Toxic Effects of 5 Organic Solvents on Euglena agilis (국내 생물 종 유글레나(Euglena agilis)를 이용한 5종 유기용매의 독성평가)

  • Acute toxicity tests for 5 organic solvents were conducted using Euglena agilis carter (E. agilis), a Korean domestic organism. Organic solvents decreased the growth rate of E. agilis in a dose dependent manner. The toxicity to E. agilis was increased in the order of chloroform>acetone${\geq}$ethanol${\geq}$methanol>DMSO based on $EC_{50}$ values from growth test. Organic solvents also induced cell motility and morphological changes of E. agilis. Especially significant effects on the cell swimming velocity, motility, and compactness were observed for chloroform at the concentration of $EC_{50}$ calculated from 96 hr growth test. Overall, toxic responses of E. agilis to test substances are comparable to or more sensitive than D. magna, M. macrocopa and V. fischeri. Our study demonstrates that E. agilis can be a putative ecotoxicity test model organism to assess domestic water quality. Results obtained from this study can be applied to establish the standard test guidelines for ecotoxicity test using E. agilis.

Toxic Effects of Metal Plating Wastewater on Daphnia magna and Euglena agilis (Daphnia magna와 Euglena agilis를 이용한 도금폐수 독성평가)

  • The ecotoxicity tests for metal plating wastewater were conducted using Daphnia magna (D. magna) and Euglena agilis (E. agilis). Evaluation for sources of toxicity was performed by 1) Correlation analysis between the concentration of individual metals in the metal plating wastewater and the toxic effects on D. magna, 2) Toxicant identification evaluation methods including graduated pH method, EDTA procedure and sodium thiosulfate procedure, 3) Comparison of toxic effect value ($EC_{50}$ or $LC_{50}$) of individual metal on D. magna and it's concentration in the metal plating wastewater. To evaluate the possibility of E. agilis, a Korean domestic organism, as a test model organism for metal plating waste water, E. agilis toxicity test was also assessed using on-line euglena ecotoxicity system (E-Tox system). Based on toxicant characterization test using D. magna, it was expected that SS, oxidants and heavy metals are responsible for toxicity of metal plating waste water. Especially Cu, Hg, and Ag were the major cationic metals that caused toxicity. E. agilis is less sensitive than D. magna based on the $EC_{50}$ value however it shows prompt response to toxic test substances. E. agilis shows even a significant effect on the cell swimming velocity within 2 min to toxic metal plating wastewater. Our study demonstrates that E. agilis test can be a putative ecotoxicity test for assessing the quality of metal plating waste water.

A Study on the Moderating Factors of the Relationship between Artwork Color Series and Visitor Satisfaction in Commercial Spaces (상업공간에서 미술품 색 계열과 방문객 만족도 관계의 조절요인에 관한 연구)

  • This study attempted to analyze the effect of the color series of artworks installed as environmental stimuli in commercial spaces on the satisfaction of visitors and the moderating effect of the relationship. To this end, based on the SOR model of Stimulate-Organism-Response applied to burial environment research in the field of environmental psychology, and the preceding research using the SOR model, artwork color series(S)-mood and spaace amenity(O)-A research framework for satisfaction(R) was developed. In the experiment, an online questionnaire was conducted for domestic college students and graduate students by producing images with two conditions depending on the case where warm colors and cold colors were installed for the color series of artworks. As a result of verifying the difference in satisfaction of respondents corresponding to the two conditions through regression analysis, it was found that the warm color(vs. cold color) of the artwork color series induces higher visitor satisfaction. In addition, as a result of verifying the controlling factors of mood and space amenity variables in this relationship of influence, a significant moderating effect was found when the positive mood of warm colors(vs. cold colors) in the artwork color series was felt higher than the average. And, of the four types of space amenity, it was found that a significant moderating effect appeared when only comfort and aesthetics were measured as moderating variables. The result of this study proves that the warm color series of artworks that stimulate the physical environment of commercial spaces has a more positive effect on the satisfaction of visitors than the cold color series, and this is reinforced by positive mood, comfort, and aesthetics. It adds understanding and provides useful implications for marketing strategies for building an effective spatial image.

  • Time series analysis is a frequently effective method of constructing model and prediction in data processing of deformation monitoring. The monitoring data sample must to be as more as possible and time intervals are equal roughly so as to construct time series model accurately and achieve reliable prediction. But in the project practice of GPS deformation monitoring, the monitoring data sample can't be obtained too much and time intervals are not equal because of being restricted by all kinds of factors, and it contains many variates in the deformation model moreover. It is very important to study the data processing method for small samples and multi-variates time series in GPS deformation monitoring. A new method of establishing small samples and multi-variates deformation model and prediction model are put forward so as to resolve contradiction of small samples and multi-variates encountered in constructing deformation model and improve formerly data processing method of deformation monitoring. Based on the system theory, a deformation body is regarded as a whole organism; a time-dependence linear system model and a time-dependence bilinear system model are established. The dynamic parameters estimation is derived by means of prediction fit and least information distribution criteria. The final example demonstrates the validity and practice of this method.

Models of Pseudomonas Growth Kinetics and Shelf Life in Chilled Longissimus dorsi Muscles of Beef

  • The aim of this study was to confirm Pseudomonas spp. as the specific spoilage organism (SSO) of chilled beef during aerobic storage and to establish a model to predict the shelf life of beef. Naturally contaminated beef was stored at $4^{\circ}C$, and the spoilage limit of Pseudomonas organisms was determined by measuring several quality indicators during storage, including the number of Pseudomonas organisms, total number of bacteria, total volatile basic nitrogen (TVBN) values, L value color scale scores and sensory evaluation scores. The beef was then stored at 0, 4, 7, 10, 15 or $20^{\circ}C$ for varying amounts of time, and the number of Pseudomonas organisms were counted, allowing a corresponding growth model to be established. The results showed that the presence of Pseudomonas spp. was significantly correlated to each quality characteristic (p<0.01), demonstrating that Pseudomonas spp. are the SSO of chilled beef and that the spoilage limit was $10^{8.20}$ cfu/g. The Baranyi and Roberts equation can predict the growth of Pseudomonas spp. in beef, and the $R^2$ value of each model was greater than 0.95. The square root model was used as follows, and the absolute values of the residuals were less than ${0.05:\;{\mu_{max}}^{1/2}$ = 0.15604 [T+(-0.08472)] (p<0.01), $R^2$ = 0.98, $\lambda^{-1/2}$ = 0.0649+0.0242T (p<0.01, $R^2$ = 0.94). The model presented here describes the impact of different temperatures on the growth of Pseudomonas spp., thereby establishing a model for the prediction of the shelf life of beef stored between 0 to $20^{\circ}C$.

  • Accurate software cost estimation is essential to both developers and customers. Most of the cost estimating models based on the size measure methods, such as LOC and FP, are obtained through size estimation. The accuracy of size estimation directly influences the accuracy of cost estimation. As a result, the overall structure of regression-based cost models applies the power function based on software size. Many growth phenomenon in nature such as the growth in living organism, performance of technology, and learning capability of human show an S-shaped curve. This paper proposes a model which estimates the developing effort by using the growth curve. The presented model assumes that the relation cost and size follows the growth curve. The appropriateness of the growth curve model based on Function Point, Full-Function Point and Use-Case Point, which are the general methods in estimating the software size have been confirmed. The proposed growth curve model shows similar performance with power function model. In conclusion, the growth curve model can be applied in the estimation of the software cost.

Practical Use of Tissue Biosensor for Safety Test of Marine Organism and Monitoring of Public Health and Environment (해양 유독생물의 독성 검사와 보건환경 모니터링을 위한 조건센서의 활용)

  • It confirmed the facilitated diffusion of $Na^+$ of frog bladder membrane which is a tissue membrane. The mechanism was explained in $Na^+$ channel model and its referred to the $Na^+$ channel obstruction ingredient which was contained in the reference to the $Na^+$ channel obstruction ingredient and son on, e.g., seaweed, shellfish, pufferfish, phytoplankton and chinese drug. Also, it introduces the result which studied from the barrier point of the application of the tissue biosensor to the trade friction on Korea or Japan pufferfish and the marine environment in the one with high dependance. It was possible for the poison quantity of small amount pufferfish toxin (TTX), paralytic shellfish poisoning (PSP) to be measured and also to measure poison quantity in the cultivation poisonous toxin phytoplankton individual. In future, as for this tissue biosensor, it expects that it is possible to contribute widely until environment watch and also monitoring to the scene.

  • Under pathological stress stimuli, dynamics of a biological system can be changed by alteration of several components such as functional proteins, ultimately leading to disease state. These dynamics in disease state can be modeled using differential equations in which kinetic or system parameters can be obtained from experimental data. One of the most effective ways to restore a particular disease state of biology system (i.e., cell, organ and organism) into the normal state makes optimization of the altered components usually represented by system parameters in the differential equations. There has been no such approach as far as we know. Here we show this approach with a cardiac hypertrophy model in which we obtain the existence of the optimal parameters and construct an optimal system which can be used to find the optimal parameters.

  • The goal of this study is to develop a biomarker used in monitoring abnormal behaviors of Japanese medaka (Oryzias latipes) as a model organism caused by hazardous chemicals. Japanese medaka was treated under appropriate sublethal concentrations of copper and the fish were subjected to copper treatment after starvation for 48 hr. The untreated individuals showed common behavioral characteristics (i.e., smooth and linear movements with small curvatures) in the movement behaviors.

  • Fluoranthene, a common polycyclicaromatic hydrocarbon (PAH), exhibits phototoxicity which may affect aquatic organisms. The eventual goal of this study is to develop a biomarker used in monitoring abnormal behaviors of Japanese medaka (Oryzias latipes) as a model organism caused by hazardous chemicals that are toxic and persistent in the ecosystem. In this study, we investigated neural toxicity of fluoranthene in Japanese medaka (Oryzias latipes) which was correlated with its behaviors.
