갓(Brassica juncea) 추출물의 항균물질이 Escherichia coli와 Staphylococcus aureus의 균체 성분의 조성 및 누출에 미치는 영향 (Effects of Antimicrobial of Leaf Mustard(Brassica juncea) Extract on Compositions and Leakage of Cellular Materials in Escherichia coli and Staphylococcus aureus)

  • To develop natural food preservatives, antimicrobial effect of the ethanol extract of leaf mustard against E. coli and S. aureus were examined in terms of compositions and leakage of cellular materials in the microorganisms treated with the extract. No effect of the concentration of ethanol extract on the fatty acid composition of E. coli and S. aureus at logarithmic phase was showen, but the content of palmitic and palmitoleic acid of E. coli slightly increased and decreased, respectively, and the content of palmitic and margaric acid of S. aureus slightly increased, when compared to each control. Ethanol extract did not affect most of the amino acids E. coli and S. aureus at logarithmic phase ; however, some of them(proline, glycine, valine and histidine of E. coli and proline, methionine and histidine of s. aureus) were elevated and some other amino acid(aspartic acid, glutamic acid, tyrosine and arginine of E. coli and aspartic acid, glutamic acid, glycine, alanine and lysine of Staph. aureus) found to be decreased. The amount of cell body protein leaked from E. coli and S. aureus increased to 1.02 and 0.22mg/g cell weight, respectively, as compared to controls. Similarly, the substances with absorbance at 260 nm from E. coli and s. aureus increased to 0.12 and 0.06mg/g cell weight, respectively.

갓으로부터 분리, 동정된 4-decanol의 항돌연변이 효과 (Antimutagenic effects of 4-decanol identified from mustard leaf)

  • 갓(Brassica juncea Cosson)의 chloroform추출물이 Salmonella typhimurium TA100균주에서 $AFB_1$의 돌연변이유발을 억제시키는 효과가있었으며, chloroform획분 중에 존재하는 화합물을 GC-MS로 분리/동정한 결과 4-decanol이 다량으로 함유되어있었다. 4-Decanol을 0.5%되게 DMSO에 녹인용액을 처리하였을 때 대조구에 비해 $AFB_1$과 MNNG의 돌연변이성을 각각 99%와 93%저해효과를 보이므로서 강한 항돌연변이 물질임이 밝혀졌다.

청갓과 적갓에 함유된 Glucosinolates의 항암 활성 및 정량 분석 (Anticancer Activity and Quantitative Analysis of Glucosinolates from Green and Red Leaf Mustard)

  • 청갓과 적갓에서 추출한 GSLs에 의한 4종류의 암세포주를 이용하여 항암 활성을 조사하였다. 또한 갓 추출물에 함유된 GSLs를 HPLC와 LC-ESI/MS로 분석한 결과는 다음과 같다. 1. 갓에서 추출한 GSLs의 항암 활성을 분석한 결과, SNU-354 (간암)>SNU-C4결정암)>SNU-251(난소암)>MCF-7(유방암) 순으로 높게 나타났다. 또한 GSLs 추출물의 처리 시간(24, 48시간)에 따르면 적갓 추출물의 항암 효과가 청갓 추출물의 항암 효과보다 큰 것으로 나타났다. 2. 갓의 주요한 GSLs 성분은 sinigrin이었으며, 청갓이 적갓보다 더 높은 함량을 나타내었으며, 또한 청갓에서만 glucoiberin이 미량으로 검출되었다. 3. 항암 활성 평가에서 적갓 GSLs 추출물이 청갓보다 더 높은 활성을 나타낸 것은 GSLs 물질 이외에 다른 물질들이 관여 했을 것으로 생각되므로 이에 대한 추가적인 연구가 필요할 것으로 사료된다.

김치종류별 식이가 Hairless Mouse 피부의 형태학적 변화에 미치는 영향 (Morphological Changes in the Skin of Hairless Mouse Fed Various Kimchi Diet)

  • 피부는 외부의 물리적 화학적 자극으로부터 신체를 보호하는 중요한 역할을 하며 노화에 따른 조직의 형태학적인 변화를 쉽게 관찰할 수 있는 기관이다. 동결건조한 배추김치, 갓김치, 부추김치를 식이에 10%씩 첨가하여 hairless mouse에 16주간 섭취시켜 피부조직에 미치는 영향을 살펴보았다. 노화가 진행됨에 따라 표피의 두께는 감소하며 본 실험에서 대조군에 비해 김치섭취군에서 유의적으로 두꺼운 것으로 나타났다. 피부 각질화에 관여하는 cytokeratin은 대조군에 비해 갓, 배추, 부추김치군의 순으로 낮았는데 전자현미경적 소견으로도 대조군에서 각질층은 중간원섬유들이 치밀하게 분포된 매우 두꺼운 각질층이 보인 반면 갓김치, 배추김치, 부추김치군은 얇은 각질층으로 배열되어 있었다. 상피기저막의 type Ⅳ 콜라겐은 갓김치섭취군에서 현저하게 증가되어 갓김치 성분들이 피부의 기저막의 노화를 억제한 것으로 사료된다. 콜라겐 함량은 노화에 따라 감소하며 대조군에서 콜라겐의 합성에 관여하는 fibroblast의 RER 발달은 뚜렷하지 않았으나 갓김치군, 배추김치군에서는 RER의 발달이 현저하여 새로운 콜라겐 생성이 활발히 이루어지는 것으로 나타났다. 이러한 결과로 보아 많은 생리활성물질 및 항산화물질이 함유되어진 김치의 섭취는 피부노화를 방지할 수 있을것으로 기대되며 특히 갓김치의 효과가 우수한 것으로 나타났다.

김치 발효에 의한 장내병원균의 생육저해효과 (Inhibitory Effect on the Growth of Intestinal Pathogenic Bacteria by Kimchi Fermentation)

  • 김치 발효가 병원성 장내세균의 생육에 미치는 여향을 조사하기 위하여 배추 및 갓 김치를 제조할 때에 6종의 병원성 장내세균을 접종하여 발효 중의 이들의 변화를 측정하고, 또 김치의 주된 재료에서 검출되는 대장균과 이들 원료로 담근 김치 중의 대장균군 균수의 변화를 측정하였다. 주된 김치 재료 중 대장균군이 가장 많이 검출된 것은 생강과 대파였으며 그 다음이 배추, 고춧가루, 마늘의 순이었다. 김치 중의 대장균군 또는 시험 병원균은 모두 발효의 진행에 의한 pH의 저하에 따라 감소하고 사멸되었다. 대장균군은 배추김치에서 pH 3.9 이하에서 검출되지 않았으며 Clostridium perfringens ATCC 13124, Listeria monocytogenes ATCC 19111, Salmonella typhimurium KCTC 1625, Staphylococcus aureus KCTC 1621, Vibrio parahamolyticus ATCC 27519, 및 Escherichia coli O157 : H7 ATCC 43894는 배추김치에서 pH 4.1, 3.7, 3.8, 3.8, 3.7, 및 3.7 이하에서 각각 검출되지 않았고 갓김치에서는 pH 4.5, 4.0, 4.2, 4.2, 4.2 및 4.1 이하에서 각각 검출되지 않았다. 또 갓 즙액 및 allyl isothiocyanate는 병원균에 대해서 뚜렷한 생육저해효과를 나타내었으며 젖산균에 대한 효과는 미약하였다. 이들 결과로부터 김치 발효는 김치의 안전성을 높이게 되면 갓의 병원균에 대한 뚜렷한 생육저해효과는 주로 갓 중에 함유된 자극성 성분인 allyl isothiocyanate 의 항균작용에 의한 것으로 추정되었다.

  • Glucosinolates(GSLs) are major secondary products($\beta$-thioglucoside N-hydroxysulfates) containing sulfates that are found mainly in Cruciferae family such as cabbage, broccoli, radish, turnip, kale, mustard and rapeseed as the important crops in agronomic and economic aspects. Especially, isothiocyanates(ITCs) have been shown to inhibit carcinogenesis with regulation cancer cell development followed by regulating target enzymes, inducing cell cycle arrest and apoptosis against various human cancer cell lines. Total desulfo(DS)-GSLs of nine type in 210 Korean leaf mustard accessions were isolated and confirmed using electrospray ionization mass spectrometric(ESI-MS/MS) method and DS-GSLs powerful library maked from several reference data. The isolated individual DS-GSLs were identified by removed $\beta$-D-glucopyranose residue ($C_6H_{11}O_5$, MW 163) from MS/MS data. Among them sinigrin(41.7%), glucoiberverin(21.7%) and gluconasturtiin(12.6%) were major components, Especially, despite gluconapin was minor component, accession K046197-1 and K046197-2 showed higher content of 4.11 and 3.31 mg/g(DW), respectively. The total GSLs contents in 210 accessions were ranged from 5.3 to 23.2 mg/g(DW) with a mean value of 13.0 mg/g(DW). As a result of principal component analysis(PCA), the individual GSLs loading plots were composed of three groups and components belonged to each group showed correlationship in quantitative pattern.

갓김치 숙성 중의 무기질, 색소, 조직, 관능 및 미생물군의 변화 (Changes in Mineral, Pigment, Texture, Sensory Score and Microflora during Fermentation of Gat(Leaf Mustard)-Kimchi)

  • In order to obtain basic data for the development of Gat-Kimchi, a salted and fermented lear mustard, changes in mineral, pigment, texture, sensory score and microflora during fermentation at 5$\pm$2$^{\circ}C$ were investigated. Changes in mineral, including iron, calcium and potassium were obviously shown and their contents were markedly decreased after 14 days of fermentation. Contents of total chlorophyll and carotenoid were slowly decreased after 6 and 10 days of fermentation, respectively and ratios of chlorophyll a/b were not changed and similar to those of other cruciferous vegetable Kimchi during fermentation. Shear force of Gat-Kimchi in rheometer during fermentation was increased. The sourness and hardness(p<0.05) of Gat-Kimchi after 24 days of fermentation were significant different in sensory evaluation with no significant difference in off-flavor, color and hotness. Compared with other Kimchi, taste of Gat-Kimchi was desirably kept for 54 days of fermentation. Total viable count and lactic acid bcateria(Genus Lactobacillus) observed to be Increased in the range of 18 to 24 days and yeasts to be gradually increased during overall period of fermentation.

Effect of Introducing Chitinase Gene on the Resistance of Tuber Mustard against White Mold

  • The objective of this research was introduction of chit42 to tuber mustard plants through Agrobacteriummediated transformation against white mold caused by Sclerotinia sclerotiorum. The binary plasmid pGisPEC1 was used in this study. Polymerase chain reaction analysis detected the transgene in 27 transformants with a transformation efficiency of 6.9%. Southern blot test was used to assess the copy number of transgene in tuber mustard plants. One, two, two, and two chit42-related bands were observed in the transformed lines TMB4, TMB7, TMB12, and TMB18, respectively. Enzymatic tests showed a significant increase in the activity of endochitinase in protein isolated from leaf tissues of chit42 transgenic 75-day tuber mustard lines. The pathogenicity of three pathogen isolates was tested on the leaves of transformed plans. The results of current study showed that expression of the gene chit42 in tuber mustard plants markedly reduced infection radius on the leaves 7 days after inoculation with the fungus.

돌산지역 갓김치의 관광 상품에 대한 관광객의 선호도에 미치는 영향 요인 평가 (Assessing Preference for Leaf Mustard Kimchi using Conjoint Model)

  • The aim of this study was to determine the relative importance of a set of attributes which influence purchase decision of leaf mustard kimchi such as color, package, taste, availability and price, and to identify tourist product factor combinations which confer the highest utility to tourists of each age group. Conjoint model was used for this study. The findings from this study were as follows. First, it was found that all age groups regarded taste and price as the very important factor. Second, it was found that the products with different attribute levels were preferred by age groups. Third, it was found that the most preferred product in each age group was significantly different by cluster. Finally, it was found that the most preferred simulation product was different by age groups.

Antimicrobial activities in the Korean Traditional Leaf Mustard, Brassica juncea Coss.

  • As part of developing natural food preservatives, the antimicrobial effect of ethanol and water extracts from the Cruciferous vegetable was examined Korean traditional Brassica juncea Coss. was used widely as an ingredient of Kimchi, a natural flavoring and spice for a long time. Antimicrobial activities were examined against 15 microorganisms which were food-born pathogens and/or food poisioning microorganisms and food-related bacteria and yeasts. Ethanol extract from leaf mustard showed an antimicrobial effect in most of the strains used in the present study. The lowest minimum inhibitory concentration (MIC) were seen in Bacillus subtilis and Bacillus natto at 10 mg/ml. MIC of water extract was 40-60 mg/ml for bacteria and yeast. Antimicrobial activity of the ethanol extract was not disappeared by the heating at $121^{\circ}C$ for 15 min and not affected by pH.
