The influences of temperature and nutritional conditions on the mycelium growth of oyster mushroom Pleurotus ostreatus (PO) and Pleurotus cystidiosus (PC) were investigated in laboratory experiment during the summer season of 2014. The results of the experiment indicated that potato dextrose agar (PDA) and yam dextrose agar (YDA) were the most suitable media for the mycelium growth of oyster mushroom PO while four media (PDA, YDA, sweet potato dextrose agar, and malt extract agar medium) were not significantly different in supporting mycelium growth of oyster mushroom PC. The optimal temperature for mycelium growth of both oyster mushroom species was obtained at $28^{\circ}C$. Mycelium growth of oyster mushroom PO was improved by carbon sources such as glucose, molasses, and at 1~5% sucrose concentration, mycelium colony diameter of mushroom PO was achieved the highest value. Whereas glucose, dextrose, and sucrose as carbon sources gave the good mycelium growth of oyster mushroom PC, and at 1~3% sucrose concentration, mycelium colony diameter of PC was achieved the maximum value. Ammonium chloride concentrations at 0.03~0.09% and 0.03~0.05% also gave the greatest values in mycelium colony diameter of mushroom PO and PC. Brown rice was found to be the most favourable for mycelium growth of two oyster mushroom species. In addition, sugarcane residue, acasia sawdust and corn cob were selected as favourable lignocellulosic substrate sources for mycelium growth of both oyster mushrooms.
The importance of biocomposites has increased owing to the changes in global consumption trends and rapid climate change. Technologies using mushroom mycelium cultivation, and molding methods for mycelial application have gained attention as potential strategies for producing eco-friendly composites. Currently, mushroom mycelia are used as raw materials for food and cosmetics; however, research on their utilization as biocomposite materials is limited. Therefore, the potential for the development of mushroom mycelium-related products and technologies is high. This review analyzes the domestic and international patent application trends related to the technologies for composite (packaging, insulation, adhesives, and leather) and food (substitute for meat) materials using mushroom mycelium, as an eco-friendly biocomposite material, to provide objective patent information that can further research and development (R&D) in this field.
버섯 균사체 혼합배양액을 이용하여 어류질병세균에 대한 항균 및, 항산화활성, 그리고 인체 암세포에 대한 항암 활성을 조사하였다. 항균활성 측정 결과 대부분의 시료에서 항균활성이 있는 것으로 나타났다. 그 중 YM broth에서 배양한 상황버섯과 운지버섯 균사체 배양 추출물이 가장 높은 활성을 띄었지만 10% 감귤추출물배지에서 배양한 상황버섯과 운지버섯 균사체 배양 추출물 또한 비교적 활성이 높아 합성배지가 고가임을 고려할 때 의의가 있다고 생각된다. 항산화 활성의 측정결과 10% 감귤추출물배지에서 배양한 상황버섯, 운지버섯, 꽃송이 버섯 균사체 배양추출물과 PD broth에서 배양한 상황버섯과 운지버섯 균사체 배양 추출물의 경우 1 mg/ml에서 90%에 가까운 라디컬 소거활성을 보였다. 항암활성 측정결과 10% 감귤추출액만을 첨가한 대조군보다 모든 실험구가 높은 활성을 나타냈으며 특히 10% 감귤추출물배지에서 배양한 상황버섯, 운지버섯 배양 추출액과 10% 감귤추출물배지에서 배양한 상황버섯, 운지버섯, 꽃송이버섯 균사체 배양 추출액이 높은 활성을 나타났다. 이러한 다양한 생리활성을 측정한 결과 최종적으로 감귤농축액이 첨가된 버섯균사체 배양 추출물들의 활성이 가장 좋은 것으로 나타났으며, 감귤의 천연배지로서의 이용가능성을 확인할 수 있었다. 따라서 감귤농축액 및 버섯균사체 혼합 배양을 이용한 다양한 사료첨가제 및 신소재의 개발이 가능할 것이다.
복령 발달단계인 균사체(mycelium), 균핵(sclerotium) 및 자실체(fruiting body)에 대한 열수(hot-water), 70% 주정(70% EtOH) 및 70% 메탄올(70% MeOH) 용매별 생리활성 성분과 발달단계별 건조시료의 베타글루칸 및 아미노산 성분함량을 분석하였다. 그 결과, 발달단계별 시료 중에서 균사체의 DPPH 라디컬 소거능, 철 환원 항산화능, 환원력, 아질산염 소거능 및 총 폴리페놀 함량이 균핵과 자실체와 비교하여 유의적 차이를 보이며 높은 것으로 나타났다. 추출용매별 결과, 균사체의 경우 열수추출물(hot-water)에서 DPPH 라디컬 소거능, 철 환원 항산화능 및 총 폴리페놀 함량이 높았으며, 균핵은 70% 주정추출물(70% EtOH)에서 철 환원 항산화능 및 총 폴리페놀 함량이, 열수추출물(hot-water)에서 DPPH 라디컬 소거능, 환원력 및 아질산염 소거능이 높았으며, 자실체는 열수추출물(hot-water)에서 DPPH 라디컬 소거능, 환원력, 아질산염 소거능 및 총 폴리페놀 함량이 다른 용매추출물에 비하여 높게 나타났다. 균사체, 균핵 및 자실체 건조시료 중의 베타글루칸 함량은 균핵에서 균사체와 자실체에 비하여 월등히 높은 함량치를 나타냈다. 총 아미노산 및 총 필수아미노산 함량은 자실체와 균사체에서 균핵과 비교하여 월등히 높았으며, 그 중 알기닌(Arg)과 페닐알라닌(Phe)이 높게 검출되었다.
Mycelium composites and leathers have versatile material properties based on their composition and manufacturing process. To prepare mycelium mat for the production of mushroom leather, several strains were mutated by gamma rays. Some mutant strains, including Lentinula edodes, Ganoderma lucidium, and Schizophyllum commune showed good hyphae growth rate and density on saw-dust media. Irradiation power (Gy), time, and height from the radiation source to the sample were examined. Based on the preliminary data obtained in this study, comprehensive research should be conducted to explore the optimal strains and culture conditions for mycelium-based leather production.
Ganoderma lucidum has been traditionally used as a medicine for treatment of bronchitis, arthritis, and high blood pressure, and it has been reported to display many biological activities including anticancer and immune activities. Since mushroom mycelium is known to have excellent biological activities together with mushroom fruiting body, studies on biological activities of mushroom mycelium have been actively conducted. Thus, the present study compared the biological activities before and after the cultivation of Ganoderma lucidum mycelium on Atractylodes rhizoma. When the radical scavenging activity was assessed by the DPPH assay, ARGL (ethanol extract of Atractylodes rhizoma mycelium fermented with Ganoderma lucidum) showed radical scavenging activity of 5.58~82.56% at concentrations of $10{\sim}500{\mu}g/assay$, while AR (ethanol extract of Atractylodes rhizoma) showed radical scavenging activity of 5.27~72.08% at the same concentrations. When measured by using the ABTS assay, ARGL showed higher radical scavenging activity than AR, which was consistent with the result obtained by the DPPH assay. In the MTT assay, the cytotoxicity of ARGL against all cell lines was higher than that of AR. In particular, the cytotoxicities of AR and ARGL against Hep3B at a concentration of $400{\mu}g/assay$ were 71.81% and 86.40%, respectively. In addition, the result obtained by the SRB assay was consistent with the result obtained by the MTT assay. According to the results mentioned above, there is a high probability that medicinal herb cultures using mycelium can be used as sources of functional foods since the cytotoxicities against cancer cells and antioxidant activities increased when the mycelium was fermented with Atractylodes rhizoma.
기존에 사용되는 1차 봉지배양 및 2차 매트생산 배양에 소요되는 기간을 단축시키기 위해 시루송편버섯 균사체에 Co-60 저준위 감마선 200 Gy 24시간 조사로 돌연변이 시킨 ToM 균사체를 활용하여 봉지배양이 생략된 박스 배양법을 시도하였다. 또한, 매트의 품질 및 물성을 강화시키기 위해 균액으로 ammonium nitrate가 3 g/L첨가된 mYMB6을 사용하였고, CaCO3를 1.5% 함유한 톱밥배지에서 균일한 품질의 균사체 매트 생산이 가능하다는 점을 확인하였으며, 기간 또한 기존의 생산방법과 비교하여 10일 이상 단축할 수 있다. 본 결과는 균사체 기반 친환경 소재 연구에 활용할 수 있을 것으로 생각된다.
본 연구는 곡물(백미, 적미, 흑미)에 상황진흙버섯 균사를 배양하여 곡물과 상황진흙버섯의 유용성분 시너지 효과를 탐색하고자 수행하였으며, 배양조건에 따른 상황진흙버섯 균사 배양곡물의 유리당, 유기산, 유리아미노산, 무기성분, 지방산을 분석하였다. 유리당의 함량을 분석한 결과 fucose, glucose, maltose 총 3종의 유리당이 검출되었으며, 대조구에서는 glucose에서만 검출되었다. 상황진흙버섯을 배양한 곡물들에서 대조구에 비해 높은 유리당 함량이 검출되었다. 상황진흙버섯을 배양한 곡물들의 유기산 함량을 분석한 결과, 4종의 유기산이 검출되었으며 유리당과 마찬가지로 상황진흙버섯을 접종한 시료구에서 대조구에 비해 높은 함량의 유기산이 확인되었다. 상황진흙버섯을 배양한 곡물들과 대조구간의 무기성분 함량 차이는 유의적이지 않았다. 16종의 아미노산이 검출되었으며, 상황진흙버섯을 배양한 곡물들에서 유리아미노산의 함량이 다량 증가함을 확인하였다. 지방산의 결과는 총 7종의 지방산이 검출되었으며 각 대조구와 상황진흙버섯을 배양한 곡물들간에 유의적인 차이는 나타나지 않았지만, 배양 원료로 사용한 백미와 유색미와의 조성에서 많은 차이가 있었다.
Schizophyllum commune has emerged as the most promising model mushroom to study developmental stages (mycelium, primordium), which are two primary processes of fruit body development. Long non-coding RNA (lncRNA) has been proved to participate in fruit development and sex differentiation in fungi. However, potential lncRNAs have not been identified in S. commune from mycelium to primordium developmental stages. In this study, lncRNA-seq was performed in S. commune and 61.56 Gb clean data were generated from mycelium and primordium developmental stages. Furthermore, 191 lncRNAs had been obtained and a total of 49 lncRNAs were classified as differently expressed lncRNAs. Additionally, 26 up-regulated differently expressed lncRNAs and 23 down-regulated between mycelium and primordia libraries were detected. Further, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that differentially expressed lncRNAs target genes from the MAPK pathway, phosphatidylinositol signal, ubiquitin-mediated proteolysis, autophagy, and cell cycle. This study provides a new resource for further research on the relationship between lncRNA and two developmental stages (mycelium, primordium) in S. commune.
본 시험은 재배과정 중 균상배지에서 분리한 세균들의 특성을 조사하여 버섯균의 생육 및 병원균과의 연관관계를 밝히고자 하였다. 정상적으로 버섯을 수확한 농가의 경우 버섯생육초기에 분리한 세균일수록 버섯균주에 대해 저해정도가 감소하였으나, 생육이 진행될수록 저해균의 수가 증가하였으나, 버섯 수확에 실패한 농가의 경우 반대의 경향을 보였다. 또한 합성배지에 분리세균을 도말한 후 느타리버섯 균의 생육을 조사한 결과 정상적 농가의 경우 살균 직후 보다 생육이 진행될수록 저해균의 수가 증가하였지만, 실패한 농가는 오히려 반대의 경향을 보였다. 그러나 톱밥배지에서는 버섯생육초기보다 후기에 분리한 세균이 저해하는 정도가 오히려 낮았다. 또한 이들 분리 세균은 분비성 물질을 통해서 버섯균의 생육과 증식에 영향을 미쳤다. 분리 미생물 중 세균성갈반병의 원인균인 Pseudomonas tolaasii을 강하게 억제하는 미생물은 거의 없었으며 소수의 균주에서 약한 길항력을 보였다. 그러나 세균성갈색무늬병을 일으키는 P. agarici에 대해서는 강한 억제력을 보이는 균주가 많이 존재하였으며, 푸른곰팡이병의 원인균인 Trichoderma harzianum을 강하게 억제하는 미생물도 배지내에 거의 존재하지 않았다. 이와 같이 배지내에는 다양한 미생물이 존재하고 이들 다양한 미생물과 버섯균의 상호작용에 대한 면밀한 연구가 이루어진다면 느타리버섯 재배과정 중 발생하는 다양한 문제점을 해결할 수 있는 좋은 방법이 될 수 있을것으로 생각된다.
