SLC6A18 유전자의 minisatellites 5 (SLC6A18-MS5)의 고혈압과의 관련성 및 진화적 의미 (Minisatellite 5 of SLC6A18 (SLC6A18-MS5): Relationship to Hypertension and Evolutional Level)

  • SLC6A18은 neurotransmitter로서 고혈압과 연관성이 보고 되었고, 유전자 내에 총 8개의 minisatellites가 존재함이 밝혀졌다. 본 연구에서 8개 minisatellites 중 가장 높은 heterozygosity를 나타내는 SLC6A18-MS5 영역에 대하여 생물정보학적 방법으로 Transfac software를 이용하여 transcription factor binding site를 분석한 결과, Pax4와 HNF4의 binding site를 발견하였다. HNF4는 당뇨병 대사에 관여하는 것으로 고혈압과의 연관성이 있을 것으로 사료된다. 그러므로 본 연구에서는 SLC6A18-MS5 영역과 고혈압과의 연관성을 조사하기 위하여, 대조군 301명과 고혈압 환자군 184명의 genomic DNA를 이용하여 대립형질의 패턴을 조사하였다. SLC6A18-MS5의 대립형질 분포와 고혈압은 직접적인 영향을 주지 않는 것으로 나타났다. 반면 높은 heterozygosity를 나타내는 SLC6A18-MS5에 친자확인 및 DNA typing 마커로서의 유용성을 알아보기 위해 20가족의 샘플을 이용하여, 감수분열 후 자손에의 분리 형태를 조사한 결과 부모에게서 자손으로 정확히 전달되는 멘델의 법칙에 의해 분리됨을 확인하였다. 또한 SLC6A18 유전자 내의 minisatellites들의 진화적 관계를 조사한 결과, 인간과 원숭이에서만 보존적으로 나타났다. 이러한 결과는 intron영역의 minisatellites 분석이 영장류의 비암호화 영역의 중요한 진화 마커로 사용될 수 있음을 나타내어, 영장류 특이적 진화를 이해하는데 도움이 될 것으로 사료된다.

  • Trehalose-tethered monomeric and dimeric berberines were synthesized in 50% and 30% from the reaction of berberrubine with 6-tosyl-${\alpha}$,${\alpha}^{\prime}$-trehalose and 6,6'-ditosyl-${\alpha}$,${\alpha}^{\prime}$-trehalose, respectively, and fully characterized by MS (HR and ESI) and NMR ($^1H$, $^{13}C$, COSY and HSQC). Spectrophotometric and spectrofluorimetric titrations indicated that compared with berberine, trehalose-tethered monomeric berberine had comparable DNA-binding affinity toward calf-thymus DNA, whereas trehalose-spaced dimeric berberine exhibited higher DNA-binding affinity. The potential application of these conjugates is also briefly discussed.

BcHSP17.6 cDNA의 도입에 의한 형질전환된 식물의 내열성 획득 (Acquisition of Thermotolerance in the Transgenic Plants with BcHSP17.6 cDNA)

  • 대만산 배추 (Brassica campestris M.)로부터 분리한 BcHSPI7.6 cDNA(내열성 유전자)를 pBKSl-l vector에 subcloning하므로서, NPTII 유전자와 P35S-HSPI7.6 cDNA를 가지는 pBKH4 재조합 플라스미드를 제작하였다. 이들 플라스미드를 갖는 A. tumefaciens LBA4404로서 담배잎 단편을 24시간 동안 공배양하므로서 감염시켰으며, 이들 유전자로 형질전환된 shoot는 $100\;{\mu\textrm{g}}/ml$의 가나마이신을 첨가한 MS-n/B 배지에서 선발하였다. 담배((Nicotiana. tabacum)의 열에 대한 치사온도는 $50^{\circ}$에서 15분 이상이었으며, BcHSPI7.6 cDNA를 갖는 형질전환된 식물체는 이 온도에서 내열성을 나타내었다. 식울체의 형질전환 여부는 ${\alpha}^{_32}P$로 표지한 BcHSPI7.6 cDNA 단편을 probe로 이용해서 Southern blot hybridization을 실시하므로서 확인하였다. BcHSPI7.6 cDNA의 발현정도는 Northern blot 분석과 이중면역확산 방법으로 확인하였다. 본 연구에서, 담배에 도입한 BcHSPI7.6 cDNA는 내열성과 관련이 있는 유전자로서, HSPI7.6 단백질은 식물체를 열에 의한 손상으로부터 방지해 주는 protector 역할을 하는 것 으로 사료된다.

Production of ${\gamma}-Aminobutyric$ Acid (GABA) by Lactobacillus buchneri Isolated from Kimchi and its Neuroprotective Effect on Neuronal Cells

  • Lactic acid bacteria that accumulated ${\gamma}-aminobutyric$ acid (GABA) in culture medium were screened to identify strains with high GAB A-producing ability. One strain, MS, which was isolated from kimchi, showed the highest GABA-producing ability among the screened strains. MS was identified as Lactobacillus buchneri based on Gram-staining, metabolic characteristics, and 16S rDNA sequence determination, Optimum culture conditions for GABA production were determined: MRS broth containing 5% MSG, 1% NaCl, and 1% glucose, at an initial pH of 5.0, the incubation temperature at $30^{\circ}C$ for 36 h. Under these conditions, MS produced GABA at a concentration of 251 mM with a 94% GABA conversion rate. Moreover, culture extracts of Lb. buchneri MS partially or completely protected neuronal cells against neurotoxicantinduced cell death.

  • 식품단백질의 물성과 기능성을 개선시켜 산업적인 가치가 매우 높은 TGase를 생산하는 S. platensis YK-2의 분자 유전학적인 연구를 위해 형질전환방법을 확립하였으며 본 연구를 위해 도입된 방법은 대장균을 plasmid DNA의 공여체로, S. platensis의 포자를 수용체로 사용하는 접합전달법이다. S. platensis를 위한 최적 접합전달조건은 50 mM의 $MgCl_2$를 첨가한 MS배지에 열처리를 하지 않은 포자와 $5{\times}10^7$의 plasmid DNA 공여체인 E. coli를 사용하는 것이다. 또 본 연구를 통해 얻어진 접합전달체의 attB site에 대한 분석을 통해 S. platensis chromosome의 pirin 상동체를 코드하는 ORF내에 attB site가 단일위치로 존재하고 있으며 이미 밝혀진 다른 방선균유래 attB site의 염기서열에 대해 77.8%~96.3%의 상동성을 나타냈다.

PAX1 Methylation Analysis by MS-HRM is Useful in Triage of High-grade Squamous Intraepithelial Lesions

  • This study is aimed to investigate the role of paired boxed gene 1 (PAX1) methylation analysis by methylation-sensitive high-resolution melting (MS-HRM) in the detection of high grade lesions in atypical squamous cells cannot exclude high-grade squamous intraepithelial lesion (ASC-H) and compared its performance with the Hybrid Capture 2 (HC2) human papillomavirus (HPV) test. In our study, 130 cases with a diagnosis of ASC-H from the cervical cytological screening by Thinprep cytologic test (TCT) technique were selected for triage. Their cervical scrapings were collected and evaluated by using PAX1 methylation analysis (MS-HRM) and high-risk HPV DNA test (HC2), followed by colposcopy and cervical biopsy. Chi-square test were used to test the differences of PAX1 methylation or HPV infection between groups. In the detection of CIN2+, the sensitivity, specificity, the PPV, NPV and the accuracy of PAX1 MS-HRM assay and high-risk HPV (HR-HPV) tests were respectively 80.6% vs 67.7%, 94.9% vs 54.5%, 83.3%, vs 31.8%, 94.0% vs 84.4%, and 91.5% vs 57.7%. The PAX1 MS-HRM assay proved superior to HR-HPV testing in the detection of high grade lesions (CIN2+) in ASC-H. This approach could screen out the majority of high grade lesion cases of ASC-H, and thus could reduce the referral rate to colposcopy.

탠덤매스에의한 체액 중 Globotriaocylceramide(Gb-3)의 측정을 이용한 한국인 고 위험도군에서의 파브리병 스크리닝 (Status of High Risk Group Fabry Disease Screening in Korea by Measuring Globotriacocylceramide in Body Fluid using Electrospray-MS/MS)

  • Fabry disease (FD) is an X-linked inborn error of glycoshpingolipid metabolism resulting from mutation in the enzyme ${\alpha}$-galactosidase A gene. The disease is an X-linked lipid storage disorder and the lack of ${\alpha}$-Gal A causes an intracellular accumulation of glycosphingolipids, mainly globotriaosylceramide (Gb-3). Measurement of Gb-3 in plasma has clinical importance for monitoring after enzyme replacement therapy for confirmed FD patients. Using electrospray ionization MS/MS we had developed, a simple, rapid, and highly sensitive analytical method for Gb-3 in plasma was used for the purpose of screening FD among high risk groups in Korean population. To date, no comprehensive results for FD screening have been performed and reported in Korea. We screened 1,100 outpatients from 13 hospitals (including clinics) to assess the incidence of FD among patients in high risk groups. For patients with borderline level amount of Gb-3, we repeated Gb-3 or performing complementary or confirmative assay with ${\alpha}$-Gal A activity and DNA mutaion analysis for confirmation diagnosis. Of 1,100 we diagnosed 3 FD with 2 classical type and 1 carrier (0.27%).

Evaluation of the inhibitory effect of Gynostemma pentaphyllum extracts on CYP450 enzyme activities using LC-MS/MS

  • Gynostemma pentaphyllum (Thunb.) Makino extract, a natural product with a history of traditional use, has gained attention for its potential health benefits. This study aimed to investigate its effects on key cytochrome P450 (CYP) enzymes using LC-MS/MS. Human liver microsomes and cDNA-expressed CYP2C8, CYP2C9, CYP2C19, and CYP3A4 supersomes were employed. Enzyme activity was assessed based on the formation of CYP-specific marker metabolites. The resulting data showed that the extract exhibited inhibitory effects on CYP2C8, CYP2C9, CYP2C19, and CYP3A4. Thus, G. pentaphyllum extract may influence the pharmacokinetics of drugs metabolized by CYP2C8, CYP2C9, CYP2C19, and CYP3A4. These findings emphasize the importance of considering potential herb-drug interactions when incorporating this extract into therapeutic regimens or dietary supplements.

Effects of Abeliophyllum distichum Nakai Flower Extracts on Antioxidative Activities and Inhibition of DNA Damage

  • The present study attempts to evaluate antioxidant activities of extracts from Abeliophyllum distichum. Nakai flower. The samples were collected in Jangyyeon-myeon, Goesan-gun, Korea and extracted with either hot-water or ethyl acetate (EtOAC). In DPPH, hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity of EtOAC extracts were 93.41%, 98.43%, and 7.38%, while those of hot-water extracts were 86.93%, 41.33% and 47.68% at 200 ${\mu}g/ml$, respectively. In ${\varphi}X$-174 RF I plasmid DNA cleavage assay, the protective effects of EtOAC and hot-water extracts against oxidative DNA damage were 82% and 17% at 200 ${\mu}g/ml$, respectively. Both extracts showed the protective effect of DNA migration by oxidative stress in intracellular DNA migration assay. Both extracts had no cytotoxity in NIH3T3 cells. Several polyphenolic compounds were identified such as 2-methoxy-benzoic acid, vanillic acid, phytol and pulegone by GC/MS. These results indicated that extracts of Abeliophyllum distichum Nakai flower showed antioxidant activities and protective activities against oxidative DNA damage and showed the possibility to be used as an effective natural antioxidants.
