 혼합되어 있었다. X-선 회절도에 의한 결정형은 모두 A형이었으며 호화개시온도는 신길전분이 $92.1^{\circ}C$로 가장 높고 한가루와 신길전분 호화액의 피크, 최저 및 최종점도가 추청과 설갱 전분보다 낮았다. 한가루 전분은 breakdown점도가 가장 컸으며 total setback점도는 가장 낮았다. 이와 같은 결과로부터 연질미 쌀가루의 전분 특성은 품종에 따라 다름을 확인하여 품종 별 쌀가루 특성과 가공성을 연구하여 전분과의 상관성을 비교해야 될 것이다.

Improvement of the Thermostability of Xylanase from Thermobacillus composti through Site-Directed Mutagenesis

  • Thermostability is an important property of xylanase because high temperature is required for its applications, such as wood pulp bleaching, baking, and animal feedstuff processing. In this study, XynB from Thermobacillus composti, a moderately thermophilic gram-negative bacterium, was modified via site-directed mutagenesis (based on its 3D structure) to obtain thermostable xylanase, and the properties of this enzyme were analyzed. Results revealed that the half-life of xylanase at $65^{\circ}C$ increased from 10 to 50 min after a disulfide bridge was introduced between the ${\alpha}$-helix and its adjacent ${\beta}$-sheet at S98 and N145. Further mutation at the side of A153E named XynB-CE in the C-terminal of this ${\alpha}$-helix enhanced the half-life of xylanase for 60 min at $65^{\circ}C$. Therefore, the mutant may be utilized for industrial applications.
