Effect of Silver Ion Solution on the Inhibition of Microcystis Growth (은이온 수용액의 Microcystis 생장 억제 효과)

  • The effect of silver ion solution on the growth of Microcystis aeruginosa UTEX 2388 (cyanobacterium) and Chlorella sp. KCTC AG20136 (green alga) was investigated using separated and mixed culture in filtered natural water and BG11 medium. In separated culture, M. aeruginosa UTEX 2388 and Chlorella sp. KCTC AG20136 were found to be sensitive to 0.01 and 0.1 mg L$^{-1}$ of silver ion, respectively. Also, the silver ion concentrations for the growth inhibition of M. aeruginosa UTEX 2388 and Chlorella sp. KCTC AG20136 in the mixed culture were same in separated culture. Cyanobacteria were more sensitive to the silver ion solution than green algae. In bloom sample, the minimal inhibition concentration of silver ion solution for the low Chl-${\alpha}$ sample (110$\sim$190 ${\mu}g$ L$^{-1}$) and high Chl-${\alpha}$ sample (1,500$\sim$1,900 ${\mu}g$ L$^{-1}$) was about 0.1 and 3.0 mg L$^{-1}$, respectively. The silver ion concentration for the inhibition of algal bloom sample was affected by the algal biomass. In order to use silver ion solution for the control of algal bloom, the silver ion concentration must be determined in consideration of a minimal effect on the environment.

Growth Inhibition of the Diatom, Chaetoceros calcitrans by Alteromonas sp. SR-14 (Alteromonas sp. SR-14에 의한 규조 Chaetoceros calcitrans 증식저해)

  • Algicidal activities of Alteromonas sp. SR-14 against Chaetoceros calcitrans were investigated at various culture conditions. The algicidal activity by Alteromonas sp. SK-14 was dependent on temperature. In mixed culture of C, calcitrans and Alteromonas sp. SR-14 at various temperatures, the algicidal activity of Alteronzonas sp. SR-14 was the highest at $20^{\circ}C$, but not showed algicidal activity above $25^{\circ}C$. With the inoculation of $10^4$ cells/ml of C. calcitrans, the diatom could not grow at the microalgal culture condition until 15 days by the simultaneous inoculation of less than 10 cells/ml of Alteromonas sp. SR-14. Alteromonas sp. SR-14 showed the strongest algicidal activity against logarithmic phase cells of C. calcitrans. During the mixed culture of C. calcitrans and Alteromonas sp. SR-14, supplementation of Conwy medium nutrients, changes of light intensity with 1,300$\~$4,600 lux and agitation with 200 rpm did not affect the algicidal activity.

Effect of Loquat(Eriobotrya japonica Lindley) Extract on Acid Production and Growth of Lactic Culture (비파(Eriobotrya japonica Lindley) 착즙액 첨가가 요구르트 균주의 산 생성 및 증식에 미치는 영향)

  • This experiment was carried out to investigate the effect of loquat (Eriobotrya japonica Lindley) extract on the acid production and growth of lactic culture in reconstituted skim milk. The supplementation level of loquat extract to reconstituted skim milk was 10%, 15% and 20%. Reconstitued skim milk containing loquat extract was fermented by single of mixed culture of Streptococcus thermophilus, Lactobacillus acidophilus and Lactobacillus casei. General compositions of loquat extract, changes of viable cell count, pH and titratable acidity during fermentation were determined. Chemical compositions of loquat extract were 91.5% moisture, 0.2% crude ash, 8.6$^{\circ}$ Brix soluble sugar, 0.34% total acid, and 4.11 in pH. Supplementation of loquat extract stimulated acid roduction and growth of lactic acid bacteria. Among supplementation levels, a group that was fermented by a single culture of Str. thermophilus with 10% loquat extract was shown the highest viable cell count (2.10${\times}$10$\^$9/ CFU/mL) at 12 hours after inoculation. When loquat extract was added to reconstituted skim milk at the level of 10%, all mixed cultures of lactic acid bacteria showed higher acid production and the number of viable cell count than 3 kinds of single cultures. Especially, the growth of mixed culture of Str. thermophilus and Lac. acidophilus was promoted by the addition of 10% loquat extract. Therefore, it was suggested to manufacture the yoghurt with the addition of 10% loquat extract and the inoculation of mixed culture of Str. thermophilus and Lac. acidophilus for on the stimulation of growth of the lactic culture.

An Experimental Study on Bacterial Adhesion onto Activated Carbon and Ceramic (활성탄 및 세라믹 재질에의 세균 부착성에 대한 연구)

  • The microbial adsorption characteristics of two different media for biological treatment were studied using attached diverse microbes onto activated carbon and ceramic. The results in the experiments of the characteristics of physical adhesion on two different media with addition of high and low concentrated substrate in the culture were observed that the efficient of adhesion onto F-400 activated carbon was higher over that of ceramic due to the surface area of media. The irradiation treatment by ultrasonication with 400 W power and 3 min retention time on the media without addition substrate conditions and subsequent mixing throughly the culture showed the highest efficiency of cell detachment on the media. Three different microbes, P. ovalis, A calcoaceticus, and B. subtillis were used for the study of the characteristics of microbial adhesion on the media. p ovalis showed the highest adhesion capability while B. subtillis showed the lowest capability adhesion onto media either addition of substrate in the culture. The mixed bacterial culture showed $10\%$ lower removal efficiency of DOC in the low concentrated substrate culture compared to the single pure culture. Whileas, it did not show significant difference between two cultures at high concentrated substrate. It was also observed same population density of microorganism by counting of microbes adhered to microbial media with an ultrasound treatment.

  • Culture has played a pivotal role in fashion from time immemorial. The objective of this research is to explore the power of cultural affiliation in fashion. The selected study setting is the Kandyan Kingdom of Sri Lanka. The Kandyan Kingdom of Sri Lanka lasted almost 3 1/2 centuries from 1474-1815. The whole period faced different foreign cultural forces. As a result of such diverse cultural influences the Kandyan dress showed a hybrid formation of Western and South Indian and Sinhalese traditional sartorial features. Sewing techniques and unsewn dress arrangement methods were mixed together. The dress became an amazing blend of Eastern and Western dress items. They combined different aspects of foreign dress together to yield a unique result. The analysis comprises observational study of actual descriptions made by observer- participants, historical records, murals of the period review of ancient literature and research papers relevant to the subject. Reliability of the data was ensured. The pictorial data were cross checked from different literary sources. Many original sources were used. Results: Culture and fashion have strong interconnection. When features of culture change, trends of fashion are gradually correspondingly changed.

Plant Regeneration from Protoplasts Isolated through Embryogenic Cell suspension Culture in Rice (벼 현탁배양을 통하여 분리된 원형질체로부터 식물체 재분화)

  • Plant regeneration was accomplished from protoplast culture of rice (Oryza sativa L. cv. Taebaeg). Embryogenic callus was induced from mature seed on MS medium containing 5 mM proline, 2.5 mg/L 2,4-D, 30 g/L sucrose in the dark at 28$^{\circ}C$ and used to establish embryogenic cell suspension culture. Suspension cells were subcultured every one week in N6 medium supplemented with 5 mM proline, 200 mg/L casein hydrolysate, 2.5 mg/L 2,4-D and amino acids of AA medium. Suspension cultures were composed of cells that were densely cytoplasmic, potentially embryogenic and were at least maintained for more than 6 months in liquid medium. Protoplasts were isolated from fast-growing suspension culture cells and cultured in a slightly modified KpR medium by mixed nurse culture. Isolated protoplasts began to divide within 5~7 days and thereafter, protoplast-derived calli were sequentially transferred to callus proliferating medium that soft agar MS medium contained 2 mg/L 2,4-D and produced distinct embryogenic cells. Microcolonies were then transferred to solid medium which consisted of MS medium containing 5 mg/L kinetin, 1 mg/L NAA, 1 mg/L ABA, 30 g/L sucrose and 10 g/L sorbitol under fluorescent light. Mulitple shoots of 4~5 per callus emerged and were transferred to hormone-free MS medium for root initiation. Thereafter, The plantlets were transferred to pots of soil to mature in the culture room.

Anaerobic dechlorinating enrichment culture on tetrachloroethene (PCE) (PCE 탈염소화를 위한 혐기성배양)

  • Starting at the beginning q the 20th century, increasing amounts of tetrach1oroethene (PCE) and trichloroethene (TCE)were manufactured due to the extensive use of these compounds in industry, in the military, and in private households, mainly as nonflammable solvents. This widespread use, along with careless handling and storage, are among the most serious contaminants of soil, sediment and groundwater. Highly chlorinated ethenes are typically not degraded through oxygenation by aerobic bacteria Since complete reductive dechlorination of PCE and TCE to ethene (ETH) has been observed in anaerobic enrichment culture, anaerobic dehalorespiring bacteria have received increased attention in the last decade. Under anaerobic conditions, these compounds con be reductively dehalogenated to less-chlorinated ethenes or innocuous ethene by microorganism through dehalorespiration. We have been studying anaerobic enrichment culture which used lactate as the electron donor for reductive dechlorination of PCE to ETH the anaerobic mixed microbial culture was enriched from the sediment sample taken from site contaminated with PCE. PCE was consistently and completely converted to ethene. In addition, the accumulation of intermediate products such as 1,2-ds-dichloroethene (cis-DCE) and vinyl chloride (VC) was observed in the anaerobic mixed microbial culture. the established dechlorinating enrichment culture was analyzed by DGGE using primers specific to DefrJ1ococcoides 16S rRNA gene sequences. In conclusion, we established the PCE dechlorinating enrichment culture and confirmed the existence of Dehalococcoides in an enrichment culture.

Addition Effect of Seed-associated or Free Linseed Oil on the Formation of cis-9, trans-11 Conjugated Linoleic Acid and Octadecenoic Acid by Ruminal Bacteria In Vitro

  • The effects of seed-associated or free linseed oil on fermentation characteristics and long-chain unsaturated fatty acids composition, especially the formation of conjugated linoleic acid (CLA) and octadecenoic acid (trans-11 $C_{18:1}$, $t-C_{18:1}$) by mixed ruminal bacteria were examined in vitro. Concentrate (1% of culture solution, w/v, as-fed basis) with ground linseed (0.6% of culture solution, w/v, DM basis) or linseed oil as absorbed onto ground alfalfa hay was added to 600 ml mixed solution consisting of strained rumen fluid and artificial saliva at the ratio of 1:1 in a glass culture jar. The culture jar was covered with a glass lid with stirrer, and placed into a water-bath ($39^{\circ}C$) and incubated anaerobically up to 24 h. Seed-associated or free linseed oil did not significantly affect the pH and ammonia concentration in the culture solution. Molar percent of acetate tended to increase while that of propionate decreased with the addition of free oil treatment throughout the incubation. Differences in bacterial number were relatively small, regardless of the form of supplements. Decreasing trends in the compositions of linoleic acid ($C_{18:2}$) and linolenic acid ($C_{18:3}$) but increasing trends of stearic acid ($C_{18:0}$), $t-C_{18:1}$ and CLA compositions were found from culture contents up to 12h incubation when incubated with both ground linseed and linseed oil. The compositions of $C_{18:0}$, $C_{18:2}$ and $C_{18:3}$ were greater but those of oleic acid ($C_{18:1}$), $t-C_{18:1}$ and CLA were smaller in a culture solution containing ground linseed than those containing linseed oil. The ratio of $t-C_{18:1}$ to CLA was lower in the culture solutions containing linseed oil up to 12h incubations as compared to those containing ground linseed.

Studies on the Biological Treatment of Waste Water from Acetaldehyde Plant (아세트 알데히드(특수산업) 폐수의 생물학적 처리)

  • In order to establish the biological treatment system which can be used for treatment of waste aster from acetaldehyde plant, it was investigated optimum nutrient requirements and growth conditions by mixed culture of Micrococcus roseus AW-6, Micrococcus luteus AW-22, Microbacterium lacticum AW-38 and Microbacterium laevaniformans AW-41 as well as the effect of coagulants and neutralization reagents. Also, it was carried out the continuous culture as well as batch culture to treat the waste water by mixed culture of these strains. The COD removal rate was reached to maximum state for 96hrs culture at pH7.0 and $30^{\circ}C$ NaOH as the neutralization reagents was the most effective, but the coagulants had no effect on the COD remonal rate and the optimum dilution times for treatment were 10 fold. The COD removal rate was also increased by supplimenting 200 ppm $NH_{2}NO_{3}$, 50 ppm $KH_{2}PO_{4}$, 15 ppm $CaCl_{2}$ and 1 ppm $MgSO_{4} \cdot 7H_{2}O $ as additional nutrients. The removal rate coefficient $K_{1}$ on the batch culture was $4.5\times 10^{-6}$, and the detention time for BOD removal rate of 85% was approximately 45hrs. The COD of waste water was reduced to 15% of its initial value by the continuous culture. The COD and BOD of the effluents were to be about 60 ppm and 40 ppm, respectively, and final pH was 7.0.

Growth Characteristics of Bifidobacteria and Quality Characteristics of Soy Yogurt Prepared with Different Proteolytic Enzymes and Starter Culture (단백분해효소와 배양방법의 종류에 따른 비피더스균의 생육특성 및 soy yogurt의 품질특성)

  • The quality characteristics of soy yogurt prepared with different proteolytic enzymes and starter culture were evaluated. In order to facilitate the growth of lactic acid bacteria and subsequent production of lactic acid, soy protein isolate(SPI) was hydrolyzed using three kinds of proteases; one extracted from Aspergillus oryzae, bromelain and ${\alpha}-chymotrypsin$. The cultural systems employed thereafter for lactic fermentations were: 1) Bifidobacterium bifidum, 2) B. bifidum and Lactobacillus acidophilus, 3) B. bifidum and Lactobacillus bulgaricus. In soy yogurt, pH was more decreased by mixed culture method than single culture method with the accumulation of lactic acid. Viable cells of lactic acid bacteria in soy yogurts were measured $10^8$ CFU/g by the single culture method while $10^9$ CFU/g by the mixed culture method except ${\alpha}-chymotrypsin$ treatment. The amount of free amino acids in soy yogurts were substaintially increased by enzyme treatment. Viscosity was decreased by enzyme treatment, resulting in higher viscosity by ${\alpha}-chymotrypsin$ treatment. Water holding capacity was found to be higher in the single culture method in case of enzyme treatment. Among the various volatile flavor components isolated and identified after enzyme hydrolysis, acetaldehyde, ethanol, diacetyl, butyl alcohol contents tended to increase by lactic fermentation.
