A Study on a New Method of Measurement of Pore Volume Distribution in Porous Materials by Adsorption of Gases

  • A new method of measurement of pore volume distribution in a porous material by adsorption of gases is proposed. In the newly proposed method the use of spherical molecules with relatively large molecular weight as adsorbates is strongly recommended to eliminate the troubles caused by stereospecificity and low boiling point of widely used $N_2$. It is also claimed that the universal t-curves that have been widely used since proposed by de Boer et al. must be replaced by the flat-surface isotherms that can be derived from the theory proposed by Chang et al. The results of analysis of pore volume distribution based on the method described in this paper are found to be reasonable and satisfactory.

Distribution Patterns of the Members of Phylum Acidobacteria in Global Soil Samples

  • The distribution pattern of the phylum Acidobacteria, a previously uncultured bacterial group, was investigated by molecular ecological analyses of global soil samples collected from pristine ecosystems across five continents. Acidobacterial 16S rDNAs were observed in almost all soil samples, and members of acidobacterial primer group A were detected in all samples that harbored the phylum Acidobacteria. Other primer groups, Y, G, and O, showed limited distribution patterns. We further divided the primer groups into acidobacterial subdivisions (class-level). Subdivisional distribution patterns were determined by comparing the observed T-RFs with theoretical T-RFs predicted by in silico digestion of acidobacterial 16S rDNAs. Consistent with the PCR results obtained with subgroup-specific primers, T-RFLP analyses showed that acidobacterial subdivision 1 belonging to primer group A was present in the majority of the soil samples. This study revealed that the phylum Acidobacteria could be globally distributed. At the subdivisional level, acidobacterial subdivision 1 might be the most widely distributed group in this phylum, indicating that members of subdivision 1 might be adapted to various soil environments, and members belonging to other subdivisions might be restricted to certain geographic regions or habitats.

효소처리에 의한 Bombyx mori fibroin 가수분해물의 분자량 및 구조 변화 (Molecular Weight and Structural Changes of Enzymatic Hydrolysate from Bombyx mori Fibroin)

  • 본 연구는 누에 실 샘으로부터 얻은 Bambyx mori 피브로인의 효소 가수분해 시 그 분자량 및 구조적 특성을 파악하고자 하였다. Gel filtration chromatography(GFC)에 의해 B. mori 피브로인을 순수 분리한 결과 회수율이 약 70% 이상으로 기존의 방법(막과 튜브 분리법)에 비해 약 40% 이상 높았다. 이는 GFC법이 고 순수 고 수율의 B. mori 피브로인을 얻을 수 있는 방법임이 확인되었다. Gel permeation chromatography에 의한 분자량 측정 결과 누에 체내 피브로인, 염화칼슘 용해 물 및 효소 가수분해물이 각각 평균분자량 108,000 Da 내외, 65,000 Da 내외 및 1,000 Da 내외임을 알 수 있었다 또한 효소 가수분해에 의한 구조적 변화를 $^{13}C-NMR$로 알아본 결과, B. mori 피브로인의 메인 골격에 해당되는 glycine, alanine 및 serine 등의 골격 피크가 관찰되어졌으며, 특히 효소가수분해 시 glycine를 의미하는 40-44 ppm 부근의 피크가 높게 나타난 것으로 보아 -G-A-G-A-반복 구조가 분해되었고, 이로 인해 분자량이 현저히 저하되었을 것으로 생각되어진다.

  • 마크로머를 이용한 음이온성 아크릴 수지를 제조하기 위하여 음이온 리빙 중합법으로 TBMA를 합성하였고 이들을 다시 MMA와 공중합 시켜 아크릴계 그라프트 공중합체를 제조하였다. 그라프트 공중합체의 음이온 site 함량을 제어하기 위하여 (TBMA)마크로머의 MMA의 weight 비율을 7/93, 10/90, 15/85, 20/80, 30/70, 40/60, 50/50가지 변화시켜 합성하였다. (TBMA)마크로머의 음이온 리빙 중합과정에서는 반응온도를 $-78^{\circ}C$로 유지하면서 n-butyllithium-diphenylethylene계 개시제 시스템을 사용하였으며 정지반응 단계에서는 말단에 이중결합을 도입하기 위하여 capping material로 benzaldehyde를 사용한 후, methacryloyl chloride와 반응시켰으나, 넓은 분자량 분포(1.4~1.5)가 나타났다. 또한 그라프트 공중합체 내의 TBMA는 p-toluenesufonic acid인 촉매로 가수분해하였고 중화제로 triethylamine을 사용한 중화 반응은 아크릴 수지의 음이온성을 부여하였다. (TBMA)마크로머의 분자량과 분자량 분포에 대한 측정은 GPC를 이용하였으며 TBMA의 가수분해 반응과 더불어 아크릴 수지의 이온화 반응을 IR과 NMR을 이용하여 분석하였다. (TBMA)수분산성 및 분산 안정성의 관점에서 그라프트 공중합체내의 (TBMA)마크로머 분자량에 대한 가지(branch)의 길이와 TBMA함량을 관련지어 검토한 결과 분자량이 작은(TBMA) 마크로머에서는 적은 TBMA 함량으로도 안정된 수분산화 현상이 일어남을 알았다.

양돈폐수의 정화처리 과정별 유기물질 성분과 분자량 분포 변화 연구 (Changes in the Organic Compounds and Molecular Weight Distribution in Pig Wastewater by Each Treatment Steps.)

  • 양돈폐수의 원수, 활성오니정화처리, 오존 처리 및 $TiO_2$처리 등 각 정화처리단계별 유기물질의 성분특성 및 분자량의 분포의 변화를 알아보기 위하여 시험을 수행하였으며 그 결과는 다음과 같다. 1. 양돈폐수 원수 중에는 acetic acid, 2-butanone, hexane, chloroform, propionic acid, butanoic acid 등 33종의 유기물질이 포함되어 있었으며 phenol 28.7%, benzoic acid 9.9%, benzeneacetic acid 13.3% 등으로 고른 분포를 보였다. 2. 오존처리 후 ethene, pentane, 2-methyl-propanal, hexane, furan 등 13종의 유기물질이 존재하였으며, hexane이 67.8%로 대부분을 차지하는 것으로 나타났다. 3. $TiO_2$ 처리를 한 후에는 ethene, hexane, chloroform, 1-decene, silane 등 12종의 유기물질이 존재하였으며, ethene 8.8%, hexane 16.2%, 1-dodecene 8.0%, 1-tetradecene 10.8%, cyclo-trisiloxane 12.6% 등의 분포를 보였다. 4. 양돈폐수 원수는 500Da 이하의 분자량이 15%, 500-1,000Da 분자량이 14% 1,000∼3,000Da 분자량인 물질은 21%를 차지하였다. 5. 활성오니처리 후 분자량 500Da 이하는 7%, 500∼1,000Da 12%, 1,000∼3,000Da 10%이었으며 난분해성 물질인 분자량 3,000∼10,000Da 24%, 10,000Da 이상 26%로 고분자 물질의 분포가 높아졌다. 6. $TiO_2$ 처리 후 분자량 3,000∼10,000Da 5%, 분자량 10,000Da 이상 5%로 급격히 감소한 반면 분자량 500Da 이하 31%, 분자량 500∼1,000Da 54%를 차지였다.

Type-Specific Human Papillomavirus Distribution in Invasive Squamous Cervical Carcinomas in Tunisia and Vaccine Impact

  • Background: High risk human papillomaviruses (HPVs) are the leading cause of cervical cancer (CC) and Pap smear screening has not been successful in preventing CC in Tunisia. HPV vaccination that targets HPV16 and 18 offers a new efficient prevention tool. Identification of HPV types in CC is thus essential to determine the impact of HPV vaccine implementation. The aim of this study is to provide specific data from Tunisia. Materials and Methods: A total of 89 histological confirmed paraffin embedded samples isolated from patients with CC diagnosed between 2001 and 2011 were collected from five medical centres from Northern and Southern Tunisia. HPV DNA was detected using a nested PCR (MY09/MY11-GP5+/GP6+) and genotyping was assessed using a reverse blot line hybridisation assay that enables the detection of 32 HPV types. Results: HPV DNA was detected in all samples. Twelve high risk types were detected; HPV16 and/or 18 were predominant, accounting together for 92.1% of all the CC cases (HPV16: 83.1%). Single infections accounted for 48.8% of the cases and were mostly linked to HPV 16 (32.6%) and less frequently to HPV 18 (2.4%). The other high risk HPV single infections were linked to HPV 35 (4.6%), 45 (4.6%), 58 (2.3%) and 59 (2.3%). Multiple infections with mixing of 2 to 4 genotypes predominately featrued HPV16 and/or 18 with HPV 35 and 45 (96.6 %) and less frequently with HPV 59, 40, 66, 73 and 58. There was no statistically significant variation in the relative distribution of HPV types with age. Conclusions: These results strongly indicate that prophylactic HPV vaccines can have a major impact in preventing CC in Tunisia.

  • 흡수, 분포, 대사, 배설 특성 및 독성을 예측하기 위한 효과적인 툴을 개발하는 것은 신약개발의 초기단계에서 NCE(new chemical entity)에 대한 가장 중요한 업무 중의 하나이다. 최근에 이런 시도중의 하나로서 ADME/T(absorption, distribution, metabolism, excretion, toxicity)관련 성질들의 예측에 support vector machine(SVM)을 이용하고 있다. 그리고 SVM은 ADME/T 성질들을 정확하게 예측하는데 많이 사용 되고 있다. 그러나 SVM 모델링에 두 가지 문제가 있다. 특성 선택(feature selection) 과 매개변수 설정(parameter setting)은 여전히 해결해야 할 과제이다. 이 두 가지 문제들은 SVM 분류의 효율성과 정확도에 결정적인 영향을 끼친다. 특히 특성 선택과 최적화된 SVM 변수의 설정은 서로 영향을 주기 때문에 동시에 다루어져야 한다. 여기서 우리는 genetic algorithm(GA) – 특성 선택에 사용 – 과 grid search(GS) method– 변수최적화에 사용 – 두 가지를 통합하는 효과적인 해결책을 제시하였다. ADME/T관련 성질 중 하나인 심장부정맥을 야기시키는 hERG 이온채널 저해제 분류 모델이 여기서 제안된 GA-GS-SVM을 위해 할당되고 테스트 되었다. 1891개의 화합물을 가지는 트레이닝 셋으로 단일 모델 3개, 앙상블 모델 3개, 총 6개의 모델을 만들었고 175개의 외부 데이터를 테스트 셋으로 사용하여 검증하였다. 데이터의 불균형 문제를 해결하기 위하여 GA-GS-SVM 단일 모델에 의한 예측 정확도와 GA-GS-SVM 앙상블 모델 예측 정확도를 비교하였으며, 앙상블모델을 사용하여 예측의 정확도를 높일 수 있었다.

Molecular Cloning, Segmental Distribution and Ontogenetic Regulation of Cationic Amino Acid Transporter 2 in Pigs

  • The goal of this study was to elucidate the expression and segmental distribution of the glomerular cationic amino acid metabolism transporter-2 (CAT-2) and thus to improve our understanding of porcine cationic amino acid transporters and amino acid absorption. Porcine CAT-2 was cloned, sequenced and characterized. The predicted amino acid sequence of porcine CAT-2 shared 86.1% and 92.1% identity with human and mouse CAT-2A, respectively. The tissue distribution patterns and ontogenic changes of CAT-2 mRNAs were determined by real-time Q-PCR. The results showed that porcine CAT-2 was highly expressed in the heart and intestinal tract (duodenum, ileum and jejunum). In addition, the mRNA of CAT-2 was found in liver, lung, kidney, brain and muscle. Within the intestinal tract, CAT-2 mRNA was most abundant in the ileum and rarely expressed in the duodenum. In the duodenum, the levels of CAT-2 mRNA reached their peak on day 7 (p<0.05) while in the jejunum, levels were low on day 1 and 7 and increased rapidly after day 26 before peaking on days 30 and 60 (p<0.05). The levels then dramatically decreased by day 90 (p<0.05). In the ileum, levels achieved their maximum on day 30 and then decreased significantly on day 60 (p<0.05).

Distribution of Polycyclic Aromatic Hydrocarbons (PAHs) in Surface Sediments inside Songsanpo and Seogwipo Harbors of Jeju Island, Korea

  • The surface sediments inside Songsanpo and Seogwipo Harbors, major harbors of Jeju Island, were collected three times (June, September and December, 2001) and analyzed for 16 polycyclic aromatic hydrocarbons (PAHs) recommended by US-EPA as priority pollutants to assess their distribution levels and their biological effects on the marine organisms, and to suggest their possible origins. The concentrations of total PAHs inside Songsanpo and Seogwipo Harbors ranged from 41 to 288 ng/g on a dry weight basis with a mean value of 121 ng/g and from 14 to 268 ng/g with a mean value of 119 ng/g, respectively, and the levels were low as compared with those in other areas of the world. The sedimentary PAHs may be correlated with organic carbon and mud content to some extent. Based on comparisons of individual and total concentrations with effects-based sediment quality guidelines, the potential for the biological effects on the marine organisms were expected to be very low. From the examinations of the four PAH origin indices, such as LMW /HMW (low molecular weight 2-3 ring PAHs over high molecular weight 4-6 ring PAHs), phenanthrene/anthracene ratio, fluoranthene/pyrene ratio, chrysene/benzo[a]anthracene ratio, it can be concluded that the sediment PAH contaminations were ascribed to both of pyrolytic and petrogenic origins.

Coalescence behavior of dispersed domains in binary immiscible fluid mixtures having bimodal size distributions under steady shear flow

  • Coalescence process of binary immiscible fluid mixtures having bimodal size distributions, prepared by mixing two pre-sheared samples at different shear rates, ${\gamma}_{pre1}\;and\;{\gamma}_{pre2}$, under shear flow at a final shear rate, ${\gamma}_f$, are examined by transient shear stress measurements and microscopic observations in comparison with the results for simply pre-sheared samples having narrow size distributions (unimodal distribution samples). Component fluids are a silicone oil (PDMS) and a hydrocarbon-formaldehyde resin (Genelite) and their viscosities are 14.1 and 21.0 $pa{\cdot}sec$ at room temperature $(ca.\;20^{\circ}C)$, respectively. The weight ratio of PDMS: Genelite was 7:3. Three cases, $({\gamma}_{pre1}=7.2sec^{-1},\;{\gamma}_{pre2}=12.0sec^{-1}\;and\;{\gamma}_f=2.4sec^{-1}),\;({\gamma}_{pre1}=0.8sec^{-1},\;{\gamma}_{pre2}=4.0sec^{-1}\;and\;{\gamma}_f=2.4sec^{-1}),\;and\;({\gamma}_{pre1}=7.2sec^{-1},\;{\gamma}_{pre2}=12.0^sec^{-1}\;and\;{\gamma}_f=7.2sec^{-1})$ the first case, transient shear stress did not show any significant difference but domains larger than the initial state are observed at short times. In the latter cases, there exist undershoot of shear stress, reflecting existence of deformed large domains, which is confirmed by the direct observation. It is concluded that coalescence between large and small domains more frequently occur than coalescence between the domains with similar size in the bimodal distribution samples.
