當歸飮子加蟾수가 皮膚癌細胞(A431)의 細胞毒性에 미치는 影響 (Cytotoxicity of water extract of Dangkwieumja ka Sumsoo on A43l Cells)

  • The purpose of this research was to investigate effect of water extract of DangKwi-Eum-Ja ka Sumsoo(DESE) on the cytotoxicity of human epidemloid cell, A431 cells. The effects of DESE on the proliferation of A431 cells, Balb/c 3T3 cells, mouse thymocytes and splenocnes were estimated by MTT colorimetric assay, and nitric oxide production from mouse peritoneal macrophage was estimated by Griess method. DESE inhibited the proliferation of A431 cells at $10{\mu}g/ml$, and did not affect the proliferation of Balb/c 3T3 cells. DESE decreased the cytotoxicity of mitomycin C or cisplatin on A431 cells, increased the cytotoxicity of mitomycin C or cisplatin on Balb/c 3T3 cells. DESE inhibited the proliferation of mouse thymocytes and splenocytes at $100{\mu}g/ml$. DESE did not affect the nitric oxide production from mouse peritoneal macrophage in vitro, but decreased the nitric oxide production from DESE-treated mouse peritoneal macrophage.

Macrophage activation by glycoprotein isolated from Dioscorea batatas

  • We demonstrate that glycoprotein isolated from Dioscorea batatas (GDB) activates macrophage function. Analysis of the infiltration of macrophages into peritoneal cavity showed GDB treatment significantly increased the recruitment of macrophages into the peritoneal cavity. In order to further confirm and investigate the mechanism of GDB on macrophage activation, we analyzed the effects of GDB on the cytokine expression including IL-$1{\beta}$, TNF-${\alpha}$, and IL-6 in mouse peritoneal macrophages. GDB increased the expression of IL-$1{\beta}$, TNF-${\alpha}$, and IL-6. Cytokine induction by GDB was further confirmed by RT-PCR and ELISA in mouse macrophage cell line, RAW264.7 cells. Treatment of RAW264.7 cells with GDB produced strong induction of NF-${\kappa}B$ DNA binding and MAPK phosphorylation, markers for macrophage activation and important factors for cytokine gene expression. Collectively, this series of experiments indicates that GDB stimulates macrophage activation.

  • The effects of genistein on murine thymocytes for inducing apoptotic cell death and phagocytic activity of peritoneal macrophage were studied in vitro. Addition of genistein (10 and 50$\mu$M) to cultured thymocytes from BALB/c mice definitely promoted DNA fragmentation. Also, cytofluorometric analysis of these cells demonstrated a reduction in mitochondrial transmembrane potential ($\Delta$Ψm). But, repeated administration of genistein (1 mg/mouse/day) to mice for 7 days did not cause any detectable DNA fragmentation. Genistein decreased lucigenin chemiluminescence and engulfment of fluorescein-conjugated E. coli particles in peritoneal macrophage. These results suggest that genistein induce an apoptosis of thymocyte via reduction in $\Delta$Ψm and decrease phagocytic activity of peritoneal macrophage in vitro.

질트리코모나스(Trichomonas waginazis)에 대한 마우스 복강 대식세포의 세포독성 (Cytotoxicity of resident and Iymphokine-activated mouse peritoneal macrophage against yrichomonas vaginalis)

  • 질트리코모나스(Trichomenas vaginalis)에 대한 마우스의 복강 대식세포 및 림포카인으 로 활성화시킨 대식세포의 세포독성을 각각 관찰하였다. 세포독성은 질트리로모나스를 3H-TdR로 label시킨 후 대식세포와 반응시켜 사멸한 원충에서 방출되는 방사능 양을 비교하여 측정하였다. 대식세포의 원충에 대한 비율을 1 : 1, 5 :1, 10 : 1, 20 : 1 및 50 : 1로 증가시키고 반응시간을 12시간 및 24시간으로 변화시켰을 때, 대식세포와 원충의 비율 10 : 1 및 24시간 반응의 경우 가장 놓은 세포독성을 보였다. 마우스 비장세포를 phytohemagglutinin으로 자극시켜 얻은 림포카인으로 활성화시킨 대식세포에서는, 아무 처리를 하지 않은 대조 대식세포와 비교하였을 때 세포독성이 유의하게 증가되었으나 세포독성이 림포카인의 희석정도와는 비례하지 않았다. 또한 질트리코모나스에 세포독성을 나타내는 세포는 주로 플라스틱에 부착하는 대식세포임을 알 수 있었다.

조각자(皂角刺)의 간암세포주(Hep G2)에 대한 세포독성, Apoptosis 및 NO에 대한 실험 (Effect of Gleditsiae Spina on Hep G2 cells cytotoxicity and Apoptosis and No)

  • In this study, antineoplastic activity against human hepatocellular carcinoma cell line(Hep G2) was tested in Gleditsiae Spina. Gleditsiae Spina was extracted with water, and the cytotoxic activity was tested using a calorimetric tetrazolium assay(MTT assay), the apoptosis was tested using a DNA electrophoresis and flow cytometry. The nitric oxide production from mouse peritoneal macrophage was tested using a Griess method. Gleditsiae Spina extracts against the proliferation of Hep G2 cells not showed cytotoxicity at the concentration of less than $100{\mu}g/ml$, and Gleditsiae Spina extracts not showed the cytotoxicity of mitomycin C and the cytotoxicity of cisplatin on Hep G2 cells. Gleditsiae Spina extracts aginist the proliperation of BALB/c 3T3 cells not showed cytotoxicity, the proliperation of mouse thymocytes and splenocytes not showed cytotoxicity at the concentration of less than $100{\mu}g/ml$. Gleditsiae Spina extracts not showed nitric oxide production from mouse peritoneal macrophage in vitro. Gleditsiae Spina was administered orally for 7 days at 300mg/kg increased nitric oxide production from mouse peritoneal macrophage.

Macrophage-Activating Factors Produced by Murine Leukemia X Fibroblast Hybrid Cells Stimulates Resistance to Mycobacterium avium Complex

  • A murine leukemia x LM fibroblast hybrid cell line with immune augmenting properties stimulated resistance to Mycobacterium avium complex (MAC) in mouse peritoneal macrophages, and in immune deficient beige mice (C57BL/6/bgj/bgj). The proliferation of MAC in mouse peritoneal macrophages was inhibited by medium conditioned by the growth of the hybrid cells (hybrid cell-CM). Under similar circumstances, media conditioned by the growth of LM cells (LM cell-CM), a mouse fibroblast cell line used as one parent in forming the hybrid cell, was exhibited no inhibitory effect. Treatment of mouse peritoneal macrophages with hybrid cell-CM, but not with LM cell-CM, stimulated the expression of each of four previously described macrophage activation antigens, suggesting that the hybrid cells formed immunomodulators in addition to those formed by LM cells. Furthermore, the morphology of the macrophages following treatment with hybrid cell-CM was clearly distinguishable from that following exposure of the cells to LM cell-CM. The therapeutic effects of hybrid cells on the progression of MAC-infection were indicated by the prolonged survival of MAC-infected immune-deficient beige mice. One hundred percent of treated animals survived more than 60 days, while untreated animals died in approximately 22 days.

Anti-inflammatory Effect of Quercus Salicina in IFN-${\gamma}$/LPS-stimulated Mouse Peritoneal Macrophage

  • Quercus salicina has been widely used as a traditional medicine for the treatment of various diseases. In macrophages, nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions in inflammation. In the present study, the inhibitory effect of methanolic extracts of Q. salicina (QSM) on NO production in LPS-stimulated mouse (C57BL/6) peritoneal macrophages was investigated. QSM suppressed NO production without notable cytotoxiciy. QSM also exhibited down-regulation of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression via attenuation of NF-${\kappa}B$ translocation to nucleus in rIFN-${\gamma}$ and LPS stimulated mouse peritoneal macrophages. The present study strongly suggest that Q. salicina may be beneficial in diseases which related to macrophage-mediated inflammatory disorders.

Cyclooxygenase 억제제 검색을 통한 항염증제 개발 연구 (New screening method for anti-inflammatory agent)

  • Gram 음성균의 세포벽 성분인 lipopolysaccharide는 각종 세포에서의 prostaglandin 합성을 증진 시키며 이는 cyclooxygenase-2의 선택적 발현에 기인한다는 사실이 이미 보고된 바 있다. 본 연구에서는 mouse peritoneal marophage를 대상으로 하여 LPS의 prostaglandin 합성 증진 작용에 대한 특성으로 검토함으로써, COX-2에 대한 선택적 저해제 검색에 이용될 수 있는지 그 가능성을 확인코자 하였다. LPS는 peritoneal macrophage에 처리시 약 8시간 정도의 lag time을 보인 후 prostaglandin 합성을 현저히 증진 시켰으며, 이는 주로 COX 활성의 증가에 기인하는 것으로 추정되었다. 또한 LPS의 작용은 항염증제인 dexamethasone에 의해서 강하게 억제 되었으며 metabolic labeling 결과 이는 COX-2의 생합성을 억제하는데 기인하는 것으로 확인되었다. 따라서 mouse peritoneal macrophage에서의 LPS에 의한 prostaglandin 합성 증진 작용은 rat alveolar macrophage와 정성적으로 동일함을 확인할 수 있었으며, 본 실험 조건은 COX-2에 대한 선택적 저해제 검색에 응용될 수 있음을 확인 하였다. 본 실험 조건하에서 비스테로이드성 항염제인 ketoprofen의 작용을 검토한 결과 ketoprofen은 COX-1에 비교적 선택적인 저해 작용을 보이는 것으로 추정 되었다.

The Anti-inflammatory Mechanism of Xanthoangelol E is Through the Suppression of NF-${\kappa}B$/Caspase-1 Activation in LPS-stimulated Mouse Peritoneal Macrophage

  • Angelica keiskei has exhibited numerous pharmacological effects including antitumor, antimetastatic, and antidiabetic effects. However, the anti-inflammatory effects and mechanisms employed by xanthoangelol E isolated from Angelica keiskei are incompletely understood. In this study, we attempted to determine the effects of Xanthoangelol E on the lipopolysaccharide (LPS)-stimulated mouse peritoneal macrophage. The findings of this study demonstrated that xanthoangelol E inhibited the production of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6, and prostaglandin $E_2$ ($PGE_2$). Xanthoangelol E inhibited the enhanced levels of cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) caused by LPS. Additionally, we showed that the anti-inflammatory effect of xanthoangelol E is through the regulation of the activation of nuclear factor (NF)-${\kappa}B$ and caspase-1. These results provide novel insights into the pharmacological actions of xanthoangelol E as a potential candidate for the development of new drugs to treat inflammatory diseases.

Enhanced macrophage function of red ginseng acidic polysaccharide(RGAP) in combination with IFN-$\gamma$

  • In this study we examined the potential for the synergistic augmentation of the activity of inflammatory mouse peritoneal macrophages by stimulation with RGAP combined with IFN-$\gamma$. The moderate augmentative effect induced by preincubation with RGAP was observed in the production of IL-1, IL-6 and NO but not TNF-$\alpha$. In addition, IFN-$\gamma$ had a low activating effect. Following preincubation with both RGAP A and IFN-$\gamma$, a marked enhancement of secretory activity and tumoricidal activity was noted in mouse peritoneal macrophages. Treatment of peritoneal macrophage with combination increased the generation of IL-1, IL-6, TNF-$\alpha$ and NO, whereas the production of reactive oxygen species were not altered. These results demonstrate the synergistic effects on macrophage function of RGAP in combination with IFN-$\gamma$ and suggest that the ability of IFN-$\gamma$ to prime macrophages to produce secretory molecules in response to RGAP may have implications for immunotherapy with this combination.
