미강에서 추출한 식이섬유 첨가가 유화형 소시지의 품질 특성에 미치는 영향 (Effects of Dietary Fiber from Rice Bran on the Quality Characteristics of Emulsion-type Sausages)

  • 본 연구는 미강 추출 식이섬유 혼합물을 첨가한 유화형 소시지의 이화학적 및 관능적 품질 특성을 조사하였다. 유화형 소시지의 수분함량은 미강 추출 식이섬유를 2%와 3% 첨가한 처리구에서 유의적으로 높았으며, 지방함량은 대조구가 가장 높았고 식이섬유 혼합물의 첨가량이 증가함에 따라 낮게 나타났다. 단백질 함량은 대조구가 높게 나타났으나, 회분함량은 대조구에 비해 처리구들이 높게 나타났다. 가열감량은 대조구에 비하여 처리구들이 낮게 나타났으며, 2% 처리구가 유의적으로 가장 낮은 가열감량을 나타내었다. 소시지의 pH, 명도와 적색도는 대조구가 처리구들과 비교하여 높게 나타났으며, 황색도는 미강 식이섬유의 첨가량이 증가할수록 높은 값을 나타내었다. 물성의 경도는 미강 식이섬유의 첨가량이 증가할수록 높은 경도를 나타내었고, 탄력성, 검성, 씹음성은 모든 처리구에서 유의적인 차이가 나타나지 않았다. 관능적 특성은 색과 연도에서 대조구, 1%, 2% 미강 식이섬유 첨가구가 높은 점수를 받았으며, 풍미, 다즙성 및 전체적인 기호도에서 대조구와 비교하여 2% 처리구가 유의적으로 가장 높은 점수를 받았다. 따라서, 유화형 소시지에 미강 추출 식이섬유 혼합물을 첨가하면 이화학적 및 관능적으로 우수한 육제품을 제조할 수 있으며, 기능성 소시지에 대한 미강의 활용 가능성을 타진하였다.

마사지가 좌골신경 절단 마우스의 골격근에 미치는 영향 (Effect of Massages on The Skeletal Muscles of Sciatic Nerve Neurectomized ddY Mice)

  • To demonstrate the effect of massages on the skeletal muscles of immobilization ddY mice models induced by right side sciatic nerve neurectomy, the cross sectional histological profiles of the muscularis (M) gastrocnemius, M. tibialis cranialis and M. tibialis caudalis were observed after 28 days of treatment of massages with the changes of body weight, thickness of hind limb and individual muscle weights. In addition, changes of serum aspartate amino- transferase (AST) and creatine phosphokinase (CK) levels were also demonstrated with diameter of individual muscle fiber and muscle fasciculata, and number of muscle fiber in each of three types of muscles located in the calf. The experimental groups were divided into five groups, 1) Sham-operated group (Sham), 2) Neurectomized but not massage control group (Control), 3) Neurectomized and massage at knee joint regions (T1), 4) Neurectomized and massage at calf regions (T2), and 5) Neurectomized and massages at achilles tendon regions (T3). The experimental animals were used 5 per groups. The results are as follow: 1. No significant changes of body weight were demonstrated in this groups during whole experimental periods compared to that of Sham group. In addition, no significant changes of body weight gains during experimental periods (Day 0 - Day 27: 28 days of treatment) were also observed. 2. The thickness of intact left side hind limb was not changes in all tested groups compared to that of Sham group. However, the thickness of right side hind limb were remarkedly decreased in Control, T1, T2 and T3 groups compared to that of Sham group. In addition, the differences of thickness of hind limb between right and left sides were also significantly increased in Control and massage groups (T1, T2 and T3) compared to that of Sham group. However, the thickness of right side of hind limb were significantly increased and the differences of thickness between both sides of hind limb were dramatically decreased in massage groups compared to that of Control group in followed order: T1 > T3 > T2. 3. Absolute and relative weight of M. gastrocnemius in the intact left side of hind limb was not changes in all tested groups compared to that of Sham group. However, the weight of right side M. gastrocnemius were remarkedly decreased in Control, T1, T2 and T3 groups compared to that of Sham group. In addition, the differences of weight of M. gastrocnemius between right and left sides were also significantly increased in Control and massage groups compared to that of Sham group. However, the weight of right side M. gastrocnemius were significantly increased and the differences of weight between both sides of M. gastrocnemius were dramatically decreased in massage groups compared to that of Control group in followed order: T3 > T1 >. 4. Absolute and relative weight of M. tibialis cranialis in the intact left side was not changes in all tested groups compared to that of Sham group. However, the weight of right side M. tibialis cranialis were remarkedly decreased in Control, T1, T2 and T3 groups compared to that of Sham group. However, the weight of right side M. tibialis caudalis were remarkedly decreased in Control, T1, T2 and T3 groups compared to that of Sham group. In addition, the differences of weight of M. tibialis caudalis between right and left sides were also significantly increased in Control and massage groups compared to that of Sham group. However, the weight of right side M. tibialis caudalis were significantly increased and the differences of weight between both sides of M. tibialis caudalis were dramatically decreased in massage groups compared to that of Control group in followed order: T3 > T1 > T2. 6. A significant increase of serum AST levels were demonstrated in Control group. However, serum AST levels of massage groups were significantly decreased compared to that of Control group in followed order: T3 > T1 > T2. In case of 73 group, the values decreased into similar levels compared to that of Sham group. 7. A significant increase of serum CK levels were demonstrated in Control group. However, serum CK levels of massage groups were significantly decreased compared to that of Control group in followed order: T3 > T1 > T2.

Development of Analysis Condition and Detection of Volatile Compounds from Cooked Hanwoo Beef by SPME-GC/MS Analysis

  • The current study was designed to optimize solid phase microextraction (SPME)-GC-MS conditions for extraction and analysis of volatile components for Hanwoo beef and to establish a tentative database of flavor components. Samples were taken from Hanwoo longissimus muscle (30 mon old steer, $1^+B$ carcass grade) at 24 h postmortem. Results indicated that the optimum adsorption time for $75{\mu}m$ CAR/PDMS fiber was 60 min at $60^{\circ}C$. Thermal cleaning at $250^{\circ}C$ for 60 min was the best practice for decontamination of the fiber. A short analysis program with a sharp oven temperature ramp resulted in a better resolution and higher number of measurable volatile components. With these conditions, 96 volatile compounds were identified with little variation including 22 aldehydes, 8 ketones, 31 hydrocarbons, 12 alcohols, 8 nitrogen- and sulfurcontaining compounds, 5 pyrazines and 10 furans. A noticeable observation was the high number of hydrocarbons, aldehydes, ketones, alcohols and 2-alkylfurans which were generated from lipid decomposition especially the oxidation and degradation of unsaturated and saturate fatty acids. This implies that these compounds can be candidates for flavor specification of highly marbled beef such as Hanwoo flavor.

Effects of Morphine on Somatosyrnpathetic Reflex and Arterial Blood Pressure Response Evoked by Stimulation of Peripheral Nerves

  • In the present study, the relationship between the somatosympathetic reflexes and arterial blood pressure responses to electrical stimulation of the peripheral nerve was investigated in cats anesthetized with ${\alpha}-chloralose$. Single sympathetic postganglionic fiber activities were recorded from the hindlimb muscle and skin nerves and also from the cervical and abdominal sympathetic chains. Effects of the morphine on responses of the sympathetic nerve and arterial blood pressure to activation of the peripheral $A{\delta}-$ and C-afferent nerves were analyzed. The following results were obtained. 1) Arterial blood pressure was depressed by peripheral AS-afferent stimulation (A-response) and was elevated during C-afferent activation (C-response). 2) Intravenously administered morphine enhanced the C-response while the A-response decreased insignificantly, Only the C-response was decreased by intrathecal morphine. 3) All the ten recorded cutaneous sympathetic fibers showed periodic discharge pattern similar to respiratory rhythm and five of them also showed cardiac-related rhythm. However, most of the muscular sympathetic fibers had cardiac-related rhythm and only four fibers showed respiratory rhythm. 4) Morphine decreased the sympathetic C-reflex elicited by the peripheral C-afferent activation and the abdominal sympathetic A-reflex was also decreased by morphine. From the above results, it was concluded that supraspinal mechanisms were involved in the enhanced arterial pressor response to peripheral C-afferent activation by intravenous morphine.

사료 내 단백질 함량 및 합성아미노산 첨가에 따른 육계의 사양성적 및 가슴육 성상에 미치는 영향 (Effects of Reduced Dietary Crude Protein Diet with Synthetic Amino Acid Supplements on the Growth Performance and Histochemical Characteristics of the Breast Meat of Male Broilers)

  • The present study was conducted to determine the effect of reduced dietary crude protein (CP) with a synthetic indispensable amino acids (AA) supplement on the growth performance, economics, and breast meat quality. A total of 450 male broilers (Ross 308) were used from the age of 7 to 28 days. On d 7, the birds were individually weighed and randomly assigned to three treatment diets with six replicate pens for each treatment in a randomized complete block design. The experimental diets were: (1) a control diet, (2) a diet with a reduction of 1% of the dietary CP with synthetic AA supplements to meet the indispensable AA requirement (OAA) of broiler chickens, and (3) a diet with a reduction of 1% of the dietary CP with synthetic Lys, Met, and Thr supplementation to exceed 5% of the indispensable AA requirement (HAA). There were no differences among the treatments on the final body weight, weight gain, and feed intake. However, the HAA diet impaired the feed conversion ratio at d 21 and during the overall feeding periods (P<0.05). The predicted total feed intake and feed price required to reached 1.5 kg BW was higher than for the birds fed the HAA diet (P<0.05). The breast meat muscle fiber cross sectional area and fiber density varied between the treatments (P<0.05). However, there were no differences in breast meat weight. In conclusion, reducing 1% of dietary CP had no adverse effects on the growth performance or breast meat yields.

액정 엘라스토머 섬유의 이해와 연구동향 (Understanding and Research Trends in Liquid Crystal Elastomer Fibers)

  • 액정 엘라스토머 섬유는 1차원 형태로서 소프트 로봇, 생체모방 구동기 등의 다양한 분야에서 광범위하게 응용되고 있다. 액정 엘라스토머는 액체의 유동성과 고체의 질서도 그리고 고무의 탄성을 포함하며 이를 바탕으로 한 자극-응답성을 가지고 있다. 특히, 형상가변 측면에서 열, 빛, 전기장, 자기장 등의 다양한 자극에 대한 응답성을 프로그래밍하여 사용하면 높은 자유도와 더불어 물건 들어올리기, 꼬임, 회전 등 다양한 움직임을 구현할 수 있다. 따라서, 액정 엘라스토머 섬유는 인공근육, 소프트로봇, 웨어러블 기술, 센싱 기술 등 다양한 분야로의 응용가능성을 가지고 있다. 이런 액정 엘라스토머 섬유의 연구는 기존 단순 섬유를 넘어서는 다양한 기능성을 포함할 수 있는 스마트 소재로서 도래한 4차 산업에서 다양한 분야에 활용도가 높다고 평가된다. 본 총설에서는 액정 엘라스토머 섬유의 구조 및 기본 특성에 대해 소개하고, 배향 기반 제작법과 이를 이용한 인공근육, 스마트 패브릭, 소프트 로봇 등, 다양한 응용에 대해 최신 연구 동향을 소개한다.

Effect of guanidinoacetic acid on the growth performance, myofiber, and adenine nucleotide of meat-type rabbits

  • Objective: This study aimed to investigate the effect of dietary guanidinoacetic acid (GAA) on the growth performance, slaughter traits, myofiber, and adenine nucleotide of meat-type rabbits. Methods: Experimental treatments consisted of control (CON) and GAA addition at 0.04% (T1), 0.08% (T2), and 0.12% (T3) of diet. A total of 240 weaned rabbits (meat-type male Chinese black rabbits) were randomly distributed into four groups with six replicates of ten rabbits each. Results: Results showed that the three doses of GAA increased (p<0.05) final body weight, carcass weight, the density and area of quadriceps femoris fiber; and T3 showed significant effects (p<0.05) on weight gain, feed/gain, and dressing percentage, and the traits of longissimus fiber, compared to CON. Dietary GAA increased (p<0.05) the meat color a* and b* in longissimus and quadriceps; and T3 showed the lowest (p<0.05) shear force of longissimus. Furthermore, GAA increased (p<0.05) the contents of adenosine triphosphate and total adenine nucleotide in longissimus and quadriceps. In longissimus adenosine triphosphate, total adenine nucleotide, and adenylate energy charges, T3 treatment was most effective (p<0.05); while T2 and T3 treatment was more effective (p<0.05) than T1 in quadriceps. Additionally, linear or quadratic responses (p<0.05) to the increased doses of GAA were found on body weight gain, meat color, total adenine nucleotide, and adenylate energy charges. Conclusion: It is concluded that GAA can be used in the rabbit diet to improve growth and carcass traits, and these are related to the high levels of muscle adenine nucleotide.

흰 쥐의 뒷다리 현수 후 집중 체중부하 트레드밀 훈련과 전기자극이 골격근 특성에 미치는 효과 (Effects of Intensive Weight Bearing Treadmill Training and Electrical Stimulation on Skeletal Muscle Properties in Hindlimb Suspended Rats)

  • The purpose of this study was to investigate the treatment effect of three interventions on the disuse atrophy of rat hindlimb after two weeks suspension. Forty-eight 11~12 weeks old female Sprague-Dawley white rats were divided into four intervention groups: 1) suspension only (S; n=10), 2) intensive weight bearing treadmill (IWBT; n=10), 3) electrical stimulation (ES; n=9), 4) 2)+3) (ES/IWBT; n=9). Another 10 rats received no intervention or hindlimb suspension and served as controls (C). After the interventions, 1) the cross-sectional area (CSA), 2) the ratio of white muscle fiber composition (WMFC), 3) isometric tetanic tension (ITT), and 4) muscle weights (MWs) were measured from the four calf muscle specimens. The results were as follows: 1. In all intervention groups, the CSAs of medial and lateral gastrocnemius (MG LG), soleus (SOL), and flexor digitorum superficialis (FDS) decreased when compared to the control (C) group (p<.05). The CSA increased in FDS and LG for the IWBT group, in SOL for the ES/IWBT group compared to the S only group (p<.05). 2. The ratios of WMFC in MG, LG, SOL, and FDS increased compared to the C group for all interventions (p<.05). The ratios of WMFC decreased in SOL and LG for the IWBT group, in SOL for the ES/IWBT group compared to the S only group, and decreased in SOL for the ES/IWBT group compared to the IWBT group (p<.05). 3. The ITT in the MG, LG, SOL, and FDS decreased compared to the C group for all interventions (p<.05). The ITT increased in MG LG/FDS, SOL, and the whole calf muscles (WCMs) in the IWBT, ES and ES/IWBT groups compared to the S only group (p<.05). 4. The MWs in MG LG/FDS, SOL, WCMs decreased compared to the C group for all interventions (p<.05). The MWs increased in MG LG/FDS and WCMs for the IWBT group, in SOL for the ES group, and in SOL for the ES/IWBT group compared to the S only group (p<.05). 5. In atrophied muscles, the IWBT group showed the best recovery and the ES/IWBT and ES groups followed in decreasing order. The most susceptible muscle to disuse atrophy was the SOL. But conversely, it showed the best recovery in the ES/IWBT group. After two weeks of hindlimb suspension, the calf muscles of rats atrophied and their isometric tension decreased. These changes were best reversed by hindlimb-focused treadmill activity. The next best results were achieved by electrical stimulation combined with the treadmill followed by only electrical stimulation. These findings indicate that full weight bearing treadmill activity alone or in combination with electrical stimulation are effective treatments for non-weight bearing induced muscle atrophy. Further study of the effect of different intensities of electrical stimulation and variations in the duration period of full weight bearing treadmill activity on disuse atrophy is recommended.

한국산 겨우살이(Viscum album coloratum)로부터 추출된 lectin의 돼지에 대한 독성 및 오제스키병 백신의 면역원성에 미치는 영향 (Toxicity of lectin extracted from Korean mistletoe (Viscum album coloratum) in piglets and its effects on the immunogenicity of Aujeszky's disease virus vaccines)

  • In the present study toxicity and immunostimulating activity of the lectin(KML-C), which was extracted from Korean mistletoe(Viscum album coloratum) were investigated in swine. To determine the toxicity, lectin was injected into thigh or cervical muscles of 4-week-old piglets(Landrace) and observed clinically and pathologically. For determination of the immnunostimulating activity, lectin($0.7{\mu}g/kg$ of body weight)-adjuvanted vaccine of Aujeszky's disease virus(ADV)(NYJ1-87) which was inactivated by 0.2% formalin was injected into the cervical muscle of antibody-negative piglets in the same age group. Subpopulation of the immune cells and serum neutralizing(SN) antibodies in the piglets were examined after vaccination, and resistance of the piglets against challenge by virulent NYJ1-87 was further examined. The results were also compared with those from piglets injected with aluminum hydroxide [$Al(OH)_3$]-adjuvanted vaccine of inactivated NYJ1-87 and NYJ1-87 vaccine without adjuvant, and the results are as follows. By injection of lectin with $30{\mu}g/kg$ of body weight to the thigh muscle, all of 12 piglets died after signs such as dyspnea, fever, systemic erythema and subcutaneous hemorrhages, and lesions pertaining to poisonous hepatitis and dysfunction of kidney were observed. By injection of lectin with $7{\mu}g/kg$ of body weight to the thigh muscle, all of 12 piglets showed signs such as edema and cutaneous hemorrhage in the injected area, lameness and depression, and lesions pertaining to poisonous hepatitis and dysfunction of kidney were observed. By injection of lectin with 1, 3 and $5{\mu}g/kg$ of body weight to the thigh muscle of each one piglet, signs such as congestion, induration and grayish coloration in the injected area, depression and inappetence were observed in all piglets. Toxic changes were also observed in the liver and kidney of piglets by lectin of 3 and $5{\mu}g$. By injection of lectin with 0.5 and $0.7{\mu}g/kg$ of body weight to the cervical muscle of each 9 piglets, all piglets were clinically normal and there were no significant changes in blood counts and chemistry values. Whereas, epithelial swelling and vacuolation of convoluted tubules were observed from one piglet injected with lectin of $0.7{\mu}g$, and necrosis and fibrosis of muscular fiber were observed in the muscle of one piglet injected with lectin of $0.5{\mu}g$. Only population of sIgM+ B lymphocytes increased among immune cells in all of 15 piglets immunized with lectin($0.7{\mu}g/kg$ of body weight)-adjuvanted vaccine, while compared to those in $Al(OH)_3$-adjuvanted vaccine and vaccine without adjuvant. No additional stimulation to the immune cells was recognized when lectin was added to $Al(OH)_3$-adjuvanted vaccine. In piglets immunized with lectin-adjuvanted vaccine, SN titers in reciprocal values for loge were 1.3-4.0 at 1-4 weeks after vaccination, which was similar to those with 1.0-3.3 by vaccine without adjuvant but lower than those with 2.0-5.7 by $Al(OH)_3$-adjuvanted vaccine. Also, no additional increase in the SN titers was recognized when lectin was added to $Al(OH)_3$-adjuvanted vaccine. Piglets immunized with lectin-adjuvanted vaccine were resistant to challenge by the virulent NYJ1-87 at 4 weeks after vaccination, and the SN titers reached to 5.0 one week after challenge, which was higher than those with 4.0 by vaccine without adjuvant but somewhat lower than those with 7.7 by $Al(OH)_3$-adjuvanted vaccine.

한우 c-fos 유전자의 염기서열 및 발현분석 (Sequence and Expression Analysis of c-fos Proto-oncogene in Korean Cattle (HANWOO))

  • c-fos 유전자는 전사조절인자로서 주로 c-jun family와 결합하여 heterodimers를 형성하며 AP-1 조절 부위를 가지는 유전자들의 전사를 조절하는 것으로 알려져 있다. 이 유전자의 발현은 myoblasts를 비롯한 여러 세포의 분화와 성장을 조절하며 최근 돼지에서 육질에 영향을 미치는 근섬유와 관련된다는 보고가 있다. 본 연구는 소에서 육질과 c-fos 유전자와의 관계를 알아보기 위한 기초자료로서 총 1,443 bp의 mRNA 염기서열을 최초로 소에서 밝혔으며 여러 조직과 기관에서의 발현양상도 살펴보았다. 한우의 c-fos 유전자의 염기서열을 사람, 돼지 및 쥐와 비교하여 본 결과 각각 89.8%, 93.5%와 87.0%의 높은 상동성을 보였다. 이 유전자의 발현은 근육중 갈비에서 가장 많은 발현량을 보였고, 조직에서는 비장에서 가장 많은 발현량을 보이는 것을 알 수 있었다. 이 연구에서 밝혀진 c-fos 유전자는 SNP의 추가분석에 의해 한우에서 육질의 향상과 관련이 있는 후보유전자로 쓰일 수 있을 것으로 사료된다.
