Ultrastructural Studies of Typhoid Cells

  • To investigate the nature of typhoid cells, three cases of clinically, serologically and histopathologically proven typhoid lesions of the small intestine and regional lymph nodes were studied light and electron microscopically, Light microscopically, typhoid cells were swollen mononuclear cells characterized by abundant amount of eosinophilic cytoplasm and frequent phagocytoses of red blood cells, bacterial clumps and other tissue debris. These cells were pyronin negative, Electron microscopically, these cells showed marked and diffuse dilatation of RER cisternae and disappearance of ordinary cytoplasmic organelles, but frequent phagocytosed materials, The meaning and reason of RER cisternal dilatation and reduction of cytoplasmic organelles were discussed, and are regarded as degenerative process due to bacterial endotoxin. Although there was hot enough cytoplasmic organelles to pinpoint the origin of typhoid cells, active phagocytosis and evidences against being either plasmacytic or lymphocytic nature favored retuculoendothelial nature of the typhoid cells.

Expression and Clinical Significance of Myeloid Derived Suppressor Cells in Chronic Hepatitis B Patients

  • We here document discovery of expression profile of myeloid derived suppressor cells (MDSCs) in chronic hepatitis B (CHB) patients and changes in the course of disease. The study population was composed of 75 outpatient HBV cases and 15 healthy control cases. Peripheral blood samples were collected for separation of mononuclear cells. Levels of MDSCs labeled with Lin-DR-CD11b+CD33+ obtained from peripheral blood mononuclear cells (PBMC), were revealed to have significant differences between the CHB and other groups. They were 0.414% for health control cases and 0.226% for CHB cases (Z=-2.356, p=0.0189). It also observed that the group of HBeAg positive cases had significant difference in MDSCs/PBMC median ($X^2=11.877$, p=0.003), compared with group of HBeAg negative cases and the healthy control group. It suggested considerable MDSCs might be involved in HBeAg immune tolerance. In addition, negative correlations between MDSCs/PBMC and parameters of ALT, AST and TBil, while positive correlation between MDSCs/PBMC and ALB parameter were found. Multiple comparisons between the four phases and health control phase again, there was a statistically sifnificant difference ($X^2=17.198$, p=0.002). Taken together, these findings may provide a new immunotherapy strategy for reduced the expression levels of MDSCs in CHB patients, through induction of an autoimmune response to virus removal.

Streptozotocin 당뇨(糖尿)마우스의 췌도세포(膵島細胞)에 미치는 Aloe vera의 영향에 대한 병리조직학적(病理組織學的) 연구(硏究) (Histopathological study on the effect of Aloe vera in the pancreatic islets of streptozotocin induced diabetic mice)

  • This experiment was carried out to investigate the influence of Aloe vera in the pancreatic islets of streptozotocin diabetic mice. Experimental diabetes was induced in ICR mice with a single injection of SZ(140mg/kg body weight, ip). The mice demonstrating hyperglycemia 48 hours after SZ injection were treated for 16 days with Aloe vera(300, 800mg/kg). Plasma glucose was measured, and for morphological studies of the islets specimens were stained with hematoxylin-eosin and by immunocytochemical methods. Then we observed the morphological changes of islets. Polymorphonuclear cells were infiltrated at the periphery of the islets 48 hours after SZ injection in SZ-treated ICR mice, but no prominent WBC infiltration was observed throughout the experiment. Blood glucose in mice treated with Aloe vera after SZ injection was higher than that of SZ injected mice, and mononuclear cells were heavily infiltrated at the islets 16 days after Aloe vera treatment(300mg/kg), and significant islets infiltration of mononuclear cells was observed 30 days after Aloe vera treatment(800mg/kg). Islets of ICR mice treated with Aloe vera after SZ injection showed severer insulitis, degranulation and necrosis of B cells than those of SZ injected mice. These studies indicate that Aloe vera in SZ injected mice increases vascular permeability and number of WBC in pancreatic islets, and potentiates destruction of B cells by cell-mediated immune system.

Nickel Increases Chemotactic Activity of Porcine Peripheral Blood Polymorphonuclear Cells

  • Nickel is a nutritionally essential trace element that plays an important role in the immune system of several animal species. The aim of this study was to examine the effect of nickel chloride on chemotactic activity of peripheral blood polymorphonuclear cells (PMNs) and whether this effect is associated with interleukin (IL)-8 and a nuclear factor-kappa B (NF-κB)-dependent pathway. Peripheral blood mononuclear cells (PBMCs) and PMNs were isolated by Percoll solution (Specific gravity; 1.080) and 1.5% dextran treatment, respectively. A modified Boyden chamber assay was used to measure the chemotactic activity of PMNs. The level of IL-8 in culture supernatant from PBMCs was measured by enzyme-linked immunosorbent assay (ELISA). Both of PBMCs and PMNs exhibited a low viability when cultured with concentration of greater than 1,000 μM of nickel chloride for 24 h. Thus, nickel chloride was used at concentration of 500 μM, which preserved cell viability. Treatment with nickel did not directly affect the chemotactic activity of PMNs. However, the chemotactic activity of PMNs was remarkably increased by culture supernatant from PBMCs treated with nickel chloride (500 μM) for 24 h. Recombinant porcine IL-8 polyclonal antibody (pAb) neutralized the enhancing effect on the chemotactic activity of PMNs by culture supernatant from PBMCs treated with nickel and this culture supernatant had higher IL-8 levels than the culture supernatant from untreated PBMCs. In addition, n-tosyll-phenylalanine chloromethyl ketone (TPCK), a NF-κB inhibitor, antagonized the enhancing effect on the chemotactic activity of PMNs by the culture supernatant from PBMCs treated with nickel. These results suggested that nickel stimulates porcine PBMCs to produce IL-8, which increases the chemotaxis of PMNs via NF-κB-dependent pathway.

The immune-enhancement effect by Falun Gong cultivation

  • Falun Gong (FG) is an advanced system of cultivation and practice, which is beneficial for both mind and body. In this study we investigated the effects of FG on the production of cytokines in FG practitioner (FGP). To study whether plasma cytokines levels were affected by FG, their levels were analyzed. The amount of $interferon-{\gamma}$ $(IFN-{\gamma})$, interleukin (IL)-2, IL-4 and IL-6 (2.5-fold for $IFN-{\gamma}$, 1.2-fold for IL-2, 2.1-fold for IL-4 and 2.5-fold for IL-6, respectively) were significantly higher in the FGP group than normal group (P<0.05). Peripheral blood mononuclear cells obtained from normal healthy control and FGP were cultured for 24 h in the presence or absence of lipopolysaccharide. The amount of $IFN-{\gamma}$, IL-2, IL-4 and IL-6 in culture supernatant was quantified. However, there were no significant differences in the level of the same cytokines between the normal and FGP group. These data suggest that FG cultivation may contribute to immune-enhancement in vivo.

Integrated Bioinformatics Approach Reveals Crosstalk Between Tumor Stroma and Peripheral Blood Mononuclear Cells in Breast Cancer

  • Breast cancer is now the leading cause of cancer death in women worldwide. Cancer progression is driven not only by cancer cell intrinsic alterations and interactions with tumor microenvironment, but also by systemic effects. Integration of multiple profiling data may provide insights into the underlying molecular mechanisms of complex systemic processes. We performed a bioinformatic analysis of two public available microarray datasets for breast tumor stroma and peripheral blood mononuclear cells, featuring integrated transcriptomics data, protein-protein interactions (PPIs) and protein subcellular localization, to identify genes and biological pathways that contribute to dialogue between tumor stroma and the peripheral circulation. Genes of the integrin family as well as CXCR4 proved to be hub nodes of the crosstalk network and may play an important role in response to stroma-derived chemoattractants. This study pointed to potential for development of therapeutic strategies that target systemic signals travelling through the circulation and interdict tumor cell recruitment.

The Effects of Seonghyangjeonggisan on Cytokines Production in the Peripheral Blood Mononuclear Cells of Acute Cerebral Infarction Patients

  • The Korean traditional medicine, Seonghyangjeonggisan (SHJGS) has long been used for acute cerebral infarction (Cl). However, scientific investigation has been carried out a little. Cytokines, involved in the regulation of inflammatory reactions and immune responses, may play an important role in the pathogenesis of Cl. The aim of this study is to investigate the effects of SHJGS on the production of various cytokines in the patients with acute Cl. Peripheral blood mononuclear cells (PBMC) obtained from the patients with acute Cl were cultured for 24hr in the presence or absence of lipopolysaccharide (LPS) and phytohaemagglutinin (PHA). The amount of TNF-α, IL-1β, IL-6 and IL-8, in PBMC culture supernatant, was significantly increased in the LPS and PHA treated cells, compared with unstimulated cells (P<0.05). This study showed that increased TNF-α, IL-1β, IL-6 and IL-8 level stimulated by LPS and PHA was inhibited by SHJGS (0.01-1 ㎎/㎖) in a dose-dependent manner but IL-8 level was not inhibited significantly at 1㎎/㎖ (P>0.05). The maximal inhibition rate of TNF-α, IL-1β, IL-6 and IL-8 by SHJGS (1㎎/㎖) was 68% (P<0.05), 53.9% (P<0.05), 45.5% (P<0.05), 46.7% (P>0.05) respectively. These results suggest that SHJGS might have anti-inflammatory effects through cytokine modulation. which might explain its beneficial effects in the treatment of acute Cl.

태아 간세포의 거핵구 집락형성 (Megakaryocyte Colony Formation of Fetal Liver Cells)

  • 목 적 : 태아 간세포는 면역학적으로 미숙하여 이식에 따른 거부반응이 잘 일어나지 않으므로 조혈모세포 이식, 특히 자궁내 이식의 좋은 공여세포로 이용할 수 있으나 아직 국내에서는 이를 위한 기초연구가 미미한 실정이다. 연구자는 태아 간 단핵세포를 이용하여 면역조직학적 염색과 체외배양 결과를 분석하여 태아 간의 거핵구 집락형성과 관련된 기초자료를 마련하고자 이 연구를 시행하였다. 방 법: 임신중절시 수집한 태아 간조직과 특발성 혈소판 감소성 자반증 및 범혈구 감소증 환아의 골수에서 단핵세포를 분리한 후 $MegaCult^{TM}-C$ 배지에서 12일간 배양 후 성장인자 첨가에 따른 거핵구 집락형성을 관찰하였고, 5일간의 Flask 배양 후 유세포 분석기를 이용하여 CD34, CD41 양성세포를 측정하였으며, methylcellulose 배지를 이용하여 $37^{\circ}C$ 5% $CO_2$ 배양기에서 배양한 후 제 7일과 14일에 성장인자 첨가에 따른 CFU-GM 집락 수를 조사하였다. 결 과 : 태아 간의 단핵세포 수는 재태주령 11주에 비해서 19주에서 더 많았으며 세포생존율은 $91.2{\pm}3.4%$였다. $MegaCult^{TM}-C$ kit를 이용하여 12일간 배양시 재태주령 11주의 태아 간이 19주에 비해서 거핵구 집락 수가 많았고, 4례 모두 성장인자의 첨가에 따른 집락증폭의 상승효과는 없었으며 배양 후 형성된 거핵구 집락 수는 태아 간, 특발성 혈소판 감소성 자반증 환아의 골수, 범혈구 감소증 환아의 골수 순이었다. 거핵구 집락 중 순수집락 수는 태아 간이 특발성 혈소판 감소성 자반증 및 범혈구 감소증 환아의 골수에 비해서 유의하게 높았으며, 순수집락 중 large CFU-Mk의 비율은 태아 간이 특발성 혈소판 감소성 자반증 및 범혈구 감소증 환아의 골수에 비해서 높았으나 유의성은 보이지 않았다. 태아 간 단핵세포를 5일간 flask 배양한 후 CD34 양성세포의 발현율은 증가하였으나 성장인자의 첨가에 따른 집락증폭의 상승효과는 TPO 투여군에서만 있었고, CD41 양성세포의 발현율 역시 배양 후 증가하였으나 성장인자 첨가군에 비해서 대조군의 발현율이 너무 높은 결과를 보였다. Methylcellulose 배지를 이용한 단핵세포 배양에서 CFU-GM 집락 수는 태아 간의 경우 배양 7일에 비하여 14일에 감소하였고, 성장인자의 첨가에 따른 집락증폭의 상승효과는 태아 간에서는 모든 군에서 보였으나 특발성 혈소판 감소성 자반증에서는 GM-CSF 투여군에서만 있었으며 범혈구 감소증 환아의 골수에서는 없었다. 결 론: $MegaCult^{TM}-C$ 배지를 이용하여 태아 간세포를 배양하고 면역조직학적 염색으로 거핵구 집락형성을 성공적으로 관찰하였으며, 태아 간에서 얻은 거핵구 집락이 특발성 혈소판 감소성 자반증 및 범혈구 감소증 환아의 골수에서 얻은 집락에 비하여 집락수나 순수집락의 분포, 집락의 크기 등에서 우수한 것으로 나타났으나 태아 간 조혈모세포 이식의 실제적인 임상적용을 위해서는 체계적인 더 많은 연구가 필요할 것으로 생각된다.
