Molecular Structure of Sorghum and Waxy Sorghum Starches

  • Amylose contents and amylopectin chain architecture of sorghmn and waxy sorghum starches were determined and compared with those of other common cereal and tuber starches. Also, in vitro digestibility of sorghum starch was estimated using a novel methodology. The absolute amylose content of sorghum starch was similar to that of com and wheat starches. The side chain length distribution patterns for sorghum and waxy sorghum amylopectin were very similar to those of com and waxy com, respectively. The $k_{cat}/K_m$ values for sorghum and potato amylopectin did not show a significant difference. The kinetic parameters could be used as novel indicators for starch digestibility.

Inhibitory Effects of Soyeum Pharmacopuncture (SPP) on LPS-induced Inflammation Related Cytokine Expressions of RAW 264.7 cells (소염약침의 LPS로 유도된 RAW 264.7 세포의 염증성 사이토카인 발현에 미치는 영향)

  • Aim This study was done to investigate whether SPP has inhibitory effects on the activation of RAW 264.7 cells. Method In tumor necrosis factor-a (TNF-a)/ interleukin-1b (IL-1b) and IL-6, the mRNA expression of molecular indicators related to inflammatory changes of the Reumatoid Arthritis (RA) were examined using quantitative real-time PCR. Results The treatment of SPP significantly suppressed the expression of proinflammatory cytokines and chemokines such as TNF-a, IL-1b, IL-6 compared with the control. The expression of NOS-II was considerably reduced, which was accompanied by a reduction in the production of nitric oxide (NO). It also reduced the expression of $TNF-{\alpha}$ in serum of Balb/c mice compared with control group. Conclusion SPP is an effective herbal material for suppressing the inflammation related cytokines of RAW 264.7 cells.

Status of Helicobacter pylori Infection among Migrant Workers in Shijiazhuang, China

  • Background: Helicobacter pylori infection leads to many upper gastrointestinal diseases. Migrant workers are the main part of floating population in China. However, up to now, their health status has not been a focus of attention. Methods: In order to assess the status of H. pylori infection among migrant workers in Shijiazhuang, over five years we interviewed 324 individuals between 2007 and 2011. Each underwent a rapid urease test to identify H. pylori infection and socio-demographic indicators were collected using a survey questionnaire. Results: Our results showed that family income (P = 0.003), dietetic hygiene (P = 0.005), education (P = 0.004) and marital status (P = 0.007) were associated with H. pylori infection. Conclusion: We found that migrant workers had little basic knowledge of H. pylori and their prevalence of infection remains high. Therefore, we need to promote education and awareness of H. pylori and to ensure access to diagnosis and treatment for infected workers.

Combined Effects of Multiple Endoplasmic Reticulum Stresses on Cytokine Secretion in Macrophage

  • Cells show various stress signs when they are challenged with severe physiological problems. Majority of such cellular stresses are conveyed to endoplasmic reticulum (ER) and unfolded protein response (UPR) serves as typical defense mechanism against ER stress. This study investigated an interaction between ER stress agents using macropage cell line Raw 264.7. When activated by lipopolysaccharide (LPS), the cell lines showed typical indicators of ER stress. Along with molecular chaperones, the activation process leads to the production of additional inflammatory mediators. Following activation, the macrophage cell line was further treated with TUN and characterized in terms of chaperone expression and cytokine secretion. When treated with TUN, the activated macrophage cell leads to increased secretion of IL-6 although expression of ER stress markers, GRP94 and GRP78 increased. The secretion of cytokines continued until the addition of BFA which inhibits protein targeting from ER to Golgi. However, secretion of cytokines was ceased upon dual treatments with BFA and TG. This result strongly implies that cells may differently deal with various polypeptides depending on the urgency in cellular function under ER stress. Considering IL-6 is one of the most important signal molecules in macrophage, the molecule might be able to circumvent ER stress and UPR to reach its targeting site.

  • Relative to the outer surface of the mastic coating, the reliability of the available waterproofing resource is determined by the ability to stabilize the structural characteristics in difficult climatic conditions. Organic components of mastic as a result of solar radiation, elevated temperatures and their alternating change, atmospheric oxidants, especially in industrial areas, have a tendency to self-polymerization and loss of low molecular weight components. This is the gradual loss of deformability and the transition to brittleness with its tendency to crack as the reasons for the gradual transition from normal to emergency operating condition.The presented mechanism of functioning of the coating surface indicates the expediency of increasing its components, able to stabilize the structure and prevent changes in deformability.Durability, hydrophobicity, water displacement, water absorption are accepted as estimating indicators. The main dependences of the influence of the lost additional components of mastic on the operational properties of the formed coating characterize the ability to provide successful resistance to environmental influences and longer stability. As a result, mastic acquires additional service life.

Circular RNA expression profiles in the porcine liver of two distinct phenotype pig breeds

  • Objective: An experiment was conducted to identify and characterize the circular RNA expression and metabolic characteristics in the liver of Jinhua pigs and Landrace pigs. Methods: Three Jinhua pigs and three Landrace pigs respectively at 70-day were slaughtered to collect the liver tissue samples. Immediately after slaughter, blood samples were taken to detect serum biochemical indicators. Total RNA extracted from liver tissue samples were used to prepare the library and then sequence on HiSeq 2500. Bioinformatic methods were employed to analyze sequence data to identify the circRNAs and predict the potential roles of differentially expressed circRNAs between the two breeds. Results: Significant differences in physiological and biochemical traits were observed between growing Jinhua and Landrace pigs. We identified 84,864 circRNA candidates in two breeds and 366 circRNAs were detected as significantly differentially expressed. Their host genes are involved in lipid biosynthetic and metabolic processes according to the gene ontology analysis and associated with metabolic pathways. Conclusion: Our research represents the first description of circRNA profiles in the porcine liver from two divergent phenotype pigs. The predicted miRNA-circRNA interaction provides important basis for miRNA-circRNA relationships in the porcine liver. These data expand the repertories of porcine circRNA and are conducive to understanding the possible molecular mechanisms involved in miRNA and circRNA. Our study provides basic data for further research of the biological functions of circRNAs in the porcine liver.

Isolation of a Bacteriocin - Producing Lactobacillus sakei Strain from Kimchi (김치에서 박테리오신을 분비하는 Lactobacillus sakei균주의 분리)

  • Bacteriocin producing lactic acid bacteria (LAB) were isolated from Kimchi by using spot-on-the-lawn method. Listeria monocytogenes, Staphylococcus aureus, and Lactobacillus plantarum were used as indicators. One isolate (P3-l) produced a bacteriocin efficiently inhibiting the growth of Listeria monocytogenes. 16S rDNA sequence and sugar utilization test identified that P3-1 was a Lactobacillus sakei strain. Accordingly, the isolate was named as Lactobacillus sakei P3-1. L. sakei P3-1 produced a bacteriocin which efficiently inhibited the growth of Listeria monocytogenes but did not inhibit other Gram positive and negative organisms tested. The bacteriocin was stable against heat, organic solvent, and pH variation and it retained 50% of activity after 10 min heat treatment at 10$0^{\circ}C$. The molecular weight of Sakacin P3-1 was estimated to be 4 kDa by SDS-PAGE.

  • The purpose of this study was to provide fundamental data for the plans to reduce occupational stress by analyzing occupational stress of medical technologist. The experimental group comprised of 193 medical technologists working in Gyeonggi province. Occupational stress was 49.74 out of 100. In detail, job insecurity showed a high value of 64.42, followed by organizational system, lack of reward, physical environment, job demand, insufficient job control, interpersonal conflict, and occupational climate with respective values of 52.46, 52.30, 51.18, 49.57, 48.43, 39.78 and 39.77. It is recommended that those in high-stress job group increase physical activity and reduce job insecurity to improve health. The development of programs to reduce occupational stress are necessary, and this study can provide objective information using biological indicators.

Vitamin D Sufficiency: How Should it be Defined and what are its Functional Indicators?

  • It has been more than three decades since the first assay assessing circulating 25 (OH)D in human subjects was performed That publication as well as several that followed it defined 'normal' nutritional vitamin D status in human populations. Recently, the wisdom by which 'normal' circulating 25 (OH)D levels in human subjects were assigned in the past has come under question. It appears that sampling human subjects, who appear to be free from disease, and assessing 'normal' circulating 25 (OH)D levels by plotting a Gaussian distribution is grossly inaccurate. There are many reasons why this method is inaccurate, including race, lifestyle habits, sunscreen usage, age, latitude, and inappropriately low dietary recommendations for vitamin D. For instance, a 400 IU/day. AI for vitamin D is insignificant when one considers that a 10-15 minute whole body exposure to peak summer sun will generate and release up to 20,000 IU vitamin $D_3$ into the circulation. Recent studies, which orally administered up to 10,000 IU/day vitamin $D_3$ to human subjects for several months, have successfully elevated circulating 25 (OH)D levels to those observed in individuals from sun-rich environments. Further, we are now able to accurately assess sufficient circulating 25 (OH)D levels utilizing specific biomarkers instead of guessing what an adequate level is. These biomarkers include intact parathyroid hormone (PTH), calcium absorption, bone mineral density (BMD), insulin resistance and pancreatic beta cell function. Using the data from these biomarkers, vitamin D deficiency should be defined as circulating levels of 25 (OH)D$\leq$30 ng/mL. In certain cases, such as pregnancy and lactation, significantly higher circulating 25 (OH)D levels would almost certainly be beneficial to both the mother and recipient fetus/infant.

Suppressive Effects of GST on Cytokine-induced Activation of Human Fibroblast-like Sinoviocytes (가미사물탕(GST)의 사이토카인으로 유도된 인간 섬유아세포양 활막 세포 활성화 저해 작용)

  • GST, an extract from 16 herbs, has been formulated and prescribed for the treatment of human rheumatoid arthritis(hRA) for many years. The present study was done to investigate whether GST has suppressive effects on activation of fibroblast-like sinoviocytes isolated from an RA patient. In tumor necrosis factor-a(TNF-a)/interleukin-1b(IL-1b) treated human sinoviocytes, The mRNA expression of molecular indicators related to pathologic changes of the sinoviocytes were examined using quantitative real-time PCR. The treatment of GST($100\;{\mu}g/ml$) suppressed the expression of proinflammatory cytokines and chemokines such as TNF-a, IL-1b, IL-6 and IL-8 compared with the control. The mRNA level of intracellular adhesion molecule-1(ICAM-1) which is known to increase in the activated sinoviocytes of RA patients, was slightly decreased by GST. The expression of NOS-II was considerably reduced, which was accompanied by a decrease in the production of nitric oxide(NO). In addition, GST considerably increased the mRNA levels of tissue inhibitors of matrix metalloproteinase-1(TIMP-1), while those of matrix metalloproteinase-3(MMP-3) were decreased. Taken together, these data suggested that GST might suppress the activation of sinoviocytes in hRA.
