Effects of Stireed Tank Bioreactor Scale-up on Cell Growth and Alkaloids Production in Cell Cultures of Eschscholtzia californica (탱크 교반형 생물반응기의 scale-up이 Eschscholtzia californica 세포생장 및 알칼로이드 생성에 미치는 영향)

  • Studies were made to investigate effects of the scale-up of stirred tank bioreactors on cell growth and alkaloids production for suspension cultures of Eschscholtzia californica. In the 1.5 L STR, cell lysis was observed at 110 rpm or higher agitation speed. The agitation speed of 30 L STR was 43.7 rpm to maintain the same shear stress developed in 1.5 L STR of 100 rpm. As a result of scale-up from 1.5 L to 30 L STR, the specific growth rate was decreased from 0.12 to 0.07 day-1. The alkaloids productivity was also decreased from 0.24 to 0.14 mg/L-day. Changes of mixing performance and oxygen transfer were studied to explain the decrease of cell growth and alkaloids production. Decreased oxygen transfer rate coefficient(KLa) and increased mixing time by the scale-up was observed at various aeration rates.

  • Turbulent in-situ mixing process is a new material process technology to get dispersed phase in nanometer size by controlling reaction of liquid/solid, liquid/gas, flow ana solidification speed simultaneously. In this study, mixing which is the key technology to this synthesis method was studied by computational fluid dynamics. For the simulation of mixing of liquid metal, static mixers investigated. Two inlets for different liquid metal meet ana merge like 'Y' shape tube having various shapes and radios of curve. The performance of mixer was evaluated with quantitative analysis with coefficient of variance of mass fraction. Also, detailed plots of intersection were presented to understand effect of mixer shape on mixing. The simulations show that the Reynolds number (Re) is the important factor to mixing and dispersion of $TiB_2$ particles. Mixer was designed according to the simulation, and $Cu-TiB_2$ nano composites were evaluated. $TiB_2$ nano particles were uniformly dispersed when Re was 1000, and cluster formation and reduction in volume fraction of $TiB_2$ were found at higher Re.

  • In this study, a comparative study was performed to predict the vapor-liquid equilibria with maximum azeotropic pressure for ethanol-benzene binary system between an equation of state model and a liquid activity coefficient model. Peng-Robinson equation of state model with a Panatiotopoulos mixing rules (PRP) was used and NRTL liquid activity coefficient model proposed by Renon was selected. The PRP model, even though it has only two binary adjustable parameters, was not inferior to the NRTL model to predict vapor-liquid equilibria for low pressure region of ethanol-benzene system and showed a better prediction capability for high pressure region of ethanol-benzene system than the NRTL model with three binary interaction parameters.

  • The effects of ion beam mixing of a SiC film coated on super alloys (hastelloy X substrates) were studied, aiming at developing highly sustainable materials at above $900^{\circ}C$ in decomposed sulfuric acid gas (SO2/SO3/H2O) channels of a process heat exchanger. The bonding between two dissimilar materials is often problematic, particularly in coating metals with a ceramics protective layer. A strong bonding between SiC and hastelloy X was achieved by mixing the atoms at the interface by an ion-beam: The film was not peeled-off at ${\geq}900^{\circ}C$, confirming excellent adhesion, although the thermal expansion coefficient of hastelloy X is about three times higher than that of SiC. Instead, the SiC film was cracked along the grain boundary of the substrate at above $700^{\circ}C$. At ${\geq}900^{\circ}C$, the film was crystallized forming islands on the substrate so that a considerable part of the substrate surface could be exposed to the corrosive environment. To cover the exposed areas and cracks multiple coating/IBM processes have been developed. An immersion corrosion test in 80% sulfuric acid at $300^{\circ}C$ for 100 h showed that the weight retain rate was gradually increased when increasing the processing time.

Effect of Vertical Mixing Scheme on Upper Ocean Simulation of the East Sea (연직혼합모수화가 동해 상층 모사에 미치는 영향)

  • This study investigates effects of three different parameterizations of vertical mixing scheme on upper ocean simulation of the East Sea, focusing on the seasonal variations of the sea surface temperature(SST) and the mixed layer depth(MLD) using an ocean general circulation model(GFDL MOM1.1). The considered vertical mixing schemes are the Laplacian scheme(L scheme) that use a constant eddy coefficient, the Mellor-Yamada scheme(MY scheme), and a new scheme(Noh scheme). The Noh scheme, a second-order turbulence closure, was developed considering recent observational evidences such as the enhancement of turbulent kinetic energy near the sea surface. During summer L scheme underestimates the SST, while MY scheme overestimates the SST, compared to climatological SST. Noh scheme produces the SST in better agreement with climatological one. During winter all schemes overestimate the SST up to $4^{\circ}C$ compared to climatological SST. Vertical profiles of the basin-mean temperature show that L scheme produces higher temperature below the thermocline than those of other schemes. The winter MLD simulated from L scheme is rather large compared to that from other schemes, but the differences in MLD during summer are not significant.

Applicability of Recycled Soil Mixed with Bentonite-Polymer for Waste Landfill Liner (순환토사의 벤토나이트-폴리머 혼합비에 따른 매립지 차수재 적용성에 관한 연구)

  • In this study, it was studied recycled soils with bentonite-polymer mixture in order to design economic landfill instead of clay liner. Recycled soil was used as SP, a sandy soil with 90.58% sand and 1.88% silt and clay. The recycled soils were mixed with 4%, 6%, and 8% bentonite by weight, and then compared with samples mixed with 2%, 3%, and 4% bentonite by weight in marine clay. Recycled soil satisfied the permeability criteria at 8%, and clay soil satisfied at 3%. In order to make a sample that satisfies the standard of the waste landfill, a permeability test was conducted by mixing 0.16%, 0.24%, and 0.28% of the polymer in a sample having 4% bentonite mixing ratio. The unconfined compression strength test was carried out at the same mixing ratio to confirm that the specification was satisfied. As the bentonite mixture ratio increased, the permeability coefficient and unconfined compression strength decreased. The strength in polymer mixing increased initially and then maintained a constant value. At 4% bentonite mixing ratio and 0.28% polymer mixing ratio, the coefficient of permeability was 1.0×10-7 cm/sec or less, and the unconfined compression strength was over 500 kPa. It was confirmed that it can be used as a mixed liner material of waste landfills.

A Study on the Mechanical Properties of Steel Fiber Reinforced Porous Concrete for Pavement Using Slag Aggregate and Fly Ash (슬래그골재와 플라이애시를 이용한 강섬유 보강 포장용 투수콘크리트의 역학적 특성에 관한 실험적 연구)

  • This study evaluates the mechanical properties of steel fiber reinforced porous concrete for pavement according to content of slag aggregate and fly ash to elicit the presentation of data and the way to enhance its function for the practical field application of porous concrete as a material of pavement. As a result, void ratio and permeability coefficient of porous concrete for pavement increased a little as mixing rate of slag aggregates increased. Void ratio and permeability coefficient increased a lot as mixing rate of fly ash decreased. As fly ash was mixed, national regulation of permeable concrete for pavement(8% and 0.1 cm/sec) was met. Compressive strength and flexural strength decreased as mixing rate of slag aggregates increased, but they increased a lot as mixing rate of fly ash increased. Even when slag aggregates were mixed 50% with 5% fly ash mixed, national regulation of pavement concrete(18MPa and 4.5MPa) was met. In addition, compared to non-mixture, flexural strength increased about 22.8% when 0.75vol.% of steel fiber was added. Regarding sliding resistance, BPN increased as mixing rate of slag aggregates increased. But BPN decreased as fly ash was mixed. Compared to crushed stone aggregates, abrasion resistance and fleers-thaw resistance decreased as mixing rate of slag aggregates increased. When fly ash was mixed, abrasion resistance and freeze-thaw resistance improved remarkably. Compared to non-mixture, 10% mixture of fly ash improved abrasion resistance and freeze-thaw resistance about 5.6% and 14.3 respectively.

Comparison of Coefficient of Consolidation and Prediction of Excess Pore Water Pressure of Agricultural Reservoir under Embankment on Soft Ground (연약지반상에 축조된 농업용저수지의 과잉공극수압 예측과 압밀계수의 비교)

  • This study was carried out to comparison of coefficient of consolidation and the prediction of excess pore water pressure in agricultural reservoir on soft clay ground. For the purpose of verification of the proposed equation, laboratory model tests and field tests were performed and excess pore water pressure was compared to those predicted with the Terzaghi's method. The predicted excess pore water pressure according to ponding was very applicable to practice because it was close to the observed data. Also, for the comparison of coefficient of consolidation, the oedometer, constant rate of strain (CRS), and Rowe cell tests were performed. The coefficient of consolidation at the Rowe cell and CRS tests showed a greate increase than in the oedometer test. The ratio of the vertical and horizontal coefficient of consolidation showed a large difference according to various tests method and mixing ratio. Therefore, it is recommended that careful attention should be paid to predicting the required consolidation period in agricultural reservoir.

A Study on the Sensitivity Analysis of Line Source Air Quality Models (移動汚染源에 대한 大氣擴散模型의 感應度 分析에 관한 硏究 (HIWAY2, PAL, CALINE3 模型을 對象으로))

  • The sensitivity analysis is a method to quantify to what extent the output of a model changes with the values of input parameters. This will lead to increase model accuracy through measurement validation. Three line source air quality models, HIWAY 2, PAL, and CALINE 3 were selected for this study. The input parameters analysed included wind speed, wind direction, stability, emission rate, mixing height, receptor distance, initial dispersion coefficient, surface roughness, and averaging time. It turned out that PAL model generally showed higher concentration than other two models, and that between CALINE 3 and HIWAY 2, CALINE 3 showed higher concentration than HIWAY 2 model near the line sources, but beyond a certain downwind distances HIWAY 2 model showed higher concentration. The modesl were very sensitive to wind speed especially in the range of 0 $\sim$ 1 m/s and to wind direction near the parallel wind to streets. In case of emission rate, the output concentration was directly proportional to these input parameters. And the sensitivity of the input parameters such as stability, mixing height, initial dispersion coefficient, surface roughness, and averaging time were not very significant.

  • This paper presents the measuring technique for local heat transfer coefficients using a copper sensor in a rod bundle with mixing vanes. A copper sensor consists of a cartridge heater and four pieces of thermocouple. The Heater is located at the center of the copper sensor and thermocouples measure the surface temperature of the copper sensor. Unheated copper sensor and heated copper sensor are able to measure the local heat transfer coefficient at the position where the heated copper sensor is installed. However the entire region of a rod bundle is actually not heated, the decay of local heat transfer coefficients measured represents overestimated value rather than an actual value. The calibration curve for local heat transfer coefficients is presented using the correction factor calculated by CFD.
