ON FARM DEMONSTRATION OF VARIOUS STORAGE METHODS FOR UREA TREATED WHEAT STRAW

  • On farm demonstration of urea treatment (5 kg urea dissolved in 60 litre water/100kg) of straw was performed at 6 different sites and treated straw was stored by three different methods i.e., plastic covered, mud plastered and existing farmers technique (mud plastered on the top and open from sides) to determine the best storage method in field. Untreated and treated samples were taken after 5 week storage period and subjected to crude protein, crude fibre and cell wall constituents analysis. In situ dry matter digestibility of straw was measured by nylon bag technique in buffalo bulls. Crude protein content increased by 100 to 153 percent in treated straw stored by different methods. Maximum increase in crude protein of treated straw was noticed in mud plastered method. Urea treatment of straw resulted in significant decrease in crude fibre contents in all the storage methods. Treatment of straw enhanced the in situ digestibility by 25-49 percent and maximum digestibility (53%) was found in mud plastered storage method. It was concluded that the mud plastered storage method for urea treated straw was found to be the best at farm level.

  • This study investigated the application of red muds, which were industrial wastes fired at 800℃, with a coloring agents. The results were summarized as following. The slumpflow, air content and unit weight volume were satisfied with each target values. The setting time was shortened on the case that 3 % of the red coloring agent and fired red mud were simultaneously replaced about 1 hour compared with the previous study which was 6 % of the red coloring agent was individually used. For the length change ratio caused by drying shrinkage and depth of neutralization on hardened concrete, they were declined when 3 % of the red coloring agent and fired red mud were simultaneously replaced compared with the previous study. On the measurement of forming colors, the case that 3 % of the red coloring agent and fired red mud were simultaneously replaced was similar to the previous study.

Preliminary Study of Seed Production of the Micronesian Mud Crab Scylla serrata (Crustacea: Portunidae) in Korea

  • Seventeen females of the mud crab Scylla serrata, from the State of Kosrae, Micronesia, were transported to the Fisheries Resources Research Institute, Gyeongsangnam-do, in oxygen-filled plastic bags. After acclimatization to a $30^{\circ}C$ holding temperature, nine females were selected for seed production trials. Spawning was hastened using eyestalk ablations; however, this may not be required in commercialscale mud crab seed production. Primary spawning produced an average of 2.4 million hatched larvae, whereas secondary spawning produced 0.4 million. About 10 days elapsed between spawning and hatching and 30 days between hatching and crablet. Mass mortalities up to 90% were observed between stages zoea 1 and zoea 2 in every trial. The highest survival rate from zoea 1 to crablet was estimated at 0.25%. Most commercial shrimp hatcheries in Korea are equipped with almost all necessary facilities and could be converted easily to mud crab hatcheries, able to run three to four times per year using hatchery technologies developed for blue crabs and Chinese mitten crabs.

  • The intertidal flat depositional environment of Southern Nam Yang Bay, west coast of Korea has been studied to understand textural, geochemical and mineralogical characteristics. The intertidal flat environment can be divided into two subenvironments, that is, the mud flat and the sand flat due to the sediment textures. From thd outer sand flat to the inner mud flat the grain size of the sediments decreases and the mud content increases. It is suggested that the intertidal flat environment is in the progradation of marsh deposits in the mud flat. The chemical composition of the sediments is related to the sediment textures. The chemical index of maturity of the mud flat sediments is higher than that of the sand flat sediments. The clay minerals of the sediments are chlorite, illite, montmorillonite and kaolinite.

Characterization and expression profiles of aquaporins (AQPs) 1a and 3a in mud loach Misgurnus mizolepis after experimental challenges

  • Two distinct cDNAs encoding aquaporins (mmAQPs 1a and 3a) were isolated and characterized from mud loach Misgurnus mizolepis. The identified mud loach AQP cDNAs encode for polypeptides of 260 and 302 amino acids. Topology predictions confirmed six putative membrane-spanning domains connected by five loops and the N- and C-terminal domains being cytoplasmic. The mud loach AQPs 1a and 3a showed broad distribution in multiple tissues including immune-responsive tissues as well as osmoregulatory tissues. Hence, the diversity of AQP distribution and expression possibly indicated its differential functions in the regulation of fluid movement in response to environmental stimuli. The transcription of mmAQP genes was differentially modulated by immune challenges. In particular, the mmAQP3a expression level in the liver was more responsive to immune challenges than that of mmAQP1a. Taken together, fish stimulation or infection resulted in significant modulation of mud loach AQP genes, suggesting potential functional roles of these proteins in piscine pathophysiological process.

Removal of Phenol from Aqueous Solutions by Activated Red Mud: Equilibrium and Kinetics Studies

  • In this work, removal of phenol from aqueous solutions by activated red mud was investigated. Scanning electron microscopy and energy dispersive X-ray spectroscopy was used to observe the morphology and surface components of activated red mud, respectively. The effects of various parameters on the removal efficiency were studied, such as contact time, pH, initial phenol concentration, and adsorbent dosage. The removal percentage of phenol was initially increased, as the solution pH increased from 3 to 7, and then decreased above neutral pH. The removal percentage of phenol was decreased by increasing the initial phenol concentrations. Adsorption results show that equilibrium data follow the Freundlich isotherm, and kinetic data was well described by a pseudo-second-order kinetic model. Experimental results show that the activated red mud can be used to treat aqueous solutions containing phenol, as a low cost adsorbent with high efficiency.

  • construct containing reporter gene(pFV4CAT) regulated by carp $\beta$-actin promoter was microinjected into the one-cell stage egg of mud loach (Misgurnus mizolepis), and was successfully expressed, possibly by the integration into the genome. Both mean hatching success and early survival of the microinjected groups were not significantly different with those of control groups (P>0.05). The incidence of transgene was ranged from 7 to 48% based on the PCR and/or Southern blot analyses with the DNA prepared from fin or blood tissue. The spatial and temporal patterns of expression of the pFV4CAT gene, measured by in situ immunohistochemical analysis peroxidase-conjugated anti-CAT antibody, were variable among the experimental individuals. These results suggest that carp $\beta$-actin promoter is effective to express other transgene in mud loach, such that this promoter can be useful in the generation of valuable transgenic mud loach.

  • The Navy has tested the holding capacity of many kinds of anchors in order to propose the design chart for the holding capacity of drag-embedment anchors. The design chart is only applicable up to the cable bottom angle 60 when load is raised to the ultimate weight. However, the anchor experiences a significant uplift force when the angle is above 60 in shallow seas. In this paper, the procedure for the estimation of the holding capacity of anchors in mud is proposed. Drag-embedment anchors do not function well when there is a significant uplift component of load in soft seafloor materials, such as mud. Under these loading and seafloor conditions, gravity anchors seems to be more efficient. However, they are too heavy for their holding capacity. Therefore, suction pile (hollow concrete block) is more beneficial to the foundntion of silt protector in shallow sea with mud seafloor materials.

  • Drilling mud was used once in the step of separating the gas and powder they were transported to a surge tank. At that time, the fine powder, such as dust that is not separated from the gas, is included in the gas that was separated from the mud. The fine particles of the powder are collected to increase the density of the powder and prevent air pollution. To remove particles from air or another gas, a cyclone-type separator generally can be used with the principles of vortex separation without using a filter system. In this study, we conducted numerical simulations of air-particle flow consisting of two components in a cyclone separator in a mud handling system to investigate the characteristics of turbulent vortical flow and to evaluate the collection efficiency using the commercial software, STAR-CCM+. First, the single-phase air flow was simulated and validated through the comparison with experiments (Boysan et al., 1983) and other CFD simulation results (Slack et al., 2000). Then, based on one-way coupling simulation for air and powder particles, the multi-phase flow was simulated, and the collection efficiency for various sizes of particles was compared with the experimental and theoretical results.
