The methanol extract of perilla leaves contained compounds to reduce the mutagenicity of aflatoxin B$_1$(AFB$_1$) in Salmonella typhimurium TA98 and 100. They were separated by solvent extractions and identified by GC and GC-MS as 2-ethoxy acetate, 1, 2, 3, 4-tetramethyl-cis-cyclobutene, two isomers of methyl 11, 14, 17-eicosatrienoate, 12-acetyl-9-octadecanoic acid, and phytol. The antimutagenicities of phytol and methyl 11, 14, 17-eicosatrienoate were dependent on the mutagens tested. Phytol reduced the mutagenicity of Trp-p-2 but not of AFB$_1$and methyl 11, 14, 17-eicosatrienoate reduced the activities both of the mutagens.
Air suspedned particulates are the most serious factor among air pollutants in Seoul and their extracts have been revealed to be pulmonary carcinogenes. Air particulates were collected at a site representative of the high traffic area of Seoul from December 1993 to May 1994. Polycyclic aromatic hydrocarbons (PAHs) and nitroarenes in organic extracts of air particulates were determined by gas chromatography and mutagenicity of particle extracts was evaluated by the pre-incubation procedure with Salmonella typhirnurium TA98 and TA 98NR. The average concentration of total suspended particulates and inhalable (less than 10 $\mu$m) particulates were 169.63 $\mu g/m^3$, 141.92 $\mu g/m^3$, respectively. The relative contribution of organic matters was highest in neutral fraction and was lowest in basic fraction. Among identified 20 PAHs, the level of PAHs with 4-benzen ring i.e., pyrene, benzo(a)anthracene and chrysene were much higher than the PAHs with higher ring numbers. The mutagenicity of air particulates was much higher in the presence of rat liver microsomai fraction (S9 mixture) than that observed in its absence, which indicates that air particulates contained both direct and indirect mutagens. Nitroarenes in this study were not almost detected but mutagenicity of their fraction was higher. These results are represent to research for the identification of nitroarenes in the forward study.
The antimutagenic properties of twenty-one strains of Bifidobacterium were examined using Salmonella typhimurium TA98 in an in vitro assay system. The mutagens utilized for testing included Trp-P-1 (3-amino-1, 4-dimethyl-$^{5}H-pyrido$ (4, 3-b) indole), benzopyrene, IQ (2-amino -3-methylimidazo [4,5-f] quinoline), and NQO. The lyophilized cells of strain showed inhibitory effect of 64, 38, 29 and 20% in average against Trp-P-1, benzopyrene, NQO and IQ, respectively. There was no marked variation between each strain or growth stage in the degree of antimutagenicity against Trp-P-1, benzopyrene and IQ. Twelve hour grown cells showed higher antimutagenicity against NQO than 5-day grown cells. The results indicate that Bifidobacterium cells had a antimutagenic effect against several well-known mutagens to some degree.
The effects of mutagens, MMS and captan, on the patterns of proteins synthesized during the early developmental stages in mice were analyzed using two-dimensional electrophoresis. Three classes of proteins were observed in terms of synthetic pattern during the preimplantation stages. The first class is synthesized from the m-RNA, which was made and preserved throughout oogenesis and activated at the fertilization. The synthesis of these proteins did not seem to be influenced by MMS. The second class, which may be stagespecific proteins synthesized by newly transcribed m-RNA, was selectively inhibited by MMS. The third class, the synthesis of which is also suppressed by MMS, is the proteins synthesized by the m-RNA transcribed in augmented fashion. While MMS inhibits protein synthesis dependent on thenew transcription, this mutagen enhances a synthesis of a few proteins which were not observed in the untreated embryos. Captan did not affect protein synthesis at morula stage.

Objectives: This study was performed in order to devise a procedure for supplementing the Special Management Materials in the Occupational Safety and Health Act and recommend candidate materials. The results are expected to be used as fundamental data for classification and criteria necessary to manage Special Management Materials in workplaces. Also, they are expected to be used as a basis for selecting target materials to nominate as additional Special Management Material. Methods: We investigated the selection standards for candidates and review ranges of data sources to nominate Special Management Materials. The substances classified as GHS(Globally Harmonized System of classification and labeling of chemicals) category 1A(known to have carcinogenic potential and reproductive toxicity for humans) or 1B(presumed to have carcinogenic potential and reproductive toxicity for humans) carcinogens and reproductive toxicants among the Controlled Hazardous Substances of the Regulation on Occupational Safety and Health Standards and substances with OELs(Occupational Exposure Limits) were inspected as the candidates for Special Management Materials. Conclusions: A seven-step procedure for selecting candidates to designate as Special Management Materials was suggested, including the setting of target chemicals for evaluation, classification of CMR(Carcinogens, Mutagens or Reproductive toxicants) by GHS classification and criteria, suggestion and selection of the candidates, and more. This study recommends 58 chemicals as qualified candidates to supplement the Special Management Materials.

Many compounds might become activated after absorption of UV light energy. In some cases, the resulting molecule may undergo further biological reaction of toxicological relevance related especially to the photo-carcinogenicity resulting from photo-genotoxicity. However, no regulatory requirements have been issued with the exception of guideline issued by the Scientific Committee of Cosmetology, Commission of the European Communities (SCC/EEC) on the testing of sunscreens for their photo-genotoxicity. Thus, the objectives of this study are to investigate the utility of photo-Ames assay for detecting photo-mutagens, and to evaluate its ability to predict rodent photo-carcinogenicity. Photo-Ames assay was performed on five test substances that demonstrated positive results in photo-carcinogenicity tests: 8-methoxypsoralen (photoactive substance that forms DNA adducts in the presence of ultraviolet A irradiation), chlorpromazine (an aliphatic phenothiazine an a-adr-energic blocking agent), lomefloxacin (an antibiotic in a class of drugs called fluoroquinolones), anthracene (a tricyclic aromatic hydrocarbon a basic substance for production of anthraquinone, dyes, pigments, insecticides, wood preservatives and coating materials) and retinoic acid (a retinoid compound closely related to vitamin A). Out of 5 test substances, 3 showed a positive outcome in photo-Ames assay. With this limited data set, an investigation into the predictive value of this photo-Ames test for determining the photo-carcinogenicity showed that photo-Ames assay has relatively low sensitivity (the ability of a test to predict carcinogenicity). Thus, to determine the use of in vitro genotoxicity tests for prediction of carcinogenicity,' several standard photo-genotoxicity assays should be compared for their suitability in detecting photo-genotoxic compounds.
Benzo(a)pyrene and bisphenol A have been classified as endocrine disrupting chemicals (EDCs), which have been of concern in toxicology and environmental sciences. Benzo(a)pyrene and bisphenol A were monitored by HPLC or GC-MS in Baekwoon and Ilwirl lakes of Kyonggi-Do province to investigate contamination levels of EDCs. During the period between June, 2000 and August, 2000, water samples were collected from four different sites of each lake once per month. Contamination levels of benzo(a)pyrene were 3.27~4.25 ppb in Ilwirl lake and 2.00~2.33 ppb in Baekwoon lake, respectively. Bisphenol A levels were detected with the range of 0.33~7.94 and 0.43~4.71 for Baekwoon lake and Ilwirl lake, respectively. pH levels were higher in Ilwirl lake than in Baekwoon lake, where the contamination was relatively lower. These data suggest that lakes in Kyonggi-Do province could be contaminated with EDCs and be subjected to the routine monitoring and water quality control.
The micronucleus test using peripheral blood reticulocytes (RETs) was evaluated in ICR mice treated with N-methyI-N-nitrosourea (MNU) and benzo(a)pyrene [B(a)P] as model clastogens. The frequency of micronucleated reticulocytes (MNRETs) in both positive compounds was similar to other results which were reported previously. On the other hand, an anticlastogenic effect of the natural antioxidant, $\beta$-carotene and one of taroholds, galangin as model anticlastogens were investigated using simultaneous treatment. Mice were treated with a model clastogen alone, or with a model clastogen and a model anficlastogen simultaneously. Both $\beta$-carotene and galangin showed anticlastogenic effects against MNU- or B(a)P-induced micronuclei in mice. However, galangin has stronger activity than $\beta$-carotene. Results from our experiment suggest that the in vivo supravital staining micronucleus test using peripheral blood is useful in the evaluation of clastogenic and anticlastogenic effects.
The single cell gel electrophoressis(SCGE) assay, also known as the comet assay, is a rapid, simple, visual and sensitive technique for measuring and analysing DNA breakage in mammalian cells. The SCGE or comet assay is a promising test for the detection of DNA damage and repair in individnal cells. It has widespread potential applications in DNA damage and repair studies, genotoxicity testing and biomonitoring. In this microgel electrophoresis technique, cells are embedded in agarose gel on microscope slides, iysed and electrophoresed under alkaline conditions. Cells with increased DNA damage display increased migration of DNA from the nucleus towards the anode. The length of DNA migration indicates the amount of DNA breakage in the cell. The comet assay is also capable of identifying apoptotic cells which contain highly fragmented DNA. Here we review the development of the SCGE assay, existing protocols for the detection and analysis of comets, the relevant underlying principles determining the behaviour of DNA and the potential applications of the technique.
To evaluate the genotoxicity of CKD-602, an anticancer agent the in viかo reverse mutation assay using Salmonella typhimurium, the Chromosome aberration assay using Chinese hamster lung (CHL) cells and the in vivo micronucleus assay using bone marrow cells of ddY mice were performed. In the reverse mutation assay, CKD-602 did not induced mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535, and TA 1537 strains with and without metabolic activation. In the chromosome aberration test using CHL cells, there was an increased incidence of structural aberrations induced by CKD-602 without metabolic activation during 24 and 48 hours, but CKD-602 did not induce chromosome aberration with metabolic activation. The in vivo induction of micronuclei was measured in polychromatic erythrocytes of bone marrow of male ddY mice. At 24 hours after treatment with CED-602 by i.p. once, there was an increased incidence of micronucleated polychromatic erythrocytes in bone marrow of ddY male mice.
