To investigate the immunostimulating and anticancer activities from hot water extract of Capsosiphon fulvescens, tumor cell toxicity, sarcoma-180 growth inhibition activity, complement system-activity, intestinal immune system and oral toxicity were performed. The extract of Capsosiphon fulvescens was prepared by hot water and precipitated by using ethanol. Partially purified extract (CFE) was obtained after dialysis and ultrafiltration. The polysaccharide compositions consisted of xylose(19.1%), fucose(15.3%), mannose(4.2%) and galactose(7.9%). The tumor cell toxicity of CFE slightly showed at high concentrations of 10-30 ${\mu}g/ml$, but inhibition ratio against mouse solid tumor was more increased for CFE of 40.1-59.4% than the control. Blood leukocyte counts increased to a maximum of 83% including liver, spleen and thymic of mouse. Immunoglobulin A binding amounts showed a high level of CFE of $2,454{\pm}113.8-2,670{\pm}133.1{\mu}g/mg$ in comparison with the control of $2,092{\pm}123.0{\mu}g/mg$. Acute toxicity of CFE was not detected at the concentration of 2,000 mg/kg in normal mouse.

To develop new potent antitumor substancces with low toxicity from natural products, especially from higher fungi of Korea, the carpophores of some wild basidiomycetes were collected and the antitimor test for their polysaccharides was done. The dried carpophores were extracted with hot water, and from the extracts, crude polysaccharides were obtained by alcohol precipitation. The alcohol precipitated crude polysaccharides were partially purified by dialysis and then used as the samples for antitumor tests. Among tested, the polysaccharide of Laccaria laccata, Trametes sanguineus, Armlliariella mellea, Clitocybe in fundibuliformis, and Xeromphalina campanella respectively showed the inhibition ratio of 75%, 72%, 94% 55%, and 47% when administered i. p. once dailly for ten days at the dose level of 20mg/kg/day into the mice implanted with $10^{6}$ cells of sarcoma 180/mouse. However, those of Craterellus cornucopiodes, Daedaleopsis confrogosa, and Coriolus sp. showed almost no activities.

This study was performed to evaluate the antitumor activities of water and ethanol (EtOH) extract of Salvia miltiorrhiza in vitro and in vivo. The proliferation of the human hepatoma (HepG2), rectum cancer (HRT-18) and colon cancer (HT-29) cells was inhibited by administration of extracts in a dose-dependent manner. Particularly, EtOH extract inhibited proliferation of the cells more effectively than water extract did. The morphology of cells induced by EtOH extract was characterized by reduction of cell size and deformatin. Oral administration of the EtOH extract (3 mg/head) to tumor-bearing mice inhibited the tumor (sarcoma-180) growth by 35% and prolonged their survival rate by 61%. The EtOH extract was shown to be nontoxic at 37.5% mg/head/day on the acute toxicity test. These studies suggest that the EtOH extract of Salvia miltiorrhiza may have antitumor activity in vitro and in vivo.
In order to investigate the anticancer activity of an anti-yeast substance (AYS), a proteoglycan produced by Rhanella aquatilis AY2000, the cytotoxicity of the AYS against cancer cells was determined in vitro. The AYS was not cytotoxic to the human Jurkat T cell or the mouse sarcoma 180 cell, but was cytotoxic to the human colon cancer TH20 cell. The AYS was increasingly cytotoxic against human colon cancer cells in a dose-dependent manner at range from 62.5 to 500 ${\mu}g/ml$. Anticancer activity by combination of the AYS and an anticancer agent was also determined. The anticancer agent combined with the AYS was shown to possess greater synergistic anticancer activity against human colon cancer cells, as compared with the anticancer agent alone.
A New lectin from baby clam, Tapes japonica, was isolated and purified through the following procedures; acetone powder, 0.15M NaCl extraction, ammonium sulfate fractionation, N-acetyl-D-galactosamine-agarose affinity column, and ion exchange Mono Q of FPLC. This lectin nonspecifically agglutinated human erythrocytes but didn't agglutinate mouse and rabbit erythrocytes. And the lectin neither stimulated human lymphocytes nor agglutinated Sarcoma 180 cells. On polyacrylamide gel electrophoresis, the lectin migrated as a major single band indicating homogeneous. A molecular weight was estimated to be about 131,000 daltons by Biogel P-300 and 125,000 daltons by SDS-PAGE without ${\beta}-mercaptoethanol$. This lectin is supposed to be a tetramer composed of heterogeneous subunits, about 30,000 and 33,000 daltons. Baby clam lectin was inhibited by EDTA and recovered agglutinating activity by $Ca^{++}\;and\;Mn^{++}$. This lectin is revealed as glycoprotein that contained about 4.2% neutral sugar.

This study was initiated in order to investigate the anticancer and immunomodulating activities of crude polysaccharides extracted in methanol, neutral saline, and hot water (hereinafter referred to as Fr. MeOH, Fr. NaCl, and Fr. HW, respectively) from the fruiting bodies of Panellus serotinus. Content of ${\beta}$-glucan and protein in Fr. MeOH, Fr. NaCl, and Fr. HW extracts of P. serotinus ranged from 22.92~28.52 g/100 g and 3.24~3.68 g/100 g, respectively. In vitro cytotoxicity tests, none of the various fractions of crude polysaccharides were cytotoxic against sarcoma 180, HT-29, NIH3T3, and RAW 264.7 cell lines at the tested concentration. Intraperitoneal injection with crude polysaccharides resulted in a life prolongation effect of 23.53~44.71% in mice previously inoculated with sarcoma 180. Treatment with Fr. HW resulted in an increase in the numbers of spleen cells by 1.3 fold at the concentration of $50{\mu}g/mL$ compared with control. Treatment with Fr. NaCl resulted in improvement of the immuno-potentiating activity of B lymphocytes by increasing the alkaline phosphatase activity by 1.4 fold, compared with control, at the concentration of $200{\mu}g/mL$. Among the three fractions, maximum nitric oxide ($13.48{\mu}M$) was recorded at $500{\mu}g/mL$ in Fr. HW. Production of tumor necrosis factor alpha, interleukin-$1{\beta}$, and interleukin-6 was significantly higher, compared to the positive control, concanavalin A, at the tested concentration. Therefore, treatment with crude polysaccharides extracted from the fruiting body of P. serotinus could result in improvement of antitumor activity.
In the previous study we described the antitumor activity of GLB, a protein-polysaccharide fraction of the growing tips of Ganoderma lucidum, against sarcoma 180 solid tu mor in ICR mice. In this study we investigated the stimulatory activity of GLB on macrophages. When analyzed using a flow cytometer, GLB ($100{\mu}g/ml$) was found to increase the phagocytic activity of the BALB/c mouse peritoneal macrophages as well as chicken macrophage BM2CL cells against FITC-labeled C.albicans by 55.2% and 21.2%, respectively. GLB also increased the spreading and the expression of MHC class II molecules of BM2CL cells as well as the mouse peritoneal macrophages. From these results, it is clear that GLB is a strong stimulator to the macrophages.
The present study was undertaken to investigate the anticancer activity of monoterepenes in the animal and the cancer cell line tests. Both of the noncyclic and cyclic monoterpenes showed significant life prolonging effects on ICR mouse with abdominal cancer induced by Sarcoma 180 cells up to 67.4% and 63.5% in case of linalool and geraniol, respectively. Linalool and geraniol also exhibited very excellent cytotoxicity against L1210 leukemic cells with $IC_{50}$/ value of 0.32 $\mu\textrm{g}$/mι in 5 days culture condition. In the presence of linalool and geraniol, the generation of $O_2$$^{[-10]}$ ion were found to be increased proportionally to the cytotoxicity arisen from these monoterpenes. Furthermore, the antioxidant enzymes activities such as superoxide dismutase (SOD) and glutathione peroxidase (GPx) responsible for the conversion of $O_2$$^{[-10]}$ ion to $H_2O$$_2$ and then to $H_2O$ augmented remarkably by linalool and geraniol. All data put together it can be postulated that monoterpenes may kill abdominal cancer cells of ICR mouse probably by activating anticancer system of the body, whereas the death of L1210 cells may be due to the detrimental attacks of reactive oxygen species (ROS) including $O_2$$^{[-10]}$ in spite of antioxidant enzymes activities to overcome the ROS attacks.
Chitin, a linked polysaccharide composed of 2-acetamido-2-deoxy-$\beta$-D-glucopytanose residues, is distributed widely in nature. It has been utilized on various application field due to the development of chitin derivatives such as chitosan, partial deacetylated chitin, carboxylmethyl chitin, sulfated chitin, and so on. Chitin and chitosan have been recently interested in antitumor and antimicrobial activities, because of a powerful tumor inhibitory effect against experimental mouse tumors. Especially, the oligosaccharides obtained by partial degradation of them exhibited a remarkable antitumor effect against sarcoma 180, MM 48 and Meth Asolid tumors and antimetastatic effect against Lewis lung carcinoma in mice. This review describes on antitumor effects of chitin, chitosan and their oligosaccharides by their mechanism of action involving enhancement of immunological system.
Purpose : In order to understand in vivo radiation damage modifying of bFGF on jejunal mucosa, bone marrow and the effect of bFGF on the growth of transplanted mouse sarcoma 180 tumor in mice. Materials and Methods : Mice were treated with $6\;{\mu}g$ of bFGF at 24 hours and 4 hours before exposing to 600 cGy, 800 cGy and 1,000 cGy total body irradiation (TBI), and then exposed to 3,000 cGy local radiation therapy on the tumor bearing thigh. Survival and tumor growth curve were plotted in radiation alone group and combined group of bFGF and irradiation (RT). Histologic examination was performed in another experimental group. Experimental groups consisted of normal control, tumor control, RT (radiation therapy) alone, $6\;{\mu}g$ bFGF alone, combined group of $3\;{\mu}g$ bFGF and irradiation (RT), combined group of $6\;{\mu}g$ bFGF and irradiation (RT). Histologic examination was peformed with H-E staining in marrow, jejunal mucosa, lung and sarcoma 180 bearing tumor. Radiation induced apoptosis was determined in each group with the DNA terminal transferase nick-end labeling method ($ApopTag^{\circledR}$ S7100-kit, Intergen Co.) Results : The results were as follows 1) $6\;{\mu}g$ bFGF given before TBI significantly improved the survival of lethally irradiated mice. bFGF would protect against lethal bone marrow syndrome. 2) $6\;{\mu}g$ bFGF treated group showed a significant higher crypt depth and microvilli length than RT alone group (p<0.05). 3) The bone marrow of bFGF treated group showed less hypocellularity than radiation alone group on day 7 and 14 after TBI (p<0.05), and this protective effect was more evident in $6\;{\mu}g$ bFGF treated group than that of $3\;{\mu}g$ bFGF treated group. 4) bFGF protected against early radiation induced apoptosis in intestinal crypt cell but might have had no antiapoptotic effect in bone marrow stem cell and pulmonary endothelial cells. 5) There was no significant differences in tumor growth rate between tumor control and bFGF alone groups (p>0.05). 6) There were no significant differences in histopathologic findings of lung and mouse sarcoma 180 tumor between radiation alone group and bFGF treated group. Conclusions : Our results suggest that bFGF protects small bowel and bone marrow from acute radiation damage without promoting the inoculated tumor growth in C3H mice. Improved recovery of early responding normal tissue and reduced number of radiation induced apoptosis may be possible mechanism of radioprotective effect of bFGF.
