Immunotoxicity of Polychlorinated Biphenyls (Aroclor 1254) in Mouse Splenocytes (마우스 비장세포에서의 Polychlorinated Biphenyls(Aroclor 1234)의 면역 독성)

  • Polychlorinated biphenyls (PCBs) has been widely used as plasticizer, insulator, lubricant, paint and ink. The persistence of PCBs in the environment and their bioaccumulation in living organism make a raise concerns regarding their toxic effects in immune system and subsequent effects on human health. However little has been known about effect of PCB, an endocrine disrupter, on splenocytes. In this study, for identifying the effect on the organs and immune cell of mice by the concentration and time of commercial PCB mixture (Aroclor 1254), each 3 mice were tested at the concentration of 3, 30, 300, 1,000 mg/kg respectively, and their organ's weight were measured in 4, 7, 14 days, respectively. Also according to concentration and time, PCB was evaluated for the effects on splenocyte viability and lipopolysaccaride (LPS) and concanavaline A (Con A)-induced splenocyte proliferation on mice spleen. In liver and lung, there were significantly defferent by concentration and time of PCB (p < 0.0001). In respect of concentration of PCB, no significant effects on mice's liver by Aroclor 1254 concentration below than 300 mg/kg were observed except at the concentration of 1,000 mg/kg doses (p < 0.0001). But there was not significant different change in mice spleen by concentration and time of PCB (p=0.2206) and the mode of weight change of spleen was different to of liver and of lung. Viabilities of splenocytes were decreased following treatment with high concentration of PCB. Also, LPS and Con A-induced cell proliferations were decreased by Aroclor 1254 at 1,000 mg/kg. These data suggest that Aroclor 1254 is the immunotoxic compound that may have an effect on mouse immune system.

  • Poncirus trifoliata (L.) Raf (Rutaceae) fruits (PTFE) has been used for the treatment of allergic disease. IgE is normally one of the least abundant immunoglobulin (Ig) isotypes in the serum of both humans and several species of experimental animals: however a number of different stimuli can result in profound increases in IgE levels relative to other isotypes. In rodents, infection with many parasitic helminths can cause approximately 100-fold elevation in IgE within 2 wks. Immunization of mice with small amounts of protein antigens on alum also results in 10-fold to fold increase in total serum IgE, much of it specific for the immunizing antigen. In this experiment, I investigated the effect of an aqueous extract of Poncirus trifoliata (L.) Raf (Rutaceae) fruits (PTFE) on a in vivo and in vitro IgE production. PTFE dose-dependently inhibited the serum levels of IgE induced by antigens. The regulation of IgE synthesis is influenced by T cells and T cell derived factors. IL -4, a T cell-derived cytokine, has been shown to stimulate murine IgE synthesis both in vitro and in vivo. Current evidence suggests that IL-4 induces IgE synthesis in the mouse by stimulating H chain isotype switch. Lipopolysaccharide (LPS) plus IL-4 cause about l00-fold increase in IgE secretion by murine B cells. The effects of PTFE on the IL-4-dependent IgE response by mouse whole spleen cells were studied. Whole spleen cells were cultured for 7 days in the presence of LPS plus IL-4 and PTFE and the supernatants were assayed for IgE. IL-4 dependent IgE production of LPS-stimulated whole spleen cells was inhibited by PTFE. Moreover, in the present study using U266Bl human IgE-bearing B cells, I found that PTFE inhibited the production of IgE activated by LPS plus IL-4. These results indicate that PTFE have antiallergic activity by inhibition of IgE production from B cells.

Comparison of Immune Promotion Effects of Water-extracted Angelicae gigantis, Rehmanniae Radix, Paeoniae japonica and Polygoni multiflori Radix (보혈 약재 (補血 藥材)인 당귀, 지황, 백작약, 하수오의 면역 촉진 효과 비교 분석)

  • Angelica gigantis (AG), Rehamaninae Radix(RR), Paenia japonica (PJ), and Polygoni multiflori Radix (PM) have been used as medicinal plants to tonify the blood. General function of the drugs have been known to nourish blood and control the heart and liver meridians. Recently, several studies have proposed mechanisms by which some oriental medicinal herbs work on the immune system. However, it is uncertain whether aqueous-extract of these drugs has immunomodulatory effect yet. In this study, I investigated the immune promotive effects of the water-extracted AG, RR, PJ and PM. The water-extracted AG, RR, PJ and PM inhibited NO production and iNOS expression in LPS stimulated RAW 264.7 macrophage cell line. Among these extracts, AG and PM induced expression of IL-2 and IFN${\gamma}$ in mouse spleen cells. In the flow cytometry analysis, PM-stimulated mouse spleen cells showed an increase in B-cell phenotype (CD45R/B220). The oral administration of Polygoni multiflori water-extracts to mice having S-180 abdominal dropsy cancer prolonged life-span more than control mice. These data suggest that among these extracts, PM has cellular and humoral immune-enhancement effect through IL-2 and IFN${\gamma}$ cytokine production, the regulation of NO production in macrophage cells and the B cell production in spleen cells.

The Human Milk Oligosaccharide 2'-Fucosyllactose Shows an Immune-Enhancing Effect in a Cyclophosphamide-Induced Mouse Model

  • The 2'-fucosyllactose (2'-FL) is the richest components in a human milk oligosaccharide. Several studies have reported that 2'-FL has beneficial effects in infants. However, there are few studies on its immune-enhancing effects. This research aimed to examine the immune-enhancing effect of 2'-FL on immunosuppression by cyclophosphamide (CCP) in ICR mice. Mice were orally administered distilled water or 0.5 mg/kg B.W. 2'-FL for 14 days. An immunocompromised mouse model was induced using CCP 80 mg/kg B.W. at 12-14 days. Using the CCP had effects on reducing their body weight, organ weight, spleen index, natural killer (NK) cell activity, and cytokines concentration and expression. This study also used concanavalin A-mediated T-cell proliferation to verify the immune-enhancing effects in the sample. Body weight, spleen index, organ weight, and cytokine levels were measured to estimate the immune-enhancing effects. The body weight at 14 days tended to increase, and the spleen weight and index significantly increased in the 2'-FL group compared to the CCP group. The NK cell activity increased in the 2'-FL group compared to the CCP group, but there was no significant difference. The concentration of interleukin (IL)-2 tended to recover in the 2'-FL group compared to the CCP group. The 2'-FL group showed a significant increase of IL-10 and IFN-gamma concentration compared to the CCP group. In addition, there was a trend of increased IL-10 mRNA expression compared to the CCP group. These results revealed that 2'-FL improved CCP-induced immunosuppression, suggesting that 2'-FL may have the potential to enhance the immune system.

Effect of Cadmium Chloride on the Immune Responses in Balb/c Mouse (카드뮴투여가 Balb/c 마우스의 면역반응에 미치는 영향)

  • This study was designed to investigate the antibody production to sheep red blood cells(SRBC) and proliferation of mitogen-stimulated spleen cells in Balb/c mice which received cadmium chloride. The mice were divided into three independent groups which were one control and two experimental groups by the cadmium treatment or not. No specific treatment was done for the control group. One of two experimental groups, which is called 'pre-treatment group' in this paper, was subcutaneously injected with low dose of cadmium chloride(0.5 mg/kg/day) for 5 consecutive days before the primary SRBC immunization. The other called 'non-pretreatment group' was only pretreated with normal saline. Both experimental groups were intraperitoneally injected with high dose of cadmium chloride(5 mg/kg) 8 hours before the primary immunization. Mice were intraperitoneally immunized twice with 2% SRBC suspension containing $10^8$ cells. The results obtained were as follows, 1. The PFG responses to SRBC were significantly increased in two experimental groups, cadmium pretreatment and non-pretreatment compared with that of control group(p<0.05). 2. The total antibody titers to SRBC in cadmium treated groups were similar to that of control group, but titers of IgG antibody were significantly elevated(p<0.01). 3. The proliferation response of spleen lymphocytes to various mitogens was suppressed in proportion to the concentration of cadmium and the degree of cadmium accumulation in liver was increased in the cadmium treated groups. These results suggest that cadmium chloride could affect on mouse immune response, especially its cell mediated immune response could be decreased while its humoral immune response could be increased, which may not be influenced by the administration methods or pretreatment of cadmium to mouse.

Effect of Herbal-acupuncture with Chaenomelis Fructus Solution at the Joksamni($ST_{36}$) of Mice with Collagen-induced Aarthritis (목과(木瓜) 약침(藥鍼)이 Collagen-induced Arthritis 억제 및 면역조절에 미치는 영향)

  • Objectives : The purpose of this study was to observe the effects of Chaenomelis Fructus herbal-acupuncture solution (ChF-HAS) at the Joksamni ($ST_{36}$) on arthritis induced by Collagen II in mice. Methods : The author performed several experimental items. The severity of arthritis, changes of mouse weight, size of the spleen and the degree of stenosis, changes of cytokine level, IgG, IgM and anti-collagen II, changes of immunocyte count, histological changes of the CIA mouse joint were analyzed and the conclusions are as follows. Results: 1. In the ChF-HA, the arthritis index, the incidence of arthritis, the degree of joint edema was significantly decreased. 2. In the ChF-HA, weight, spleen size and stenosis rate was low and maintained as the normal group was. 3. In the ChF-HA, cytokine level, IgG, IgM and anti-collagen II were significantly decreased. 4. In the ChF-HA, on changes of immunocyte count were maintained to the levels of normal group. 5. In histological changes of the CIA mouse joint, the cartilage destruction and synovial cell proliferation were decreased. Conclusions: These results suggest that ChF-HA at the $ST_{36}$ has an important role to control the immune reactions and suppress inflammatory response on the collagen induced rheumatoid arthritis. This study can be a significant supporting evidence that ChF-HA is chosen to be the principal therapy for clinical practice of the rheumatoid arthritis in the future.

Studies on the Aggregation of H-Y Antibody-Sexed and Bisected Rabbit Embryo (H-Y항체에 의한 토끼배의 성 감별과 이등분 절단 토끼배의 융합에 관한 연구)

  • These experiments were carried out to examine the development capacity of sexed and then bisected embryo from 8-cell to morula stage. Antisera to histocompatibility-Y(H-Y) antigen were prepared in inbred SD female rat by repeated immunization of spleen cell or testis supernatant from males of same strain. Male and female embryos were separated by delaying development of embryos against H-Y antibody. After sexing, rabbit embryos were bisected and aggregated. The results obtained from the these experiments were summuerized as follows: 1. When mouse and rabbit 8-, 16-cell and morular embryos were culature in H-Y antiserum, the ratio of embryo which has developed to hatching blastocyst was 53.4, 46.3 and 57.4% in mouse embryos, and 49.0, 52.0 and 61.0% in rabbit embryo, respectively. The ratio of mouse and rabbit embryos developed to hatching blastocyst showed nearly natural sex rate(50%), except rabbit mourla showed a little higher ratio(61.0%) as compared to natural sex ratio. 2. When rabbit demi-embryos from 8-, 16-cell embryo and morula were cultured, the percentage of demi-embryos was 70, 68 and 58% without zona pellucida removed, and 62, 69 and 51% with zona pellucida. The rate of aggregation was higher in 8- and 16-cell demi-embryos than in morula demi-embryo. 4. When sexed-demi-embryo was aggregated with another demi-embryo with demi-embryo with same sex, the rate of embryo developed to blastocyst was 60, 50 and 25%, respectively. Eight-cell demi-embryo showed highest rate. In conclusion, it showed that H-Y antiserum which was made by rat spleen cell enabled sexing rabbit embryos. And when rabbit sexed 8-, 16-cell and morula demi-embryo were aggregated, they were developed to eu-blastocyst which suggested the potential of sexed embryo manipulation.

Characterization of Monoclonal Antibodies against Heavy and Light Chains of Flounder (Paralichthys olivaceus) Immunoglobulin

  • Flounder (Paralichthys olivaceus) Immunoglobulins (Igs) were purified from the serum of mouse IgG-immunized flounder by using affinity chromatography. Under denaturing conditions in SDS-PAGE, the flounder Igs appeared to be composed of 2 heavy (H) chains (72 and 77 kDa) and two light (L) chains (26 and 28 kDa). Monoclonal antibodies (MAbs) were produced by the fusion of myeloma cells (SP2/0) with Balb/c mouse spleen cells that were previously sensitized against affinity-purified flounder Igs. In a Western blot analysis, the produced MAbs, FIM511, FIM519, and FIM562 recognized both the 72 and 77 kDa H chains, 26 kDa, and 28 kDa L chain, respectively. Mouse antiserum against flounder Igs reacted more strongly with the L chain of 28 kDa than with 26 kDa, suggesting that the 28 kDa molecule is more immunogenic than the 26 kDa L chain molecule. In a FACS analysis, the ratios of the Ig+ cell population in the flounder head kidney and spleen cells were 49% and 24%, respectively. Unexpectedly, however, the ratios of the Ig+ B-like cell population in the flounder were not significantly augmented, even after the immunization of an immunogenic antigen. This suggests that the humoral immune response in fish could be considerably different from that in mammals. The produced MAbs in this study would be useful in characterizing flounder Ig+ B-like cells and in developing flounder Ig detecting an immunoassay system.

Study on Morphological Changes and TUNEL Reaction of Apoptotic Cells in Mouse Liver by Apoptosis Induction

  • We investigated the morphological changes and TUNEL reaction of apoptotic cells in the liver of D-galactosamine (20 mg/mouse) and lipopolysaccharide (5 $\mu\textrm{g}$/mouse)-treated 30 mice (BALB/c), and in additioa also of apoptotic cells in kidney and spleen. The livers and other some organs of mice at 6, 12, 24, 48 and 72 hrs after treatment were collected and were fixed with 10% neutral formalin and paraffin sections were stained with hematoxylin-eosin or terminal deoxynucleotidly transferase-mediated dUTP nick end labeling (TUNEL) method. Morphological changes in apoptotic hepatocytes were chondensation of nuclei and density of cytoplasms, then the margination and pyknosis of chromatin, the formation of half-moon- or horse-shoe- or ship-like shapes of condensed chromatin mass, lastly formation of apoptotic bodies, disappearance of nuclear envelopes, decrease of stainability, then lysis and disappearance of apoptotic bodies. TUNEL positive reactions of hepatocytes were appeared first moderate in uncondensed hepatocytes, severe in condensed hepatocytes, moderate in chromatin-marginated hepatocytes. These reactions also were appeared moderate in hepatocytes with half-moon- or horse-shoe- or ship-like pyknotic chromatin mass or apoptotic bodies, and mild or negative in hepatocytes with lysed apoptotic bodies or with disappeared nuclear envelopes. Consequently these results suggested that TUNEL positive reactions of hepatocytes appeared at more early stages than appearance of chromatin condensation and disappeared at more early stage than disappearance of histological findings of apoptosis. We also confirmed that the differentiation of apoptotic cells from normal healthy cells of Kupffer cells and vascular endothelial cells in liver, reticular cells and lymphocytes in spleen and epithelial cells of tubules and ducts in kidney was impossible in H-E preparations but was possible in TUNEL preparations.

Effects of Orostachys Japhonicus Herbal-Acupuncture on Transferred Hepatic Cancer of Mouse Induced by Colon26-L5 Human Colon Cancer Cells (와송(瓦松) 약침(藥鍼)이 mouse의 간전이 암모델에 미치는 영향)

  • To study the effects of anti-cancer, anti-metastasis and immune response improvement effects of herbal-accupunture with Orostachys japhonicus A.Berger, infusion solution put into Kansu(BL18) of mouse induced by Colon26-L5 human colon cancer cells, which are corresponding to humanbody. We observed the change of body weight, surviving number, median surviving time, increase of life span, changes in amount of leukocyte, erythrocyte, platelet, total protein, creatinine, glucose and LDH, weight of spleen and kidney, histological analysis on tissue metastasis of liver, splenic cell proliferation, the expression of cytokine gene, the number of CD4+, CD8+, CD9+ and NK cell, and concluded like this. The results were obtained as follows ; 1. In acute and sub-acute cytotoxicity experiment, significantly signs were not appeared in all groups. 2. Antimetastatic experiment in vitro and in vivo showed that Orostachys Japhonicus A.Berger Herbal-acupuncture at Kansu(BL18) has antimetastatic effects. 3. The spleen cells proliferation of the experimental groups treated with Orostachys Japhonicus A.Berger infusion solution extract has increased significantly compared with that of the control group. 4. As compared with control, the population of total T cell, helper T cell, cytotoxic T cell and macrophage were increased. 5. The production of Th 1 type cytokines from splenocyte and cytokines which is associated with anti-tumor activity form macrophage were increased significantly. Above the results revealed that herbal-accupunture with Orostachys Japhonicus A.Berger infusion solution has effects of anti-cancer, anti-metastasis and immune response improvement.
