Candida albicans의 시험관 희석법에 의한 항균력 검사시 배지가 항균력에 미치는 영향 (Effect of Culture Medium on Results of Maerobroth Dilution Antifungal Susceptibility Testing of Candida albicans)

  • A total of 42 strains of Candida albicans were examined for susceptibility to three antifungal agents, amphotericin B(AMB), 5-fluorocytosine(5-FC), and ketoconazole(KTZ), using defined medium, synthetic amino acid medium-fungal(SAAM-F), supplemented yeast nitrogen base(SYNB) and undefined medium Sabouraud's dextrose broth(SDB) and Kimmig broth media. A tube dilution method was used with minimum inhibitory concentrations(MICs) determined after incubation for 24 hour and 48 hours. All testes were performed in duplicate. In general, MICs were more reproducible after 48 hour of incubation. Forthermore, MICs determined after incubation for 48 hours were significantly higher than those determined after 24 hours. The actural MICs obtained with the different antifungal agents were clearly influenced by the test medium used. The rank order of AMB MICs according to the test medium was as follows: SAAM-F>SYNB>SDB>Kimmig broth. With 5-FC, the following pattern was observed: SYNB>SAAM-F>SDB>Kimmig borth. For ketoconazole, the MICs according to the test medium was SAAM-F>SDB>SYNB> Kimmig broth. In amphotericin B, the MICs mean value with the test medium was as follows: SDB, 0.24 mcg/ml; Kimmig broth, 0.29 mcg/ml; SYNB, 0.21 mcg/ml and SAAM-F, 0.15mcg/ml. The actural value of 5-FC was; SDB, 37.20 mcg/ml; Kimmig broth, 67.41mcg/ml; SYNB, 21.29 mcg/ml and SAAM-F, 24.61 mcg/ml and in ketoconazole, the MICs value was; SDB, 1.83 mcg/ml; Kimmig broth, 4.08 mcg/ml; SYNB, 1.95 mcg/ml and SAAM-F, 1.41 mcg/ml. The results of this investigation suggested that broth dilution susceptibility testing of yeast and yeast-like fungi are best performed with an incubation period of 48 hours. Furthermore, medium composition can significantly influence the results of such testing.

Antimicrobial Effects of Linalool and ${\alpha}$-Terpineol against Methicillin-Resistant Staphylococcus aureus Isolated from Korean

  • Methicillin-resistant Staphylococcus aureus (MRSA) is one of the important causative microbes for nosocomial infection and has been isolated from the dental environment. The purpose of this study was to investigate the antimicrobial activity of linalool and ${\alpha}$-terpineol against MRSA isolates from a Korean population. In the experiments, we determined the minimum inhibitory concentrations (MICs) and minimum bactericidal concentrations (MBCs) of these two compounds against 18 strains of MRSA. The data revealed that the $MIC_{90}/MBC_{90}$ values of linalool and ${\alpha}$-terpineol against MRSA were >12.8 mg/ml and 6.4 mg/ml, respectively. These results indicate that ${\alpha}$-terpineol has more potent antimicrobial activity against MRSA than linalool and may have utility as an anti-MRSA cleansing agent for dental instruments and dental unit chairs.

Evaluation of Pharmacological Activities of Ethanol Extracts Prepared from Selected Korean Medicinal Plants

  • In this study, 23 ethanolic extracts from 20 medicinal plants were evaluated for biological activities. Results revealed that of 23 samples, seven samples have demonstrated good antimicrobial activity. Minimum inhibitory concentrations were 0.4-2.0 mg/mL, while minimum bactericidal concentrations were mostly high 0.8-2.0 mg/mL for selected seven samples. Five samples revealed > 70 mg gallic acid equivalent (GAE)/g of total phenolic contents. Among test samples, six samples exhibited > 80% inhibition of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical and only two samples exhibited > 80% inhibition of 2,2'-Azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radicals. A total of five test samples revealed Trolox equivalent antioxidant capacity more than $1000{\mu}m/g$. The MTT assay indicated that eight test samples exhibited > 90% viability of murine macrophage cells (RAW 264.7) at $250{\mu}g/mL$ and suppressed iNOS mRNA expression at transcriptional level when stimulated by lipopolysaccharide (LPS). Some medicinal plants revealed promising results, and so they have prospective for further more inclusive studies.

Anti-Salmonella Activity of Lemongrass Oil Alone and in Combination with Antibiotics

  • The effects of Cymbopogon citratus essential oil (Lemongrass oil) and its main component, citral (84.30%), on antibiotic-susceptible and -resistant strains of Salmonella enteritidis and S. typhimurium were assessed. C. citratus oil and citral significantly inhibited all strains of the two Salmonella species examined, with minimum inhibiting concentrations (MICs) ranging from 0.5 mg/ml to 8.0 mg/ml. The combined effects of C. citratus oil and citral (84.30%) were evaluated using a checkerboard microtiter assay. Essential oil fractions of C. citratus and citral exhibited strong synergistic or additive effects with streptomycin or kanamycin against S. typhimurium strains with fractional inhibitory concentration (FIC) indices in the range of 0.28 to 1.00. In conclusion, a combination of streptomycin and lemongrass oil or its main component, citral, may be useful for reducing the minimum effective dose of antibiotic required for the treatment of resistant S. typhimurium infections.

스노우베리 추출물의 항균 활성에 관한 연구 (A Study on the Antimicrobial Activity of Snowberry Extract)

  • 본 연구는 스노우베리 추출물과 발효 추출물을 대상으로 피부 상재균인 Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Pseudomonas aeruginosa, Candida albicans를 이용하여 천연화장품 소재로서의 항균 활성을 비교 평가하였다. 이를 위한 스노우베리 추출물의 항균 활성은 10-200mg/mL 농도에서 생육저해환(paper disc diffusion method)과 최소저해농도(Minimum inhibitory concentration)를 평가하였다. 스노우베리 추출물과 발효 추출물의 생육저해환을 확인하였을 때, 스노우베리 추출물의 경우 뿌리 추출물에서만 100mg/mL, 200mg/mL의 농도에서 항균력을 나타내었고, 발효 추출물의 경우 200mg/mL에서 항균력을 나타내는 것을 확인하였다. 그러나 스노우베리 추출물과 발효 추출물의 4종의 세균에 대한 항균력은 확인할 수 있었으나, 진균인 Candida albicans에서는 항균력을 확인할 수 없었다. MIC 결과 스노우베리 잎, 뿌리 추출물에서 발효의 경우 줄기 추출물에서 각 균에 대한 최소저해농도가 100mg/mL, 200mg/mL로 확인되었다. 이러한 결과는 스노우베리가 피부 개선을 위한 항균 소재 및 방부제로서의 활용 가치가 있다고 사료된다.

유백피(Ulmi cortex)의 항균활성 (Antimicrobial Activity of Ulmi cortex Extracts)

  • The solvent extracts of Ulmi cortex, which were extracted by using several solvents with different polarities, were prepared for utility as a natural preservatives. The antimicrobial activities and cell growth inhibitions were investigated to each strain with the different concentrations of Ulmi cortex extracts. Methanol extract showed the highest antimicrobial activity. The methanol extract was represented the broad antimicrobial activities for the gram positive and negative strains. Minimum inhibitory concentrations (MIC) for each strains were appeared to around 0.3mg/ml at each of Bacillus cereus, Bacillus subtilis, and Staphylococcus aureus. The cell growth inhibitions were not shown on Lactobacillus bulgaricus, Lac tobacillus plantarum, and Bifidobacterium bifidum, but greatly on the Clostridium butyricum. The meth anol extracts were further reextracted sequentially with hexane, chloroform, ethyl acetate, and butanol for purifying crude methanol extracts. The extract, which was reextracted by butanol, showed the highest antimicrobial activity.

소 유방염 원인균에 대한 은이온의 향균력 평가 (Evaluation of Bactericidal Capacity of Silver Ion against Bovine Mastitis Pathogens)

  • 소 유방염은 주로 세균 감염에 의한 유선조직에 염증이 생기는 질병으로 유방염의 치료와 억제를 위해서는 1차적으로 세균을 사멸시키는데 있다. 본 연구에서는 소 유방염의 원인이 되는 다양한 세균들에 대한 은 이온의 항균효과와 적용 농도를 측정하였다. 유방염 원인균에 대한 은 이온의 최소 억제 농도 (minimal inhibitory concentrations, MICs)와 최소 살균 농도 (minimum bactericidal concentrations, MBCs)를 측정하였으며, 주사전자현미경 (scanning electron microscopy, SEM)을 이용하여 은 이온에 의한 세균의 형태 변화를 확인하였다. 다양한 유방염 원인균에 대한은 이온의 최소 억제 농도와 최소 살균 농도는 1.9에서 15.6 ${\mu}g$/ml로 세균에 따라 차이가 보였으며, 은이온 처리 후 주사전자현미경 촬영 결과 Staphylococcus aureus와 Escherichia coli의 세포벽이 함몰되거나 파괴되는 것을 볼 수 있었다. 이러한 결과는 유방염 원인이 되는 다양한 원인균에 대해 은이온의 살균 효과와 그 적용 농도를 보여주는 것으로, 소 유방염의 치료와 방어를 위한 임상적인 적용 가능성을 보여주는 것이다.

Biological Characterization of Periconicins, Bioactive Secondary Metabolites, Produced by Periconia sp. OBW-15

  • Periconicin A and B, two new fusicoccane diterpenes originally isolated from the cultures of endophytic fungus Periconia sp. OBW-15, were tested by several biological assays. Periconicin A was consistently more active than periconicin B. In an antifungal activity assay, periconicin A showed potent inhibitory activity against the agents of human mycoses, including Candida albicans, Trichophyton mentagrophytes, and T. rubrum, with minimum inhibitory concentration (MIC) in the range of 3.12- 6.25 $\mug$ /ml. In a plant growth regulatory activity assay, periconicins inhibited hypocotyl elongation and root growth of Brassica campestris L. and Raphanus sativus L. At concentrations below 1 μg/ml, however, both compounds accelerated root growth by 110- 135%. From these results, it is apparent that a methyl group positioned in a cyclopentane ring may play an important role in plant and fungal growth inhibitory activity.

천연 항균제처리를 병용한 과채류의 자연 저온저장기술 개발에 관한 연구 (Inhibitory Effects of Natural Antimicrobial Agenton Postharvest Decay in Fruits and Vegetables under Natural Low Temperature)

  • In order to prevent the postharvest decay and to promote the freshness retention of fruits and vegetables grapefruit seed extract(GFSE), natural microorganism control agent, was applied to the preservation of fresh fruits and vegetables. Freshfruits and vegetables treated with GFSE and stored in polyethylene film (0.1mm) at 1$0^{\circ}C$-15$^{\circ}C$ of natural low temperature low kept better qualities in color and texture than the GFSE -not- treated control. The treatment using GFSE ina 250ppm to 500ppm concentration seemed to be an effective one for the control of Botrytis cinerea isolated in red wine grapes. After 4 weeks of storage the firmness rate of cucumbers treated with the dilute GFSE was four times higher than that of non-treated ones. GFSE showed effective inhibitory action towards plant pathological bacteria and fungi which were involved in the decay of fruits and vegetables. Minimum inhibitory concentrations of GFSE towards them were in the range of 250ppm to 500ppm .Direct visualization of microbial cells and spores using electron microscopy showed microbial cells and fungal spores the function of which was destroyed by treating with the dilute solutions of GFSE. It was observed that GFSE would reduced disease damages and have bactericide & fungicide properties during the storage of such fruits and vegetables as egg plant, wild edible greens , kumquat, and kiwi fruit.

수용성 프로폴리스의 항균성 (Antimicrobial Activity of Water Soluble Propolis)

  • In this study, the minimum inhibition concentration(MIC), growth inhibition activity, and colony forming inhibitory activity of water soluble propolis against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae and Salmonella enteritidis were tested. The MICs of the water soluble propolis against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, and Salmonella enteritidis were 312.5 ppm, below 156.3 ppm, 625 ppm, 10,000 ppm, above 10,000 ppm, 10,000 ppm, above 10,000 ppm, above 10,000 ppm, 10,000 ppm, and above 10,000 ppm, respectively. The growth inhibition concentrations against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, and Klebsiella pneumoniae were 156.3 ppm, below 156.3 ppm, 625 ppm, 5,000 ppm, 10,000 ppm, 10,000 ppm, 10,000 ppm, 10,000 ppm, and 5,000 ppm, respectively. However, 10,000 ppm did not inhibit the growth of Salmonella enteritidis. Finally, the colony forming inhibitory activities against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, and Salmonella enteritidis were 98.0%, 99.8%, 69.8%, 98.1%, 62.0%, 63.1%, 79.5%, 61.9%, 79.6%, and 0.0%, respectively.
