Chemical Characterization of Neutral Extracts Prepared by Treating Pinus radiata Bark with Sodium Bicarbonate

  • A neutral extract (NE), that is soluble in cold water and has excellent antioxidant activity, from Pinus radiata pine bark was prepared by sodium bicarbonate treatment, and its chemical characteristics were investigated. NE was prepared by treating P. radiata bark with 0.8% NaHCO3 aqueous solution with a 5 : 1 liquor-to-bark ratio at boiling temperature for 1 h, resulting in 44% yield and final pH of 6.66. The yield of NE was 11% higher than that of the hot water extract (HWE) due to the increase in the solubility of polyphenols, the main component in the bark, by NaHCO3 treatment. NE was characterized through FT-IR, NMR, and MALDI TOF MS analyses. The results indicated that NE is mostly composed of proanthocyanidins (PAs) consisting of procyanidin (PC) units. The acetylated neutral extract (Ac-NE) had weight average molecular weight (${\bar{M}}w$) of 5,300 Da. The Ac-NE had wide molecular weight distribution and its polydispersity (${\bar{M}}w/{\bar{M}}n$) was 6 times higher than that of pure PA. The antioxidant activity of NE was determined by 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay and showed that NE had comparable antioxidant activity with pure PA.

  • Degradation of the starch-polyethylene composite film by an activated sludge treatment and a soil burial test was examined. The treated films had deformed structures which were resulted from the removal of starch during the treatments. The added auto-oxidant caused the degradation of the polyethylene matrices and the crosslinking reactions after the oxidation of polyethylene chains as well. As a result of soil burial test, the degradation reaction of the composite film containing corn oil as an auto-oxidant was, however, prevalent : number- and weight-average molecular weight of polyethylene decreased, while the molecular weight distribution increased.

  • The distributions of 3 disperse dyes with different molecular structures among polyurethane (PU) and polyester (PET) in dyeing PET-PU blend and which dye structure contaminated less PU portion were studied. The amounts of dyes left substrates after reduction clearing and washing were measured to prove which dye structure had an effect on improving washing fastness. Disperse dye with high molecular weight showed similar distribution onto PU and polyester by dying both substrates with similar rate and less contamination to polyurethane while other dyes distributed to PU faster than to PET had.

  • Characteristics of pyrolysis and liquid product distribution of ABS plastics have been studied in the thermogravimetric(TG) reactor and bomb microreactor. Pyrolysis reactions were performed at temperature $400\sim450^{\circ}C$ and yield of each pyrolytic product was obtained by the weight measurement method. The molecular weight distributions of liquid products were determined by the GC-SIMDIS method. It was observed that solid residue which could not be detected in the thermogravimetric experiments was significantly formed in the batch-type microreactor. It was found that the yield and average molecular weight of liquid products were decreased with the increase of reaction temperature and time. but the formation of styrene monomer was significantly increased. The chain-end scission rate parameters were determined to be 54.1kcal/mole far ABS by the Arrhenius plot.

  • Liquid-phase catalytic degradation of waste high-density polyethylene (HDPE) over ZSM-5 (powder type (PW)) and ZSM-5+binder (granule type (GR)) has been investigated with a stirred semi-batch operation at 400°C. Two ZSM-5 catalysts with a different crystal size were synthesized and also each ZSM-5 (25%) Catalyst was mixed with a same binder (kaolin: silica sol: alumina = 55%:10%:10%). The performance of prepared catalysts that has different physicochemical properties was discussed with the cumulative amount distribution, molecular weight distribution and also paraffin, olefin, naphthene and aromatic (PONA) distribution in liquid product. These liquid product quality and distributions were changed depending on the physicochemical properties of the catalyst. Moreover, the characteristic of ZSM-5 in the catalyst was strongly influenced on the activity and PONA distribution in liquid product.

Changes in Molecular Weight Distribution and Enzyme Susceptibility of Rice Starch by Extrusion-cooking and Simple Heat-treatment (압출조리와 단순 열처리에 의한 쌀전분의 분자량 분포변화와 효소민감성에 관한 연구)

  • The changes in molecular size distribution of rice starch during extrusion cooking and simple heating of rice flour were compared and the effect of subsequent enzyme treatment on the molecular size was examined. A single-screw extruder was used with varing feed moisture contents ($17{\sim}29%$) and barrel temperatures ($100{\sim}150^{\circ}C$). An aluminium capsule immersed in oil bath ($100{\sim}200^{\circ}C$) was used for the simple heat treatment of rice flour. In case of extrusion cooking the mechanical energy input varied sharply at around 23% moisture content of the feed. At the feed moisture content of $17{\sim}23%$, a significant molecular size reduction of rice starch was observed by the gel permeation chromatography using Sephacryl S-1000 gel. The intact starch molecules of above $4{\times}10^{7}$ dalton were largely disintergrated by extrusion cooking of rice flour containing the moisture content less than 23%. It was mostly degraded further into the molecules having below $5{\times}10^{6}$ dalton by ${\alpha}-amylase$ treatment. But at the feed moisture content above 26% the starch did not show molecular size reduction either by extrusion cooking or by subsequent enzyme treatment. On the contrary little changes in molecular size of starch was occured by simple heating of rice flour containing the moisture less than 20%. but slight size reduction was observed at the moisture content above 23%, where the effect of ${\alpha}-amylase$ was also observed.

Application of Coliform Bacterial Plasmid as a Trophic Indicator (콜리형 세균 Plasmid의 영양단계 지표 활용에 관한 연구)

  • This present study has been carried out to examine the correlation between the distribution of the coliform bacterial plasmids and the viability test against heavy metals in the upper stream of Han river(Gapyung; clean water) and the its basin (Anyang Cheon; polluted water). And the distribution of plasmids were examined to be used as trophic indicator for analysis between the clean and polluted waters. 1. A total of 110 isolates were analyzed for the presence of plasmids by means of the boiling method and agarose gel techniques. Plasmids were significantly more frequent in the strains which had been isolated from the clean water (14.3%). Also, there were much higher multiplicity of plasmids at the polluted water(41.0%), compared with the clean water (33.0%). By the comparision between molecular weight of bacterial plasmids in the clean water and those of polluted water, there were no significant differences of the clean water from the polluted water, to such extent as 30.0%, 28.6%, respectively, in frequency for occurrence of high molecular weight plasmids iver 35.8% Mdal. 2. Each isolate was carried out the resistance test for mercury(10-5M), nickel(10-3M) and arsenic (0.1%). At the polluted site, the survival ability of the plasmid-carrying straings(Hg, 31.0%l Ni, 5.7%; As, 65.7%) was higher than that of the non plasmid-carrying strains(Hg, 12.1%; Ni, 3.0%; As, 54.6%). This trend was more remarkable in the clean site (plasmid-carrying strains: 16.7%,-,-). As a result it is suggested that plasmids could be used as an indicator of a certain types of water pollution. In addition, heavy metals might have inflyenced, some extent, to the distribution of plasmids in the environment which has been surveyed in the present study.

  • The variations of the glass transition, melting peaks, molecular weight and its distribution (polydispersity index: PI) due to the annealing temperature and time have been investigated using the thermoplastic segmented polyurethanes (TPUs) and its blends based on the contents of hard segment. The position of the melting peak and its magnitude have been increased with the annealing temperature and time. This may be arised from the rearrangement of the microdomain structure due to the long-range or short-range segmental motion, the order-disorder transition of non-crystalline microphase, the variation of the domain size or the degree of disorder of crystalline structure by given different thermal histories. The annealing temperature and time affected the molecular weights and polydispersity : the number and weight average molecular weights were increased, while the polydispersity index (PI) deceased at certain temperatures : for TPU-35 at $135^{\circ}C$, for TPU-44 at $170^{\circ}C$ and for TPU-53 at $180^{\circ}C$. The temperatures which give the variations in molecular weights and in PIs are consistent with the annealing temperatures of which $T_3$ solely exists for each sample. Thus it is suggested that the chain dissosiation and recombination simultaneously occur at the above mentioned temperature for each sample.

Gelatinization Properties and Molecular Structure of Waxy Rice Starches Isolated from Korean Japonica and Indica Cultivars (국내산 자포니카와 인디카 품종 찹쌀전분의 호화특성과 분자구조)

  • Gelatinization properties and the molecular structure of Korean waxy rice starchesisolated from two japonica types, Sinseonchal, and Dongjinchal, as well as an indica type, Hangangchal 1 were investigated. Sinseonchal is preferred cultivar for making Korean traditional rice cakes and cookies. Sinseonchal starch was the highest in crude protein, amylopectin, damaged starch contents, and water binding capacity among the cultivars tested. The initial pasting temperature ($72.75^{\circ}C$), peak (360.54 RVU), breakdown (162.21 RVU) and setback (30.83 RVU) viscosities of Sinseonchal had the highest values (p<0.05). Onset and peak temperatures by differential scanning calorimeter were also the highest in Sinseonchal. The molecular weight of Sinseonchal amylopectin was 5.46 107higher than those of the other cultivars, but its peak height and area were the lowest among them. The amylopectin peak by HPSEC showed a shoulder in the lower molecular weight portion and its relative area decreased in the following order; Sinseonchal > Dongjinchal > Hangangchal 1. On the branch chain length distribution of amylopectin, the proportion of DP13-24 and DP25-36 showed reverse trends, with higher japonica type amylopectin in DP13-24.

Structure and Properties of Starch on Rice Variety (계통이 다른 쌀 전분의 구조적 성질)

  • The molecular structure of rice starch was investigated using Korean rice[3 varieties of Japonica type and 3 varieties of Tongi type(Japonica-Indica breeding type)]. The λmax of iodine complex and inherent viscosity of Japonica type were higher than those of Tongil type. $\beta$-Amylolysis limit of the starches was not different between the two rice types. In the distribution of molecular weight of rice starch, the molecular size of amylose and amylopectin for Japonica type were smaller than those for Tongil type. The chain of rice starch distributed F1 of above DP 55, F2 of DP 40~50 and F3 of DP 15~20, and the ratio of F3 against F2 for Japonica type was higher than that of Tongil type. The results suggest that rice of Japonica and Tongil type was different molecular structure of starch.
