Sooty Mould Infection on Mulberry-Management

  • Black sooty mould fungus was observed on the upper side of the mulberry leaves caused by the Capnodium sp. This fungus develops with the utilization of the honeydew dropped by the whiteflies. Few selected insecticides like Monocrotophos, Chloropyriphos and Nuvan were tried to control the whitefly incidence and followed by the application of Maida (wheat flour paste) and Starch solution separately to control the incidence of the Capnodium on mulberry. It is found that a significant control of the whitefly incidence with the application of Nuvan (2 $m\ell$/L) and followed by Chloropyriphos (2 $m\ell$/L) and Monocrotophos (1.6 $m\ell$/L) and also a significant control of sooty mould infection were recorded with Starch and Maida application.

Comparison and Analysis of Insecticide Resistance of Two Spotted Spider Mite (Tetranychus urficae) among Apple Orchards (사과과수원 점박이응애의 약제 저항성 비교 분석)

  • Six insecticides, monocrotophos (24%, Lq), milbemectin (1%, Ec), tebufenpyrad (10%, Ec), propargite (30%, Wp), dicofol (42%, Ec), and fenpropathrin (5%, Ec) were studied for their $LC_{50}s$ to local two-spotted spider mite (TSSM) strains collected at apple orchards in Chungju, Kunwi and Sobo in Korea. Monocrotophos and fenpropathrin were not effective due to resistance development, but milbemectin, tebufenpyrad, dicofol and propargite were effective to TSSM. $LC_{50}$ values to TSSM strains showed the same distribution pattern among apple orchards. However, TSSMs from different apple orchard were clustered into different groups.

  • For the control of Corythucha ciliata, the insecticidal activity of five commercial pesticides (Monocrotophos 25%SL, Phosphamidon 50%SL, Imidacloprid 20%DC, Thiamethoxam 15%DC and Acetamiprid 20%SL) were examined with trunk injection method in Seoul, Sanbon and Chungju. The insecticidal activity of five commercial pesticides was >88%. The insecticidal activity of Monocrotophos maintained 31 days later after trunk injection when treated leaves were supplied to this pest. This result indicates that one application of trunk injection is enough to control this pest, and this method can save the control cost compared with ground application which should be applied two or three times per year to control this pest. To make the model of treatment dosage at each D.B.H class (cm), $1.0m{\ell}/DBH$, $1.5m{\ell}/DBH$ and $2.0m{\ell}/DBH$ pesticide dosage was used. At <20cm, the insecticidal activity of $1.0m{\ell}$ dosage was >95%, and $1.5-2.5m{\ell}$ dosage was needed for the effective control at 30~50cm. More than $2.5m{\ell}$ dosage was effective at >50cm. Spray method was used in laboratory to select pesticides for ground application. Three commercial pesticides (Ethofenprox 20%EC, Ethofenprox 10%WP and Cyfluthrin 1%EC) were used, and all pesticides showed >95% insecticidal activity.

  • A series of experiments were carried out to evaluate the differences in susceptibility of the 13-aphidicidial insecticides in the green peach aphids(Myzus persicae) collected from 13 different localities. The aphids were reared and increased on the potted tobacco plants in the laboratory. The susceptibility to insecticides was compared at the $LC_{50}$ levels with the resistant ratio by a leaf-dip method with a laboratory strain reared for 2 years in the laboratory without exposure to insecticides. The susceptibility to the insecticides was greatly varied with the local strains. The local strains demonstrated relatively high resistance to the insecticides over the laboratory strain at the $LC_{50}$ level; in resistant fold, 2.3 to 519.0 to cypermenthrin, 2.3 to 494.5 to decamethrin, 2.8 to 442.4 to fenvalerate, 2.5 to 170.6 to formothion, 1.5 to 494.5 to decamethrin, 2.8 to 442.4 to fenvalerate, 2.5 to 170.6 to formothion, 1.5 to 231.8 to phosphamidon, 3.1 to 42.1 to monocrotophos, 1.0 to 30.9 to phenthoate+dimethoate, 1.8 to 21.0 to heptanophos, 2.1 to 24.8 to oxydemeton-methyl, 1.0 to 24.9 to thiometon, 1.6 to 4.7 acephate, 0.8 to 4.1 to pirimicarb.

Monitoring of Pesticide Residues in Special Products (지역특산품 중 잔류농약 실태조사)

  • This experiment was carried out to analyze for pesticide residues in 17 different types of the special of geographical indication. We purchased 3 cereal grains, nuts and seeds, 3 fruits, 8 vegetables, mushrooms and other plants (Korean medicines) mainly at the agricultural cooperative's joints markets. Total 209 pesticides including multi-analysed pesticides (204) and single-analysed pesticides (5 ; acephate, methamidophos, monocrotophos, omethoate, vamidothion) were analysed with a GC/MS/MS, an HPLC/UVD (PDA) and a GC/FPD. No. 83 method and single-analysed method (Screening of multi-pesticide residue in the special products of geographical indication) of Korea Food Code was selected for validation in recovery and interferences of matrice. The results were as follows: among the selected 17 the special products, the residual pesticides were detected in 8 types of the special products (40 in 302 samples, detection ratio; 13.2%). All of the samples were not detected over MRLs, but tebuconazole, procymidone and isoprothioran were detected with considerable high frequency. These results could be used as KFDA official methods for the analysis of pesticide residues in foods and reference data will be provided to the related institutions.

A Study on Residual Pesticides in Commercial Fruits & Vegetables (시중 유통 과채류 중의 잔류농약에 관한 연구)

  • The 88 kinds of pesticide residues were analyzed in 12 kinds(289 samples) of fruits and vegetables brought at department store & mart in Kwangju from Feburary to September in 1999. The detection rate of pesticide residue in 12 kinds(289 samples) of fruits and vegetables was 21.1%, and the rate exceeds standard was 5.2%. The order of highly exceed rate in fruits and vegetables were perilla leaf, lettuce, spinach and chickery. The order of pesticide which founded frequently in fruits and vegetables were procymidone, endosulfan, vinclozoline, chlorpyrifos, cyprermethrin, pirimiphos-M, fenvalerate, fenarimol, and monocrotophos. And that of pesticide which exceed highly standard were procymidone, vinclozolin, endosulfan, pirimiphos-M, fenarimol, monocrotophos, etc. Eight different washing methods were compared for removal efficiencies of residual pesticides(procymidone, vinclozoline) on the perilla leap, the lettuce, the spinach, the strawberry, and the minitomato. Residual pesticides such as procymidone and vinclozoline were removed most efficiently by ultrasonic washing.

Inheritance if Resistance in Dicofol-selected Strain of the Two-spotted Spider Mite, Tetranychus urticae Koch (Acarina: Tetranychidae), and its Cross Resostance (Dicofol 저항성 점박이응애의 저항성 유전과 교차저항성)

  • Dicofol susceptible two-spotted spider mite(S) which was selected by 150 times of dicofol treatment showed 27 5-fold increase in resistance as compared to S strain. Inheritance of resistance and crossresistance in selected strain($R_d$) were investigated. There were differences oi susceptibility in the dicofol concentmtion-moltaliity relationships between $F_1$ progenies (R♀, XS♂, R♂, XS♀) obtained from reciprocal cross with the S strain and $R_d$, strain. Degree of dominance of the $F_1$ progenies (R♀, XS♂, R♂, XS♀) were -0.29 and -0.72 in adult females and -0.42 and -0.93 in adult males, respectively. These results suggest that inheritance of dicofol resistance is controlled by an incomplete recessive. $R_d$ strain exhibited a high level of cross resistance to amitraz, and also showed cross resistance to pyrethrotd acaricides, acrinathrin and bifenthnn. $R_d$ strain showed non-cross reslstancz to abamectin, chlolfenson, clofentezine, cyhexatm, fenbutatin oxlde, fenpyroximate, hexythiazox, monocrotophos, propargite and tetradifon, but negatively correlated cross-resistance to azocyclotin and fenbutatin oxide belonging to tin acaricides.

  • Simple diagnostic kits for monitoring insecticide susceptibility of beet armyworm, Spodoptera exigua (Hiibner) were developed and applied to the field populations. The operation of the kits was based on the correlations between enzyme activities of esterase (EST) and acetylcholinesterase (AChE) and the insecticide susceptibilities. Four different kinds of diagnostic kits (ED, EM, AD, and AM) were designed and classified by diagnostic enzymes (E for esterases and A for acetylcholinesterase) and inhibitors (D for dichlorvos and M for monocrotophos). Diagnostic inhibitor concentrations were 1 mM for ED, 10 mM for EM, 100 mM for AD, and 100 mM for AM. Resistant larvae which were not inhibited by the diagnostic amounts of insecticides developed positive staining (red color), but susceptible~ s howed negative (no color). An insect was used for both EST and AChE diagnostic kits, but different in their samples: hemolymph for EST and the head for AChE. These four diagnostic kits were applied to 1 1 different populations which showed variations of insecticide susceptibilities. Four kits were different in the capability discriminating the insecticide susceptibilites according to insecticides: ED to bifenthrin, AD to methomyl, and ED and AM to chlorpyrifos-methyl. These diagnostic devices can be used for insecticide-resistance management program for this insect pest. It also provide a technical guide to insect pest management for farmers, directors, and researchers.

  • The acute oral, intraperitoneal and subacute toxicity of monocrotophos, an insecticide, was studied in albino rats. The acute oral $LD_{50}$ values for female and male rats were 5.0mg/kg and 8.7mg/kg, respectively. Conversely the intraperitoneal $LD_{50}$ values for female and male rats were 4.9mg/kg and 6.0mg/kg, respectively. Plasma cholinesterase (ChE) activity in rats treated with a single dose was the most depressed 4 hours after administration, but returned to normal within 72 hours of administration. Significant depression of body weight gain and food consumption was observed in rats receiving the 3.5mg/kg/day dose level for 28 days as compared with the controls. Additionally plasma ChE activity depression was also observed.

Simultaneous Analysis of 17 Organophosphorous Pesticides in Blood by Automated Head Space-SPME GC/MS (HS-SPME-GC/MS에 의한 혈액중 17종 유기인계 농약의 동시분석법)

  • HS-SPME-GC/MS was studied and optimized for the determination of 17 orgarnophosphorous pesiticides (OPPs: chlorpyrifos, chlorpyrifos-methyl, demeton-s-methyl, diazinon, dimethoate, EPN, fenitrothion, fenthion, malathion, methidathion, monocrotophos, parathion, phenthoate, phosphamidon, sulfotep, terbufos, triazophos) in blood. Optimum SPME parameters were selected: choice of SPME fiber (85 ${\mu}m$ polyacrylate), pH effect (0.5 N HCl), salt effect ($Na_2SO_4$, 0.2 g; 20%), headspace incubation temperature ($80^{\circ}C$), headspace incubation time (1 min), headspace adsorption time (30 min) and GC desorption time (2 min). These parameters were optimized using HS-SPME autosampler coupled with gas chromatography-mass spectrometry (GC-MS). Method validation was carried out in terms of linearity, limit of detection (LOD), limit of quantitation (LOQ) and recovery in blood. The assay was linear over 0.5~5.0 mg/l ($r^2$=0.955~1.000). Limit of detection (LOD) and limit of quantitation (LOQ) in blood were determined 0.03~0.3 mg/l (S/N=3) and 0.1~1.1 mg/l (S/N=10), respectively. Relative recovery with 0.5, 1 and 5 mg/l (in blood) were 90.8%, 98.5% and 94.1%, respectively. This method will be applied to the determination of the orgarnophosphorous pesticides in postmortem blood. The proposed protocol can be an attractive alternative to be used in routine toxicological analysis.
