Host-Mediated Assay를 이용한 감궁탕의 돌연변이원성 평가 (Evaluation of Mutagenicity with Gamgung-tang Using Host-Mediated Assay)

  • Mutagenicity of Gamgung-tang (GGT) was tested using in vitro S-9 mixture in vitro host-mediated assay with Salmonella typhimurium. In the previous reports, GGT was tested for the safety using Ames(-S-9), Bacillus subtilis Rec, and umu gene expression mutagenicity tests. Mutagenic activity in any assays we tested was not found. In this report, we further investigated safety of GGT after metabolic activation in vivo. Ames test with S-9 mixture and host-mediated assay with Salmonella typhimurium TA98 were used to identify metagenic property of GGT. GGT was administered 3 times with i.m. to Balb/c mice did not induced mutagenic effect in Salmonella typhimurium TA98 recovered from the liver after 3.5h with i.p. treatment. Over the entire dose range $(3{\sim}150mg/mouse)$ tested no toxicity was detected to the bacterial cells. These results suggest that there was no DNA damage and mutagenicity by GGT.

환경성 변이원에 대한 Genistein의 항돌연변이성 (Antimutagenic Effect of Genistein toward Environmental Mutagen)

  • This study was carried out to determine the antimutagenic effects of genistein on the somatic mutagenicity induced by aflatoxin B1 (${AFB}_1$), using Drosophila wing spot test system. Mutagen alone or mutagen with genistein were administered to the heterozygous(mwh/+) third instar larvae by feeding, and somatic cell mutations were detected in adult fly wing hairs. Genistein did not show any mutagenicity with the feeding concentrations of 5~15% in the test system. As the feeding concentrations of genistein increased, genistein inhibited the mutagenicity induced by AFB1 (14.6%~62.2% inhibition rate), while as the concentrations of AFB1 increased, small much spots that arise mostly from chromosome deletion and nondisjunction were more strongly suppressed by genistein than the large mwh spots from chromosomal recombination. In each group of different AFB1 concentrations, the rate of inhibition for total mwh spots was dependent on the dose of genistein. These results indicate that genistein have inhibitory effect on the mutagenicity induced by a mtagen, ${AFB}_1$. It seems to suggest that genistein may exert inhibitory effects to mutagenic and/or carcinogenic properties of DNA damaging agents.

볶은 보리 및 참깨의 갈색도와 돌연변이 유발성 (Browning and Mutagenicity of Roasted Barley and Sesame Seeds)

  • 본 연구는 현재 국내에서 사용되고 있는 보리차용 보리와 참깨의 볶음정도와 아울러 Salmonella typhimurium TA98과 TA100 균주를 이용한 Ames test에 의한 돌연변이 유발성 여부를 실험하였다. 시판용 보리차는 갈색도로 보아 볶음정도가 다양함을 알 수 있었고 업소용 보리차는 가정용 보리차에 비해 5배 정도로, 참기름용 참깨는 깨소금용 참깨보다 4배 정도 더 볶아진 것으로 평가되었다. 볶은 보리와 볶은 참개의 수용성, 에탄올 용해성 및 에테르 용해성 성분에 대한 Ames test 결과는 모두 최고의 시험농도에서 돌연변이 유발능이 있다고 단정하기는 어려웠다.

  • 국내 화장품 업계에서 실제 사용되고 있는 합성타르색소인 적색 204호 , 적색215호, 등색 203호의 3가지 색소에 대하여 유전독성시험을 행하였다.Chinese hamster lung(CHL) 세포에서의 염색체이상시험과 ddY 마우스를 이용한 소핵시험 및 초파리 날개를 이용한 체세포돌연변이 재조합 시험을 실시하였다. 그 결과 염색체이상시험과 소핵시험에서는 적색 204, 적색 215, 등색 203호 에서는 유전독성을 나타내는 변이원인으로는 작용하지 않는 것으로 나타났고 초파리 날개를 이용한 체세포돌연변이 시험에서는 각각 100mg/ml에서 single small spot의 출현빈도가 p<0.05의 유의수준에서 대조군에 비해 증가양상을 보였고, twin spot의 빈도 수는 적색 204호의 경우는 50mg/ml 농도에서, 적색 215호의 경우는 100mg/ml 농도에서 p<0.05의 유의수준에서 증가양상을 보여 약한 돌연 변이 원성을 나타내었다.

비 마약성 진통제 DA-5018의 변이원성 연구 (Mutagenicity of DA-5018, a Non-narcotic Analgesic Agent)

  • DA-5018, a non-narcotic analgesic agent, was examined for mutagenicity in the reverse mutation test on bacteria, chromosomal aberration test on cultured mammalian cells and micronucleus test on mice. The reverse mutation test was performed by a plate incorporation method with or without a metabolic activation system(S9 mix) using Salmonella typhimurium strain TA100, TA1535, TA98 and TA1537. DA-5018 did not significantly increase revertant colonies in any of the test strains under any conditions at concentrations ranging from 0.0049 to 1.25 mg/plate, compared with the vehicle control. In the chromosomal aberration test using cultured Chinese Hamster Lung(CHL) cells, DA-5018 did not increase the number of aberrant cells in the presence or absence of S9 mix at concentrations of 0.016 mM/plate to 0.25 mM/plate, compared with the vehicle control. In the micronucleus test, male ICR mice were given DA-5018 intraperitoneally at a dose level of 0.55, 1.10 and 2.20 mg/kg. The incidence of bone marrow micronucleated polychromatic erythrocytes in the DA-5018 treated mice was not significantly different from that of the vehicle control. These results indicate that DA-5018 does not have mutagenic potential under the present test conditions.

  • DA-3002, an authentic recombinant human growth hormone(rhGH), was examined for mutagenicity in the reverse mutation test on bacteria, in the chromosomal aberration test on cultured mammalian cells and in the micronucleous test on mice.   • The inhibitory effects of various extracts from Chinese pepper on the mutagenicity and the growth of MG-63 human osteosarcoma cells were studied. Chinese pepper was extracted with methanol and then the methanol extract was further fractionated by using hexane, chloroform, ethyl acetate and butanol. The methanol extract of Chinese pepper revealed the strong antimutagenic activity on the aflatoxin B1(AFB1) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Ames mutagenicity test and SOS chromotest.

Ames, Rec 및 umu Assay를 이용한 황기의 안전성평가 (Evaluation of Safety with Astragali Radix : Ames, Rec and umu Assays)

  • Water extract from Astragali Radix (AR) was tested for the safety using Ames, Bacillus subtilis Rec, and umu gene expression mutagenicity tests. Mutagenic activity in any assays we tested was not found. In Ames test, Salmonella typhimurium TA98 and TA 100 were used to identify mutagenic property, and the number of histidine revertants was measured. In the Recassay, Bacillus subtilis ${H-17(Rec^+)\;and\;M-45(Rec^-)}$ strains were used to test DNA damage activity. In the SOS umu test, Salmonella typhimurium TA1535 containing plasmid pSK1002 was used as a test strain, and we monitored the levels of umu operon expression by measuring the ${\beta}-galactosidase$ activity. From the results, there was no DNA damage and mutagenicity of AR. Hepatotoxicity of AR to female ICR mice was also monitored by the measurements of s-GOT, s-GPT, LDH activities after oral feeding for 15 days. AR was not shown any significant changes of s-GOT, s-GPT and LDH activities in mice sera.

  • 국내에서 실제사용되고 있는 22가지의 식용 및 외용색소에 대하여 유전독성실험을실시하였다. salmonella typhimurium을 이용한 유전자 복귀돌연변이 시험(Ames test)과 Chiness hamster lung cell을 이용한 염색체이상 시험을 시행한 결과 Ames test에서는 등색 203호 (D&C Orange No.17)가 대사활성계의 존재 유무와 관계없이 돌연변이 유발성을 보였고, 적색 204호 (D&C Red No.9)는 대사활성계의 존재하에서 돌연변이 유발성을 나타내었다. 염색체이상 시험에서는 적색104-1호 (Phroxin B)의 시약급과 적색 215호 (D&C Red No. 37)가 의양성을 나타내었다.

항암제 취급간호사의 항암제 체내흡수에 관한 조사 (Biological Monitoring on the Absorption of Antineoplastic Drugs in Nursing Personnel)

  • Antineoplastic agents may exhibit effects not only in patients therapeutically exposed, but also in health workers who prepare and administer these drugs. This study was done to clarify whether nurses who handle anticancer drugs show signs of drug absorption. The experimental group was 14 nurses handling anticancer drugs at three medical wards of a hospital in J city ; the control group was 12 psychiatric nurses at the same hospital. The test materials were the nurses' 24hr urine specimens, which were concentrated by XAD-2 column chromatograpy. Tester strains were TA98(±S9mix), TA100(±S9 mix), TA1535(±S9 mix) and TA1537(±S9 mix) : the salmonella mammalian microsomal test (Ames test) was used for the urinary mutagenicity assay. The results are summarized as follow : 1. In qualitative analysis of the results, both experimental group and control group showed 15.4% urine toxicity. 2. The experimental group revealed significantly higher urinary mutagenicity both in the activation method test and non-activation method test of the tester strains TA98, TA100 and TA1535. In the case of TA1537, the two groups showed no difference in the non-activation method test, but the activation method revealed a difference. 3. In urinary mutagenicity of the experimental group by ward career, there was a significant difference between the group with more than 20 months experience and the group with less than 20 months on the tester strains TA98, TA100, and TA1537. No Significant difference was found between two groups by the tester strain TA1535.
