The Effect of $Ca^{2+}$ on the Interaction of Anionic-Nonionic Surfactant Mixture with Iodine (음이온성-비이온성 혼합계면활성제와 요오드간의 상호작용에 미치는 $Ca^{2+}$의 영향)

  • In the presence of SDS(sodium dodecyl sulfate), effect of $Ca^{2+}$on the interaction of NP-40EO[nonylphenol-(ethylene oxide)40] with iodine in aqueous solution were investigated by UV-visible spectrophotometer. In the presence of SDS, the intensity of interaction peaks were decreased and markedly increased by the addition of $Ca^{2+}$. Decrease of interaction peaks is caused by decrease of EO number to be interact with iodine per unit surface area of mixed micelle to the insertion of SDS and increase is attributed to the compactness of micelle in the presence of $Ca^{2+}$. These phenomena may be explained by the fact that the linear EO (ethylene oxide) chains, to be free in aqueous solution, could form a pseudo-crown ether structures of forming with $Ca^{2+}$ion.

  • In this study the solubility of cholesterol was examined with ginseng saponin and chemical saponin used in the preparation of reaction mixture of 0.15M NaCl solution. It was shown that the solubility of cholesterol varied in the concentration of ginseng saponins and in the temperature and that the osmotic pressure and the surface tension was amenable to changes in the concentration of ginseng saponins and the incubation period of the reaction mixture. From the results, it was revealed that the solubility of cholesterol was dependent upon the incubated times and the concentration of ginseng saponins in 0.15mM NaCl solution and that ginseng saponins have the cholesterol solubilizing effect as evidenced in the physicochemical properties of mixed micelle formation and surface-activity.

Characterization of Emulsion Properties for D-limonene (D-리모넨 오일의 유화특성)

  • Microemulsion is prepared by the method of phase inversion emulsification with d-limonene that is environmental friendly substance and nontoxic to human body as dispersed phase. Emulsifier used for preparation of microemulsion is nonionic surfactants, polyoxyethylene nonylphenyl ether. Stability of prepared microemulsion was estimated by the various method of ξ-potential, hydrodynamic diameter and electric conductivity. When d-limonene is emulsified by NP series, microemulsion is most stable and narrowly distributed at HLB value of 12.3(either one emulsifier or mixed emulsifiers). Stability of microemulsion is increased as the amount of emulsifiers is increased at same HLB value of 12.3. In the case of using the same amount of emulsifiers, number of produced micelle are relatively large as hydrodynamic diameter is small. Therefore, the state of microemulsion is stable and the electric conductivity is increased. One can determine that higher electric conductivity value means that microemulsion has more micelles and is more stable.

  • The ultra pH-sensitive polymeric mixed micelles based on poly(L-histidine) chemistry and constructed from block copolymers containing polyHis, present four functionalities as decreasing pH: ligand exposure at pH 7.0, micelle destabilization below pH 6.8, enhanced DOX release and endosomal membrane disruption. The first functionality is expected to endow tumor pH specificity to nonspecific ligands and the rest ones may help to treat solid tumors that are hard-to-treat by conventional chemotherapy (resistant tumors). The concept was proven in vitro studies and in vivo model.

  • Three kinds of surfactant systems - cationic surfactant (system 1), combinition of two cationic surfactants (system 2), and combination of two cationic surfactant and non-ionic surfactant (system 3) - for the simultaneous removal of nitrate and phosphate by micellar-enhanced ultrafiltration (MEUF) were investigated. The highest removal efficiencies of nitrate and phosphate were observed in system 2, which were 90 % of nitrate and 72 % of phosphate. The COD of permeate in system 3 was the lowest, because the added non-ionic surfactant made critical micelle concentration (CMC) lower than that of other surfactant systems. In all systems, the flux decline was similar.

  • The effects of phosphatidic acid(PA) on the activity of phospholipase D were examined in detail. The enzyme activity was examined in the liposome system containing phosphatidylcholine and PA, which was suspended in a desired buffer solution by ultrasonication. The substrate of large unilamella vesicle (LUV) state by ultrasonication was more effective on the enzyme activity than that of multilamella vesicle(MLV) by water-bath type sonication. The most effective molar ratio of PC-PA liposome for enzyme activity was found to be 1:0.7. The other optimum conditions were found 5 mM $Ca^{2+}$ ion, pH 6.6, and incubation temperature of $27^{\circ}C. K_m \;and \;V_{max}$ values were estimated to be 1.43 mM and 0.8 $nmole/min/{\mu}g$ protein respectively. These properties in a PC-PA liposome system were compared with those in a PC-SDS mixed micelle system. The effects of other phospholipids and organic phosphates on the enzyme activity were also examined.

  • Silybin is the main component of Cardus marianus extracts (Silymarin) originated from Silybum marianum, called as milk thistle. It has a hepato-protective effect and is used clinically for the treatment of liver disease. But it is water-insoluble and is poorly absorbed from the gastrointestinal tract, resulting in very low oral bioavailability. Polymeric mixed-micelle precursor formulation containing surfactants, co-solvents, and block-co polymers with Cardus marianus extracts was made to enhance the dissolution rate of silybin and encapsulated with soft gelatin capsule.   • This study is to form the self assembly of cubic crystalline phase to penetrate into the skin epidermis. The various performance synthesized diglyceryl phytylacetate (DGPA) having hydroxyl group (-OH) and 4 methyl chains with phytyl group was carried out as an amphoteric lipid such as emulsifying power, self assembly. Emulsifying activity of DGPA was very stabilized on only 1% of small content, it could make a W/O emulsion containing high internal phase incorporated with water. Cubic liquid crystal structure with DGPA on three-phase diagram was formed, when mixed DGPA, dimethicone (2CS), and water. Through three-phase diagram forming the cubic liquid crystal area, hexagonal structure zone, and mixing water phase and hexagonal structure area, reversed micelle area were respectively certified. Its structure was proved by the SAXS (small angled x-ray scattering) analysis. As an application, formation of cubosome containing 10% of magnesium ascorbylphosphate and 5% of pyridoxine tris-hexyldecanoate was encapsulated. Occlusive effect of cubosome had above 1.7 times better than reversed micelle. From using poloxamer of dispersing agent, phase structure recovered from W/O emulsion to cubic liquid crystal phase when storage in $33^{\circ}C$ incubator. Therefore, our this study is expected to be as epidermal-dermal skin absorbers in skin care cosmetics and pharmaceuticals industries as raw materials to form a cubic crystal phase through a more in-depth research to DGPA having amphoteric lipid property.

  • Direct measurement of the kinetics of free fatty acid transfer between phospholipid model membrane is technically limited by the rapid nature of the transfer process. Separation of membrane-bound fatty acid by centrifugation has shown that although the equilibrium distribution of free fatty acid is determined by this method, fatty acid transfer occurs too rapidly for accurate kinetic measurements. Recently fluorescence resonance energy transfer(FRET) assay has been developed to examine transfer of fatty acids between membranes. Donor membranes which has fluorescent fatty acid, anthroyloxy fatty acid(AOFA), is mixed with acceptor membranes which has non-interchangeable fluorescent quencher, nitrobenzo-xadiazol(NBD), using stopped flow apparatus. As the fluorescent fatty acids transfer from donor membrane to acceptor membrane, fluorescence intensity would be decreased and the rate and degree of fatty acid transfer can be analyzed. Fatty acid transfer between micelles is more complicated because of bile salt. Therefore in experiments with micelles, fluorescence self quenching assay is used. At high concentrations, a fluorophore tends to quench its own fluorescence causing a reduction in fluorescence intensity. Donor micelles contained self quenching concentrations of fluorophore and acceptor micelles had no fluorophore. Upon mixing of donor and acceptor micelles, the rate of transfer of the fluorophore from the donor to the acceptor was measured by monitoring the release in self quenching when its concentration in donor decreased over time.

Emulsion Properties of Pseudo-Ceramide PC104/Water/Polyoxyethylene Cholesteryl Ether and Polyoxyethylene Cetyl Ether Mixtures.

  • The formation of emulsions and micelles in water/ceramide PC104/CholE $O_{20}$/C$_{16}$E $O_{20}$ and water/ceramide PC104/CholE $O_{20}$ mixtures was investigated through the phase behavior studies. The phase diagrams showed the existence of micelle and emulsion regions in both systems. The mixed surfactant system (CholE $O_{20}$/C$_{16}$E $O_{20}$) showed the wider micellar and emulsion regions than the single surfactant system (CholE $O_{20}$). From FT-IR measurements, it was found that the polyoxyethylene (POE) groups of surfactants formed the hydrogen bonds with amido carbonyl group in ceramide PC104. This result indicated that the hydrophilic part (EO) of surfactants could stabilize the lamellar structure and emulsion of ceramide PC104. The mixed surfactant system (CholE $O_{20}$/C$_{16}$E $O_{20}$) resulted in the smaller emulsion droplet size due to the effect of curvature at the interface, thus further increasing emulsion stability. With the penetration of $C_{16}$E $O_{20}$into the interfacial layer of surfactants in emulsion, the curvature of the interface might be altered for the formation of smaller emulsion droplets. The mixed surfactant system could incorporate up to 4 wt. % of ceramide PC104 into emulsion more than single surfactant system.ystem.m.
