유전공학 Porcine Somatotropin의 투여가 돼지의 건강에 미치는 영향 (Effect of Recombinant Porcine Somatotropin(rPST) Administration on Pig Health)

  • Safety of recombinant porcine somatotropin administration on pig was studied using 32 Landrace x Yorkshire crossbred pigs. The starting body weight ranged from 55.5kg to 65.3kg. Eight pigs were allotted to each low dose group of sustained releasing rPST(SL), high dose group of sustained releasing rPST(SH), daily injection group of rPST(DI), and control group(C). Pigs in SL group and SH group were injected subcutaneously twice in 3 week-interval with 1000$\mu\textrm{g}$ and 2000$\mu\textrm{g}$ of sustained releasing rPST per kg body weight, respectively. Pigs in DI group were injected intramuscularly with 100$\mu\textrm{g}$ of rPST everyday for 6 weeks. Blood was collected from anterior vena cava just before the first treatment, and at four weeks and six weeks of experiment. Hematological parameters and blood chemical parameters indicating liver function, kidney function, electrolyte metabolism, mineral metabolism and lipid metabolism were determined. Necropsy and urinalysis were performed after final blood collection. The results were summarized as follows, and it is concluded that rPST administration does not affect pig health negatively. 1. rPST administration did not affect kidney function as manisfested by BUN, creatinine and urinalysis. 2. rPST administration did not affect liver function as manisfested by total protein, albumin, serum AST activity serum ALT activity serum ALP activity, serum LDH activity, serum GGT activity and serum SDH activity. 3. rPST administration did not affect skeletal muscle, cardiac muscle and brain as manifasted by serum AST activity and serum LDH activity. 4. rPST administration increased blood glucose level within normal range. 5. rPST administration did not affect lipid metabolism as manisfested by triglyceride, cholesterol, and phospholipid concentrati on. 6. rPst administration dia not affect mineral metabolism as manisfested by calcium, phosphorus, magnesium and iron concentration. 7. rPST administration did not affect electrolyte metabolism as manisfested by Na, K, chloride concentration. 8. rPST administration did not affect erythrocyte count, leukocyte count, thrombocyte count, and plasma fibrinogen level.

흰쥐에서 연령이 골격근의 당 대사에 미치는 영향 (Effect of Age on Glucose Metabolism of Skeletal Muscle in Rats)

  • 연령에 따른 내당능의 감소 발생여부를 Sprague-Dawley종 흰쥐 수컷을 실험동물로 하여 연구한 결과는 다음과 같다. 연령에 따른 몸무게 변화는 1개월에서 2개월 사이에는 급격한 증가를 보였으며 2개월에서 4개월까지는 완만한 증가를 그 후 8개월까지는 미미하였다. 절식상태에서 혈당량(mg/dl)은 성장기군에서 $92{\pm}8.9$였으며 성숙기군에서는 $106{\pm}13.6$으로 성장기군에 비해 높았으며 인슐린 농도 또한 성숙기군에서 높았다. 체중 100mg당 180mg의 당을 부하한 내당능 검사에서도 성숙기군에서는 성장기군에 비해 당내성의 감소현상을 나타내었다. 당부하 검사시 동시에 측정한 혈장 인슐린 농도는 첫 30분에는 성장기군에서 높았으나, 120분에는 성숙기군에서 높은 값을 나타내었다. Soleus근의 당 섭취능은 성숙기군에서 성장기군보다 낮았으나 통계적으로 유의한 차이는 없었다. 인슐린 첨가에 의한 당 섭취능의 변화양상은 성숙기군과 성장기군 사이에 특별한 차이를 발견할 수 없었다.

조기 이유한 흰쥐에서 유단백질의 섭취수준과 조성비가 기관성장과 단백질대사에 미치는 영향 (Effects of Milk Protein levels and Casein/Whey Ratios on Organ Growth and Protein Metabolism in Early Weaned Rats)

  • This study was conducted to investigate the effects of protein levels and casein/whey ratios on organ growth and protein metabolism in early weaned rats. Premature rats weaned by the 17th day were fed six semipurified synthetic, isocaloric and gel diets that contained three levels (low, medium and high) and two different combinations(casein/whey ; 80 : 20 or 20 : 80) of milk protein for 8 days. On the 25th day postpartum, frest weigth and DNA, RNA and milk protein contents in brain, liver, kidney and muscle were determined to ascertain organ and cellular growth. Futher, with a view to ascertain protein metabolism and renal functions, serum total protein, $\alpha$-amino N, urea N, and creatinine and creatinine and urinary urea N, creatinine and hydroxproline were determined. Total DNA contents of brain, liver and kidney, which may represent as an index of cell numbers in those organs were significantly decreased in the rats fed diets containing low level protein regardless of casein/whey ratio. However, as fat as the rats fed high protein diets were concerned, their fresh weight, protein contents and GFR of kidney were significantly increased. Furthermore, nitrogen components, $\alpha$-amino N, urea N and creatinie in serum and urine were also increassed. Another observation was that high casein/whey ratio significantly facilitated accumulation of porteins in muscle and kidney and urinary hydorxyproline excretion, not affecting the DNA content of those organs. This study showed that low(8%) or high(32%) contents of protein had less desirable effects either on protein metabolism or on organ cellular growth in prematurely weaned rats, whereas there were no effects on general growth and bone strength.

Identification of the Food Sources-Metabolism of the Pacific Oyster Crassostrea gigas using Carbon and Nitrogen Stable Isotopic Ratios

  • In order to understand food sources-metabolism for the pacific oyster (Crassostrea gigas), the stable isotope ratios of carbon (${\delta}^{13}C$) and nitrogen (${\delta}^{15}N$) of its gut, gill, and muscle as well as potential food sources (particulate organic matter, sedimentary organic matter, benthic microalgae, seagrass detritus) were determined in Dongdae Bay. Average ${\delta}^{13}C$ and ${\delta}^{15}N$ values reflect that oysters primarily fed on sedimentary organic matter as opposed to suspended organic matter during summer and winter seasons. However, the relatively enriched $^{15}N$ values of particulate organic matter (>$250{\mu}m$) and sedimentary organic matter in the summer may be due to the photosynthetic incorporation of $^{15}N$-enriched nitrogen (DIN) or the spawning events of bivalves. Specific oyster tissues (gut, gill, and muscle) revealed different metabolic pathways, which were determined through analysis of ${\delta}^{13}C$ and ${\delta}^{15}N$ in each organ. The present results suggest the determination of carbon and nitrogen stable isotopes to be a useful approach in ecological research related to the food sources- metabolism of Crassostrea gigas.

Ginsenoside Rg5 promotes muscle regeneration via p38MAPK and Akt/mTOR signaling

  • Background: Skeletal muscles play a key role in physical activity and energy metabolism. The loss of skeletal muscle mass can cause problems related to metabolism and physical activity. Studies are being conducted to prevent such diseases by increasing the mass and regeneration capacity of muscles. Ginsenoside Rg5 has been reported to exhibit a broad range of pharmacological activities. However, studies on the effects of Rg5 on muscle differentiation and growth are scarce. Methods: To investigate the effects of Rg5 on myogenesis, C2C12 myoblasts were induced to differentiate with Rg5, followed by immunoblotting, immunostaining, and qRT-PCR for myogenic markers and promyogenic signaling (p38MAPK). Immunoprecipitation confirmed that Rg5 increased the interaction between MyoD and E2A via p38MAPK. To investigate the effects of Rg5 on prevention of muscle mass loss, C2C12 myotubes were treated with dexamethasone to induce muscle atrophy. Immunoblotting, immunostaining, and qRT-PCR were performed for myogenic markers, Akt/mTOR signaling for protein synthesis, and atrophy-related genes (Atrogin-1 and MuRF1). Results: Rg5 promoted C2C12 myoblast differentiation through phosphorylation of p38MAPK and MyoD/E2A heterodimerization. Furthermore, Rg5 stimulated C2C12 myotube hypertrophy via phosphorylation of Akt/mTOR. Phosphorylation of Akt induces FoxO3a phosphorylation, which reduces the expression of Atrogin-1 and MuRF1. Conclusion: This study provides an understanding of how Rg5 promotes myogenesis and hypertrophy and prevents dexamethasone-induced muscle atrophy. The study is the first, to the best of our knowledge, to show that Rg5 promotes muscle regeneration and to suggest that Rg5 can be used for therapeutic intervention of muscle weakness and atrophy, including cancer cachexia.

Biotechnologies for Improving Animal Metabolism and Growth - A Review

  • Biotechnology will play critical role in improving animal productivity. Animal growth rate and muscle deposition potential can be greatly improved by the application of biotechnology and biotechnological products. Administration of recombinant somatotropin (ST) or other compounds such as IGF-1 and growth hormone-releasing peptides (GHRPs) can enhance growth rate and carcass lean percentage. Gene transfer offers a powerful approach to manipulate endocrine system and metabolic pathways toward faster growth and better feed efficiency. Biotechnology is also extensively used for improving metabolism and activity of gut microorganisms for better nutrient digestibility. Knockout of growth-inhibiting genes such as myostatin results in considerable acceleration of body weight and muscle growth. Animal growth can also be improved by the use of gene therapy. Immunomodulation is another approach for efficient growth through controlling the activity of endogenous anabolic hormones. All the above aspects will be discussed in this review.

Effects of Brazilin Glucose Metabolism in Isolated Soleus Muscles from Streptozotocin Induced Diabetic Rats

  • The present study was performed to evaluate the hypoglycemic mechanism of brazilin. Brazilin significantly reduced plasma glucose level in streptozotocin induced diabetie rats and this effect seems to be mediated by extrapancratic effects rather than by pacreatic effect because no significant changes were observed in plasma insulin levels. The rates of glycogen synthesis, glycolysis and glucose oxidation in soleus muscle were markedly increased following brazilin treatment to diabetic animals. Glucose transport seemed to be increased by the treatment of brazilin. Brazilin did not affect insulin binding to muscles from streptozotiocin induced diabetic rats. These results suggest that potentiation of periopheral glucose utilization may be one of the major causes of hypoglucemic action of brazilin.

식이 중 아연과 단백질의 수준이 성숙쥐의 지질대사에 미치는 영향 (Effect of Different Dietary Zinc and Protein Levels on Lipid Metabolism)

  • The effect of different levels of Zn (0, 30, 300ppm) and protein(7, 20, 40%) in the diet upon lipid metabolism was investigated in Sprague-Dawley male rats weighting 180.54$\pm$29.08g(n=450 fed one of nine diets in a 3$\times$3 factorial design for 5 weeks. The reults obtained were summarized as following. 1) Total lipid contents in serum and liver were tended to be lower in LZn group than CZn and Hzn groups. Those of LP group were higher than CP and HP groups. 2) HDL-cholesterol and total cholesterol contents in serum were significantly affected by dietary Zinc level and were increased as dietary Zinc level increase. 3) Total cholesterol in liver and muscle were tended to be decrease as dietary Zinc level increase. Those in LP group were higher than CP and HP groups. 4) Zinc contents in plasma, liver, muscle and testis were tended to be lower in LZn group than CZn and HZn groups. 5) Protein contents in plasma and liver lower in LZn group than CZn and HZn groups when dietary protein level was 7% and 20%. Those in LP group were lower than those in CP and HP groups. 6) Cu contents in plasma, liver, muscle and testis were tended to be decreased as dietary Zinc level increase.

Expression of Tumor Necrosis Factor (TNF)-z${\alpha}$ from Cells Undergoing Death by FADD

  • Apoptosis of vascular smooth muscle cell is observed in the vascular diseases such as atherosclerosis and restenosis. The death of vascular smooth muscle cells can be induced by cytokines and activation of Fas-pathways. It is widely accepted that apoptosis occurs without inflammation. There are, however, reports that apoptosis is not silent. Vascular smooth muscle cells dying by Fas-pathway secreted inflammatory cytokines including monocyte chemoattractant protein-1. This study have investigated whether apoptosis is associated with potent inflammatory cytokine tumor tumor necrosis factor (TNF)-${\alpha}$. The cells which undergo apoptosis by expressing FADD in the absence of tetracycline expressed and secreted TNF-${\alpha}$. When the level of TNF-${\alpha}$ transcript was investigated, dying smooth muscle cells exhibited transcriptional activation of TNF-${\alpha}$. The data indicate that dying vascular smooth muscle cells contribute to inflammation by expressing inflammatory cytokines. The present study suggests that apoptosis could not be silent in certain pathological situations.
