Effect of Black Garlic and Gaeddongssuk (Artemisia annua L.) Extracts on the Lipid Profile and Hepatic Antioxidant Enzyme Activities of Exercised Rats (흑마늘 및 개똥쑥 추출물의 급이가 강제운동 시 흰쥐의 체내 지질 성분 및 간조직의 항산화효소 활성에 미치는 영향)

  • To develop functional products based on black garlic, a black garlic extract (BG) of 7 brix, a gaeddongssuk extract (GS) of 0.7 brix and two types of mixtures (MBS-I, black garlic 7 brix : gaeddongssuk 0.7 brix; MBS-II, black garlic 14 brix : gaeddongssuk 1.4 brix, 93:7, v/v) were supplemented to rats training on a treadmill for 4 weeks. Body weight from the training did not decrease during the experimental period. Serum albumin content significantly increased in the groups fed an experimental diet compared to the control. The BUN content significantly decreased in BG and MBS-II groups compared to the control. AST and ALP activities significantly decreased in the groups fed an experimental diet compared to the control. Serum triglyceride and total cholesterol levels in MBS-I and MBS-II groups significantly decreased compared to the control. Lipid levels in the serum and liver tissue were not significantly different between the MBS-I and MBS-II groups. The lipid peroxide content in the serum and liver tissue was significantly reduced in the groups fed all extracts compared to the control; the serum and liver lipid contents was lowest in the MBS-I and MBS-II groups, respectively. Hepatic catalase activity in the GS and MBS groups increased by 1.8~2.3 times compared to the control. SOD and GSH-px activities significantly increased from treatment with the extracts by 1.3~1.5 times and 1.2~1.7 times, respectively. These results indicate that a mixture of BG and GS extracts has higher biological activity than a single supplementation of BG or GS extract. Therefore, the addition of gaeddongssuk to black garlic (MBS-I and MBS-II) is effective as a defense material against oxidative stress. MBS-I may be especially effective for its biological activities.

Effect of Soybean Meal and Soluble Starch on Biogenic Amine Production and Microbial Diversity Using In vitro Rumen Fermentation

  • This study was conducted to investigate the effect of soybean meal (SM) and soluble starch (SS) on biogenic amine production and microbial diversity using in vitro ruminal fermentation. Treatments comprised of incubation of 2 g of mixture (expressed as 10 parts) containing different ratios of SM to SS as: 0:0, 10:0, 7:3, 5:5, 3:7, or 0:10. In vitro ruminal fermentation parameters were determined at 0, 12, 24, and 48 h of incubation while the biogenic amine and microbial diversity were determined at 48 h of incubation. Treatment with highest proportion of SM had higher (p<0.05) gas production than those with higher proportions of SS. Samples with higher proportion of SS resulted in lower pH than those with higher proportion of SM after 48 h of incubation. The largest change in $NH_3$-N concentration from 0 to 48 h was observed on all SM while the smallest was observed on exclusive SS. Similarly, exclusive SS had the lowest $NH_3$-N concentration among all groups after 24 h of incubation. Increasing methane ($CH_4$) concentrations were observed with time, and $CH_4$ concentrations were higher (p<0.05) with greater proportions of SM than SS. Balanced proportion of SM and SS had the highest (p<0.05) total volatile fatty acid (TVFA) while propionate was found highest in higher proportion of SS. Moreover, biogenic amine (BA) was higher (p<0.05) in samples containing greater proportions of SM. Histamines, amine index and total amines were highest in exclusive SM followed in sequence mixtures with increasing proportion of SS (and lowered proportion of SM) at 48 h of incubation. Nine dominant bands were identified by denaturing gradient gel electrophoresis (DGGE) and their identity ranged from 87% to 100% which were mostly isolated from rumen and feces. Bands R2 (uncultured bacterium clone RB-5E1) and R4 (uncultured rumen bacterium clone L7A_C10) bands were found in samples with higher proportions of SM while R3 (uncultured Firmicutes bacterium clone NI_52), R7 (Selenomonas sp. MCB2), R8 (Selenomonas ruminantium gene) and R9 (Selenomonas ruminantium strain LongY6) were found in samples with higher proportions of SS. Different feed ratios affect rumen fermentation in terms of pH, $NH_3$-N, $CH_4$, BA, volatile fatty acid and other metabolite concentrations and microbial diversity. Balanced protein and carbohydrate ratios are needed for rumen fermentation.

Changes of Blood Gases, Plasma Catecholamine Concentrations and Hemodynamic Data in Anesthetized Dogs during Graded Hypoxia Induced by Nitrous Oxide (아산화질소에 의한 점진적 저산소가스 흡입이 혈중 가스치와 Catecholamine치 및 혈역학에 미치는 영향)

  • The sympathoadrenal system plays an important role in homeostasis in widely varing external environments. Conflicting findings, however, have been reported on its response to hypoxia. We investigated the effect of hypoxia on the sympathoadrenal system in dogs under halothane anesthesia by measuring levels of circulating catecholamines in response to graded hypoxia. Ten healthy mongreal dogs were mechanically ventilated with different hypoxic gas mixtures. Graded hypoxia and reoxygenation were induced by progressively decreasing the oxygen fraction in the inhalation gas mixture from 21%(control) to 15%, 10% and 5% at every 5 minutes, and then reoxygenated with 60% oxygen. Mean arterial pressure, central venous pressure and mean pulmonary arterial pressure were measured directly using pressure transducers. Cardiac output was measured by the thermodilutional method. For analysis of blood gas, saturation and content, arterial and mixed venous blood were sampled via the femoral and pulmonary artery at the end of each hypoxic condition. The concentration of plasma catecholamines was determined by radioenzymatic assay. According to the exposure of graded hypoxia, not only did arterial and mixed venous oxygen tension decreased markedly at 10% and 5% oxygen, but also arterial and mixed venous oxygen saturation decreased significantly. An increased trend of the oxygen extraction ratio was seen during graded hypoxia. Cardiac output, mean arterial pressure and systemic vascular resistance were unchanged or increased slightly. Pulmonary arterial pressure(PAP) and pulmonary vascular resistance(PVR) were increased by 55%, 76% in 10% oxygen and by 82%, 95% in 5% oxygen, respectively(p<0.01). The concentrations of plasma norepinephrine, epinephrine and dopamine increased by 75%, 29%, 24% in 15% oxygen and by 382%, 350%, 49% in 5% oxygen. These data suggest that the sympathetic nervous system was activated to maintain homeostasis by modifying blood flow distribution to improve oxygen delivery to tissues by hypoxia, but hemodynamic changes might be blunted by high concentration of nitrous oxide except PAP and PVR. It would be suggested that hemodynamic changes might not be sensitive index during hypoxia induced by high concentration of nitrous oxide exposure.

Development of Protein-rich Food Mixtures for Infants and Growing Children in Korea -(4) Nutritional and Biochemical Evaluation of Formulas F-P-5, F-P-6, F-P-7 and storage stability of F-P-4- (유유아(乳幼兒) 및 성장기아동을 위한 영양식품 개발에 관한 연구 -(4) F-P-5, F-P-6 및 F-P-7의 영양학적, 생화학적 검토 및 그 저장성-)

  • From the previous studies, F-P-4 formula was found to be comparable to full fat dry milk in its nutritive value and feeding performance. However, an attempt was made in order to make sure whether or not any possibility might exist, by which further improvement of nutritive quality and simultaneous reduction of product costs may be achieved. Using F-P-4 as a control, modifications were made in new formulas, F-P-5, F-P-6 and F-P-7 by reducing FPC, eliminating yeast from the mixture, and by enriching with methionine as needed. In particular, F-P-7 is completely free of FPC, hydrogenated oil and yeast. Yet, levels of total protein and fat were kept equal to those of F-P-4 in all formulas. An animal feeding test for all formulas using 10 female rats per group for 8 weeks and an infant feeding trial for F-P-5 and F-P-6 with 5 of each female infants under age of one for one month were conducted along with F-P-4 as a control. Almost the same results were obtained with F-P-4, 5 and 6, but F-P-7 showed the lowest body weight gain. FER of F-P-5 and 6 was 0.20 as was with F-P-4, while that of F-P-7 was 0.16. Acceptability to infants was excellent; growth, appearance and biochemical data were normal. As an example F-P-4 packed in 0.04mm polyethylene bags was used for storage study at $25^{\circ}C$ and relative humidity of $65{\sim}85%$ for 8 months. Although viable bacterial counts and vitamin C contents were reduced, peroxide and TBA values were increased gradually during such storage. Since there are also significant changes in color and organoleptic quality, the expected shelf life under the given conditions is considered to be about 2 months and thus further works are needed both on the product and packaging in order to improve the storage stability. Either elimination of yeast form F-P-4, that is F-P-5, or partial replacement of FPC with methionine, that is F-P-6 may well reduce material costs about 10%. Considering blending process of ingredients, F-P-5 is thus found to be the best formula developed. While F-P-7 free of FPC is inferior in its nutritive quality than that of others, but significantly superior than of rice. Furthermore, the material cost of the product can be reduced about 20% from that of F-P-4. And thus this vegetable blend is considered to be useful as a low cost supplementary food mixture for growing children.

Intergrowth and Interlayering of Muscovite, Chlorite, and Biotite in a Garnet Zone Metamorphic Rock of the Ogcheon Belt, South Korea (옥천대의 석류석데 변성암에서 산출되는 백운모, 녹니석 및 흑운모의 Intergrowth와 Interlayering)

  • Muscovite, chlorite and biotite in metapelites of the Ogcheon Hetamorphic Belt are studied using electron probe microanalysis (EPMA), backscattered electron images (BEI) of scanning electron microscopy (SEM) and lattice fringe images of transmission electron microscopy (TEM). These minerals are observed to be intergrown under a polarized light microscope and are apparently interlayered below EPMA resolution; EPMA data often indicate mixtures of phyllosilicates such as muscovite/chlorite (M/C), biotite/chlorite (B/C), muscovite/pyrophyllite/chlorite (M/P/C). biotite/pyrophyllite/chlorite (B/P/C) or biotite/muscovite/chlorite (B/M/C). BEI observations show that the three minerals (muscovite, chlorite and biotite) are mixed at various scales in a grain through the garnet zone, and the interlayering of the three minerals are observed from TEM lattice fringe images and selected area electron diffraction patterns. The result of TEM observations reveals that 7-$\AA$ layers (serpentine, precursor of chlorite) are interlayered within 10-$\AA$ layers (muscovite) at 100~200 $\AA$ scale as well as M/C in the chlorite zone. The 7-$\AA$ layers become smaller in size and less frequent in the biotite tone, and 10-$\AA$ layers are interlayered with chlorite (14 $\AA$) at an individual layer scale. The 7-$\AA$ layers are no longer observed in the garnet zone, and 10-$\AA$ layers (biotite) are interlayered with chlorite (B/C) at 50~100 $\AA$ scale. Relatively large scale (1000~2000 $\AA$) of intergrowth is also frequently observed from the garnet zone samples. However, rocks from all three metamorphic zones show interlayering of a few units of 7-, 10- and 14-$\AA$ layers with each other at TEM observations. The result of this study implies that metamorphic minerals such as muscovite, chlorite and biotite form through disequilibrum mineral reactions resulting in inhomogenious phases.

Geochemistry and Mineralogical Characteristics of Precipitate formed at Some Mineral Water Springs in Gyeongbuk Province, Korea (경북지역 주요 약수의 지화학과 침전물의 광물학적 특성)

  • Mineralogical characteristics of secondary precipitate formed at some mineral water springs in Gyeongbuk Province, Korea were studied in relation to water chemistry. The chemical water types of mineral water springs are mostly classified as $Ca-HCO_3$ type, but $Na(Ca)-HCO_3$ and $Ca-SO_4$ types are also recognized. Ca, Fe, and $HCO_3\;^-$ are the most abundant components in the water. The pH values of most springs lie in 5.76${\sim}$6.81, except Hwangsu spring having pH 2.8. Saturation indices show that all springs are supersaturated with respect to iron minerals and oxyhydroxides such as hematite and goethite. The result of particle size analysis shows that the precipitate is composed of the composite with various sizes, indicating the presence of iron minerals susceptible to a phase transition at varying water chemistry or the mixtures consisting of various mineral species. The particle size of the reddish precipitate is larger than that of the yellow brown precipitate. Based on XRD and SEM analyses, the precipitate is mostly composed of ferrihydrite (two-line type), goethite, schwertmannite, and calcite, with lesser silicates and manganese minerals. The most abundant mineral fanned at springs is ferrihydrite whose crystals are $0.1{\sim}2\;{\mu}m$ with an average of $0.5\;{\mu}m$ in size, characterized by a spherical form. It should be interestingly noted that schwertmannite forms at Hwangsu spring whose pH is very low. At Shinchon spring, Gallionella ferruginea, one of the iron bacteria, is commonly found as an indicator of the important microbial activity ascribed to the formation of iron minerals because very fine iron oxides with a spherical form are closely distributed on surfaces of the bacteria. A genetic relationship between the water chemistry and the formation of the secondary precipitate from mineral water springs was discussed.

Thermal-Denaturation of File Fish Myofibrillar Protein and Protective Effect of Sucrose, Sorbitol and Amino Acids (말쥐치 근원섬유단백질의 열안정성과 및 가지 첨가제의 영향)

  • Thermal-denaturation of myofibrillar protein of dorsal skeletal muscle from file fish was investigated by measuring denaturation constant($K_D$) and thermodynamic parameters at various temperatures. The protective effects of sucrose, sorbitol and amino acids when added individually or combined were also discussed. The denaturation rate as reflected in inactivation of myofibrillar protein Ca-ATPase was followed the first order reaction. The $K_D$ values at $25^{\circ}C,\;30^{\circ}C,\;and\;35^{\circ}C$ were $19.52{\times}10^{-5},\;112.25{\times}10^{-5},\;and\;247.20{\times}10^{-5}$, respectively. The activation energy of the reaction at $30^{\circ}C$ was 43 kcal/mole. The protective effects of sucrose, sorbitol, glycine, alanine and Na-glutamate were increased with the concentration but the effects of sorbitol and Na-glutamate decreased beyond 1.0 mole. Basic amino acids such as arginine and lysine did not revealed any protective effect on the thermal denaturation. In case of mixed addition, the effects of Na-glutamate to glycine, sorbitol to glycine, and sorbitol to sucrose or sorbitol to Na-glutamate were enhanced 1.2 to 7.0 times as much as that of control (ratio of mixing; 1:1, range of concentration; 0.5 to 1.25 mole). Under the frozen condition at $-20^{\circ}C$, two mixtures such as Na-glutamate to glycine and sorbitol to sucrose apparently revealed the protective effects.

  • The combinatorial approach has been applied to discover and optimize the composition of the novel or enhanced materials. In this study, we screened the optimum composition of the system SrO-Gd$_2$O$_3$-Al$_2$O$_3$ doped with $Tb^{3+}$ by a polymerized-complex combinatorial chemistry method. Mixtures with compositions of Sr, Gd and Al component that is in the range from 0 to 1 in about 0.05 increments could be tested. The sample powders were synthesized by a polymerized complex method. To prepare appropriately polymeric precursors, we used the metallic nitrates, citric acid and ethylene glycol. The luminescence properties of the synthesized powders are investigated using the UV and VUV (Vacuum-UV: 147 nm) photoluminescence spectrometer. In addition, the crystallinity and morphology of powder were monitored by X-ray diffraction spectrometer and scanning electron microscopy. In result of VUV PL works, there are good luminescent samples with the composition of 0.595 < x < 0.733 and 0.016 < y < 0.017 in Gd1-x-yAlxTbyO$\delta$ and 0.049 < x < 0.064 and 0.02 < y < 0.039 in $Sr_xAl_{1-x-y}Tb_yO_$\delta$$, their materials can be applicable to plasma display panels as the green phosphor.

Treatment of Artificial Sewage Using a Zeolite Column (제올라이트 칼럼에 의한 인공생활하수처리)

  • Constructed wetlands typically cost less to build and operate, and require less energy than standard mechanical treatment technology but they have similar performance to centralized wastewater treatment plants. Therefore, they were constructed especially many in rural areas, where are small villages but not industries. Plantless column tests were performed to investigate the possibility on using zeolite as a filter medium of constructed wetland for the wastewater treatment. Removal efficiency was $COD_{Cr}$ 94.63% T-P 41.41% and $NH_4^+-N$ 99.75% at hydraulic load 314 $L/m^2{\cdot}d$ and filtering height 100 cm filled with a zeolite mixture. This zeolite mixture consisted of 1 : 1 by volume of zeolites in the diameter range of 0.5 to 1 mm and 1 to 3 mm. Accordingly, hydraulic load 314 $L/m^2{\cdot}d$ was considered as optimal. Three zeolite mixtures were used to determine the optimal mixing ratio by volume of a zeolite (A) in the diameter range of 0.5 to 1 mm to a zeolite (B) in the diameter range of 1 to 3 mm 1 : 3, 1 : 1 and only B in A to B by volume were tested at hydraulic load 314 $L/m^2{\cdot}d$ and filtering height 100 cm $COD_{Cr}$ removal efficiency was more than 89% at mixing ratios of 1 : 3 and 1 : 1 in A to B. That of T-P ranged 56.42 to 58.72% and, that of T-N and $NH_4^+-N$ was 87% and 99% regardless of mixing ratios of two zeolites. Removal efficiency was lower at the column filled with only B. Removal efficiency was better at Inter medium filled with mixing ratio 1 : 1 in A to B than with the other mixing ratios. Thus, it was found that the mixture of mixing ratio 1 : 1 in A to B was appropriate far Inter medium of constructed wetland Removal efficiency was higher in down-flow than in up-flow, and all contaminants were removed most in 20 cm filter height near feeding area.

Determination of Optimum Fermentation Condition for Alcohol-Fermented Feeds with Wet Brewer's Grains (맥주박을 이용한 알코올 발효사료의 최적 발효조건 규명)

  • To determine the optimum fermentation condition, the 50:50 feed mixture of both crushed corn and wet brewer’s grain were anaerobically fermented at various conditions. Three supplementation levels(0, 2%, and 5%) of molasses, five supplementation levels(0, 0.1%, 0.5%, 1.0%, and 2.0%) of yeast and five different incubation temperatures(4$^{\circ}C$, 10$^{\circ}C$, 30$^{\circ}C$, 40$^{\circ}C$ and 50$^{\circ}C$) were tested to determine the optimum fermentation conditions. During fermentation, alcohol concentration, live yeast cell number (LYCN) and pH values of the mixture were analyzed. Alcohol concentrations of the fermented feed mixtures were proportionally increased with increasing level of molasses supplementation. After 24hr fermentation, alcohol concentrations at 0.1%, 0.5%, 1.0% and 2.0% of yeast supplementation was 1.9%, 2.4%, 3.1% and 3.1%, respectively. These results indicate a proportional increase of alcohol concentration with the increasing supplementation of yeast (P<0.05) up to 1%, thereafter showing plateau at 2% supplementation. LYCN was relatively high with more than 0.5% yeast supplementation, whereas the value was significantly low(P<0.05) at 0.1% yeast supplementation. Alcohol production was significantly higher at 30$^{\circ}C$ and 40$^{\circ}C$ than other incubation temperatures (P<0.05), with the highest concentration at 30$^{\circ}C$. pH was rapidly decreased until 24 hours of incubation with steeper decrease at lower temperatures(P<0.05). Based on there results, optimum fermentation conditions for the alcoholic-fermented feed production could be achieved with 5% supplementation of molasses, 1% supplementation of yeast, and 24hr long incubation at 30$^{\circ}C$.
